To evaluate effects of ligninolytic enzyme type on the mycelial response and ligninolytic enzyme production during interspecific interactions among wood-rotting fungi, 4 fungal strains, Trichophyton rubrum LKY-7, Trichophyton rubrum LSK-27, Pycnoporus cinnabarinus, and Trichoderma viride, were selected. Regarding ligninolytic enzyme production, LKY-7 secreted laccase and manganese peroxidase (MnP), P. cinnabarinus secreted only laccase, and LSK-27 secreted only MnP in glucose-peptone medium, while T. viride did not produce any ligninolytic enzymes. In the co-culture of LKY-7 with P. cinnabarinus, the formation of aerial mycelium was observed and the enhancement of laccase activity owing to interspecific interaction appeared to be very low. In the co-culture of LKY-7 and P. cinnabarinus with LSK-27, a hypha-free clear zone was observed, which resulted in deadlock, and increased laccase or MnP activity was detected at the interaction zone. The interaction responses of LKY-7, P. cinnabarinus, and LSK-27 with T. viride were characterized by the formation of mycelial barrages along the interface. As mycelial barrages were observed at the T. viride territory and no brownish pigment was observed in the mycelial barrages, it is suggested that laccase and MnP are released as part of an offensive response, not as a defensive response. The co-culture of P. cinnabarinus with T. viride lead to the highest enhancement in laccase activity, yielding more than 14-fold increase in laccase activity with respect to the mono-culture of P. cinnabarinus. MnP activities secreted by LKY-7 or LSK-27 was generally low in interspecific interactions.
Keratinophilic fungus (KPF), a type of dermatophytes, is usually present as normal flora on the skin of humans and animals but can produce ring worm-like dermatophytosis by invading the skin in infected individuals. They are distributed worldwide, but their occurrences vary distinctively in accordance with the geographical location and environmental change. Because these fungi grow by degrading keratin, they are abundantly found on the skin, hair, and nails, which are rich in keratin. To investigate the presence of keratinophilic fungi in the soil, we selected a popular beach in South Korea, Haeundae Beach, where numerous people gather each year during the summer holidays. Hundred soil samples were analyzed using the hair-baiting technique, among which, a total of 23 colonies of KPF were identified from 21 soil samples. The identified KPF were Microsporum gypseum (43%), Chrysosporium spp. (35%), Trichophyton ajelloi (13%), and Microsporum cookie (9%). This study confirmed that pathogenic fungi can be found in places crowded by many people. Further research and continuous data collection are needed to confirm the distribution of pathogenic KPF.
An endospore-forming, rod-shaped bacterium was isolated from forest soil samples collected at the Taebaek mountain of Gangwon province, Korea, and taxonomically characterized by physiological, biochemical and phylogenetic methods. Its 16S rRNA sequences showed the maximum similarity of 97% with B. amyloliquefaciens. In addition, the isolate BCNU 2002 was determined to have the ability to produce enzymes such as amylase, protease, gelatinase and catalase. The in vitro antifungal activity of Bacillus sp. BCNU 2002 was also examined against human pathogenic fungi such as Aspergillus niger, Candida albicans, Epidermophyton floccosum, Saccharomyces cerevisiae, Trichophyton mentagrophytes and Trichophyton rubrum. A maximum production level of antifungal substances of Bacillus sp. BCNU 2002 was achieved under aerobic incubation at $28^{\circ}C$ for 7 days in LB broth. BCNU 2002 showed strong antifungal activities against T. mentagrophytes and T. rubrum with the range of percentage inhibition from 56.25 to 63.23%. It was also confirmed that ethylacetate extract of cultured broth showed a strong antifungal activity against A. niger, C. albicans, S. cerevisiae and T. rubrum by agar diffusion method. The peptide fraction also exhibited broad antifungal spectrum against various pathogenic fungi. The minimum inhibitory concentration values for active extracts ranged between 125 ${\mu}g$/mL and 1000 ${\mu}g$/mL.

