쌍자엽 식물에 있어서 잎의 발달에 관한 과정은 매우 복잡하게 얽혀있지만, 이를 조절하는 메커니즘에 대해서는 아직 잘 알려져 있지 않다. 모델식물인 애기장대의 잎의 발달의 기초과정을 해명하기 위하여 잎의 형태이상 돌연변이체를 이용한 접근방법을 시도하였다. 본 연구에서 잎이 작고 가는 lanceolata1 돌연변이체를 선별하였고, 이원인유전자의 기능을 밝히기 위하여 유전학적 및 해부학적인 연구를 실시하였다. lan1-7 돌연변이체는 잎 뿐만아니라 줄기도 가늘어지는 표현형을 나타내었고, 이는 세포수준에서 세포크기와 세포수의 감소에 의한 조절되고 있음이 밝혀졌다. 이 결과는 LAN1 유전자가 잎의 발달과정에서 세포분열과 세포신장을 제어하고 있다는 사실을 시사하고 있다. lan1-7 돌연변이체와 35S-AG 형질전환체와의 F1 세대의 표현형 분석을 통하여, AG 유전자가 잎이 가늘고 위로 말리는 모양을 제어하고 있다는 사실이 판명되었다. 이 결과로 MADS-box유전자가 LAN1 유전자에 의해서 제어되고 잎의 발달과정에 관여하고 있다는 사실이 판명되었다.
환경 스트레스 신호 인지부터 스트레스 반응 유전자의 발현에 이르는 신호전달 네트워크에 있어서, 스트레스 반응 프로모터의 cis-조절 요소와 거기에 결합하는 다양한 전사인자는 환경 스트레스에 대한 식물의 적응을 조절한다. 애기장대 AP2/ERF 전사인자 패밀리 중 그룹 VII ERF는 RAP2.12, RAP2.2, RAP2.3, AtERF73/HRE1, AtERF71/HRE2 유전자를 포함하며, 저산소 스트레스 반응에서 중요한 역할을 하는 것으로 알려져 있다. 본 연구에서는 HRE1 프로모터의 저산소 반응 부위를 동정하였다. 이를 위해 1,000 bp, 800 bp, 600 bp, 400 bp, 200 bp, 100 bp, 그리고 50 bp HRE1 프로모터 부위를 포함하는 벡터를 제작하여 프로모 터 활성을 분석한 결과, -200에서 -100 프로모터 부위가 HRE1의 저산소 반응에서 중요함을 확인하였다. HRE1의 -200에서 -100 프로모터 부위에는 저산소 반응 cis-조절 요소로 알려진 ERF-결합 부위와 DOF-결합 부위가 존재하는데, 이는 HRE1의 발현이 ERF 전사인자와 DOF 전사인자에 의해 조절될 수 있음을 시사한다. 전체적으로, 본 연구를 통해 HRE1 의 저산소 스트레스 반응에는 -200에서 -100 프로모터 부위에 존재하는 cis-조절 요소가 중요한 역할을 한다는 것을 확인하였다.
