A Study on the Characteristics of Pine-tree Mushroom(Tricholoma matsutake Sing.) Pickle for the Standard Recipe (냉동 자연 송이버섯의 피클 조리법 표준화를 위한 연구)

  • This study investigated the rheological and sensory characteristics of pickle with frozen pine mushroom. P3(Developed pickle seasoning) was the best by preference among three kinds of pine mushroom pickle seasoning. For flavor and functionality, pine mushroom pickle was processed by using three kinds of method(P3-1: P3+cinnamon 10 g, P3-2: P3+licorice 10 g and P3-3: P3+licorice 5 g+cinnamon 5 g). As a result, the product from P3-3(P3+licorice 5 g+cinnamon 5 g) was the best preferred pine mushroom pickle. The pH value of P3-3 was 2.15, 42.9 degrees Brix, and its color value was L(54.65), a(-1.61), b(17.87). Its texture level was higher than that of other products, but it would be lowered on storage. Until the 28th day of storage, microorganisms in pine mushroom pickle seasoning were detected less than 30 CFU/mL.

Mycorrhizal Formations and Seedling Growth of Pinus desiflora by in vitro Synthesis with the Inoculation of Ectomycorrhizal Fungi

  • The mycelia were directly isolated from eight species of fungal basidiocarps, confirmed to the ectomycorrhiza in the roots from the fields(forestry); Suillus bovinus, Paxillus involutus, Lactarius hysginus, Russula fragilis, Lepista nuda, Lyophyllum shimeji, Tricholoma matsutake, and Russula integra. The mycelia were pure-cultured with several transferring in various agars, and inoculated to the roots of pine(Pinus densiflora) seedling by in vitro method. After ten months growth under artificially aseptic conditions, all pine seedlings inoculated were stimulated at the growth-height, whereas those not inoculated were nearly dead. Also, the ramifications of ectomycorrhizal pine roots formed in the synthetic in vitro systems and were various according to the different mycelia. Synthesis of ectomycorrhiza were clearly confirmed in ten months growth, but not distinguished at this moment. It was clearly proved that the mycelia isolated caused the ectomycorrhizae in the roots of pine seedlings.

  • Ectomycorrhizal fungi are associated with plants roots and acquire significant amounts of nitrogen sources from the soil. For artificial cultivation, mass production of ectomycorrhizal fungi in liquid media is required. We studied the edible ectomycorrhizal mushrooms Hygrophorus russula, Ramaria fumigata, Sarcodon aspratus, and Tricholoma matsutake. All strains except S. aspratus NIFoS 2031 grew generally well on modified Melin-Norkran's (MMN) medium compared to on other media. All strains analyzed in this study showed significantly higher growth on organic nitrogen. Specifically, two strains of H. russula significantly responded to both tryptone and neopeptone media. Among different species and strains, there were clear differences in the capacity to grow on animal-based organic nitrogen sources.

  • ABSTRACT$.$Seven species of mushrooms (Sarcodon asparatus, Agaricus bisporus. Calocybe gambosa, Len tin us ,edodes.Ramariabotrytis.Pleurotus ostreatus and Tricholoma matsutake) produced in Korea were investigated on their mineral contents mainly by using Inductively coupled plasma-technique. The obtained results are summerized as follows: 1) Fe, N a and Zn are relativel highly contained in almost all mushrooms tested in this study. 2) Harmful elements such as Cd, Hg. Pb and As, were found only in trace amounts. 3) Sb, Se and Sn were not detected in all mushrooms.hrooms.

