Effect of Dietary Protein and Magnesium Levels on Serum Lipid Contents and Enzyme Activities in Rats (단백질과 마그네슘 공급수준이 흰쥐의 혈청중 지질함량 및 효소활성에 미치는 영향)

  • To study the effects of dietary protein and magnesium level on serum lipid contents and enzyme activities, male Sprague-Dawley rats were divided into six groups. Six groups of animals were fed six experimental diets different with protein(8% and 20%) and magnesium levels (Mg-free, 400mg/kg diet and 800mg/kg diet). The serum lipid contents and enzyme activities of rats fed different dietary protein and magnesium levels were examined. Serum triglyceride contents was significantly decreased as the dietary magnesium level was increased. Serum phospholipid contents was significantly higher in magnesium deficient group than those in the other groups. Serum alkaline phosphatase activity was significantly decreased in magnesium dificient group compared with other groups, but serum glutamic oxaloacetic transaminase and glutamic pyruvic transaminase activities was significantly increased in magnesium dificient group than other groups. In conclusion, serum triglyceride contents in low protein level was remarkably decreased as the dietary magnesium level was increased.

  • Solanum lyratum (Solanaceae) has been used as a traditional analgesic, antipyretic and hepatoprotective agents in Korea. In this study, we investigated the hepatoprotective effect of ethylacetate extract of Solanum lyratum (SL) on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of SL (150, 300 mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase levels. Heat-processed ginseng saponin ameliorates the adenine-induced renal failure in rats

  • To evaluate the effect of the saponin of heat-processed ginseng (Sun ginseng, SG), we investigated the protective effect of SG total saponin fraction against adenine-induced chronic renal failure in rats. SG saponin significantly decreased the levels of urea nitrogen and creatinine in the serum, but increased the urinary excretion of urea nitrogen and creatinine, indicating an improvement of renal function. SG saponin also inhibited adenine-induced kidney hypertrophy and edema. SG saponin reduced serum glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, and lactate dehydrogenase activities increased by adenine. Based on these findings, the ameliorating effect of SG on chronic renal failure may result from its saponin.

$CO_2$ Evolution in Peroxisomes Isolated from Spinach Leaves at Various Ages (시금치 잎의 엽영에 따라 분리한 Peroxisome에서의$CO_2$방출)

  • The rates of photorespiration and total CO2 fixation depending on leaf ages of spinach (Spinacia oleracea L.) were investigated. Metabolic rates of glycolate and glyoxylate in isolated peroxisomes were also measured. The rate of photorespiration and total CO2 fixation ability increased with the maturing of leaf, but decreased with senescence. Activities of enzymes involved in the peroxisomal photorespiratory pathway such as catalase, glycolate oxidase, NADH-glyoxylate reductase and glutamate-glyoxylate transaminase were highest in the mature leaf, but also decreased with aging of leaf. Glutamate-glyxolate transaminase activity significantly decreased with senescence, especially. the metabolic rate of glycolate was observed to be lower than that of glyoxylate in isolated peroxisomes. Glycolate seemed to be metabolized mainly to glycine, however, it also oxidized to CO2 when glycolate was supplied as a substrate for glycine synthesis instead of glyoxylate. The conversion rates of glycolate and glyxylate into CO2 increased with the senescence of leaves.

Studies on the CO2 Fixation Patterns Following the Chloroplast Development in Maize Leaves (옥수수엽의 엽록체발달에 따른 CO2 고정양상에 관한 연구)

  • These studies were undertaken to determine the $CO^2$fixation patterns following the chloroplast development in maize leaves. At the early stage of chloroplast development $^{14}C$ was incorporated into aspartate (41%) and malate (22%) respectively. Whereas the incorporation of $^{14}C$ into malate was higher than that of aspartate as chloroplast developed. Activity of NADPH-dependent malate dehydrogenase was increased throughout chloroplast development, but that of aspartate transaminase was not. Much incorporation of $^{14}C$ into aspartate at the early stage of chloroplast development and into malate at later stage of chloroplast development lead us to conclude that NADPH-dependent malate dehydrogenase activity is closely associated with chloroplast development, but activity of aspartate transaminase is not.