애기장대에서 AtERF71/HRE2는 핵에서 전사인자로 작용하여 하위 유전자의 발현을 증가시키는 역할을 수행함으로써 저산소와 삼투 스트레스 반응에 관여할 것으로 여겨지는 유전자이다. 본 연구에서는 AtERF71/HRE2에 의해 직, 간접적으로 발현이 조절되는 하위 유전자를 알아보기 위해 AtERF71/HRE2 과발현체를 대상으로 마이크로어레이 실험을 수행하였다. 야생형에 비해 AtERF71/HRE2 과발현체에서 발현이 2배 이상 증가한 기능이 알려진 유전자는 AtERF71/HRE2 자신을 제외하고 161개였다. 161개 유전자 중 전사인자와 DNA-결합 단백질 등과 같은 전사조절자가 24개로 확인되어, AtERF71/HRE2는 하위 전사조절 유전자의 발현 조절을 통해 더 많은 유전자의 발현을 조절하는 상위 전사인자로서의 기능을 가질 것으로 추정되었다. 161개 유전자 중 15개 유전자를 대상으로 RT-PCR을 수행하여 마이크로어레이 결과의 신뢰성을 검증하였다. Genevestigator 데이터베이스 분석 결과, 161개 유전자 중 51개 유전자는 저산소 및 삼투 스트레스에 의해 발현이 증가하는 것으로 확인되었다. RT-PCR 분석 결과 AtERF71/HRE2 과발현체에서 발현이 증가한 15개 유전자 중 3개 유전자가 저산소에 의해 발현이 증가하였고, 다른 3개 유전자가 삼투 스트레스에 의해 발현이 증가하였으며, 이러한 결과는 이들 유전자가 AtERF71/HRE2에 의해 매개되는 저산소 또는 고염 스트레스 신호전달의 하위 유전자일 수 있음을 의미한다. 또한 본 연구의 마이크로어레이 분석 결과는 AtERF71/HRE2가 저산소 및 삼투 스트레스 반응뿐만 아니라 다른 환경 스트레스 반응과 식물 발달 조절에도 관여할 수 있음을 시사한다.
원예작물의 생육을 저하시키는 각종 병충해로 인한 과도한 농약과 화학비료의 사용은 환경오염뿐만 아니라 작물의 생산량에도 큰 영향을 미치고 있다. 식물생명공학기술을 이용하여 농약이나 화학비료 사용량을 획기적으로 줄일 수 있는 식물체의 개발, 즉 형질전환을 이용한 분자 육종기술은 병충해 내성 농작물을 개발하여 과도한 화학비료의 사용에 따르는 여러가지 문제점들을 극복할 수 있는 대안으로 대두되고 있다. 본 연구에서는 모델식물인 애기장대에서 분리한 식물생체방어 신호전달에 관련된 AtCBP63 유전자를 감자에 과발현시켰고, 이러한 형질 전환 감자에서 병저항성에 관여하는 유전자인 PR-2, PR-3, PR-5 유전자들의 발현이 증가되어 지속적으로 식물 방어 기작이 활성화되어 있음을 확인하였다. 또한, 감자에서 무름병 (soft rot disease)을 일으켜 막대한 피해를 유발하는 병원성 세균인 Erwinia carotovora subsp. Carotovora (Ecc)를 이용하여 AtCBP63 유전자를 과발현한 감자에 감염시켰을 때, 병 저항성이 증가한다는 사실을 검증하였다. 앞으로, 다양한 곰팡이 균에 대응하여 AtCBP63 유전자를 과발현한 감자에 저항성을 검증하고자 한다.
한발에 대한 식물체의 내성은 수분이 부족한 환경에서도 안정적으로 작물의 생산성을 유지하기 위해 필요한 유용한 특성 중 하나이다. 우리는 애기장대의 $H^+$-pyrophosphatase (AVP1)를 발현하도록 형질전환된 배추가 내건성과 관련되어 있는 몇몇 생리적 척도에 있어 향상됨을 검증하였다. 관수중단 처리로 조성된 토양수분 결핍 조건에서 AVP1 발현 식물체는 비형질전환체와 비교하여 비록 외형적 위조의 정도로는 그 차이를 구별할 수 없었지만 형질전환체가 재식된 토양의 수분포텐셜이 비형질전환체 재식 토양에 비해 더 빠르게 낮아졌다. 이는 형질전환체 잎의 상대적 수분함량이 비형질전환체에 비해 더 높은 것과 연관되어 있는 것으로 사료된다. 또한 건조스트레스 환경에서 비형질전환체에 비해 형질전환체는 광계II 양자수율이 높은 반면 전해질누출과 활성산소족의 하나인 $H_2O_2$와 3,3'-diaminobenzidine의 반응산물이 적었다.
