Changes of Phenol Compounds according to Storing Years in Soybean

  • The objective of this study was to determine the role of storing years with the variation of total phenol and individual phenolic compounds in soybean (Glycine max L.) seeds. The total phenol content varied from 0.36 to 0.42% over four years, with the highest value (0.42%) found at storage for two and three years. Among the nine soybean varieties examined, Daweonkong had the highest total average phenol content (0.58%). The total content of 11 phenolics varied from 730.0 to 1812.8 $\mu\textrm{g}\;\textrm{g}^{-1}$ over storage for four years, and the highest concentration (1812.8 $\mu\textrm{g}\;\textrm{g}^{-1}$) was found at storage for two years. Myeongjunamulkong (1465.4 $\mu\textrm{g}\;\textrm{g}^{-1}$) had the highest mean content among the nine soybean varieties. The total content of 11 phenolic compounds measured in this study occupied from 20.96 to 47.73% of the total phenol contents. The highest total phenol contents were in seeds with black coats (5279.4 $\mu\textrm{g}\;\textrm{g}^{-1}$), while the highest concentration of individual phenolic compounds were in seeds with green coats (1419.5 $\mu\textrm{g}\;\textrm{g}^{-1}$). Our study suggests that it may be feasible to improve soybean varieties with high functional substances such as phenolic compounds.

Changes of Biological Activities of Rhizome and Leaves of Polygonatum sibiricum Redoute according to Steaming Time and Temperature (층층갈고리둥굴레 (Polygonatum sibiricum Redoute) 뿌리줄기와 잎의 증숙 시간과 증숙 온도에 따른 생리활성 변이 연구)

  • Background: The aim of this study was to analyze the total phenol and total flavonoid contents and antioxidant activity of steam-treated leaves and rhizomes of Polygonatum sibiricum Redoute. In addition, we aimed to confirm their potential use as cosmetic materials by investigating their anti-aging and skin-whitening activity. Methods and Results: The leaves and rhizomes of P. sibiricum were treated with steam at different temperatures for different durations, and the antioxidant activity (DPPH and ABTS radical scavenging activity) and total phenol and total flavonoid contents of each sample were tested. The steam temperature and treatment duration siginificantly affected the antioxidant activity and, total phenol and total flavonoid content of the leaves and rhizome of P. sibiricum. Treating the P. sibiricum samples with steam at 120℃ for 12 h, yielded higher total phenol and total flavonoid contents. Comparatively, the samples treated with steam at 120℃ for 12 to 24 h showed significantly higher antioxidant activity. Further, the steamed samples of P. sibiricum demonstrated collagenase and tyrosinase inhibition activity, which indicated their anti-aging and skin-whitening properties. The samples steamed at 120℃ for 12 h, exhibited higher collagenase and tyrosinase inhibition activity. Conclusions: Leaves and rhizomes of P. sibiricum steamed at 120℃ for 12 h, showed highest antioxidant activity and, total phenol and total flavonoid contents than all other samples. Our results indicate the potential of using P. sibiricum as a cosmetic material by confirming its excellent anti-aging and whitening activity.

Comparison of Total Phenol, Flavonoid Contents, and Antioxidant Activities of Orostachys japonicus A. Berger Extracts (와송(Orostachys japonicus A. Berger) 추출물의 총 페놀, 플라보노이드 함량 및 항산화 활성 비교)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of Orostachys japonicus A. Berger. Orostachys japonicus A. Berger have been known to contain functional materials such as kaempferol, hydroquinone, methyl gallate, quercetin, gallic acid etc. To identify the main functional materials of Orostachys japonicus A. Berger, the contents of flavonoid and phenol were measured. We extracted Orostachys japonicus A. Berger powder from four solvents such as chloroform:methanol (CM, 2:1, v/v), distilled water (DW), 70% methanol, 70% ethanol. After that, this study determined tannin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power and reducing power of Orostachys japonicus A. Berger extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of Orostachys japonicus A. Berger extracts was higher in the order of 70% ethanol, 70% methanol, DW and CM (p<0.05). The results showed that antioxidant activity of Orostachys japonicus A. Berger extracts supposed to affect by the total phenol and flavonoid contents.

