Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI.

  • Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [$Ca^{2+}$]i elevation from anti$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface $Fc{\varepsilon}RI$ expression and the binding between the cell surface $Fc{\varepsilon}RI$ and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the $Fc{\varepsilon}RI$ ${\alpha}$ chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface $Fc{\varepsilon}RI$. This mechanism may occur through $Fc{\varepsilon}RI$ expression inhibition.

Characterization of Protein Kinases Activated during Treatment of Cells with Okadaic Acid

  • Six renaturable protein kinases that utilize the myelin basic protein (MBP) as a substrate were activated during prolonged exposure of cardiac myocytes to okadaic acid (OA). We characterized the substrate preference and activation of these kinases, with particular emphasis on 3 novel kinases-MBPK-55, MBPK-62 and MBPK-87. The transcription factors c-Jun, Elk, ATF2, and c-Fos that are used to assess mitogen-activated protein kinase activation were all poor substrates for these three kinases. MAPKAPK2 was also not phosphorylated. In contrast, Histone IIIS was phosphorylated by MBPK-55 and MBPK-62. These protein kinases were activated in cultured cardiac fibroblasts, H9c2 cardiac myoblasts, and Cos cells. High concentrations (0.5 to $1\;{\mu}M$) of OA were essential for the activation of the protein kinases in all of the cell types examined, whereas calyculin A [an inhibitor of protein phosphatase 1 (PP1) and PP2A], cyclosporin A (a PP2B inhibitor), and an inactive OA analog all failed to activate these kinases. The high dose of okadaic acid that is required for kinase activation was also required for phosphatase inhibition, as assessed by immunoblotting whole cell lysates with anti-phosphothreonine antibodies. A variety of chemical inhibitors, including PD98059 (MEK-specific), genistein (tyrosine kinase-specific) and Bisindolylmaleimide I (protein kinase C-specific), failed to inhibit the OA activation of these kinases. Thus, MBPK-55 and MBPK-62 are also Histone IIIS kinases that are widely expressed and specifically activated upon exposure to high OA concentrations.

  • Younggaechulgam-tang has been used for treating skin diseases. In this study, I investigated the immunosuppressive effect of Younggaechul-tang in the human T cell line MOLT-4 cells. MOLT-4 cells were stimulated with the phytohemagglutinin (PHA) and phorbol 12-myristate 13-acetate (PMA) + A23187. The secretion appeared to be greater when cells were stimulated with PHA than with PMA + A23187. Younggaechulgam-tang had no affect proliferation stimulated by PHA. I showed that IL-2 secretion and expression by PHA stimulated MOLT-4 cells were inhibited by Younggaechugam-tang treatment. Maximal inhibition rate of IL-2, TNF-${\alpha}$ secretion was 80$\%$ and 30$\%$, respectively. Younggaechulgam-tang also inhibited nuclear translocation of p65 subunit of nuclear factor-kB and nuclear factor of activated T cells (NFAT). In conclusion, these results suggest that Younggaechulgam-tang may contribute to the immunosuppressive oriental drug clinically.

U-937 세포에 있어서 세라마이드에 의한 c-jun 유전자 발현의 조절 (Ceramide-Mediated c-jun Gene Expression in U-937 Cells)

  • Ceramide has been suggested as an important mediator of the effects of extracellular agonists on cell growth inhibition, differentiation, apoptosis. However the biochemical sign aling mechanism involved in transducing the effects of ceramide on leukemia cell differentiation is still unclear. In these respects, we examined the regulatory effects of ceramide on c-jun gene expression during differentiation. In U-937 cells. ceramide increased c-jun mRNA levels in a time-dependent manner. The half life, of c-jun mRNA was 30 min. In contrast, inhibition of protein synthesis with cycloheximide in the absence, of transcription with actinomycin D increased the half-life of c-jun mRNA in ceramide-treated U-937 cells to more than 90 min. In order to examine whether ceramide-inhibited c-jun gene expression is regulated through ceramide-activated protein phosphatase (CAPP), a direct target for the action of ceramide, okadaic acid were treated to the cells. Okadaic acid inhibited enhancement of c-jun mRNA induced by C2-ceramide in a dose-dependent manner. These results suggested that ceramide increases c-jun mRNA level during differentiation in U-937 cells and regulates the gene expression on posttranscriptional level. In addition, we provide the evidence that CAPP is involved in ceramide-induced c-jun gene expression in U-937 cells.

