Acute toxicity of heavy metals( mercury, cadmium lead, chromium and copper ), organophosphours insecticides( EPN, 2, 4- D, and parathion ), and other chemicals( arsenic, cyanide, and phenol ) to Daphnia magna was analyzed. Acute toxicity of heavy metals, organophosphours insecticides and other chemicals on Daphnia magna, LC$_{50}$ was showed 0.07-0.73mg/1, 0.22-1.94mg/t and 1.73-39.92mg/1 respectively. Daphnia magna to most of toxic chemicals was sensitive and then it seems to be useful except phenol in toxic test of water. Toxic chemicals were classified as Group 1 which had a high LC$_{50}$ value and the low increase rate of toxicity according to the increase of concentration, Group 2 which had a high LC$_{50}$ value and the high increase rate of toxicity, Group 3 which had a low LC$_{50}$ value and the low increase rate of toxicity. To Daphnia magna, lead, chromium, EPN, and parathion were included in Group 1; Mercury and copper in Group 2; Arsenic and cadmium in Group 3; Cyanide 2,4- D, and phenol in Group 4.
A rapid, inexpensive enzymatic method is proposed for indirect water quality testing in terms of heavy metal toxicity. The activity of glucose-6-phosphate dehydrogenase was applied for heavy metal toxicity test as an effective criterion in water quality. The toxicity of Pb (lead) and Cd (cadmium) for water flea, Moina macrocopa, were evaluated for 2-8 days with variables of mobilization ability. And the reproduction impairment of Moina macrocopa were investigated as the parameter of chronic toxicity test for Pb and Cd. As a result, the EC$_{50}$ for immobilization of Moina macrocopa were Pb and Cd were 1.6749 and 0.4683, respectively. The values of reproductive impairment to Moina macrocopa for Pb and Cd were 9.5938 and 8.3264 in EC$_{50}$ A significant alteration of G6PDH (Glucose-6-phosphate dehydrogenase) activity of Moina macrocopa was observed when Cd and Pb were treated in media. The results obtained indicate that G6PDH activity of Moina macrocopa can be used as an indicative parameter in aquatic toxicity tests for heavy metals.als.
Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are fully flurorinated organic compounds which are highly persistent in environment and accumulated in organism. These chemicals are released to the environment at their manufacture, during their use in industrial and consumer applications and from disposal after their use. The purpose of this study was to determine the effects of PFOA and PFOS on the freshwater flea (Moina macrocopa). Acute toxicity test and chronic toxicity test were performed for 2 days and 10 days, respectively. Acute toxicity was assessed on the basis of mortality, while chronic toxicity was assessed by fecundity as well as mortality. The acute toxicity studies on PFOA and PFOS showed that the values of $LC_{50}$ were $73.9\;mg/L\;and\;27.7\;mg/L$ respectively. In the chronic toxicity test, fecundity was reduced significantly at 24.1 mg/L of PFOA and 9.3 mg/L of PFOS, respectively. Conclusively, the results of this work suggest that Moina macrocopa could be a suitable model organism for screening and assessing of environmental pollutants in water.
해양 기원의 발광박테리아 Vibrio fischeri를 이용한 발광박테리아 독성시험법은 1980년대 초반에 소개된 이후 많은 연구자들에 의해 이용되어오다 1998년 국제공인시험법으로 등재되어 지금은 전 세계적으로 이용되고 있다. 이 시험법은 테스트하고자하는 화학물질이나 환경시료에 발광박테리아를 30분간 노출시킨 후 나타나는 발광량 감소의 정도로서 독성 유무나 강도를 평가하는 방법으로서 신속성, 정밀성, 민감성, 경제성이 뛰어나다는 평가를 받고 있으며, 시험생물의 유지/배양없이 상용화된 키트 제품을 이용하여 독성 시험을 수행할 수 있다. 본 연구에서는 해양성 발광 박테리아 독성평가 키트로 판매되는 상용화제품(N-Tox)을 이용하여 수행된 결과를 제시하였다. 시험법 개발 및 표준화를 위한 시험항목으로는 시료 주입량, 발광량과 같은 기본 항목 테스트와 표준독성물질을 이용한 정도관리 테스트, 화학물질 및 현장시료에 대한 테스트가 포함되었다. 이 시험법은 자동화되어 누구나 쉽게 수행이 가능하면서도, 시험 결과 독성물질에 대한 용량-반응 관계가 확실하게 평가되고, 정밀도가 뛰어나며, 현장시료 독성평가에 이용할 수 있을 정도의 민감성을 지닌 것으로 입증되었다. 따라서 아직 생물검정이 보편화되지 않은 국내에서 편리하게 이용할 수 있는 표준독성시험방법의 하나로서 활용 가능하다고 사료된다.
Objectives: This study was performed to check for reversibility in the changes induced by a 13-week, repeated, dose toxicity test of Sweet Bee Venom (SBV) in Sprague-Dawley (SD) rats. Methods: Fifteen male and 15 female SD rats were treated with 0.28 mg/kg of SBV (high-dosage group) and the same numbers of male and female SD rats were treated with 0.2 mL/kg of normal saline (control group) for 13 weeks. We selected five male and five female SD rats from the high-dosage group and the same numbers of male and female SD rats from the control group, and we observed these rats for four weeks. We conducted body-weight measurements, ophthalmic examinations, urinalyses and hematology, biochemistry, histology tests. Results: (1) Hyperemia and movement disorder were observed in the 13-week, repeated, dose toxicity test, but these symptoms were not observed during the recovery period. (2) The rats in the high-dose group showed no significant changes in weight compared to the control group. (3) No significant differences in the ophthalmic parameters, urine analyses, complete blood cell counts (CBCs), and biochemistry were observed among the recovery groups. (4) No changes in organ weights were observed during the recovery period. (5) Histological examination of the thigh muscle indicated cell infiltration, inflammation, degeneration, necrosis of muscle fiber, and fibrosis during the treatment period, but these changes were not observed during the recovery period. The fatty liver change that was observed during the toxicity test was not observed during the recovery period. No other organ abnormalities were observed. Conclusion: The changes that occurred during the 13-week, repeated, dose toxicity test are reversible, and SBV can be safely used as a treatment modality.