  • This study was conducted to examine the physiology of pine mushroom mycelia cultured with various media for artificial culture of pine mushroom. The results obtained were as follows: 1) Among the various media, the medium composed of honey, boiled pine mushroom and soil extract fluid, fibrous root extract fluid, dry yeast, $KH_2PO_4$ inositol, folic acid, and biotin was the best for the growth of pine mushroom mycelium. 2) The optimum temperature for germinating pine mushroom spore and for culturing pine mushroom mycelium, was $24^{\circ}C$ and the optimum pH was 4.5. 3) There was no significant difference in growth between the mycelium separated from the tissue of pine mushroom sporophore and that separated from the spore. 4) No noticeable effect was found on the growth if such salts as $ZnSO_4$, $MnSO_4$, $MgSO_4$, $CaCl_2$ and ferric citrate were added to the Hamada's medium. 5) The addition of fibrous root extract promoted the growth of pine mushroom mycelium. 6) As a carbon source of artificial media, honey was more effective than glucose. 7) The culture infiltration of Mortierlla growing often in Fairy Ring was good for the growth of mycelium compared with the control. 8) The addition of fibrous root extract, inositol, biotin, and folic acid to artificial culture media was greatly effective in growth. When the temperature was lowered $19^{\circ}C$ after mycelium has appeared, the formation of primordium was observed.

Effects of Climatic Condition in September on Pine-mushroom(Tricholoma matsutake) Yield and a Method for Overcoming the Limiting Factors in Korea (한국(韓國)에서 9월(月)의 기상인자(氣象因子)가 송이 발생(發生)에 미치는 영향(影響)과 그 극복방안(克服方案))

  • Relationships between pine-mushroom(Tricholoma matsutake) yield and important climatic factors for the yield(such as monthly precipitation, relative humidity, temperature, and radiation percentage) were examined to find out limiting factors for pine-mushroom production and to develop a method for overcoming the factors by analyzing the yield and climate data for 17 years collected from 18 main regions of pine-mushroom production. Although there were variations among the production regions, climatic condition of September was the most significant factor for pine-mushroom yield in general, and the degrees of importance of each climatic factors were different among the production regions. Mean minimum temperature of September was positively correlated with pine-mushroom yield($$r^2{\geq_-}0.41$$) at the 1% level, of which were 9 regions such as Youngduck, Uljin, Samchuck, Bongwha arid so on. In these regions, vegetation control was expected to be effective for pine-mushroom production by allowing much sunlight penetration to the pine stand, which may increase soil temperature and keeping the temperature around the fungal colony in soil. Precipitation during September was positively correlated with pine-mushroom yield($$r^2{\geq_-}0.41$$) at Namwon, Moonkyung and Sangju. Thus, irrigation around fungal colony in dry soil during September would be effective for enhancing mushroom yield at the regions. Pine-mushroom yield of 1994 was quite low, similar to that of 1993, due to serious drought. In this period, we could manifest the possibility of enhancing pine-mushroom yield by irrigation for overcoming drought at Moonkyung and Keochang regions.

Survey on distribution and vegetation environment of Tricholoma matsutake in Kyonggi province, Korea (경기도지역 송이버섯 분포 및 식생환경조사)

  • We carried out survey of a pine mushroom picking district and its vegetation environment for increase and maintainment of its production in Kyonggi province. The district which we could pick up pine mushroom among the nineteen surveyed district were sixteen such as Geoleun ri Gangcheon myeon Yeoju gun Kyonggi do, etc. and total yields were 470~600 kg in Kyonggi province. The temperature of atmosphere and earth in Yeoju and Bonghwa were droped to $19^{\circ}C$ at 5~7th september and maintained from $12^{\circ}C{\sim}18^{\circ}C$ during the a pine mushroom picking period. The density of a pine tree per $100m^2$ was 5.1~27.1 trees, the years of it was 15~50years old and height was 4~11 meter. The pine tree in a pine mushroom picking district was competed with oak trees, azaleas, lacquers, etc. The depth of organic materials accumulated on the pine mushroom mycelium layer was 4.5 cm in Yeoju. The soil texture of a pine mushroom picking district was almost sandy, soil pH was 4.3~5.1 and organic material content was 3.91~8.28%.