  • Using a flue- cured tobacco variety, KF 109, effect of growth regulators(fatty alcohol and C- MH) on the change of protein, DNA, and RNA were investigated. Generally, inhibition of DNA synthesis was observed soon after become notably reduced when checked on 14 days after the treatment. Fatty alcohol treatment appeared to alleviate the inhibition of DNA synthesis caused by the C - MH treatment. It was also observed that in the tips DNA content increased slightly at the early stage after the C - MH treatment but evident reduction of it was resulted from 7th day after the treatment. RNA content in cutters and tips was increased initially but variable transcription inhibitory activities - not so obvious as was observed in DNA synthesis - according to leaf positions were shown thereafter. Ripening of leaves probably due to senescence was advanced by the treatment of the growth regulators. DNA content in root was relatively higher in plants treated with the growth regulators while it was clearly decreased in stalk, However, RNA contents in tissue of stalk and root was not different with that of foliage. Increase of protein content in foliage as well as in stalk was evident 14 days after dual treatment of fatty alcohol and C - MH.

한방 처방에서 이뇨 효과에 대한 실험 논문 분석 및 고찰 (Analysis of experimental articles of Korean medicinal prescriptions on the diuretic effect)

  • Objective : Diuretics are effective on the patients with hypertension and heart failure, as well as edema by regulating the function of kidney, which is key organ to maintain the balance of the different electrolytes in the body. The present review article is designed to review the diuretic effect of Korean medicinal prescription base on the experimental studies.Method : For this purpose, every article related to ‘diuresis’ and 'Korean medicinal prescription' were analysed from articles which published at domestic and international journals.Results : 1. Representative Korean medicine prescriptions showed diuretic effect along with electrolyte excretion were Oryeong-san, Paeryung-tang, Hwangryungbokryung-tang and Daeganghwal-tang. 2. Some Korean medicinal prescription including Oryeong-san, Jueryeong-tang, Sipcho-tang distribute diuresis through the inhibition of renin-angiotensin-system. 3. Oryeong-san, Bojungchiseup-tang had a diuretic effect on the down-regulation of aquaporin water channel in the renal collecting duct.Conclusion : Korean medicinal transcriptions have a diuretic effect through several types of mechanism such as along with electrolytes excretion, inhibition of renin-angiotensin system, and down-regulation of water channels.

ERK Activation by Fucoidan Leads to Inhibition of Melanogenesis in Mel-Ab Cells

  • Fucoidan, a fucose-rich sulfated polysaccharide derived from brown seaweed in the class Phaeophyceae, has been widely studied for its possible health benefits. However, the potential of fucoidan as a possible treatment for hyperpigmentation is not fully understood. This study investigated the effects of fucoidan on melanogenesis and related signaling pathways using Mel-Ab cells. Fucoidan significantly decreased melanin content. While fucoidan treatment decreased tyrosinase activity, it did not do so directly. Western blot analysis indicated that fucoidan downregulated microphthalmia-associated transcription factor and reduced tyrosinase protein expression. Further investigation showed that fucoidan activated the extracellular signal-regulated kinase (ERK) pathway, suggesting a possible mechanism for the inhibition of melanin synthesis. Treatment with PD98059, a specific ERK inhibitor, resulted in the recovery of melanin production. Taken together, these findings suggest that fucoidan inhibits melanogenesis via ERK phosphorylation.

