Growth and Yield Response of Transgenic Rice Plants Expressing Protoporphyrinogen Oxidase Gene from Bacillus subtilis

  • Transgenic rice plants expressing a Bacillus subtilis protoporphyrinogen oxidase (Protox), the last shared enzyme of the porphyrin pathway in the expressed cytoplasm or the plastids, were compared with non-trangenic rice plants in their growth characteristics such as tiller number, plant height, biomass, and yield. Transgenic rice plants of $\textrm{T}_3$ generation had 8 to 15 % and 25 to 43% increases in tiller number compared to non-transgenic rice plants at 4 and 8 weeks after transplanting(WAT); similar values were observed for $\textrm{T}_4$ generation at 4 and 8 WAT. However, the plant height in both $\textrm{T}_3$ and $\textrm{T}_4$ generations was similar between transgenic rice plants and non-transgenic rice plants at 4 and 8 WAT. Transgenic rice plants had 13 to 32% increase in above-ground biomass and 9 to 28% increase in grain yield compared to non-transgenic rice plants, demonstrating that biomass and yield correlate with each other. The increased grain yield of the transgenic rice plants was closely associated with the increased panicle number per plant. The percent of filled grain, thousand grains and spikelet number per panicle were similar between transgenic and non-transgenic rice plants. Generally, the growth and yield of transgenic generations ($\textrm{T}_2$, $\textrm{T}_3$, and $\textrm{T}_4$) and gene expressing sites (cytoplasm-expressed and plastid-targeted transgenic rice plants) were similar, although they slightly varied with generations as well as with gene expressing sites. The transgenic rice plants had promotive effects, indicating that regulation of the porphyrin pathway by expression of B. subtilis Protox in rice influences plant growth and yield.

The Function of ArgE Gene in Transgenic Rice Plants

  • We carried out to study the function of ArgE in transgenic rice plants, which were confirmed by PCR analysis and hygromycin selection. Transgenic rice plants were with selectable marker gene(HPT) inserted in genome of the rice. Southern analysis with hpt probe confirmed by two restriction enzymes that copy numbers of the selectable gene was introduced into the plant genome. We displayed that the relationship between drought stress and ArgE gene with the overexpressing rice plants. From this result, we observed that the degree of leaves damage has no difference in control and transgenic lines. The total RNAs were extracted from 6 weeks-seedling in normal condition in order to examine their expression levels with ArgE-overexpressed transgenic rice. In particular, expression patterns of genes encoding enzymes involved in abiotic stress, including drought and salt stresses. OsGF14a and OsSalt were investigated by reverse transcription-PCR(RT-PCR). Expression levels of the OsSalt gene decreased significantly in transgenic rice plants compared to control plant. However, ion leakage measurement did not demonstrate any leaves damage change between control and ArgE transgenic plants exposure to mannitol treatment. These results suggest that expression of the ArgE is not involved in tolerance for drought stress in rice but may playa role of signaling networks for salt-induced genes.

Disease Occurrence in Transgenic Rice Plant Transformed with Silbene Synthase Gene and Evaluation of Possible Horizontal Gene Transfer to Plant Pathogens

  • Genetic engineering is being used to enhance disease resistance and nutritional value of crops including rice plant. Considering the fast-growing agricultural biotechnology and rapidly increasing global area of transgenic crops, the risk evaluation on environment is necessary. In this study, we surveyed the difference of disease occurrence between transgenic rice variety, Iksan526 transformed with peanut stilbene synthase gene and non-transgenic rice varieties, Dongjin and Nampyeong in the field. Moreover, the possibility of gene transfer from transgenic rice to bacterial and fungal pathogens was investigated. The results of this study indicated that there was no significant difference in the occurrence and severity of the diseases between Iksan526 and Dongjin or Nampyeong. In addition, the results suggested that rice pathogen, such as Xanthomonas oryzae pv. oryzae, Rhizoctonia solani and Magnaporthe grisea did not take up stilbene synthase and bar genes under natural conditions. Moreover the transformed DNA was not transferred to the pathogens even in repetitive contacts.