Two methods, a Ceriodaphnia algal uptake suppression test (CAUST) and a new toxicity test based on temperature control (TTBTC) which are based on feeding behaviour and temperature control, respectively, were developed and compared for the adoption as the better methodology for short-term toxicity screening. As previously published by Lee et aI., (1997), the CAUST method is based on the feeding behaviour of C. dubia and requires as little as 1 hour of contact time between C. dubia neonates and toxicant. However, even though CAUST requires only 1 hour of contact time, this method still take many hours for the preparation and measurement. Before the test starts, neonate digestive tracts were cleared by feeding yeast to the daphnids, Neonates were then exposed to toxicant, followed by addition of Scenedesmus subspiatus into the bioassay vessels. Daphnids were examined under the bright-field microscope with the presence of algae (indicated by a green colored digestive tract) or the absence of algae. Uptake indicated no toxic effect, whereas, absence of uptake indicated toxic inhibition. Unlike CAUST, the newly developed method (TTBTC) is based on just temperature control for the toxicity test of C. dubia. Initially, neonates are exposed to toxicants while the temperature of water bath containing media increased to $35.5^{\circ}C$. After 1.25 hour of contact time, the number of the daphnids, either live (no toxic effect) or dead (toxic effect), is counted without the aid of any instrument. In both methods, median effective concentrations ($EC_{50}$ values) were computed based on the results over a range of dosed toxicant concentrations. It showed that TTBTC was as sensitive as the standard 48-hour acute bioassay and CAUST. TTBTC and CAUST were much more sensitive than the I-hour I.Q. test and 30-minute Microtox. This study indicates that TTBTC is an easier and more rapid toxicity test than the standard 48-hour acute bioassay and even CAUST.
Kyoaesamultang(KAT) has been used as an important prescription for various diseases including threatened abortion, associated with pregnancy in traditional medicine. In oder to identify the safety of KAT, this study was designed to determine mutagenicity and hepato-toxicity. In Rec-assay, Bacillus subtills H-17($Rec{^+}$) and M-45($Rec{^-}$) strains were used to clarify the DNA damage property. In Ames test, Salmonella typhimurium TA98 and TA100 were used for mutagenicity testing. In SOS umu test, Salmonella typhimurium TA1535 containing plasmid pSK1002 was used as a tester strain, and the levels of umu operon expression were monitored by measuring the $\beta$-galactosidase activity. From tested results, KAT did not show DNA damage and mutagenicity. On the other hand, hepato-toxicity of KAT to female ICR mice was monitored by the measurements of s-GOT, s-GPT and LDH activities after oral feeding for 15days. KAT showed 34% increase of s-GOT and s-GPT activities, also exhibited 35% increase of LDH activity in mice sera.
1) Puerariae Radix butanol ext. (100, 200, 400mg/kg, p.o. single treatment) alone partly showed blood pressure decreasing effect in SHRs and increasing effect of urinary volume in normal rats. 2) Cadmium nitrate (10mg/kg, s.c. single treatment) induced toxicity such as body weight decreasing effect, antidiuretic effect and muscle relaxant effect such as pull-up test, traction test and rota rod test in rats. However, Puerariae Radix butanol ext. (100, 200, 400mg/kg, p.o. single treatment) showed antidotal effects on the above and also in acute toxicity test when coadministered with both of them. 3) Cadmium nitrate (1mg/kg, s.c. 7 days consecutive treatment) did not showed toxicity in body weight change, blood pressure, change, serum biochemical parameters in rats. Puerariae Radix butanol ext. (100, 200, 400mg/kg, p.o. 7 days consecutive treatment) did not also showed any antidotal effects when coadministered with both of them for 7 days.
Acute and subacute toxicity of Artemisia asistica Nakai Aqua-acupuncture Solution (ANAS) were studied in ICR mice. In acute toxicity test, mice were injected intraperitoneally with single dose of $1{\times}$, $5{\times}$, $10{\times}$ ANAS, and toxicological responeses were observed for consecutive 14 days. Mortality, body weight changes, organ weight, and serum chemistry were performed. The mortality and body weight changes of mice treated with $1{\times}$ and $5{\times}$ ANAS were not affected during the experimental periods. With the $10{\times}$ ANAS treatment, there were dead animals and changes of body weight, organ weight and serum biochemical values were observed during the experimental period. In subacute toxicity test, mice were injected intraperitoneally with doses of $1{\times}$, $10{\times}$ ANAS for 14 days. No difference was found between control and $1{\times}$ ANAS treated group in mortality, changes of body weight and organ weight, and serum biochemical values. However, Dead animals, changes of body weight and organ weight, and increased serum biochemical values were observed with $10{\times}$ ANAS treated groups. These results suggest that $1{\times}$ ANAS causes no toxicity in acute and subacute toxicity tests. However $10{\times}$ ANAS causes toxicity in both tests.
