Alcohol Fermentation of Opuntia ficus Fruit Juice

  • Prickly pear juice(PPJ) extracted from prickly pear fruit (Opuntia ficus-indica)was used as raw material for the production of alcoholic beverages. Prickly pear juice (PPJ) had 0.88 oBrix of soluble solid, pH 3.96 and 0.14% of total acidity. Alcohol fermentation of 25% PPJ including 22 oBrix of sugar and 1$\times$106 of inoculum was suitable for alcohol fermentation indicating the rapid decrease of sugar content. The 22 oBrix of sugar in PPJ (25%)GJ (50%) mixture was changed to 6.5 oBrix after fermentation at 3$0^{\circ}C$ for 7 days. The alcohol content was 9.2% (w/v). PPJ (70%)/GJ (30%) mixture produced alcoholic beverage with 6.9% alcohol content resulting in the gradual decrease of soluble for 7 days. On the other hand, PPJ (50%)/GJ(50%) mixture carried out completely the alcohol fermentation at 22$^{\circ}C$ for 6 days and enhanced the red color of alcoholic beverages.

Physicochemical Properties of Citrus miyakawa wase Produced in Cheju, and Citrus Jam-making (제주산 궁천조생의 특성과 젤리화 식품의 제조)

  • Soluble solids and total carbohydrates of Citrus miyakawa wase harvested middle of November 1993 Topyung-Dong, Seogwipo-si, and Cheju were 10.7 and 8.57%, and carbohydrate was consisted of about 1/2 sucrose, 1/4 glucose and 1/4 fructose, respectively. Acid content of citrus Juice was 1.04%, and citric acid was 74.27% of total organic acids. Fruit weight, peel thickness, soluble solids, pH, hardness and edible part ratio had a good correlation in linear function with increasing fruit size. Total carbohydrate, reducing sugar and vitamin C of citrus jam made on optimum conditions were 65.33%, 27.98%, and 51.40mg/100g , respectively. Microbial growth on the products were not recognized at 3$0^{\circ}C$ for a month. Compared with other related citrus jellying products, the test sample was excellent in appear ance, taste and total preference degree on sensory evaluation.

Optimization of Extraction Conditions for Soluble Ginseng Components Using Microwave Extraction System under Pressure (가압형 마이크로웨이브 추출장치를 이용한 가용성 인삼성분의 추출조건 최적화)

  • Microwave extraction system equipped with closed vessels was applied to confirm its capability of extracting ginseng solubles including saponin, which was monitored to optimize extraction con ditions by response surface methodology. Total yield increased with the decrease in particles size and more extraction steps. Soluble components were completely extracted by operating the system within 6 min per one step, which should be repeated by 3 or 4 times. Optimized conditions for maxi mum extraction of response variables(total yield, crude saponin) were 40~50% of ethanol concentration, about 140oC of extraction temperature, and within 6 min of the extraction time. The extraction effi ciency of total yield was dependent on the decrease of ethanol concentration, while crude saponin content was favored on the higher degree of ethanol concentration. According to the temperature guideline, below 90oC of extraction temperature, the optimal ranges of extraction conditions were predicted as 30~50% of ethanol concentration and 2.5~6.5 min of extraction time. Estimated values of total yield and crude saponin were in good agreement with experimental values.

  • This study was investigated to know changes of the cell wall components, cell wall degrading enzyme activities and contents of soluble protein of strawberry during ripening and softening. The contents of water soluble substances were slightly increased during ripening, but the contents of alcohol-insoluble substances were not changed. The contents of pectin were not changed at green mature and turning stage, while decreased after mature stage. The contents of alkali-soluble hemicellulose and cellulose were increased during ripening and softening. The contents of water-soluble and saltsoluble protein were not changed, but the content of cell wall protein was slightly decreased during ripening. The content of total protein was increased at turning stage, it is not changed after turning stage. $\beta$-Galactosidase activity was increased during ripening, and pectinmethylesterase activity was decreased at turning. Phenylalanine ammonia-lyase activity was changed up to mature stage, but decreased at overripening stage. Polygalacturonase and cellulase activities were not detected at all of ripening stages.

Compositional Differences of Ojeok-san (Wuji-san) Decoctions Using Pressurized or Non-pressurized Methods for Variable Extraction Times

  • This study compared Ojeok-san (Wuji-san in Chinese) decoctions produced using different extraction methods for variable times. Decoctions were extracted in pressurized or non-pressurized conditions for 60, 120, and 180 mins. We investigated the Ojeok-san extract yield, the total soluble solid content, the hydrogen ion concentration (pH), and the reference compound content. The extract yield and the total soluble solid content were higher in decoctions produced by non-pressurized extraction; both were proportional to the extraction time. The pH tended to decrease as the extraction time was increased in decoctions produced using both methods. After 60 and 180 mins, the levels of albiflorin, paeoniflorin, nodakenin, naringin, and neohesperidin were significantly higher in decoctions extracted using the non-pressurized method compared with those extracted using the pressurized method. After 120 mins, only cinnamaldehyde was extracted in a greater amount in pressurized decoctions compared with non-pressurized decoctions. The levels of paeoniflorin, ferulic acid, nodakenin, naringin, hesperidin, neohesperidin, and glycyrrhizin increased with time in non-pressurized decoctions. This study showed that the use of pressurized and non-pressurized extraction methods for different times affected the composition of Ojeoksan (Wuji-san) decoctions.