Improved Inhibition of Human Immunodeficiency Virus Type 1 Replication by Intracellular Co-overexpression of TAR and RRE Decoys in Tandem Array

  • Intracellular expression of RNA decoys, such as TAR or RRE decoy, has been previously shown to protect immune cells from human immunodeficiency virus type 1 (HIV-1) replication by inhibiting the binding of the HIV-1 regulatory protein to the authentic HIV RNA sequence. However, HIV-1 challenge experiments of primary human T cells, which express the RNA decoy, demonstrated that the cells were only transiently protected, and hence, more improved protocols for HIV-1 inhibition with the RNA decoys need to be developed. In this report, in order to develop a more effective RNA decoy, we analyzed and compared the ability of a series of RNA decoy derivatives in inhibiting HIV-1 replication in CEM cells. Using an improved tRNA cassette to express high levels of RNA decoy transcripts in cells, we found that co-expression of both TAR and RRE decoys, in the form of an aligned sequence in a single transcription cassette, much more potently blocked cells from HIV-1 than the expression of only one kind of RNA decoy. This observation will have an important implication for experiments involving optimization of clinical applications in RNA decoy-based gene therapy against HIV-1.

Transglutaminase 2 Promotes Autophagy by LC3 Induction through p53 Depletion in Cancer Cell

  • Transglutaminase 2 (TGase 2) plays a key role in p53 regulation, depleting p53 tumor suppressor through autophagy in renal cell carcinoma. We found that microtubule-associated protein 1A/1B-light chain 3 (LC3), a hallmark of autophagy, were tightly associated with the level of TGase 2 in cancer cells. TGase 2 overexpression increased LC3 levels, and TGase 2 knockdown decreased LC3 levels in cancer cells. Transcript abundance of LC3 was inversely correlated with level of wild type p53. TGase 2 knockdown using siRNA, or TGase 2 inhibition using GK921 significantly reduced autophagy through reduction of LC3 transcription, which was followed by restoration of p53 levels in cancer cells. TGase 2 overexpression promoted the autophagy process by LC3 induction, which was correlated with p53 depletion in cancer cells. Rapamycin-resistant cancer cells also showed higher expression of LC3 compared to the rapamycin-sensitive cancer cells, which was tightly correlated with TGase 2 levels. TGase 2 knockdown or TGase 2 inhibition sensitized rapamycin-resistant cancer cells to drug treatment. In summary, TGase 2 induces drug resistance by potentiating autophagy through LC3 induction via p53 regulation in cancer.

인체 폐암세포에서 인삼사폐탕에 의한 Cdk inhibitor p27의 발현 증가 및 pRB의 인산화 억제 (Induction of Cdk inhibitor p27 and Inhibition of pRB Phosphorylation by Insamsapye-tang Treatment in Human Lung Cancer A549 Cells)

  • We investigated the effects of Insamsapye-tang (ISSPT) water extract on the cell proliferation of human lung carcinoma A549 cells. ISSPT treatment resulted in the inhibition of cell proliferation in a concentration-dependent manner. This anti-proliferative effect of A549 cells by ISSSPT treatment was associated with morphological changes such as membrane shrinking and cell rounding up. DNA flow cytometric histograms showed that population of G1 phase of the cell cycle was increased by ISSPT treatment in a concentration-dependent manner. ISSPT treatment induced the levels of tumor suppressor p53 protein and cyclin-dependent kinase (Cdk) inhibitor p27 without significant alteration of cyclins and Cdks expression. In addition, ISSPT treatment resulted in down-regulation of phosphorylated retinoblastoma protein (pRB). However, the levels of p130, the pRB family protein, and transcription factors. E2F-1 and E2F-4. were remained unchanged. The present results indicated that ISSPT-induced inhibition of lung cancer cell proliferation is associated with the blockage of G1/S progression and the induction of apoptosis, and we suggest that ISSPT will be an effective therapeutic agent on human lung cancer.
