  • The Chinese medicinal plant Artemisia annua is the source of the antimalarial compound artemisinin. By the way, Artemisin annula was known have endoperoxide ring structure is included and has anti-malarial effect. Malaria and Toxoplasma gondii (T. gondii) is belong to Apicomplexa genera. So, confirmed whether we go compound 1,5-bis(4-methoxyphenyl)-6,7-dioxa-bicyclo[3.2.2]nonane that have endoperoxide ring structure and there is anti-toxoplasmosis effect. The efficacy of 1,5-bis(4-methoxyphenyl)-6,7-dioxa-bicyclo[3.2.2] nonane alone was examined in vitro and in a murine model of acute toxoplasmosis. In vitro studies were peformed with HeLa cell cultures, with quantification of Toxoplasma growth by a cell proliferation assay. Selectivity of 1,5-bis(4-methoxyphenyl)-6,7-dioxa-bicyclo[3.2.2]nonane was 4.9 in vitro cell proliferation assay, this is higher than sulfadiazine (selectivity was 1.63). For in vivo studies, mice were acutely infected intraperitoneally with 10$^5$ tachyzoites of the virulent RH strain and then treated perorally for 4 days from 6 hours postinfection. Efficacy was assessed by sequential determination of parasite burdens in peritoneal cavity. in vitro, 1,5-bis(4-methoxyphenyl)-6,7-dioxa-bicyclo[3.2.2]nonane inhibited Toxoplasma growth at a concentration of 150mg/kg of body weight per day, the inhibition ratio was estimated to be 85.72%.

Sterculic Acid and Its Analogues Are Potent Inhibitors of Toxoplasma gondii

  • Toxoplasmosis is a serious disease caused by Toxoplasma gondii, one of the most widespread parasites in the world. Lipid metabolism is important in the intracellular stage of T. gondii. Stearoyl-CoA desaturase (SCD), a key enzyme for the synthesis of unsaturated fatty acid is predicted to exist in T. gondii. Sterculic acid has been shown to specifically inhibit SCD activity. Here, we examined whether sterculic acid and its methyl ester analogues exhibit anti-T. gondii effects in vitro. T. gondii-infected Vero cells were disintegrated at 36 hr because of the propagation and egress of intracellular tachyzoites. All test compounds inhibited tachyzoite propagation and egress, reducing the number of ruptured Vero cells by the parasites. Sterculic acid and the methyl esters also inhibited replication of intracellular tachyzoites in HFF cells. Among the test compounds, sterculic acid showed the most potent activity against T. gondii, with an $EC_{50}$ value of $36.2{\mu}M$, compared with $EC_{50}$ values of $248-428{\mu}M$ for the methyl esters. Our study demonstrated that sterculic acid and its analogues are effective in inhibition of T. gondii growth in vitro, suggesting that these compounds or analogues targeting SCD could be effective agents for the treatment of toxoplasmosis.

  • 본 연구는 우리나라에서의 정신질환 환자에 관한 Toxoplasma 항체 가의 분포를 조사한 성적이다. 대상은 국립서울정신병원에 입원중인 환자 573명이며 대조군은 울산중공업의 근로자중 신체검사로 건강인으로 판점된 76명이 다. 검사시기는 1983년 7월~8월이 었으며 검사방법으로는 간접 Latex응집반응용 kit("Eikon" E-ST 06)을 입수해서 사용하였다. 1. 대조군에서는 76명중 항체가 1 : 64 1예(1.3%)가 있었다. 2. 정신과환자는 573명 중 양성반응자 11예 (1.9%)이었으며 이중에서 정신분열환자군은 495명 중 3예(1.8%), 그리고 우울중환자 27명 중 2예(7.4%)가 양성이었다. 3. 실험군의 양성반응자중 남자는 367명 중의 8예(2.2%). 그리고 여자는 206명 중 3예(1.5%)애서 양성반응을 나타내었다. $\ulcorner$본 연구를 수행함에 있어서 여러가지 지도를 아끼지 않으셨던 일본 Jikei 의과대학 소림교수와 항원을 제공해주신 "Eikon" 회사측에 심심한 사의를 표합니다. $\lrcorner$

제주도 고등학교 학생의 톡소포자충 항체 양성률 및 감염 위험요인 (Seroprevalence and Risk Factors of Toxoplasma gondii Infection in High School Students in Cheju Province)

  • Objectives : To assess the seroprevalence and risk factors of toxoplasmosis in high school students in Cheju Province, Korea. Methods : A total of 4,570 high school students from 18 schools in Cheju Province were investigated for Toxoplasma gondii antibodies(IgG) by enzyme linked immunosorbent assay (ELISA). Risk factors for toxoplasmosis, such as place of residence, type of house, contact with cats and other pets, and rare meat consumption, were examined by questionnaire. Results . The overall antibody positive rate was 5.5% and ranged from 2.6 to 11.5% by school. There was no significant difference between males and females. Statistical analyses of the questionnaire data indicated that the risk factors for seropositivity were: (1) birth place (Cheju/others), (2) place of residence (rural/urban), (3) dietary habits (vegetarian/non vegetarian), (4) eating rare meat, (5) exposure to pets and (6) hepatitis B. Conclusion : We confirmed that the prevalence of the anti-Toxoplasma gondii antibody in a population of high school students in Cheju Province was to the previously reported prevalence.