  • A study on the variation of the air temperature in the pine mushroom box containing dry ice for the preservation of the freshness of the pine mushroom, Tricholoma matsutake (Ito et Imai) Sing. produced in Korea was made in Seoul, Korea during the period of August 1 to 30, 1975. A safe shipment of 5 kg (net weight) of fresh mushrooms with freshness preserved for 24 hours in August required the placement of 4.5 kg of dry ice-a half in the upper portion and the other half in the middle portion-in an airtight container (mushroom box). The container used for the shipment test, made of 3mm-thick double-layer cardboard, was of the dimensions : $0.33{\times}0.4{\times}0.5=0.066(m^3)$. As the temperature fell, decreased amounts of dry ice were needed : 3 to 2 kg in September and 1.5 to 0.3 kg in October. With this preservation method, large quantities of fresh mushrooms were exported to Tokyo ana Osaka, Japan without damage in the process of transportation.

Isolation and Taxonomical Characterization of Strain KM1-15 with Antibiotic Activity from Pine Mushroom (Tricholoma matsutake) Basal Soil (송이 자실체 기저부 토양으로부터 항균활성을 가지는 KM1-15 균주의 분리 및 분류학적 특성)

  • Two hundred and sixty-eight bacterial strains were isolated from pine mushroom (Tricholoma matsutake) basal soil. In the course of screening for antifungal activity against seven plant pathogenic fungi (Alternaria panax, Botrytis cinerea, Colletotrichum gloeosprioides, Fusarium oxysporum, Phytopthora capsici, Pythium ultimum, Rizoctonia solani) of isolates, strain KM1-15 showed strong antibiotic activity against Alternaria panax and Colletotrichum gloeosprioides. In determining its relationship on the basis of 16S rDNA sequence, KM1-15 strain was most closely related to Bacillus $koguryoae^T$ (AY904033) (99.62%). When assayed with the API 50CHE Kit, unlike Bacillus koguryoae, it is positive for utilization of L-arabinose, cellobiose, inulin, and D-turanose. Results of cellular fatty acid analysis showed that major cellular fatty acids were 15:0 anteiso (35.78%) and 17:0 anteiso (17.97%). In particular, hydroxyl fatty acids such as 13:0 iso 3-OH, 14:0 iso 3-OH, 15:0 iso 3-OH, and 17:0 iso 3-OH were only restricted to strain KM1-15. DNA G+C content was 43.7 mol% and quinone system was MK-7 (100%) in strain KM1-15.

Use of Sprinkler System for Production Forest Management of Pine Mushroom (Tricholoma matsutake) (살수장치(撒水裝置)를 이용(利用)한 송이산 관리(管理)에 관(關)한 연구(硏究))

  • In order to prevent the pine mushrooms, Tricholoma matsutake, from being damaged by the pine needle gall midges, Thecodiplosis japonensis, and thereby, to increase their production and improve their quality, a sprinkler system was installed on the mushroom field. A low-concentration insecticide (deltamethrin 1% EC, x2,000) was sprayed once at insects' most active time every day during the period of insects' adult occurrence and thereafter, the irrigation by ground water spraying was periodically enforced. Such a test was conducted at Yangyang-Gun, Kwangwon-do, Korea for 2 years from 2000 through 2001. The pine needle gall midges generally emerged for about 40 days from late May to early July. 50% emergence of them was about June 6, and peak emergence (more than 80%) was early or mid-June. Gall formation rate was 3.5% on average with this ground insecticide spraying, while 51.3% when not treated. Control effectiveness of this insecticide spraying was 92.3%, which was higher than 82.5% by the conventional injection of insecticide into tree stems. Pine mushrooms emerged for about 35 days from mid-September through earlier October, and around 80% of them did for about 15 days from late September through early October. As a result of the periodic ground water-spraying (30 mm per week) for 2 months (from August to October), the production of mushrooms increased by 74.3% (110% in terms of weight), with their quality improvement. The mushrooms produced from the treated stand by the spraying system were priced 8,670,000 wons per hectare, and thus, the net income deducting the facility and management cost was 4,310,000 wons, about 5% higher than value from the control stand. It was analyzed that this treatment was significantly cost effective when the facilities are used more than 5 years.
