Removal of Oil from Soil Using Nonionic Surfactant : The Effects of Middle Phase Formation and Dynamic Interfacial Tension (비이온 계면활성제를 사용한 토양으로부터 오일의 제거에 관한 연구 : 중간상생성 및 동적 계면장력의 영향)

  • The soil remediation by non-ionic surfactant solutions ($C_{12}H_{25}O(CH_2CH_2O)_5H$ and Triton X-100) was studied. Depending on the amounts and use of co-surfactants, MPT(phase inversion temperature), dynamic interfacial tension, and the detergency efficiency of the surfactant solutions in soil were investigated. The oils used were kerosene, n-hexadecane, and paraffin oil. With respect to a higher detergency efficiency, a lower interfacial tension and the MPT was very important. The $C_{12}H_{25}O(CH_2CH_2O)_5H$ was better than Triton X-100 on the oil removal from the soil and the effect of oil kinds was kerosene>paraffin $oil{\geq}n-hexadecane$. The co-surfactant, n-dodecanol, reduced the MPT compared to no addition of this, whereas it did not enhance the detergent efficiency.

Determination of Tb(III) in aqueous solution by fluorescence spectrometry (형광분광법에 의한 수용액 중의 Tb(III) 정량)

  • Methods to determine terbium(III) ion in aqueous solution by measuring the enhanced fluorescence intensity of terbium(III)-terephthalic acid(TPA) complex ion have been studied. The optimum analytical conditions for pH, excitation wavelength and concentration of TPA were found to be 6.0, 260nm and $4.0{\times}10^{-4}M$, respectively. The fluorescence intensity of the terbium(III) complex ion was further increased with addition of trioctylphosphine oxide (TOPO). In this case Triton X-100 was used to dissolve TOPO in aqueous solution. When TOPO was used, the optimum analytical conditions for pH, excitation wavelength, and concentrations of TPA, TOPO and Triton X-100 were found to be 4.5, 285nm. $4.0{\times}10^{-4}M$, $5.0{\times}10^{-5}M$, and 0.05%, respectively. Under the optimum experimental conditions, calibration curve for Tb(III) was linear over the range from $4.0{\times}10^{-8}M$ to $4.0{\times}10^{-5}M$ and the detection limit was $4.0{\times}10^{-8}M$. When TOPO was used, the concentration range of linear response, and the detection limit were $4.0{\times}10^{-9}M$ to $2.0{\times}10^{-6}M$, and $4.0{\times}10^{-9}M$, respectively. Effects of interferences from various cations for the determination of terbium(III) ion were also investigated.

  • The characteristics of demulsification of petroleum emulsion by Streptomyces sp. 8321 were investigated. Demulsification ability of Streptomyces sp. 8321 appeared to be confined within the spores. Spore surface hydrophobicity was increased with culture age stimulating the demulsification ability. Over $1.1{\times}10^8spores/ml$ completely demulsified kerosene-0.2% Triton X-100 (2:1) emulsion. Among the low viscosity hydrocarbons, hydrocarbons with longer chain such as n-hexadecane and diesel were more rapidly demulsified. However, only 20-30% of the emulsion with high viscosity hydrocarbons was demulsified after 24 hours. Oil-in-water emulsions made by Corexit, Finalsol and BP series surfactants were completely demulsified within one minute. Demulsification rate ($t_{1/2}$) of oil-in-water emulsions made by Corexit 7664, 8667, Triton X-100 and Tween 80 decreased as their concentration increased. In case of water-in-oil emulsion made by Seagreen, $t_{1/2}$ was over 24 hours. Therefore, demulsification ability of Streptomyces sp. 8321 was more effective on oil-in-water emulsions.