경남지역의 돼지톡소플라즈마병 감염실태 조사 (Prevalence of Toxoplasma gondii infection from domestic pigs in Gyeongnam province)

  • Toxoplasma gondii is a species of parasitic protozoa in the genus Toxoplasma. The definitive host of T. gondii is the cat, but the parasite can be carried by the vast majority of warm-blooded animals, including humans. It is often found in the tissues of food animals including pigs and sheep. To determine the regional prevalence of infection with T. gondii, bloods (n=300) from domestic pigs and tissues (n=200) from slaughter pigs in Gyeongnam province were tested using an enzyme-linked immunosorbent assay (ELISA) and a polymerase chain reaction (PCR) for detection of antibody and antigen. A total of 115 sero-positive pigs were identified for a prevalence rate of 38.3%. Of the 50 herds from domestic pigs tested, 34 had at least one sero-positive pig for a herd prevalence rate of 68.0%. Sero-positive rates of pigs in fattening farm were higher than that of pigs in breeding company. Sero-positive rates of sows were higher than that of growing pigs. Seasonally, sero-positive rates of pigs were highest in winter (80.0%) and lowest in spring (23.8%). According to farm size, sero-positive rates of pigs were higher in small size farms (${\leq}$2,000) than that of big size farms (>2,000). However, none of the bloods (n=300) from domestic pigs and tissues (n=200) from slaughter pigs were positive for T. gondii specific DNA by PCR.

ELISA를 이용한 돼지 톡소플라스마병의 조기 진단에 관한 연구 (Use of the enzyme-linked immunosorbent assay for the detection of toxoplasmosis in swine)

  • This study was conducted to evaluate the possibility of application of a microenzyme-linked immunosorbent assay(micro-ELISA) for the serodiagnosis of specific toxoplasma antibodies in swine sera and this test was performed as a microplate system by coating the polystyrene plates with toxoplasma soluble antigen, incubated serially diluted sera, then added horse radish peroxidase labelled goat anti-swine IgG(r) conjugate followed by o-phenylenediamine as substrate. The color development by enzyme-substrate reaction was determined by the photometric reading [ELISA reader at 490nm (OD)] and visual reading. The soluble antigen was prepared from the tachyzoites in mouse peritoneal cavity. A total of 1,200 swine sera from pig slaughter-house and a total of 116 swine sera from pig breeding station (S-C farm) were tested for the detection of antibodies to Toxoplasma gondii. The results obtained were summarized as follows: 1. The optimal reactions of indirect ELISA for the test sera were determined by the dilution of antigen 1:256 and 1:3,200 of horse radish peroxidase conjugate [anti-swine IgG(r)]. 2. The specific togoplasma antibody(IgG) in pigs infected with Tp artificially were detected as the serum titers of 1:64 or 1:128 at one week postinfection. 3. Of a total of 1,200 swine sera from pig slaughter-house 505 samples of sera were detected as positive (42.1%) and of a total of 116 swine sera from S-C pig breeding station 68 samples of sera as positive (58.6%). 4. The specific antibody(IgG) detection rates against a total of 1,200 test sera from pig slaughter-house were not significant between male (43.1%) and female (40.7%). 5. The indirect ELISA was proved to be a sensitive and specific procedure for the serodiagnosis of swine toxoplasmosis and also evaluated as an effective screening test for the large scale of test samples in laboratory.

Inhibitory Effects of Toxoplasma Antigen on Proliferation and Invasion of Human Glioma Cells

  • Objective: Currently available therapies for human malignant gliomas have limited efficacy. Toxoplasma gondii, an obligate intracellular protozoan parasite, and Quil-A are nonspecific, potent immune stimulants. T. gondii is shown to have antitumor activity in some types of cancers. Therefore, this study is undertaken to evaluate the antitumor effect of Toxoplasma lysate antigen (TLA), alone or in combination with Quil-A, on human glioma U373MG and U87MG cells. Methods: The in vitro effects of TLA alone or in combination with Quil-A on the proliferation, invasion, and apoptosis of glioma cells were tested using MTT, Matrigel invasion, and DNA fragmentation assays, and the in vivo effects on the growth of gliomas were evaluated in athymic nude mice transplanted with glioma cells. Results: Treatment with TLA resulted in the suppressed proliferation and invasion of both U373MG and U87MG cells, in a dose-dependent manner. In addition, at high concentration, TLA induced glioma cell apoptosis. When TLA was administered in the mouse glioma model, malignant glioma growth was decreased. The combined treatment of TLA with Quil-A significantly inhibited the proliferation and invasion of cultured cells as well as tumor mass of implanted mice. Conclusion: TLA inhibits the proliferation and invasion of glioma cells in vitro and in vivo, and these antitumor effects of TLA are significantly enhanced by the addition of Quil-A.