The analogs of cyclic hydroxamic acids containing N-OH group have been proposed to play effective laccase-mediators in kraft pulp bleaching with laccase/mediator system. The existing mediators (NHA, 1-HBT, VA), the best laccase-mediators reported so far, and selected several analogs of cyclic hydroxamic acids were evaluated as a laccase-mediator for kraft pulp bleaching. It was found that NHA was the most effective mediator for the Trychophyton sp. LKY-7 laccase (TrL) in kraft pulp bleaching with TrL/mediator system, increasing about 10% ISO of brightness and decreasing about 2.8 of kappa number after alkaline-peroxide bleaching. Of the cyclic hydroxamic acidstested, the NHP.1(N-hydroxypyridone analog) was shown to enable TrL to effectively degrade lignin in kraft pulp bleaching, demonstrating approximately similar effect with that of NHA. However, the effect of substituent patterns of cyclic hydroxamic acid analogs in kraft pulp bleaching was not observed. The inhibitions of NHA and NHP.1 on TrL were not exhibited in TrL/mediator system. As a new mediator for TrL, NHP.1 was considered to be able to use in kraft pulp bleaching with TrL/mediator system.
This study was undertaken to investigate the activity or the water extract of Polygonum aviculare Linne in vitro. Some of the purely isolated strains or dermatophytes were inoculated on Sabouraud's dextrose agar medium containing different concentrations of the Polygonum extracts and their growth was observed for about two weeks at room temperature. Then we measured the sizes of fungal colony grown in various conditions and compared them with those of Sabouraud's medium as control to determine fungistatic effectiveness of the extract. For additional study, slide cultures on Sabouraud's dextrose medium and Sabouraud's media containing 3ml/10ml extract were performed with Epidermophyton floccosum to observe the growth of hyphae, sporulation and other mycological findings. 1. The growth of Epidermo-phyton floccosum was completely inhibited in the media containing 3ml/l0ml Polygonum extract. 2. Trichophyton rubrum, Trichophyton mentagrophytes and microsporum canis were completely inhibited in several strains of each specimen and a moderate inhibitory effect was observed in all of another strains in the media containing the 3ml/10ml extract. 3. In the slide culture of Epidormophyton floccosum the hypha was thin and more desiccated. The characteristic macroco-nidia formation was not observed on the media containing 3ml/10ml Polygonum extract as compared with those findings of Sabouraud's dextrose control medium.
In this study, we evaluated antifungal and antioxidant activities of condensed liquid of volatile organic compounds generated during the drying process of Chamaecyparis obtuse. Condensed liquid of volatile organic compounds were extracted with hexane and ethyl acetate, respectively. The extracts were used for antifungal activity at range from 100 to 500 mg/L Antifungal activity of extracts increased as the extracts concentration increased, the activity against tested dermatophytes was high at 500 mg/L. Among extract solvents, antifungal activity was higher at hexane extract than the ethyl acetate. Especially, the antifungal activity against Trichophyton rubrum was higher than other tested dermatophytes. Antioxidant activity was 90% above 25 mg/L regardless of the extractive solvents, while the hexane extract showed high antioxidant activity below 25 mg/L. Major compounds of hexane extract were torreyol, alpha-cadinol, and tau-cadinol. However, major compounds of ethyl acetate extract were alpha-amorphene, alpha-cadinol, and gamma-cadinene.
Many nucleoside compounds such as 5-halogen substituted uridine, 5'-amino-5'-deoxyuridine conjugates of amino acid, peptide, and penicillin G, 5'-monophosphate uridine derivatives and 5'-monophosphate uridine-fatty acid derivatives were chemically synthesized and their antifungal, antibacterial, and antitumor activities were tested. 5-Bromo-2',3'-O-isopropylideneuridine(6) inhibited the growth of Trichophyton rubrum at $0.2{\mu}$g/ml of MIC. 5'-Amino-5'-deoxyuridine-penicillin G(19), 5'-amino-5'-deoxyuridine-cyclo(Phe-Asp)(20), and 5-iodo-5'-amino-5'-deoxyuridine- penicillin G(22) had antibarterial activity(MIC was $6.25{\mu}$g/ml against S. aureus) and the latter two nucleoside compounds were the most antitumor derivatives(their $IC_{50}$ against L5178Y murine lymphoma cell was $6.5{\mu}$g/ml).
