Studies on the Anticoccidial Efficacy of an Unique Polyether Ionophorous Antibiotic, Maduramicin Ammonium in Comparison with Salinomycin Sodium and Monensin Sodium for Broiler Chicks (폴리에텔계 항생제인 Maduramicin ammonium, Salinomycin sodium 및 Monensin sodium이 육계에 있어서 항콕시듐 효능과 증체에 미치는 영향에 관한 연구)

  • Battery trial with 240 broiler chicks of Hubbard strain was conducted for a period of 2 weeks in order to compare the anticoccidical efficacy of polyether ionophorous antibiotics ; Maduramicin ammonium, Monensin and Salinomyc in sodium. The criteria used in these anticoccidial efficacy studies were anticoccidial index, growth rate, feed efficiency, mortality, lesion score and the number of oocysts produced after artificial inoculation with 70,000 sporulated oocysts of Eimeria tenella(90%) and E.necatrix (10%) to each bird. The result obtained are summarized as follow: 1. All groups medicated anticoccidial feed additives improved body weight gain and feed efficiency. However, it was found that the group medicated with Maduramicin showed better body weight gain (352.5 and 648.8 g) and feed efficiency(1.603 and 1.680) during the first and the second week experiments, 2. The mortality rate(4.2%) and lesion scores (1.72) of Maduramicin medicated group, from artificial coccidiosis were comparatively lower than those of other two medicated groups, 3. It was also found that oocyst output (0.25 ${\times}$ 10$^4$) in Maduramicin medicated group were lower than those of other two groups. 4. Anticoccidial indexes during the first week were 177.9 in Maduramicin medica-group, 158.7 in Salinomycin medicated group, 141.6 in Monensin medicated group and 78.0 in infected, nonmedicated group as compared with 200.0 in noninfected, nonmedicated group (NNC) 5. Anticoccidial indexes during the second week were 201.1 in Maduramicin group 184.0 in infected, nonmedicated group as compared with 200.0 in noninfected, nonmedicated group (NNC).

A Criteria on Nitrate Concentration in Soil Solution and Leaf Petiole Juice for Fertigation of Cucumber (Cucumis sativus L.) under Greenhouse Cultivation (시설 오이의 관비재배를 위한 토양용액과 엽병즙액중 질산태 농도 기준 설정)

  • To develope a technique for efficiently managing fertilizer for cucumber, a quick test method to quantify nitrate content in soil solution and leaf petiole juice using a simple instrument was investigated. Among the nitrate analyzing instruments such as compact ion meter, nitrate ion meter, and test strip with reflectometer, the paper test-strip used in conjunction with a hand-held reflectometer was most closely correlated with ion chromatography method in nitrate content, and then it would be suggested with a tool that a farmer can use rapidly, conveniently and accurately for nitrate analysis in a field. Nitrate content in soil solution collected by porous cup was very variable on the lapsed time after drip irrigation and the sampling positions such as soil depth and the distance from dripper. As a result, a significant correlation between nitrate contents of soil solutions and 2M KCl soil extract was not found. However, nitrate content in soil solution extracted with a volume basis (soil:water=1:2) showed the highly significant correlation with that in 2M KCl extract. Nitrate contents of cucumber leaf petiole juices was greatly different between upper and lower leaves. Eleven to sixteen positioned-leaf would be a proper sampling position to determine nitrate content in leaf petiole for evaluating nutrient state by plant tissue analysis. From the secondary regression equations between nitrate contents of soil and petiole juice and the yield of cucumber, nitrate levels for real time diagnosis were estimated as $400mg\;l^{-1}$ soil solution by porous cup. $300mg\;l^{-1}$ in a soil volume extraction, and $1400mg\;l^{-1}$ in petiole juice from spring to summer season. In addition, the maximum yield of cucumber fruit in pot test was obtained in nitrate $1500mg\;l^{-1}$ level of petiole juice, which was similar to nitrate $1400mg\;l^{-1}$ in greenhouse trial.