Drying Condition Affects Total Phenol Contents and Antioxidant Activities of Hizikia fusiformis

  • This study was performed to evaluate the effect of drying methods on the polyphenol contents and antioxidant activities of Hizikia fusiformis. The seaweed was dried using four different conditions: vacuum drying (VD) at $20^{\circ}C$ and $30^{\circ}C$ and hot-air drying (HD) at $40^{\circ}C$ and $60^{\circ}C$. After drying, the total phenol content, DPPH and nitrite scavenging activities of the water extracts were determined. The total phenol contents were significantly (p<0.05) higher in the vacuum dried samples than the hot-air dried samples. DPPH radical was scavenged 82.67% and 83.45% by VD-20 and VD-30, and 70.44% and 71.23% by HD-40 and HD-60, respectively. At pH 1.2, Hizikia fusiformis scavenged 87.43%, 88.45%, 72.14% and 73.45% of nitrite by the treatment VD-20, VD-30, HD-40 and HD-60, respectively. Interestingly, the nitrite scavenging activity was much more reduced at pH 3.0 than at pH 1.2. Through these experiments, we were able to show that the drying conditions affect the total phenol contents and antioxidant capacities of Hizikia fusiformis.

Studies on the Phenol Compounds in Smoke Total Particulate Matters of Flue-cured Tobacco Leaves (황색종 잎담배의 연기응축물중 페놀 화합물에 관한 연구)

  • This study was carried out to quantitatively determine phenol compounds in smoke total particulate matter(TPM) to evaluate the qualititive characteristics of flue-cured tobacco varieties(NC82, KFl14 and KFl18). After collecting The TPM by using smoking machine, the concentration of phenol compounds were analyzed by gas chromatography as their trimethylsilyl derivative on a fused capillary column bonded with a 5% PE ME siloxane stationary phase. In all the flue-cured tobacco varieties, the major phenol components were monohydroxy compounds. The order of the highest concentration of total phenol compounds in TPM was NC82, KF114 and KFl18 but the contents of dihydroxy compound in the KFl18 was higher than those of NC82 and KF114.

Development of Soybean Snacks and Analysis of Bioactive Compounds (조미 콩 스낵 개발 및 생리활성물질 분석)

  • The purpose of this study was to analyze the bioactive compounds of soybean snacks and to carry out a sensory evaluation of those variations which had various seasoning mixing ratio. Five soybean snacks were developed for these experiments. Proximate composition, the contents of total phenol compounds and isoflavone, and sensory characteristics of the soybean snacks were measured. There were significant differences in amounts of isoflavones and total phenolic compounds of soybean snacks($p$ <0.05). The seasoned soybean snacks had a higher level of daidzein and genistein contents than did control group. The total phenol compound contents of the control group was 210.3 mg gallic acid equivalents/100 g, whereas those for individual soybean snacks(A~E) were 152.3, 160.2, 162.4, 158.4 and 164.6 mg gallic acid equivalents/100 g, respectively. In sensory evaluation, there were significant differences($p$ <0.05) in taste, texture, and overall acceptability of the soybean snacks.

Impacts of Various Solvent Extracts from Wild Haw(Crataegus pinnatifida Bunge) Pulpy on the Antioxidative Activities (야생산사(Crataegus pinnatifida Bunge)과육 용매별 추출물의 항산화 효과)

  • Haw(Crataegus pinnatifida Bunge) belongs to the rose family and has beneficial health effects such as anti-obesity and gastroprotective activities. This study compared the antioxidative activities of various solvent extracts from haw. After removing seeds, pulpy was extracted with 70% methanol, 70% ethanol, chloroform :methanol(CM, 2:1, v/v), n-butanol, and ethyl acetate(EA). Total phenol contents were 71.46, 70.41, 47.63, 47.49 and 45.95 mg/g in the 70% methanol, 70% ethanol, CM, n-butanol and EA extracts, respectively. Total flavonoid contents of CM and n-butanol extracts were higher compared to other extracts at 1.63 mg/g and 1.59 mg/g, respectively, whereas EA extract had the lowest flavonoid content at 1.12 mg/g. Corresponding to total phenol contents, among $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay, and superoxide dismutase(SOD)-like ability, 70% methanol and 70% ethanol extracts showed higher antioxidative activities than the other extracts. However, the lowest activities were n-butanol extract in $NO_2$ radical scavenging activity as well as EA extract in antioxidant activity by ${\beta}$-carotene bleaching assay and SOD-like ability. These results suggest that haw extracts are available as a new natural antioxidant, and its activities are attributed to antioxidant substances such as phenolic compounds and flavonoids.