Hepatoprotective activity of methanol extracts of Glinus oppositifolius and Trianthema decandra against paracetamol induced liver damage

  • The plants, Trianthema decandra and Glinus oppositifolius are commonly used by tribal people in India for the treatment of liver diseases. Hepatoprotective activity of methanol extracts of Glinus oppositifolius and Trianthema decandra at the dose of 250 and 500 mg/kg body weight administered orally was evaluated against paracetamol induced liver damage in rats. Biochemical parameters such as serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, serum alkaline phosphatase, bilirubin, total serum protein, lipid peroxide and glutathione content of the liver were estimated to determine liver function and metabolism. From the biochemical observations, it was concluded that methanol extracts of Glinus oppositifolius and Trianthema decandra significantly restored the altered biochemical parameters towards normal condition in paracetamol induced liver damage.

  • This study was aimed to investigate the effects of Ginseng ethanol extract on Diabetes induced by alloxan in comparison with that of insulin. The blood sugar, blood urea nitrogen, glutamic oxaloacetic transaminase and cholesterol were measured by spectrophotometry. The results were obtained as follows. 1. Ethanol extract of Ginseng increased the blood sugar level in normal rats. 2. Ethanol extract of Ginseng restrained the increase of blood sugar in diabetes induced by alloxan. 3. Ethanol extract of Ginseng acted to oppose a rapid decrease of blood sugar by insulin. 4. Blood urea nitrogen, glutamic oxaloacetic transaminase and cholesterol level in blood were not affected by Ginseng.

A Case Study of Poisoning Symptoms Caused by Misusing Xanthii Fructus, Treated with Korean Medicine (도꼬마리 열매 복용으로 발생한 중독증상(中毒症狀)의 한방치료 1례)

  • This is a clinical report of one patient with mental disorientation, abnormal behavior, spasm, and other symptoms caused by poisoning with Xanthii Fructus, who recovered completely with treatment. The patient was treated with Korean medical treatments, such as herbal medicine and acupuncture. After treatment, various symptoms caused by the Xanthii Fructus poisoning disappeared completely and laboratory findings, such as liver function tests (aspartate transaminase and alanine transaminase), were also close to the normal ranges. This case shows that Korean medicine therapy can be applicable in treating various symptoms caused by Xanthii Fructus poisoning. It is considered that further research is needed to confirm the safe use of Xanthii Fructus.

  • Water extracts of 21 traditional Korean herbal drugs were prepared, and a dose range of 100 mg/kg to 400 mg/kg was administered orally into mice once a day for five days. Changes of serum enzyme activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), blood urea nitrogen (BUN), alkaline phosphatase, body weight changes and histo pathological examination of various organs were investigated. Water extract of Ephedra Herba caused severe body weight loss at a dose of 100 mg/kg and death from a dose level of 200 mg/kg by oral administration. Angelica koreanae Radix and Anthrisci Radix showed a slight body weight loss and damages to liver and kidney.

  • cis-Dichlorodiammineplatinum(cisplatin) is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. So, to evaluate the effects of 2(3)-tert-butyl-4-hydroxyanisole(BHA) on cisplatin nephrotoxicity in rats, both compounds were given intraperitoneally. Remarkable protective effects of BHA against nephrotoxicity of cisplatin were observed when BHA was administered to rats 1hr after cisplatin injection. On the other hand pretreatment with BHA 1hr prior to cisplatin did not reduce weight loss, blood urea nitrogen and creatinine levels. Hepatotoxicity induced by combination treatment of cisplatin and BHA was evaluated by measuring serum glutamic pyruvate transaminase and serum glutamic oxalate transaminase. Combination treatment did not affect the levels of SGPT and sGOT except 1hr pretreatment. The present results indicate that BHA may provide protection against cisplatin nephrotoxicity, when it is given 1hr after cisplatin.