Sequential Chemotherapy and Radiation Therapy for Advanced Nasopharyngeal Carcinoma (진행된 비인강암의 화학요법 및 방사선 치료)

  • Between January 1985 and July 1992, 52 patients with locally advanced nasopharyngeal carcinoma were studied retrospectively for the effectiveness of sequential chemotherapy and radiation therapy. The male to female ratio was 3.3:1 with a median age of 41 years. Forty patients had squamous cell carcinoma and the remaining 12 had undifferentiated carcinoma. Seven patients had stage III disease and the remainder had stage IV disease at time of presentation. All patients were treated two courses of chemotherapy followed by radiation therapy. Chemotherapy consisted of either CVB (cisplatin, vincristine and bleomycin) or CF (cisplatin and 5-FU). Total radiation dose to the primary site ranged from 6000 cGy to 7500 cGy. Neck nodes were given booster treatment to maximum of 7000 cGy, depending on the extent of disease. Local control, overall survival and disease-free survival rates were analyzed. The complete response (CR) rate to chemotherapy was $15\%$ and the partial response (PR) rate was $46\%,$ for overall major response rate of $61\%.$ The CR rate was $87\%$ after radiation therapy. Median follow-up time was 51 months. The overall survival and disease-free survival rates at 36 months were $54\%\;and\;49\%,$ respectively. Median time to relapse was 15 months. The patterns of initial relapse in CR patients was as follows: locoregional failure only, 12 patients; distant metastasis only,11: both,2. Cox's multivariate regression model revealed that nodal status was the single most important independant prognostic factor influencing disease-free survival (p=0.001). Comparision of these results with other published reports with radiation therapy alone showed that a high rate of initial response to chemotherapy did not translate into local control or survival. At present time radiation therapy alone remains the standard treatment for locoregional cancer of the nasopharyngeal cancer. More controlled clinical trials must be completed before acceptance of chemotherapy as a part of treatment of advanced nasopharyngeal carcinoma.

Radiotherapy for Oral Cavity Cancer (구강암의 방사선치료)

  • Eighty five patients of oral cavity cancer, treated with radiation at the Department of Therapeutic Radiology, Korea Cancer Center Hospital, during the period from March 1985 to September 1990 were analyzed retrospectively. Among 85 patients, 37 patients were treated with radiation only and 48 patients were treated with radiation following surgery. And 70 patients received external irradiation only by $^{60}Co$ with or without electron, the others were 7 patients for external irradiation plus interstitial implantation and 8 patients for external irradiation plus oral cone electron therapy. Primary sites were mobile tongue for 40 patients, mouth floor for 17 patients, palate for 12 patients, gingiva including retromolar trigone for 10 patients, buccal mucosa for 5 patients, and lip for 1 patient. According to pathologic classification, squamous cell carcinoma was the most common (77 patients). According to AJC TNM stage, stage I + II were 28 patients and stage III+IV were 57 patients. Acturial overall survival rate at 3 years was $43.9\%,$ 3 year survival rates were $60.9\%$ for stage I + II, and $23.1\%$ for stage III+IV, respectively. As a prognostic factor, primary T stage was a significant factor (p<0.01). The others, age, location, lymph node metastasis, surgery, radiation dose, and cell differentiation were not statistically significant. Among those factors, radiation plus surgery was more effective than radiation only in T3+T4 or in any N stage although it was not statistically sufficient (p<0.1). From those results, it was conclusive that definitive radiotherapy was more effective than surgery especially In the view of pertainig of anatomical integrity and function in early stage, and radiation plus surgery was considered to be better therapeutic tool in advanced stage.

Modification of Late Radiation Response of Rat Salivary Glands by Pentoxifylline and Diltiazem (쥐의 타액선 방사선조사 후 만성반응에 Pentoxifylline과 Diltiazem이 미치는 영향)

  • Purpose : To elucidate the effects of pentoxifylline and diltiazem on the late response of the salivary glands of the rat after irradiation. Materials and Methods : Sixteen Sprague-Dawley rats were divided into 4 groups : (a) irradiation alone (b) irradiation with pentixifylline (PTX) (c) irradiation with diltiazem (DTZ) (d) irradiation with both PTX and DTZ. Irradiation was given in a single fraction of 16 Gy using 4 MV photon energy through an anterior port encompassing the left side of the salivary gland leaving the right side of salivary gland as a control. PTX, 20 mg/kg and/or DTZ, 50 mg/kg were infused intraperitoneally before irradiation, Two rats from each group were sacrificed on the 10th week and the rest was sacrificed on the 16th week after irradiation. Histopathologic examinations were undertaken for each section and the proportion of vacuolated cells out of the total number of cells under light microscopic fields was calculated. The statistical significance in the difference of the proportion of the vacuolated cells among the experimental groups was evaluated by a $x^2$-test. Results : Irradiated salivary glands of the 10th week group revealed markedly increased number of vacuolated cells compared to those of unirradiated control. The proportion of vacuolated cells was significantly reduced in both the PTX group (p value=0.001) and the combined PTX and DTX group compared to those of irradiation alone group. The DTZ alone group did not reveal the significant reduction of vacuolated cells compared to those of irradiation alone group (p value, >0.05). The 16th week groups revealed similar findings to those of the 10th week group, but the degree of chronic inflammatory cell infiltrates and interstitial fibrosis was increased and the number of acinar cells was reduced compared to those of the 10th week group. Conclusions : PTX significantly reduced the late radiation response of salivary glands, but DTZ did not reduce the same degree as PTX did. Taking the positive results of this study into consideration, it seems reasonable to apply PTX into the clinical trial for the head and neck irradiation to reduce the late radiation sequelae of salivary glands in the near future. At the same time the further experiment to clarify the subcellar mechni는 involved in PTX should be preceded.