Changes in Antioxidative Activity and Total Polyphenols of Crude and Defatted Grape Seed Extract by Extraction Condition and Storage (포도종자의 처리별 추출조건 및 저장에 따른 항산화활성 및 총페놀함량의 변화)

  • This study was conducted to determine the yield, free radical scavengering effect and total phenol contents of various solvent fractions on the crude and defatted grape seed extract during storage. The optimal condition for the extraction yield, free radical scavengering effect and total phenol contents was $90\%$ ethanol for 6 hour at $70^{\circ}C$. The extraction yield for crude and defatted grape seed at optimal condition was $8.9\%\;and\;9.16\%$, respectively. Also, the strongest free radical scavengering effect with $41.52\;{\mu}g/mL$ was observed in $95\%$ ethanol of defatted grape seed extracted for 6 hour at $70^{\circ}C$. Similar result was observed in total phenol contents of defatted grape seed. The ethyl acetate fraction obtained from ethanol extract of defatted grape seed showed the strongest RC50($12.35\;{\mu}g/mL$) compared to other organic fractions. Free radical scavengering effect of crude and defatted grape seed extracts treated with alkali condition(pH 10) was reduced compared to that of acidic condition(pH 2) during storage far 1 month at $50^{\circ}C$. Overall, more stronger free radical scavengering effect and higher total phenol contents in defatted grape seed extracts was observed than that of crude grape seed.

Antioxidant Activity of Extracts from Akebia quinata Decne

  • Antioxidant activity of Akebia quinata Decne was evaluated. Water extract (0.5 g/50 mL) of flowers and leaves of A. quinata were prepared and total phenol contents and radical scavenging activity of the extracts was determined for antioxidant activity. The total phenol contents of extracts from A. quinata flowers (FAQ) and leaves (LAQ) were $30.05{\mu}M\;and\;20.23{\mu}M$, while the radical scavenging activity of FAQ and LAQ were 60.51 % and 52.97%, respectively. In addition, the effect of FAQ and LAQ extract on DNA damage induced by $H_2O_2$ in human lymphocytes was evaluated by comet assay. The FAQ and LAQ showed strong inhibitory effect against DNA damage induced by $200{\mu}M$ of $H_2O_2$. These results suggest that water extracts of A. quinata Decne flowers and leaves showed significant (p<0.05) antioxidant activity and protective effect against oxidative DNA damage.

Browning Degree of Various Apple Cultivars for Minimal Processing (최소 가공용 사과의 품종별 갈변정도)

  • This study was carried out to select apple cultivars suited for the fresh cutting or minimal processing as investigating correlation between total phenol and ascorbic acid as browning factors and the enzymatic browning degree. In soluble solid-acid ratio, 'Gamhong' and 'Arisoo' were highest value of 71.69 and 71.52 compared with other cultivars. And the change of lightness (${\Delta}L$) and color (${\Delta}E$) in 'Gamhong' and 'Arisoo' showed lower than other cultivars while 'Arkansas black' was highest change of lightness and color as ${\Delta}L$ 8.27, ${\Delta}E$ 13.86 respectively. Total phenol and ascorbic acid contents in 'Arkansas black' were highest such as 334.3 mg GAE/100 g and 9.22 mg/100 g, respectively. And polyphenol oxidase (PPO) activity also was the highest such as 14.43unit in 'Arkansas black'. The correlation test showed browning of apple was significantly dependent on ${\Delta}L$, ${\Delta}E$, total phenol, ascorbic acid contents and PPO activity (p<0.01). On the contrary, no significant correlation had been determined between the browning and ascorbic acid oxidase (AAO) activity. Thus, it seemed that the browning of apple was not only due to oxidation of ascorbic acid but also total phenol. It was suggested that 'Gamhong' and 'Arisoo' were suitable for fresh cutting or minimal processing.
