Methylmercury (MeHg), one of the heavy metal compound, can cause severe damage to the central nervous system in humans. Many reports have contributed MeHg poisoning to contaminated foods and release into the environment. Despite many studies on the pathogenesis of MeHg-induced central neuropathy, no useful mechanism of toxicity has been established. To find genes differentially expressed by MeHg in neuronal cell, we peformed forward and reverse suppression subtractive hybridization (SSH) method on mRNA derived from neuroblastoma cell line, SH-SY5Y treated with solvent (DMSO) and 6.25 uM (IC$\sub$50/) MeHg. Differentially expressed CDNA clones were sequenced and the mRNAs were re-examined on Northern blots. These sequences were identified by BLAST homology search to known genes or expressed sequence tags (ESTs). Analysis of these sequences has provided an insight into the biological effects of MeHg in the pathogenesis of neurodegenerative disease and a possibility to develop more efficient and exact monitoring system of heavy metals as common environmental pollutants.

Toxicologic Pathology, 25: 529-531, 1997. Male and female F344 rats, approximately 19 weeks old, from prechronic toxicity studies performed for NTP/NIEHS over a period of several years at different laboraories located throughout the US. The rats were supplied by 2 different production colonies located in the eastern and western areas of the US. Salmonella typhimurium, causing mouse typhoid, infects hosts such as macrophage cells, and proliferates in intracellular vacuoles causing infected cells to trigger numerous genes to respond against the infection. In this study, we tried to identify such genes in RAW264.7 cells by using the PCR screening method with degenerate primers. Fourteen genes were found to be differentially expressed after a 4 h infection in which the expression of 8 genes increased while expression of the others decreased. Most of the genes were involved in proinflammatory responses such as cytokines production and cell death. The mutation in msbB gene encoding the myristoyl transferase in lipid A of lipopolysaccharide (LPS) resulted in much lower toxicity to the inoculated animals. We compared the expression of the identified genes in wild-type and msbB-mutated S. typhimurium infections and found that Lyzs encoding lysozyme type M was differentially expressed. This gene is quite likely to be related to bacterial survival in the host cells.
Differentially expressed genes by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) were identified in order to evaluate them as dioxin-sensitive markers and crucial signaling molecules to understand dioxin-induced toxic mechanisms in human bronchial cells. Gene expression profiling was analyzed by cDNA microarray and ten genes were selected for further study. They were cytochrome P450, family 1, subfamily B, polypeptide 1 (CYP1B1), S100 calcium binding protein A8 (calgranulin A), S100 calcium binding protein A9 (calgranulin B), aldehyde dehydrogenase 1 family, member A3 (ALDH6) and peroxiredoxin 5 (PRDX5) in up-regulated group. Among them, CYP1B1 was used as a hallmark for dioxin and sharply increased by TCDD exposure. Down-regulated genes were IK cytokine, interferon-induced protein with tetratricopeptide repeats 1 (IFIT1), nuclease sensitive element binding protein 1 (NSEP1), protein tyrosine phosphatase type VI A, member 1 (PTP4A1), ras oncogene family 32 (RAB32). Although up-regulated 4 genes in microarray were coincided with northern hybridization, down-regulated 5 genes showed U-shaped expression pattern which is sharply decreased at lower doses and gradually increased at higher doses. These results introduce some of TCDD-responsive genes can be sensitive markers against TCDD exposure and used as signaling cues to understand toxicity initiated by TCDD inhalation in pulmonary tissues.
Rusty root, the browning disease on ginseng, decreases quality and value. Recent studies indicated that endophytic bacteria could be a possible cause of rusty root. Actinomycetes antagonistic to the rusty-root-causing bacteria were isolated from soil. Twenty nine out of 932-isolates of Actinomycetes from soil showed antibacterial activity against Agrobacterium tumefaciens and Pseudomonas veronii an endophytic isolate in ginseng. The strongest antibacterial strain(ATO4O104) was classified based on 16S rDNA sequence. The Actinomycetes strain, ATO4O104, isolated in soil of USA volcano national park was identified as Streptomyces adephospholyticus. To test plant toxicity, radish seeds were sprouted with the culture of S. adephospholyticus and it did not show any harmful effect. The butanol partition out of n-hexane, ethyl acetate, butanol, and water partions showed the highest antibacterial activity.

