생쥐 흉선세포의 증식과 생존력에 미치는 인삼 사포닌의 영향 (Effect of Ginseng Saponin on the Proliferation and Viability of Murine Thymocyte, in vitro)

  • 면역 작용에 중요한 역할을 하는 림프구의 일종인 T세포가 성장 및 증식하는 기관인 흉선의 흉선세포증식과 생존력에 미치는 인삼 saponin의 영향과, 흉선 세포의 증식에 관여하는 Interleukin2(IL2)에 미치는 인삼saponin의 영향과,IL2의 생성에 영향을 미치는 Adenosin Deaminase(ADA)에 대한 인삼 saponin의 영향 등을 알아보기 위하여 생쥐의 흉선세포를 사용하여 다음과 같은 결과를 얻었다. 여러 농도의 인삼 saponin에서 배양한 전체 흉선세포수와 살아있는 흉선세포수의 변화는 $10^{-5}$%농도에서 대조군보다 증가하였으나 $10^{-1}$%농도군에서는 감소되었다. 여러 농도의 IL2에 인삼 saponin$10^{-5}$%농도를 첨가하여 배양한 전체 흉선세포수는 1.5%,3%및 6%농도군에서 9시간 또는 12시간배양시 증가하였으며,6% 실험군은 24시간과 48시간 배양시에도 유의성있게 증가하였다. 그러나 살아있는 흉선세포수는 대조군과 별차이가 없었다. 마지막으로 흉선세포의 ADA활성도에 미치는 인삼 saponin의 영향은 $10^{-5}$%농도에서 25%의 유의성있는 활성 증가가 일어났다.

  • A human hepatoma cell line, hep G2, was used to investigate the mechanism of serum cholesterol reduction by ginseng total saponin, ginsenoside-$Rb_1$, - $Rb_2$, and non-saponin fraction (ether extraction). Hep G2 cells were incubated in 10 $\mu\textrm{g}$/ml of cholesterol containing serum free-RPMl1640 medium with various concentration of ginseng components. The amounts of cholesterol in Hep G2 cells were decreased to maximum 51% in total saponin or two ginsenoside-treated groups while there was 137% increase in cholesterol level of control group as compared with that of normal group. Nonsaponin groups did not show the same effect. In order to elucidate the observed changes in the amount of cholesterol, the activity of amyl CoA : cholesterol acyltransferase (ACAT) in groups showing remarkable reduction in cholesterol amount, i.e., total saponin 10-6%, ginsenoside-$Rb_1$ $10^{-4}$%, ginsenoside-$Rb_2$, $10^{-4}$%, and non-saponin fraction $10^{-4}$%, was assayed using [1-$^{-14}C$%]oleic acid as enzyme substrate. The activity of ACAT was increased in all groups tested as compared with that of control group except for non-saponin group cultured in water soluble cholesterol containing medium. The serum cholesterol lowering effects of ginseng components can partially be attributed to the increased hepatocellular ACAT activity.

인삼 조사포닌의 조제 방법 개선 (Improved Method for the Preparation of Crude Ginseng Saponin)

  • This stuffy was carried to establish a new efficient method for the preparation of edible crude ginseng saponin. The conventional butanol extraction and resin adsorption methods were compared for the contents of total crude ginseng saponin and major ginsenosides. Seventy- percent methanol extract was applied to Diaion HP-20 column and the resin was washed with Hn and eluted with absolute methanol. The methanol elute was dried in vivo and analyzed for its ginsenosides. Use of ethanol instead of methanol to make edible crude ginseng saponin gave a similar result. Butanol extraction was performed by the conventional method. The final aqueous layer from butanol extraction was passed through Diaion HP-20 column followed by elution with methanol and Diaion HP-20 passed fraction was extracted with butanol to recover remaining components, respectively, in order to determine saponin loss. TLC and HPLC qualitatively and quantitatively monitored Ginsenosides, respectively. Loss of ginsenosides was higher in butanol extraction method than in Diction HP-20 adsorption method. In addition, saponin fractions prepared by Diction HP-20 adsorption method showed higher content of each ginsenoside, showing 8.2% higher purity than that of butanol extracted fraction. From these results, we propose the resin adsorption method as a new efficient measure for the preparation of crude ginseng saponin, which is edible by using spirit instead of methanol.