이원적 전사유도 시스템(binary trans-activation system)은 도입유전자의 발현을 조절하는 기작(mechanism) 중에 하나로, 목적 유전자의 발현이 전사활성 인자를 가지고 있는 식물체와의 교배를 통해서만 발현되는 시스템이다. 본 연구에서는 이원적 전사유도 시스템을 원예 작물의 우수한 유전자원 및 신품종 보호 방법으로 이용하고자, 배추에서 이 시스템의 기능을 검정하였다. 배추작물에서 이원적 전사유도 시스템의 이용가능성을 검정하기 위하여 activator construct(35SLhGBart)와 reporter construct(pOpGUS1300)를 작성하였고 공동형질전환방법으로 배추에 형질전환하였다. 두 종류의 카세트가 도입된 형질전환체는 항생제를 이용하여 선발하였으며, 재분화된 신초의 GUS 유전자 발현으로 이 시스템의 활성을 확인하였다. 또한 이 시스템을 조직 특이적으로 유도하기 위하여 애기장대의 자성 배우체 특이적 프로모터를 이용하여 activator construct(795LhGBart)를 작성하여 애기장대에 형질전환 하였다. 공동형질전환된 애기장대는 자성 배우체에서 조직 특이적인 발현을 나타냈다. 이러한 결과는 이원적 전사유도 시스템이 목적유전자의 발현을 배추의 $F_1$ 종자에서 선택적으로 유도하는 방법으로써 우수한 유전자원 및 신품종 보호에 이용될 수 있다는 것을 보여주는 것이라고 생각된다.
The Arabidopsis ${\omega}$-3 fatty acid desaturase (AtFAD7) catalyzes the synthesis of trienoic fatty acids (TA). A transgenic tobacco line, T15, was produced by a sense AtFAD7 construct and showed a cosuppression-like phenotype, namely extremely low TA levels. The sequence similarity between AtFAD7 and a tobacco ortholog gene, NtFAD7, was moderate (about 69%) in the coding sequences. AtFAD7 siRNAs accumulated at a high level, and both AtFAD7 and NtFAD7 mRNAs are degraded in T15 plants. The low-TA phenotype in T15 was dependent on a tobacco RNA-dependent RNA polymerase6 (NtRDR6). We also produced tobacco RNAi plants targeting AtFAD7 gene sequences. The AtFAD7 siRNA level was trace, which was associated with a slight reduction in leaf TA level. Unexpectedly, this RNAi plant showed an increased NtFAD7 transcript level. To investigate the effect of translational inhibition on stability of the NtFAD7 mRNAs, leaves of the wild-type tobacco plants were treated with a translational inhibitor, cycloheximide. The level of NtFAD7 mRNAs significantly increased after cycloheximde treatment. These results suggest that the translational inhibition by low levels of AtFAD7 siRNAs or by cycloheximide increased stability of NtFAD7 mRNA. The degree of silencing by an RNAi construct targeting the AtFAD7 gene was increased by co-existence of the AtFAD7 transgene, where NtRDR6-dependent amplification of siRNAs occurred. These results indicate that NtRDR6 can emphasize silencing effects in both cosuppression and RNAi.