We prepared to total RNA from SH-SY5Y cells treated with solvent (DMSO) and $6.25\;{\mu}M\;(IC_{50})$ MeHg and performed forward and reverse SSH. Differentially expressed cDNA clones were screened by dot blot, sequenced and confirmed that individual clones indeed represent differentially expressed genes with real time RT-PCR. These sequences were identified by BLAST homology search to known genes or expressed sequence tags (ESTs). Analysis of these sequences may provide an insight into the biological effects of MeHg in the pathogenesis of neurodegenerative disease and a possibility to develop more efficient and exact monitoring system of heavy metals as ubiquitous environmental pollutants.
Aqueous extract of Morus alba L. blocked the toxic effect of paraquat on E. coli growth. The active components in the extract may be capable of crossing the cell membranes and protect against superoxide toxicity in E. coli, The extract inhibited $FeSO_4/H_2O_2$ induced lipid peroxidation in rat liver homogenate and protected against t-butyl hydroperoxide caused Ac2F cell damage. Moreover, the extract showed inhibitory effect on phospholipase $A_2$ activity in a dose dependent manner. Antiinflammatory effect of the extract was further investigated using the carrageenin-induced oedema model. A single adminstration of the extract (3g/kg body, p.o.) was more effective than indomethacin. These results suggest that the isolation and identification of the active components would have significant therapeutic application to inflammation associated with oxygen radicals.
The anti-diabetic effect of dried-silkworm Dongchunghacho water extracts was investigated in streptozotocin (STZ)-induced diabetic rats. Seven-week-old SD rats were randomly assigned to six groups: normal control (NC) group, diabetic control (DC) group, water extracts of Paecilomyces japonica grown on a dried-silkworm (PJ-DS, 50 mg/kg) group, PJ-DS (250 mg/kg) group, water extracts of Cordyceps militaris grown on a dried-silkworm (CM-DS, 250 mg/kg) group, and water extracts of dried-silkworm (DS, 250 mg/kg) group. These groups were orally provided with sample dissolved in water respectively for four weeks after an injection of STZ (60 mg/kg, ip) followed by identification of diabetic control (DC) group. After four weeks, body weight of all diabetic groups was significantly lower than that of the normal control (NC) group, but among diabetic groups there were no significant differences. Blood glucose levels of the PJ-DS (50 mg/kg), PJ-DS (250 mg/kg), CM-DS (250 mg/kg) and DS (250 mg/kg) groups were reduced compared to the DC group by 3.0 percent, 18.5 percent, 6.9 percent and 13.1 percent, respectively. Concentration of total cholesterol and triglyceride in blood plasma were to some degree decreased in PJ-DS (250 mg/kg) and DS (250 mg/kg) groups compared to the DC group. According to these results dried-silkworm Dongchunghacho water extracts (PJ-DS) boost protection against STZ-induced toxicity.
A proteomic analysis of the proteins in mouse brain that were differentially expressed in response to $TiO_2$ nanoparticles was conducted to better understand the molecular mechanism of $TiO_2$ nanoparticle-induced brain toxicity at the protein level. A total of 990 proteins from mouse brain were resolved by two-dimensional gel electrophoresis. A comparative proteomic analysis revealed that the expression levels of 11 proteins were changed by more than 2-fold in response to $TiO_2$ nanoparticles: eight proteins were upregulated and three were downregulated by $TiO_2$ nanoparticles. In addition, the activities of several antioxidative enzymes and acetylcholine esterase were reduced in $TiO_2$ nanoparticle-exposed mouse brain. The protein profile alterations seem to be due to an indirect effect of $TiO_2$ nanoparticles, because $TiO_2$ nanoparticles were not detected in the brain in this investigation.
Metabolism study of the dye, benzidine, was performed by gas chromatography-mass selective detector (GC/MSD) in the urine of rats orally administered 100 mg/kg benzidine. Urine samples were collected in metabolic cages for 0-24, 24-48, and 48-72 hrs. Ten ml of the urine was extracted with XAD-2 resin and the XAD-2 column was eluted with methanol. After evaporation, benzidine and its metabolites were extracted with diethyl ether (for non-conjugated fraction). For conjugated metabolites, $\beta$-glucu-ronidase was added to the aqueous layer that was incubated for 1 hr at 5$0^{\circ}C$ and the aqueous layer was extracted as in non-conjugated fraction. Aliquot of trimethylsilylated derivatives was applied to the GC/MSD. The mutagenicity of benzidine and its acetylated metabolites was tested by histidine/reversion assay. Five metabolites observed and confirmed either by electron impact and chemical ionization modes of the GC/MSD, or authentic compounds were monoacetyl-, diacetyl-, hydroxyacetyl-, hydroxydiacetyl-, and hydroxy-benzidine. Monoacetyl-benzidine was more potent than benzidine in histidine/reversion assay. This data indicates that monoacetylation of benzidine may be one of the metabolites produced in metabolic activation process.