Expression of GFP Gene in the Porcine Preimplantation Embryos after ICSI with DNA/Sperm Complex

  • The possibility of producing transgenic embryos expressing the green fluorescence protein (GFP) gene have been evaluated after transfer of exogenous gene into the porcine zygote cytoplasm using the intracytoplasm sperm injection (ICSI) as gene delivery method. For DNA binding to sperm heads, 0.05% Triton X-100 or Lipofectin was used. After injection of the sperm bound to DNA by means of Lipofectin or Triton X-100 triturate, the blastocyst formation rates on day 6 were not significantly different from that of ICSI only group (18.8, 19.2 and 25.3%). In terms of GFP expression, more embryos were in GFP form in Triton X-100 group than in Lipofectin group (40.6 vs 36.4%), while percentage of non-mosaic embryos expressing the GFP gene in all blastomere was higher (P<0.05) in Lipofectin group than in Triton X-100 group (4.2 vs 0.9%). ICSI embryos derived from sperm treated with Lipofectin/DNA complex was transferred into 3 recipients and were collected by uterine flushing on days 5, 7 and 15 after embryo transfer, and then GFP expression was observed by a fluorescence microscopy. Over 26% of the collected embryos were normally expressed GFP gene. These results suggest that foreign gene transfer method with DNA bound sperm caused minimal damage to structure of oocytes that can result to full development of porcine embryos. This was confirmed in this study when the embryos that were transferred after ISCI of DNA bound sperm had a normal development and gene expression until preimplantation.

  • Subsurface contamination of industrial hazardous organic substances is a serious social issue. Decomposing the hydrophobic organic compounds in the subsurface is technically difficult and the compounds can last as long-term contaminant sources of groundwater once they are sorbed on the soil. Although the danger of contaminated subsurface has long been recognized little was known about the effective remediation technique. Focusing on the remediation of the p-Cresol and 3, 5-Dichlorobiphenyl among subsurface contaminants, this paper studies the surfactant-enhanced desorption technique. Nonionic surfactant(Triton X-100) and anionic surfactant(SDS ) were used as desorbing solvents for extracting organic compound sorbed on soil particles. Sorption characteristics of soils and organic compounds were analyzed and the applications of surfactant solution were studied through batch tests and the flexible-wall permeameter tests. As a result of the sorption isotherm tests, a log-log linear relation was obtained between the linear-partition coefficient, $K_p$ and the octanol-water partition coefficient, $K_{ow}$ of each organic compound. The result of the batch test also showed that Triton X-100 at 0.5% of solution desorbs the 3, 5-Dichlorobiphenyl 28 times more than the water in the batch tests. The surfactant-enhanced subsurface remediation technique becomes more effective when the contaminants are hydrophobic and hard to be decomposed.

The Preparation of Reversed Micelle Containing Water Soluble Collagen Solution and Their Application on Lip Make up Products (콜라겐 수용액을 함유하는 역미셀의 제조 및 립 제품에 응용)

  • Water-soluble collagen (30 wt%) was entrapped within oil phase of lipstick using reversed micelle method to improve the moisture capacity of the lipstick. Reversed micelles containing collagen were prepared using caprylic/capric triglyceride as external phase and polyoxyethylene (10) octylphenyl ether (Triton X-100) and 1-dodecanol as surfactant and co-surfactant, respectively. The formation of reversed micelle encapsulating collagen was confirmed by measuring electric conductivity and UV-vis spectrum using methylene blue (MB). The stability and moisture capacity of the lipstick containing 20 wt% collagen encapsulated reversed micelles were observed by measuring rheology property, moisture content and amino acid content. The molecular ratio (W, water-pool) of water to surfactant (Triton X-100) in the most stable reversed micelle was ${\leq}$ 10. The hardness of the lipstick had no difference with that of the lipstick without reversed micelle, and the moisture content was increased to 59% and the amino acid content was 92.7%.

  • Multivariate models were developed for the simultaneous spectrophotometric determination of copper (II), nickel (II) and zinc (II) in water with 1-(2-thiazolylazo)-2-naphthol as chromogenic reagent in the presence of Triton X-100. To overcome the drawback of spectral interferences, principal component regression (PCR) and partial least square (PLS) multivariate calibration approaches were applied. Performances were validated with several test sets, and their results were then compared. In general, no significant difference in analytical performance between PLS and PCR models. The root mean square error of prediction (RMSEP) using three components for $Cu^{2+}$, $Ni^{2+}$ and $Zn^{2+}$ were 0.018, 0.010, 0.011 ppm, respectively. Figures of merit such as sensitivity, analytical sensitivity, limit of detection (LOD) were also estimated. High reliability was achieved when the proposed procedure was applied to simultaneous determination of $Cu^{2+}$, $Ni^{2+}$ and $Zn^{2+}$ in synthetic mixture and tap water.