Combined Transcatheter Arterial Chemoembolization and Local Radiotherapy for Unresectable Hepatocellular Carcinoma (절제불가능 원발성 간암에서 경간동맥 항암 색전술과 국소 방사선의 병용요법)

  • Purpose : The best prognosis for hepatocellular carcinoma can be achieved with surgical resection. However, the number of resected cases is limited due to the advanced lesion or associated liver disease. A trial of combined transcatheter arterial chemoembolization(TACE) and local radiotherapy(RT) for unresectable hepatocellular carcinoma(HCC) was prospectively conducted and its efficacy and toxicity were investigated. Materials and Methods : From 1992 to 1994, 30 Patients with unresectable HCC due either to advanced lesion or to associated cirrhosis were entered in the study Exclusion criteria included the presence of extrahepatic metastasis, liver cirrhosis of Child's class C, tumors occupying more than two-thirds of the whole liver, and an ECOG scale of more than 3. Patient cHaracteristics were : mean tumor size $8.95\pm3.4cm$, serum AFP+ in all patients, portal vein thrombosis in all patients, liver cirrhosis in 22 patients, and UICC stage III and IVA in 10 and 20 patients, respectively. TACE was performed with the mixture of Lipiodol(5ml) and Adriamycin(50mg) and Gelfoam embolizatin. RT(mean dose $44.0\pm9.3Gy$) 10 days with conventional fractionation. Results : An objective response was observed in 19 patients($63.3\%$). Survival rates at 1 2, and 3 years were $67\%,\;33.3\%$ and $22.2\%$, respectively. Median survival was 17 months. There were 6 patients surviving more than 3 years. Distant metastasis occurred in 10 patients, with 8 in the lung only and 2 in both lung and bone, Toxicity included transient elevation of liver function test in all patients, fever in 20, thrombocytopenia in 4, and nausea and vomiting in 1. There was no treatment-related death. Conclusion : Combined TACE and RT appear to produce a favorable response and survival results with minimal toxicity.

Sequential Chemoradiotherapy for Stage I/II Nasal Natural Killer/T Cell Lymphoma (I/II 병기 비강 Natural Killer/T Cell 림프종에 대한 순차적 항암화학요법과 방사선치료)

  • Purpose: Authors would report the results of sequential CHOP chemotherapy (cyclophosphamide, adriamycin, vincristine, and prednisone) and involved field radiotherapy (IFRT) for early stage nasal natural killer/T-cell Iymphoma (NKTCL). Materials and Methods: Fourteen among 17 patients, who were registered at the Samsung Medical Center tumor registry with stage I and II nasal NKTCL from March 1995 to December 1999 received this treatment protocol. Three to four cycles of CHOP chemotherapy were given at 3 weeks' interval, which was followed by local IFRT including the known tumor extent and the adjacent draining lymphatics. Results: Favorable responses after chemotherapy (before IFRT) were achievable only in seven patients (5 CR's+2 PR's: 50%), while seven patients showed disease progression. There were six patients with local failures, two with distant relapses, and none with regional lymphatic failure. The actuarial overall survival and progression-free survival at 3 years were 50.0% and 42.9%. All the failures and deaths occurred within 13 months of the treatment start. The factors that correlated with the improved survival were the absence of 'B' symptoms, the favorable response to chemotherapy and overall treatment, and the low risk by international prognostic index on univariate analyses. Conclusion: Compared with the historic treatment results by IFRT either alone or followed by chemotherapy, the current trial failed to demonstrate advantages with respect to the failure pattern and survival. Development of new treatment strategy in combining IFRT and chemotherapy is required for improving outcomes.

