본 연구는 최근 이용압력이 높은 서울시 초안산근린공원을 대상으로 숲길의 분포와 훼손 실태를 파악하여 향후 도심 내 숲길 정비 방향을 설정하기 위한 목적으로 수행하였다. 초안산근린공원 숲길(25,964m)의 훼손 현황 중 노폭 2m 이상 20.3%, 침식 깊이 30cm 이상 36.3%, 훼손이 심한 형태인 나지침식이나 근계노출, 암석노출 구간 47.0%이었고, 환경피해도 등급 IV 이상이 70.6%로 숲길 훼손이 심각한 상태이었다. 훼손이 심한 숲길 노선은 대부분 주능선을 따라 형성된 주요 숲길과 저지대 급경사 숲길, 주요 숲길과 연결된 샛길이었다. 연구결과에 기초하여 초안산근린공원 숲길의 복원유형을 숲길 확산방지, 숲길 지반안정, 숲길 노면정비, 숲길 폐쇄 후 식생복원, 유지 등 5가지로 구분하였고, 유형별 복원방안을 제안하였다. 숲길 확산방지는 나지가 노출되어 있는 숲길 폭원 확대를 방지하고, 숲길 지반안정은 숲길 내 침식 및 뿌리노출을 방지하며, 숲길 노면정비는 훼손이 심각한 숲길의 노면을 목재데크, 목재계단, 노면 포장 실시 등을 통하여 보호하기 위한 계획을 수립하였다.
혈근초(Sanguinaria canadensis) 뿌리에서 유래된 benzophenanthridine alkaloid의 일종인 sanguinarine은 항균, 항산화 및 항암작용 등 다양한 생리활성을 지니고 있는 것으로 알려져 있다. 비록 TRAIL이 암세포에서는 apoptosis를 유도하지만 정상세포에서는 세포독성을 나타내지 않는다는 큰 장점으로 제 2 임상 단계에서 유의적인 성과를 이루었지만, TRAIL 저항성을 극복해야 하는 큰 어려움이 남아있다. 본 연구실에서는 TRAIL이 세포독성을 나타내지 않는 범위의 sanguinarine과 혼합처리에 의하여 TRAIL 저항성 AGS 위암세포에서 apoptosis를 유발하였음을 보고한 바 있으며, 본 연구에서는 sanguinarine의 TRAIL 저항성 관련 극복에 대한 추가적인 기전 연구를 실시하였다. 본 연구의 결과에 의하면, sanguinarine과 TRAIL의 혼합처리는 각각의 단독 처리에 비하여 AGS 세포의 증식억제 및 apoptosis 유도의 상승 효과가 있었으며, 이를 MTT assay, agarode gel 전기영동, 염색질 응축 현상 및 flow cytometry 분석을 통하여 확인하였다. 또한 sanguinarine과 TRAIL의 혼합처리는 DR5의 발현을 증가시켰으며, ROS의 생성을 촉진시켰다. 그러나 동일 조건에서 MAPKs 신호 전달계에는 큰 영향을 주지 않았다. 아울러 ROS 생성을 인위적으로 차단하였을 경우, sanguinarine과 TRAIL의 혼합처리에 의한 생존도 저하가 유의적으로 회복되었다. 이러한 결과는 sanguinarine과 TRAIL이 DR5의 발현 증가와 ROS의 생성을 촉진시킴으로서 apoptosis 신호를 활성화하였음을 의미하는 결과로서 TRAIL 저항성 극복을 위한 sanguinarine 활용의 유용성을 보여주는 것이다.

Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is a member of the TNF family and potent inducer of apoptosis. TRAIL has been shown to effectively limit tumor growth in vivo without detectable cytotoxic side effects. Interferon (IFN)-${\gamma}$ often modulates the anti-cancer activities of TNF family members including TRAIL. We previously reported that IFN-${\gamma}$ enhanced TRAIL-induced Apoptosis in HeLa cells without the unknown mechanism. In this study, we investigated whether IRF-1 involves in IFN-${\gamma}$-enhanced TRAIL-induced apoptosis. We exposed HeLa cells to IFN-${\gamma}$ for 12 hours and then treated with recombinant TRAIL protein. No apoptosis was induced in cells pretreated with IFN-${\gamma}$, and TRAIL only induced 30% apoptosis after 3 hours treatment. In HeLa cells pretreated with IFN-${\gamma}$, TRAIL induced cell death to more than 75% at 3 hours, showed that IFN-${\gamma}$-pretreatment enhanced HeLa cell death to TRAIL-induced apoptosis. To investigate the functional role of IRF-1 in IFN-${\gamma}$-enhanced TRAIL-induced apoptosis, IRF-1 was overexpressed by using an adenoviral vector AdIRF-1. IRF-1 overexpression increased apoptotic cell death and significantly enhanced apoptotic cell death induced by TRAIL when infected cells were treated with TRAIL. Our findings show that IFN-${\gamma}$ enhances TRAIL-induced apoptosis by IRF-1 in HeLa cells.