인삼의 총사포닌, Ginsenoside-$Rb_1$, Ginsenoside-$Rb_2$와 Lovastatinul 의한 Hep G2 세포의 HMG CoA Reductase 및 LDL수용체 mRNA 발현 유발효과의 비교 (Comparison of the Effects of Ginseng Total Saponin, Ginsenoside-$Rb_1$, Ginsenoside-$Rb_2$ and Lovastatin on the Expression of mRNAs for HMG CoA reductase and LDL Receptor)

  • The effects of ginseng total saponin, ginsenoside-Rb, and -Rb, on the reduction of chmlesterol level and the myNA expression rates of HMG CoA reductase and LDL receptor in Hep G2 were investigated and compared with that of lovastatin, a competitive HMG CoA reductase Inhibitor. The amounts of cholesterol in Hep G2 decreased in total saponin-and ginsenoside-treated groups as compared with that of control group, while there was no significant reduction in lovastatin-treated group. The mRNA expression rates of HMG CoA reductase increased in total saponin and gin- senoside groups except for ginsenoside-Rb, (10-3%) group and decreased in lovastatin group com- pared with that of control group. The mRNA expression rates of LDL receptor generally increased In all of the test groups except for total saponin (10-5%) group compared with that of control group. Because the ginseng components tested were more effective in the reduction of cholesterol level in Hep G2 than lovastatin and induced the gene expression of LDL receptor, we suggest the possibility that they could be used as a replacement agent for lovastatin which can not be prescribed especially to patients with hepatic diseases.

홍삼 사포닌이 수태중인 흰쥐의 항산화 효소활성에 미치는 영향 (Effect of Tota1 Saponin from Red Ginseng on Acvtivities of Antioxidant Enzymes in Pregnant Rats)

  • 수태는 많은 신체적인 기능들의 높은 에너지 요구 및 산소요구량 증가로 인하여 생리적인 변화를 수반한다. 때문에 산소 섭취량 및 이용이 증가하여 산화적인 스트레스의 증가를 기대할 수 있다. 수태중에 발생되는 free radical에 대하여 홍삼사포닌 투여가 간 항산화효소의 활성에 미치는 영향을 연구하였다. 수태중에 superoxide dismutase(SOD)의 활성은 전반적으로 감소하는 경향을 나타냈으며, 사포닌 투여군은 대조군에 비하여 유의한 변화가 관찰되지 않았다. 그리고 glutathione peroxidase(GPX) , gluthatione reductase (GRD)와 catalase의 활성도는 수태중에 감소하는 경향을 나타냈으며, 반면에 사포닌 투여군은 대조군 비하여 GRD 및 catalase의 활성도에 유의한 변화를 나타냈다. 사포닌 투여군의 GPX의 활성도는 대조군에 비하여 감소하는 경향이 다소 적게 나타났다. 수태중 대조군의 glutathione-S-transferase (GST) 활성도는 항상성을 유지하기 위해 증가하는 경향을 나타냈으며, 사포닌 투여군도 대조군에 비하여 이 효소의 활성도가 더욱 증가하는 경향을 나타냈다. 분만 후 대조군 및 사포닌 투여군은 정상수준으로 회복되었다. 수태와 같은 생체변화에도 불구하고 수태한 흰쥐의 간 항산화 효소활성에 대한 사포닌의 영향은 산소독성에 대하여 모체간을 보호해주는 생리적 항상성의 적응메카니즘에 의하여 조절되는 것으로 보인다.