A Study on the UV-cut Properties of Cotton Fabrics Treated with UV-absorber (자외선 흡수제 처리 면직물의 소비성능 개선(제1보) - 자외선 차단성능에 관한 연구 -)

  • The influence of ultraviolet(UV)-ray in sun light on human skin has been noted. Textiles can provide protection against harmful UV-radiation. Normally UV-absorbing finishes are used to get better protection. The purpose of this study is to evaluate the UV-cut properties of cotton fabrics treated with UV-absorber. 2,2-dihydroxy-4,4-dimethoxbenzophenone, as UV-absorber was applied to 100% cotton fabric. Reagents added in finishing solution were Triton X-100, polyethylene glycol 400, and $MgCl_2{\cdot}6H_2O$, and C.I. Direct Red 81. Both untreated and treated cotton fabrics were exposed to a xenon arc lamp for 20 and 80 hours. UV absorption spectra of finishing solutions and UV transmission spectra of fabrics were measured by the UV/VIS spectrophotometer. The results of this study can be summarized as follows. The results of this study can be summarized as follows. Absorption and the related transmission spectra were modified in a controlled way with UV-absorber. Absorption effect of UV-absorber was improved by adding Triton X-100, PEG 400, and $MgCl_2{\cdot}6H_2O$ in finishing solution. The UV absorption of finishing solution was in the following order: U/D/T/P/M>D/T/P/M> D/T> D/P, D>U/T/P/M>U/T>T/P/M>T. The UV transmittance of cotton fabrics was remarkably decreased by the application of UV-absorber and additives. The UV-cut properties were most improved by the application of U/D/T/P/M.

Characterization of Extracellular Cholesterol Oxidase Produced from Soil Microorganism (토양 미생물로부터 생산된 Extracellular Cholesterol Oxidase의 특성)

  • Cholesterol oxidase catalyses the conversion of cholesterol to 4-cholesten-3-one. This enzyme has been used for clinical assay of human serum cholesterol and for reduction of cholesterol level in foods and feeds. In order to search the microorganism which has a high extracellular and stable activity of cholesterol oxidase, soil microorganisms were screened. As a result, the one with the highest extracellular cholesterol oxidase activity was obtained and named as the BEN 115. The BEN 115 strain was identified as one of the Nocardia species based on our taxonomic studies. The cholesterol oxidase from this strain was shown to have two bands of extracellular proteins on SDS-PAGE and Western blot. Their molecular masses were estimated to be about 55 and 57 kDa, respectively. In addition, this cholesterol oxidase was considerably stable at the broad range of pH $3.5{\sim}9.5$ and at the temperature of $25{\sim}55^{\circ}C$. The optimum pH and temperature of this cholesterol oxidase were pH 5.5 and $35^{\circ}C$, respectively. The activity of extracellular cholesterol oxidase could be enhanced 1.6 to 2.0 folds by the addition of nonionic detergent such as Triton X-114, Triton X-100, or Tween-80 into the culturing broth. The substrate specificities against campesterol, sitosterol and stigmasterol were measured to be 50%, 50%, and 27%, respectively, compared to the cholesterol. These results suggest that Nocardia sp. BEN 115 may be useful as a microbial source of cholesterol oxidase production.

Characterization and Partial Purification of Storage Protein-i Receptor in the Fat Body of Hyphantria cunea (미국흰불나방 지방체에서 저장단백질-1의 수용체의 특성과 부분정제)

  • In vitro tissue culture of fat body of Hyphantria cunea in the medium containing [35S]-methionine reveaied that storage protein-i (SP-1) is taken up into fat body of prepupae and 1-day-old pupae. Using Western blotting and ligand binding method, we were able to identify the protein band of the SP-1 receptor protein. For the partial purification, the membrane proteins of fat body cells were solubilixed with 1% Triton X-1OO and applied to anion exchange chromatography. The results revealed the molecular weight of the receptor protein to be about 80 kl)a in SDSPAGE, and the P1 was estimated to be about 6.1. The mobility of the receptor protein in 8D8-PAGE was highly dependent on both temperature during electrophoresls and the condition of samples whether they were in reducing or nonreducing.