본 연구에서는 플라보노이드 계열 중의 하나인 luteolin을 이용하여 TRAIL에 저항성을 가지는 T24 방광암세포에서 TRAIL 저항성 극복 가능성을 조사하였다. 본 연구의 결과에 의하면 luteolin 및 TRAIL 각각 단독 처리 시 세포증식에 전혀 영향을 미치지 못한 농도의 복합 처리 시 세포증식억제 효과가 크게 증가하였음 알 수 있었다. 이러한 증식억제와 연관된 aspoptosis 유도는 caspase-8의 활성화에 의한 tBid의 발현 증가와 pro-apoptotic 인자인 Bax의 발현 증가로 인한 caspase-9 및 -3의 활성화로 이어지는 type II apoptosis에 의한 것이라 추측되며, 이러한 가정은 각각의 caspase 선택적 저해제를 이용하여 재확인 하였다. 본 연구결과는 TRAIL에 저항성을 보이는 암세포에 luteolin이 감수성을 높이는데 효과적일 수 있으며, 암세포에 대한 combination therapy를 위한 기초자료로 활용성이 높을 것으로 사료된다.
TRAIL은 다양한 암세포에서 apoptosis를 유발하는 것으로 알려져 있으나 간암세포를 포함한 일부 암세포에서 TRAIL 저항성이 획득된 것으로 보고되어지고 있다. 대두의 대표적인 생리활성 물질인 isoflavonoid계열 genistein은 이미 많은 암세포에서 apoptotic 효능을 가진 것으로 알려져 있으나 TRAIL에 의한 apoptosis 유도에 미치는 영향과 기전에 대한 연구는 여전히 미비한 실정이다. 본 연구에서는 TRAIL 저항성을 가진 Hep3B 간암세포에서 TRAIL에 의한 apoptosis 유도를 genistein이 더욱 상승시킬 수 있음을 보고하고자 한다. 본 연구의 결과에 의하면, Hep3B 세포에 세포독성을 보이지 않는 범위의 genistein에 의한 TRAIL 유도 apoptosis 상승효과는 미토콘드리아의 기능 손상과 연관성이 있었다. 또한 genistein과 TRAIL 복합처리에 의한 apoptosis 유도는 p38 MAPK 활성 저하로 더욱 상승하였으며, 이는 Bid의 truncation 증가, pro-apoptotic 단백질인 Bax의 발현 증가와 anti-apoptotic Bcl-2의 발현 감소 및 미토콘드리아에서 세포질로의 cytochrome c 유출의 증가와 연관성이 있었다. 또한 p38 MAPK 억제제는 genistein 및 TRAIL 복합처리에 의한 caspase의 활성 증가와 PARP 단백질의 단편화를 촉진시켰으며, 이는 미토콘드리아의 기능적 손상 증가에 의한 것임을 알 수 있었다. 따라서 본 연구의 결과는 genistein이 TRAIL에 의한 apoptosis 유도를 효과적으로 증가시킬 수 있으며, 이러한 과정이 p38 MAPK 의존적으로 이루어짐을 알 수 있었다.
이 연구에서는 개정 교육과정의 수학 I 에서 다루어지는 그래프의 지도 목표가 7차 교육과정의 이산수학에서 그래프의 지도 목표와 같지 않음을 확인하였다. 이를 바탕으로 수학 I 에서 그래프와 행렬 단원의 내용 요소인 경로의 교수학적 변환 방식과 학생들의 이해 정도를 분석하였으며, 그 결과 개정 교육과정의 수학 I 에서 경로는 trail로 정의되지만 교과서에 따라서는 이를 path로 해석한 경우가 있으며 학생들 역시 경로를 trail보다는 path로 이해하려는 경향이 있음을 파악할 수 있었다. trail로서 경로의 정의는 오일러 회로와 해밀턴 회로를 다루는 체계에서 보다 의미를 가지며 개정 교육과정의 수학I에서는 trail로서 경로의 가치를 찾는 것이 쉽지 않다.