A water soluble antifungal antibiotic, Solumycin, was separated from the culture broth of Streptomyces sp. LAM-593, isolated from soil, by butanol extraction, alumina-, 1st and 2nd Sephadex LH-20 column chromatography. The substance was pale yellow crystal which gave a single spot at Rf value 0.24 with ethanol-ammonia water-water (8:1:1), 0.46 with butanol-ethanol-water (5:1:4), 0.84 with 50% methanol on silica gel TLC. It was dissolved well in water, methanol and acidic aq. butanol but not in ethanol, acetone, ethyl acetate, chloroform. acetic acid etc., and gave positive Fehling and Molish reaction. The UV spectrum in methanol showed absorption at 342, 361, 380, and 404 nm. The antibiotic was active against fungi such as Candide, Cryptococcus, Saccharomyces, Trichophyton and Trichosporon, but not to bacteria such as Bacillus, Escherichia and Staphylococcus.

A new wood-degrading fungus Trichophyton rubrum LKY-7 secretes a high level of laccase in a glucose-peptone liquid medium. The production of laccase by the fungus was barely induced by 2,5-xylidine. The laccase has been purified to homogeneity through three chromatography steps in an overall yield of 40%. The molecular mass of the purified laccase was about 65 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The purified laccase had the distinct blue color and had basic spectroscopic features of a typical blue laccase: two absorption maxima at 278 and 610 nm and a shoulder at 338 nm. The N-terminus of the laccase has been sequenced, revealing high homology to laccases from wood-degrading white-rot fungi such as Ceriporiopsis subvermispora. The enzyme had a "low" redox potential (0.5 V vs normal hydrogen electrode), yet it was one of the most active laccases in oxidizing a series of representative substrates/mediators. Compared with other fungal laccases, the laccase has a very low Km value with ABTS [2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid] as a substrate and a very high Km value with violuric acid as a substrate. The laccase has the isoelectric point of 4.0. The laccase had very acidic optimal pH values (pH 3-4) while it was more stable at neutral pH than at acidic pH. The laccase oxidized hydroquinone faster than catechol and pyrogallol. The oxidation of tyrosine by the laccase was not detectable under the reaction conditions. The laccase was strongly inhibited by sodium azide and sodium fluoride. fluoride.
The aim of this study was to investigate the terpenoids of Total Volatile Organic Compounds (VOCs) released during drying of Cryptomeria japonica using the thermal extractor (TE). Considering the drying process of C. japonica, temperatures of TE were set at $27^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$, $100^{\circ}C$, and $120^{\circ}C$, respectively. As the result, the emission factors of VOCs and terpenoids were increased as temperature increased. The amount of terpenoids included in VOCs emission factors were 87.5%, 81.6%, 83.6%, 90.1%, and 97.3% depending on above temperatures, respectively. Especially at$100^{\circ}C$ and $120^{\circ}C$, the amount of terpenoids were measured more than 90%. ${\delta}$-cadinene was the highest yield at each temperature and 32 types of terpenoids were collected. Emitted terpenoids were classified into the sesquiterpene group which consists of 15 carbon sources. These 32 sesquiterpenes were used for determining the useful bioactivity such as antifungal activity by the agar dilution. As the result, they showed the antifungal activity against Trichophyton rubrum, Trichophyton mentagrophytes, Microsporum gypseum. The 5,000 ppm concentration of terpenoids showed a strong activity with 100% against the 3 fungi. At the 1,000 ppm concentration of terpenoids, the antifungal activities against three fungi were 95.2%, 98.7%, and 97.3%, and their activities were a little inhibited at 100 ppm concentration.