  • A preservation test of Toxoplasma gondii tachyzoites for considerable time was tried to obtain simple and economical methods using various suspending media at $4^{\circ}C$ rather than serial passage of the parasite in mice. The preservation period was a term that the tachyzoites were detected from the peritoneal fluid of mice after inoculation of 2$\times$$10^5$ organisms preserved according to the lapse of time. The numbers of tachyzoite per 1\textrm{mm}^3$ of the peritoneal fluid with 2$m\ell$ of the saline solution taken in 4days after inoculation were presented as percentage in proportion to the control. The numbers. of tachyzoite per 1\textrm{mm}^3$ of the peritoneal fluid of the control were consisted of the average number of the tachyzoites of 10 mice inoculated with 2$\times$$10^5$ organisms by serial mouse passage. The tachyzoites could be preserved for 26 days in the suspending medium of saline solution at $4^{\circ}C$ Ringer's solution for 18 days, Hank's solution for 28 days, and egg-glycerine solution for 50 days.

강남성모병원과 제주의료원 내원 환자의 간접 Latex 응집 반응에 의한 Toxoplasma 항체가 (Toxoplasma antibody titers by indirect latex agglutination test in patients of Kangnam St. Mary's Hospital and Cheju Medical Center)

  • 강남성모병원의 일반 내원 환자 1,019명, 내원 환자 중 천식으로 진단된 1,030명, 그리고 제주의료원에 내원 한 환자 780면 등 총 2,829명에 대하여 Toxoplasma 항체가를 측정하기 위해 간접 latex 응집반응을 실시하였다. 간접 latex 응집반응의 양성 항체가를 1 : 32로 볼 때, 강남성모병원 일반 내원 환자에서는 19명이 양성으로 1.86%(남자 550명 중 12면으로 2.18%, 여자 469명 중 7명으로 1.49%), 천식 혹은 알레르기 환자에서는 11명이 양성으로 1.07%(남자 470명 중 4명으로 0.85%, 여자 560명 중 7명으로 1.25%)이었으며, 제주의료원 내원환자 에서는 45명으로 5.77%(남자 530명 중 33명으로 6.23%, 여자 250명 중 12명으로 4.80%)이었다. 연령적 분포로는 강남성모병원 일반 환자와 제주의료원 내원 환자의 남자에서 50대에 높은 빈도를 보였고, 전체 여자에서는 일반적으로 30대, 40대 및 60대에서 높은 빈도를 보였다. 양성 항체가를 보인 75명의 항체가별 분포로는 1 32가 32명으로 42.7%, 1 : 64가 21명으로 28.0%, 1 : 128이 8명으로 10.7%, 1 256이 6명으로 8.0%, 1 : 512가 3명으로 4.0%, 그리고 1 : 2,048 혹은 그 이상의 항체가를 보유한 사람은 5명으로 6.7%였다.

Anti-Toxoplasma Effects of Methanol Extracts of Feijoa sellowiana, Quercus castaneifolia, and Allium paradoxum

  • Objectives: The currently available agents for use against toxoplasmosis have serious limitations. Thus, the aim of the present study was to investigate the anti-Toxoplasma gondii (T. gondii) activities of methanol extracts of Feijoa sellowiana (F. sellowiana) (leaves and fruits), Quercus castaneifolia (Q. castaneifolia) (fruits), and Allium paradoxum (A. paradoxum) (leaves) in vitro and in vivo. Methods: Vero cells were treated with different concentrations (from 0 to $400{\mu}g/mL$) of the above extracts or with pyrimethamine at a dose of 50 mg/mL (positive control). Then, the viabilities of the T. gondii-infected cells were measured by using colorimetric MTT (3-(4, 5-dimethylthiazol-2-yl) 2, 5-diphenyltetrazolium bromide) assays. In addition, the survival rates of mice acutely infected with $2{\times}10^4$ RH strain tachyzoites of T. gondii were examined in vivo after intraperitoneal injection of the extracts at doses of 100 and 200 mg/kg/day for 5 days. Results: In the in vitro anti- T. gondii assay, the $IC_{50}$ values were 12.77, 180.2, 74.73, 213.2 and $163.8{\mu}g/mL$, and the selectivity indices were 6.05, 1.31, 0.35, 0.69 and 1.30 for the F. sellowiana (leaves and fruits), Q. castaneifolia, and A. paradoxum extracts and pyrimethamine, respectively. Moreover, the mice treated with F. sellowiana (leaves and fruits) achieved better results in terms of survival than the others (P < 0.05). Conclusion: The results of the current study indicate that methanol extract of F. sellowiana has significant anti-Toxoplasma activity. Further study should be conducted to investigate the potential bioactivity of this extract through bioactivity-guided fractionation.