DNA methylation is an epigenetic mechanism for gene silencing. In Arabidopsis, MET1 is the primary DNA methyltransferase that maintains CG DNA methylation. Plants having an overall reduction of MET1 activity, caused by a met1 mutation or a constitutively expressed MET1 antisense gene, display genome hypomethylation, inappropriate gene and transposon transcription, and developmental abnormalities. However, the effect of a transient reduction in MET1 activity caused by inhibiting MET1 expression in a restricted set of cells is not known. For this reason, we generated transgenic plants with a MET1 antisense gene fused to the DEMETER (DME) promoter (DME:MET1 a/s). Here we show that DME is expressed in leaf primordia, lateral root primoridia, in the region distal to the primary root apical meristem, which are regions that include proliferating cells. Endogenous MET1 expression was normal in organs where the DME:MET1 a/s was not expressed. Although DME promoter is active only in a small set of cells, these plants displayed global developmental abnormalities. Moreover, centromeric repeats were hypomethylated. The developmental defects were accumulated by the generations. Thus, not maintaining CG methylation in a small population of proliferating cells flanking the meristems causes global developmental and epigenetic abnormalities that cannot be rescued by restoring MET1 activity. These results suggest that during plant development there is little or no short-term molecular memory for reestablishing certain patterns of CG methylation that are maintained by MET1. Thus, continuous MET1 activity in dividing cells is essential for proper patterns of CG DNA methylation and development.
Background: Cytochrome P450 enzymes catalyze a wide range of reactions in plant metabolism. Besides their physiological functions on primary and secondary metabolites, P450s are also involved in herbicide detoxification via hydroxylation or dealkylation. Ginseng as a perennial plant offers more sustainable solutions to herbicide resistance. Methods: Tissue-specific gene expression and differentially modulated transcripts were monitored by quantitative real-time polymerase chain reaction. As a tool to evaluate the function of PgCYP736A12, the 35S promoter was used to overexpress the gene in Arabidopsis. Protein localization was visualized using confocal microscopy by tagging the fluorescent protein. Tolerance to herbicides was analyzed by growing seeds and seedlings on Murashige and Skoog medium containing chlorotoluron. Results: The expression of PgCYP736A12 was three-fold more in leaves compared with other tissues from two-year-old ginseng plants. Transcript levels were similarly upregulated by treatment with abscisic acid, hydrogen peroxide, and NaCl, the highest being with salicylic acid. Jasmonic acid treatment did not alter the mRNA levels of PgCYP736A12. Transgenic lines displayed slightly reduced plant height and were able to tolerate the herbicide chlorotoluron. Reduced stem elongation might be correlated with increased expression of genes involved in bioconversion of gibberellin to inactive forms. PgCYP736A12 protein localized to the cytoplasm and nucleus. Conclusion: PgCYP736A12 does not respond to the well-known secondary metabolite elicitor jasmonic acid, which suggests that it may not function in ginsenoside biosynthesis. Heterologous overexpression of PgCYP736A12 reveals that this gene is actually involved in herbicide metabolism.
This study describes an efficient protocol for Agrobacterium tumefaciens-mediated transformation of two subgroups of genotype AAA bananas (Musa acuminata cv. Pei Chiao and Musa acuminata cv. Gros Michel). Instead of using suspension cells, cauliflower-like bud clumps, also known as multiple bud clumps (MBC), were induced from sucker buds on MS medium containing $N^6$-Benzylaminopurine (BA), Thidiazuron (TDZ), and Paclobutrazol (PP333). Bud slices were co-cultivated with A. tumefaciens C58C1 or EHA105 that carry a plasmid containing Arabidopsis root-type ferredoxin gene (Atfd3) and a plant ferredoxin-like protein (pflp) gene, respectively. These two strains showed differences in transformation efficiency. The EHA105 strain was more sensitive in Pei Chiao, 51.3% bud slices were pflp-transformed, and 12.6% slices were Atfd3-transformed. Gros Michel was susceptible to C58C1 and the transformation efficiency is 4.4% for pflp and 13.1% for Atfd3. Additionally, gene integration of the putative pflp was confirmed by Southern blot. Resulting from the pathogen inoculation assay, we found that the pflp transgenic banana exhibited resistance to Fusarium oxysporum f. sp. cubense tropical race 4. This protocol is highly advantageous to banana cultivars that have difficulties in setting up suspension cultures for the purpose of quality improvement through genetic transformation. In addition, this protocol would save at least 6 months in obtaining explants for transformation and reduce labor for weekly subculture in embryogenic cell suspension culture systems.