설악산국립공원 내설악지구의 6개 주요 등산로에 대해 이용강도에 따른 등산로 및 주변 환경훼손상태를 조사하였다. 총 29.2km, 132개 지점에서 등산로 상태를 조사한 바, 등산로폭, 나지노출폭, 등산로물매 등에서 이용강도에 따른 차이가 인정되었다. 훼손유형의 출현빈도는 암석노출, 뿌리노출, 종식, 분기 등의 순으로 이들 훼손지점의 등산로상태는 건전한 지점과 뚜렷한 차이를 보였으며, 또한 각 등산로에서의 건전지점들은 훼손지점들에 비해 대체로 표고가 낮은 곳에 나타났다. 전반적으로 등산로 훼손에 미치는 이용영향은 외설악 지구 등산로에 비해 훨씬 적었다. 주연부 상층식생의 우점종은 등산로별로 달랐으며 하층식생은 계곡부에서는 조록싸리, 생각나무, 사면부에서는 철쭉꽃이 우점종이었다. 종다양도 및 주연부 하층식생의 피도는 이용강도가 낮은 오세암등산로에서 가장 높았으며, 등산로간 하층식생의 종구성은 대체로 이질적이었다.
연구배경: 정상세포는 보호되고 종양세포에 독성을 보인다고 알려진 TNF유전자족으로 새로이 확인된 TRAIL이 폐암세포에서 보이는 아포프토시스 효과를 확인하고, 아포프토시스로부터 세포를 보호하는 전사인자 $NF-{\kappa}B$가 TRAIL에 의하여 활성화 되는 정도를 평가하여 MG132의 $NF-{\kappa}B$활성억제가 TRAIL 유도성 아포프토시스를 감작시키는지를 확인하기 위하여 본 연구를 시행하였다. 방법: A549(wt p53) 및 NCI-H1299(null p53) 폐암세포주를 사용하였다. 세포독성 검사는 MTT assay를 이용하였고 아포프토시스는 Annexin V assay와 FACS 분석을 이용하였다. $NF-{\kappa}B$ 전사활성은 luciferase reporter gene assay를 이용하였고 $I{\kappa}B{\alpha}$ 분해는 western blot을 이용하였으며, TRAIL에 의해 활성화된 $NF-{\kappa}B$와 DNA 결합은 electromobility shift assay와 anti-p65 antibody를 이용한 supershift assay로 확인하였다. 결과: 1) TRAIL 100 ng/ml 농도에서 wild-type p53인 A549 폐암세포는 34.4%, p53 null인 NCI-H1299 폐암세포는 26.4%의 세포사를 관찰하였다. 2) Luciferase reporter gene assay로서 TRAIL에 의한 $NF-{\kappa}B$의 활성이 A549 $IgG{\kappa}B-luc$세포에서 2.45배 증가하고 NCI-H1299 $IgG{\kappa}B-luc$세포에서는 1.47배 증가함을 관찰하여 TRAIL에 의하여 $NF-{\kappa}B$가 활성화됨을 확인하였다. 3) MG132의 전처치로 TRAIL에 의한 $NF-{\kappa}B$의 활성이 A549 세포와 NCI-H1299 세포에서 각각 기저수준의 0.24, 0.21배로 강력히 억제되었다. 4) TRAIL단독으로 30% 전후의 세포독성이 MG132 전처치 후 TRAIL을 투여하면 두 세포주 모두에서 80% 이상의 세포독성이 관찰되어 MG132가 TRAIL유도성 아포프토시스에 감작효과가 있음을 확인하였다. 결론: 이상의 결과로 TRAIL에 상대적인 내성을 보이는 폐암세포주에서 MG132가 $NF-{\kappa}B$ 활성억제로서 TRAIL유도성 아포프토시스를 강화시키는 효과가 있음을 확인할 수 있었다. 따라서 본 연구는 향후 폐암치료에 있어서 TRAIL유도성 아포프토시스가 이용될 수 있는 가능성을 확인한 기초자료가 된다고 생각된다.

Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is a homo-trimeric cytotoxic ligand. Several studies have demonstrated that incorporation of artificial trimerization motifs into the TRAIL protein leads to the enhancement of biological activity. Here, we show that linkage of the isoleucine zipper hexamerization motif to the N-terminus of TRAIL, referred as ILz(6):TRAIL, leads to multimerization of its trimeric form, which has higher cytotoxic activity compared to its native state. Size exclusion chromatography of ILz(6):TRAIL revealed possible existence of various forms such as trimeric, hexameric, and multimeric (possibly containing one-, two-, and multi-units of trimeric TRAIL, respectively). Increased number of multimerized ILz(6):TRAIL units corresponded with enhanced cytotoxic activity. Further, a high degree of ILz(6):TRAIL multimerization triggered rapid signaling events such as activation of caspases, tBid generation, and chromatin condensation. Taken together, these results indicate that multimerization of TRAIL significantly enhances its cytotoxic activity.