Effect of Pitamin as an Antibiotics Replacement for Organic livestock Feed Additives in Broiler Chickens (유기사료원료로서 소나무껍질 추출물 피타민의 브로일러에 대한 항생제 대체효과)

  • Pitamin is a component of pine bark extract that exhibits antimicrobial activity and a variety of physiological effects. This study was earned out to investigate the effects of dietary pitamin as an organic livestock feed additive in broiler chickens. A 35 day trial was conducted to determine the influence of dietary premix containing 5% pitamin; investigated parameters included blood lipids, growth performance, quality characteristics of carcasses, and changes of caecal microbials in broiler chickens. Chickens were randomly divided into groups that were untreated (control), treated conventionally with antibiotics in the absence of premix, received 0.1 % or 0.2% premix containing 5% pitamin. Plasma lipids were lower in groups fed diets with pitamin premix (p<0.05). The body weight gain from broiler chickens fed with the diet containing 0.1% pitamin premix and antibiotics was similar, and was significantly higher than that of the other groups (p<0.05). The weight of breast muscle and thigh meat of carcasses was similar, and was higher than that of the control group (p<0.05). Abdominal fat and thymus index from chickens receiving either pitamin-supplemented premix was significantly lower and increased, respectively, that of the antibiotic and control groups (both p<0.05). The chickens on the pitamin premix-supplemented diets evidenced significantly higher caecal levels of Bifidobacterium species as compared with the chickens on the control diet (p<0.05). These results suggest that feeding a diet supplemented with a 0.1% premix containing 5.0% pitamin for 35 days maintains the production of broiler chickens at a level comparable to that obtained from the use of antibiotics.

  • Objectives In this study, we divided Korean asthma patients into excess syndrome or deficiency syndrome groups according to clinical phenotype. Genetic analysis was conducted to investigate the association of exonic SNPs in the CD46 gene polymorphism with the clinical phenotype based on the differentiation syndrome of the bronchial asthma patients.Methods There were 95 healthy patients (control group) and 53 asthma patients. (The deficiency syndrome group included 24 and the excess syndrome group 29). We searched the exonic areas of the CD46 gene in the NCBI website SNPs with <0.01 minor allele frequency (MAF) and <0.01 heterozygosity. We finally selected two SNPs: rs138843816, Ser13Phe and rs7144, 3’-UTR. Hardy-Weinberg equilibrium was calculated using SNPStats.Results There were significant differences in the codominant 1 model and the dominant model between the healthy group and the asthma group. There were significant differences between deficiency syndrome group and the excess syndrome group in the genotype frequencies and in the codominant 1 model, the dominant model, and the log-additive model. The allele frequency of rs7144C showed a significant difference between the deficiency syndrome group and the excess syndrome group. Two-SNP haplotype analysis showed a significant difference in frequency in the deficiency syndrome group and in the excess syndrome group. There were significant differences between the healthy group and the excess syndrome group in the codominant 1 model, the dominant model, and the log-additive model. The frequency of the rs7144 C allele exhibited a significant difference in the demonstration. SNP haplotype analysis between the healthy group and the excess syndrome group showed a significant difference in the frequency of the CT haplotype and the CC haplotype.Conclusions The results indicate that two CD46 SNPs (rs138843816, Ser13Phe and rs7144, 3′–UTR) might be associated with the symptomatic excess syndrome in Korean asthma patients.

Effects of Roughage Feeding Type during the Growing and Early-Fattening Periods on Growth Performance, Feed Efficiency and Carcass Characteristics in Holstein Steers (육성 및 비육전기의 조사료 급여형태가 홀스타인 거세우의 성장 발육, 사료이용성 및 도체특성에 미치는 영향)

  • Twenty four Holstein steers fed commercial concentrates with rice straw(T1), mixed grass hay(T2), or wet brewer’s grains based TMR ad libitum(T3) were used to investigate the effects of roughage feeding type during the growing and early-fattening periods on growth performance, feed efficiency and carcass characteristics. Growth performance and feed efficiency were investigated during the feeding trial conducted from 6 to 23 mo of age, and carcass characteristics were investigated after slaughter. Body weight at slaughter and daily gains were 751 to 802 kg(mean 782 kg) and 1.064 to 1.159 kg/d(mean 1.122 kg/d), respectively, showing that those for T2 were highest followed by T3. Compared with T1, the daily gain for T2 was improved by 8.9%. The TDN intakes per kg gain were 6.87, 7.17 and 7.15 kg for T1, T3 and T2, respectively, indicating that feed efficiency for T2 was improved(approx. 5%) compared with T1. The feeding treatments did not change dressing percentage(mean 60.8%), back fat thickness(6.68 mm) and rib-eye area(84.0$cm^2$). Neither meat quality characteristics nor physicochemical properties were significantly different among the treatments. However, economic analysis accounting for 882.8, 991.7 and 943.8 thousand won per head for T1, T2 and T3, respectively, indicated that T2 and T3 increased(7-12%) total net income compared with T1. Therefore, it may be concluded that high quality meat of Holstein steers can be produced if they are fattened until 23 mo of age and that feeding mixed grass hay as roughage or TMR containing mainly wet brewer's grains during the growing and early-fattening periods can improve growth performance compared with feeding rice straw.