Freshness Extension of ‘Fuji’Apple to Packaging materials (포장재가 ‘후지’사과의 신선도에 미치는 영향)

  • This study was measured weight loss, total ascorbic acid, titratable acidity, and soluble solids content to investigate the effect of EPS(expanded polystyrene foam, foaming rate 55 times V/W) box, LDPE film pouches and double wall corrugated fiberboard box during storage at 20$\^{C}$. Weight loss of apple packed with corrugated paperboard box after 40 days storage was 5.7%, arid those of LDPE, CE(zeolited coated film) film and EPS box were 0.2-0.5%. Ascorbic acid content of apple packed with LDPE, CE, EPS was higher than that of control. Titratable acidity and total soluble solid content of control were changed 40% and 10%, but the EPS were changed 27% and 4%, respectively. Overall appearances of ‘Fuji’apple packed with LDPE, CE and EPS were better than that of control.

Effects of High Phosphorus Supply on Zn and Cu Uptake by Mulberry(Morus alba L.) (고농도(高農度) 인산(燐酸) 수경액(水耕液)중에서 뽕나무의 Zn과 Cu 흡수(吸收))

  • Water culture studies were conducted in the greenhouse with mulberry plants to investigate the nutrient uptake, especially Zn and Cu, under high phosphrous concentration. Mulberry plants were grown with five phosphorus levels(0, 0.2, 0.5, 2.0, 5.0 mM). Leaves and roots were analyzed for water content, total nitrogen, P, K, Ca, Mg, Fe, Mn, total Zn, soluble Zn, Cu, Cl, $NO_3HPO_4$ and $SO_4$. Dry matter increased upto 2.0mM phosphorus level, and then decreased. Water content, total nitrogen, P, K, and Fe in leaves increased with increasing phosphorus level. Total Zn content in leaves showed little change, whereas soluble Zn increased and Cu decreased with increasing phosphorus level. With increasing phosphous level. $SO_4$ and Cl decreased and then sharply increased above 2.0mM phosphorus. Lower uptake of Cu and higher uptake of $SO_4$ and Cl suggest a cause of mulberry yield decline with high accumulation of soil phosphorus.

Comparison of phytochemical composition and physiological activity of 'Hongsan' and 'Hansan', a new variety of garlic (마늘 신품종 홍산과 한산의 식물화학적 구성 및 생리활성 비교)

  • This study was performed to investigate the physiological activity of Hongsan garlic and Hansan garlic, which were registered in the plant breeders' right in 2016. The content of mineral components, except sodium and sulfur ingredients, was found to be high in Hansan garlic. In addition, organic sulfur components were high in Hansan garlic. Moreover, the total soluble solid content, reducing sugar content, and alpha glucosidase inhibitory activity were high in Hansan garlic. Acidity, pH, total phenol, and flavonoid content were high in Hongsan garlic. Both varieties showed higher inhibitory activity to gram-positive bacteria than to gram-negative bacteria. The antimicrobial activity of Hongsan garlic was slightly higher than that of Hansan garlic. These results are expected to provide useful information to farmers who grow the above garlic varieties.

  • The present study was designed to investigate biosynthetic patterns of polysaccharides under catabolic repression and derepression in Saccharomyces uvarum. Correlation coefficients between polysaccharide synthesis and polyphosphate accumulation were examined, according to the culture phase and under various phosphate concentrations (free, limited, sufficient). During catabolic derepression, biosynthesis of glycogen was enhanced. rapidly and highly in the cells grown on minimal medium, compared with those grown on the complete medium. Acid soluble glycogen type was the main component of total glycogen and alkali soluble glycogen was synthesized in small amount, after 24 hr culture, at the time of almost exhaustion of sugar in the medium. Total glycogen was accumulated highly in proportion to the amount of phosphate added to the medium. It could be postulated that type 'C' isoenzyme among ALPase was directly or indirectly correlated with the glucan synthesis. Mannan synthesis indicated maximal amount at the early exponential phase and stationary phase, and also acid soluble sugars at the stationary phase. Correlation coefficient between the mannan synthesis and poly-p-'C' accumulation, and also between mannan synthesis and phospholipid content indicated 0.866 and 0.726, respectively.

Changes in Photosynthetic Rate and Protein Content in the Leaf during the Senescence of Tobacco Plant (Nicotiana tabacum L) (담배의 노화과정 중 광합성 및 단백질 함량의 변화)

  • This study was carried out to obtain the basic data which include the change of the photosynthetic rate and protein content according to growth stage in the process of senescence of tobacco plant The photosynthetic rate was the maximum with 26.31$\mu$mol.CO2/m2.sec and stomatal resistance was the minimum with 0.2552cm/sec at 15th days after leaf emergence. However, after 50 days the photosynthesis was very little occurred. During leaf developments the number of chloroplast was increased and reached at the maximum at 25th days after emergence of leaf, thereafter, it was decreased gradually. The content of protein increased continuously and showed the highest value at 15th days after leaf emergence. The degradation rate of soluble protein was more rapid than that of insoluble protein at early stage of senescence. The range of decrement in the insoluble protein was low at late stage of senescence. The content of Rubisco, the key enzyme of photoamthesis, corresponded to about 50% of soluble protein and reached to the maximum at 150 days after leaf emergence. As the senescence progressed, the content of large subunit(UV) of Rubisco showed a tendency to decrease more rapidly than that of small subunit(SSU). The total amount of amino acids was the highest at 15th days after leaf emergence.