Agrobacterium-mediated Transformation of Rice 'Ilmibyeo' using HPT Selection Maker Gene

  • This study was conducted to produce the transgenic plant of rice. We obtained Agrobacterium AGL1 harbaring pCambial 300 vector with HPT gene. We carried out PCR analysis of 22 ea putative transgenic rice to investigate transformed lines. The 3 ea transgenic lines were detected insertion of HPT gene. Transgenic lines selected from PCR analysis were performed by Southern blot. From Southern blot, we obtained that two transgenic lines detected single band. We are going to study the method improving of cotransformation as well as transformation efficiency in rice.

GUS Expression by CaMV 35S and Rice Act1 Promoters in Transgenic Rice

  • To determine the patterns and the levels of expression of the cauliflower mosaic virus (CaMV 35S) promoter and the rice actin 1 (Act1) promoter in rice, transgenic rice plants containing CaMV 35S-$\beta$-glucuronidase (GUS) and Act1-GUS constructs were generated and examined by fluorometric and histochemical analyses. The fluorometric analysis of stably transformed calluses showed that the activity of the rice Act1 promoter was stronger than that of the CaMV 35S promoter in rice cells. In a histochemcial study of the transgenic rices, it was shown that the GUS activity directed by the CaMV 35S promoter was localized mainly in parenchymal cells of vascular tissues of leaves and roots and mesophyll cells of leaves. These results are similar to those of potato, a dicot plant. In contrast, rice plant transformed with Act1-GUS fusion construct revealed strong GUS activity in parenchymal cells of vascular tissue, mesophyll cells, epidermal cells, bulliform cells, guard subsidiary cells of leaves and most cells of the root, suggesting that the rice Act1 promoter is more constitutive than the CaMV 35S promoter. It was also confirmed that in both types of transgenic rice little or no staining was localized in metaxylen tracheary elements of vascular tissue from leaves or roots. These results indicate that the rice Act1 promoter can be utilized more successfully for expression of a variety of foreign gene in rice than the CaMV 35S promoter.

Inhibitory Effects of Resveratrol and Piceid against Pathogens of Rice Plant, and Disease Resistance Assay of Transgenic Rice Plant Transformed with Stilbene Synthase Gene

  • Resvestrol has been known to inhibit bacterial and fungal growth in vitro, and can be accumulated in plant to concentrations necessary to inhibit microbial pathogens. Hence, stilbene synthase gene has been used to transform to synthesize resveratrol in heterologous plant species to enhance resistance against pathogens. In the present study, we investigated the antimicrobial activities of resveratrol and piceid to bacterial and fungal pathogens, which causing severe damages to rice plants. In addition, disease resistance was compared between transgenic rice varieties, Iksan 515 and Iksan 526 transformed with stlibene synthase gene and non-transgenic rice varieties, Dongjin and Nampyeong. Minimum inhibitory concentration of resveratrol for Burkolderia glumae was 437.5 ${\mu}M$, and the mycelial growth of Biplaris oryzae was slightly inhibited at concentration of 10 ${\mu}M$. However, other bacterial and fungal pathogens are not inhibited by resveratrol and piceid. The expression of the stilbene synthase gene in Iksan 515 and Iksan 526 did not significantly enhanced resistance against bacterial grain rot, bacterial leaf blight, sheath blight, and leaf blight. This study is the first report on the effect of resveratrol and piceid against pathogens of rice plant, and changes of disease resistance of transgenic rice plants transformed with stilbene synthase gene.

제초제 내성 형질전환 벼와 잡초성 벼의 표현형질 비교 (Phenotype Comparison between Herbicide Tolerant Transgenic Rice and Weedy Rice)

  • 제초제 내성 형질전환 벼와 모본(동진벼) 및 잡초성 벼 두 계통(화성앵미 1과 광양앵미 12)의 표현형질을 비교하였다. 형질전환 벼와 모본의 질적형질은 모두 유사하였지만, 까락의 유무, 지엽 자세 및 종실 색은 형질전환 벼와 잡초성 벼 사이에 큰 차이가 있었다. 양적형질 중에서 초장, 분얼 수 및 지상부 건물중은 형질전환 벼나 모본에 비해 잡초성 벼에서 유의하게 우세했다. 형질전환 벼 및 모본의 주당 종실중과 천립중은 잡초성 벼에 비해 유의하게 무거웠다. 형질전환 벼는 모본 및 잡초성 벼에 비해 탈립이 용이하지 않았으며, 형질전환 벼의 배유 아밀로스 및 단백질 함량은 잡초성 벼와 유의한 차이가 있었다. 이와같은 형질전환 벼와 잡초성 벼 표현형질의 비교 연구는 형질전환 벼의 잡초화 가능성 예측에 활용될 수 있을 것이다.

  • Sucrose-phosphate synthase (SPS) is a key regulatory enzyme in sucrose synthesis. To investigate the role of SPS in carbon partitioning, we produced transgenic rice plants overexpressing a cyanobacterial SPS from Synechocystis sp. PCC 6803. The gene was expressed under the control of the maize Ubil promoter in transgenic plants. Southern and Northern blot analyses confirmed the integration and the expression of the transgene in four transgenic rice lines. All of the four transgenic! lines analyzed showed abnormal vegetative and reproductive developments. Analysis of SPS activities and primary metabolites in the transgenic rice plants will be presented.

Physiological Evaluation of Transgenic Rice Developed for Drought Tolerance

  • Evaluation of physiological performance of trehalose-producing transgenic rice line was conducted to investigate drought tolerance at early growth stage. Under artificially induced drought condition of 8% polyethylene glycol 6000, this transgenic rice line had leaf photosynthetic rate of 11.08 uml CO$_2$ $m^{-2}s^{-1}$, leaf transpiration rate of 8.38 mmol $H_2O$ $m^{-2}s^{-1}$ and leaf water potential of -1.12 MPa after 96 hours of treatment. Nakdongbyeo, the parent of this tyansgenic rice line, had photosynthetic rate of 15.42 $\mu$mol CO$_2$ $m^{-2}s^{-1}$, leaf transpiration rate of 8,04 mmol $H_2O$ $m^{-2}s^{-1}$ and leaf water potential of -0.88 MPa. The other variety used in this experiment for comparison, IR 72, showed higher values than both tyansgenic rice line and variety Nakdonbyeo on all three parameters; leaf photosynthetic rate of 20.61 $\mu$mol CO$_2$ $m^{-2}s^{-1}$, leaf transpiration rate of 12.88 mmol $H_2O$ $m^{-2}s^{-1}$, and leaf water potential of -0.82 MPa. So this transgenic rice line did not show superior performance in leaf transpiration rate, leaf photosynthetic rate and leaf water potential compared to variety Nakdongbyeo. This result along with visual observation on leaf rolling and drying during the experimental period indicated poor physiological performance of this transgenic rice line. Further studies on metabolic status of stress-induced trehalose, along with study on physiological response of this transgenic rice line during drought stress would shed more light on overall physiological performance of this transgenic rice line.

Overproduction of 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) confers resistance to the herbicide glyphosate in transgenic rice

  • Plants expressing Agrobacterium sp. strain CP4 5-enolpyruvylshikimate-3-phosphate synthase (CP4 EPSPS) are known to be resistant to glyphosate, a potent herbicide that inhibits the activity of the endogenous plant EPSPS. In order to develop herbicide-resistant rice, we prepared transgenic rice plants with CP4 EPSPS gene under the control of CaMV 35S promoter for over-expression. A recombinant plasmid was transformed into rice via Agrobacterium-mediated transformation. A large number of transgenic rice plants were obtained with glyphosate and most of the transformants showed fertile. The integration and expression of CP4 EPSPS gene from regenerated plants was analyzed by Southern and northern blot analysis. The transgenic rice plants had CP4 EPSPS enzyme activity levels more than 15-fold higher than the wild-type plants. EPSPS enzyme activity of transgenic rice plants was also identified by strip-test method. Field trial of transgenic rice plants further confirmed that they can be selectively survived at 100% by spay of glyphosate (Roundup$^{(R)}$) at a regular dose used for conventional rice weed control.