  • The effects of total ginseng saponin. extracts of Panax ginseng C.A. Meyer, on the differentiation of F9 teratocarcinoma stem cells were studied. F9 stem cells cultured in the presence of ginseng saponin together with dibutyric cAMP became parietal endoderm - like cells. Moreover, the expressions of differentiation marker genes. laminin. type IV collagen. and retinoic acid $receptor-{\beta}(RAR{\beta})$ were increased after treatment of ginseng saponin. Among various ginsenosides purified from crude ginseng saponin, $Rh_1\;and\;Rh_2$ caused the differentiation of F9 cells most effectively. Since ginsenosides and steroid hormone show resemblance in chemical structure. we studied the possibility of the involvement of a steroid receptor in the differentiation process induced by ginsenosides. According to Southwestern blot analysis, a 94 kDa protein regarding as a steroid receptor was detected in F9 cells cultured in the medium containing ginseng saponin. Based on these data, we suggest that ginseng saponin, especially ginsenosides $Rh_1\;and\;Rh_2$ cause the differentiation of F9 cells and the effects of ginsenosides might be exerted via binding with a steroid receptor or its analogous nuclear receptor.

인삼 사포닌 분획이 세포벽에 미치는 영향 (A Study on The Effect of Ginseng Saponin Fraction on Cell Wall)

  • In this experiment, observations were made on the effects of ginseng saponin, one of the major components of Korean ginseng (Panax ginseng, C. A. Meyer) root, on the membranes of microorganism (E. coli K-12), the concentration of intracelluar and extracellular cycle AMP therein, and uptake of U-14C-glucose. When the E. coli were grown on media containing 0.1% ginseng saponin, the growth was faster than for that of the control by about 30 minutes. The lysis of E. coli grown on the ginseng saponin medium increased to about double that of the control in the stationary phase. And the amount of protein and lipopolysaccharides in the outer cell meberances increased 25% and 80% respectively in comparison with the control. By electron microscope observation, it was shown that the periplasmic region of the E. coli grown on the ginseng saponin medium was widened it was observed that the cellular cyclic AMP content of the E. coli increased significantly to the hightest levels between the late exponential phase and early stationary phase. The total cyclic AMP content of E. coli grown on the ginseng saponin medium decreased about 50% when compared to that of the control.

  • 인삼의 잎, 줄기, 뿌리 절편에 Agrobatriumrhizogenes(stain $A_4$를 접종하여 형질이 전환된 조직(모상근)을 유도, 배양한 후 saponin 고생산능 clones을 선발하였으며, 아울러 형질전환 여부를 dot blot hybridization 과 opine 분석으로 확인하였다. 여러 부위에서 유도된 모상근은 모두 암조건의 MS 배지에서 활발히 생장하였으며, 이들 중 HB3 clone을 이용하여 동질화딘 모상근 clon으로 배양한 후 saponin 고생산능 clon을 선발한 결과 HB2-10 clone이 최대의 생장을 보였으며 총 saponin함량은 0.55wt%로 나타났다. 그러나 총 saponin함량이 가장 높은 clone은 비교적 생장이 느린 HB3-2 clone이었으며 0.74wt%의 함량을 나타내었다. 아울러 dot blot hybridization 결과 Ri-T-DNA가 식물체의genome 내로 삽입되어 있었으며, opine확인결과 모든 모상근 clone으로부터 argropine과 mannopine이 검출되었다.

진세노사이드 Rd와 사포닌 대사물인 compound K의 항지질과산화 효과 (Anti-lipid Peroxdation Effect of Ginsenoside Rd and Its Metabolite Compound K)

  • To study on antioxidant effects in the liver of 40-week-old mouse, the sample were orally pretreated 5mg/kg/day for 5 days with red ginseng saponin components(total saponin, protopanaxadiol saponin, protopanaxatriol saponin, ginsenoside-Rd, ginsenoside-Re, compound-K) for 5 days. The ability of saponin to protect the mouse liver from oxidative damage was examined by determining the activity of superoxide dismutase(SOD), glutathione peroxidase(GPx) and the contents of glutathione, the level of malondialdehyde, The only protopanaxadiol among the ginseng saponin fractions was significantly increased the hepatic SOD activity(p<0.01). The red ginseng saponin induced a slight increase of GPx activity, especially ginsenoside Rd, compound K and protopanaxatriol treatments significantly increased its activity. The content of glutathione was significantly increased by total saponin, protopanaxadiol and ginsenoside Rd(p<0.01), but the oxidized glutathione level was lowered in all the red ginseng saponin. Finally, the level of malondialdehyde was significantly decreased by ginsenoside Rd and protopanaxadiol. In conclusion, protopanaxadiol and ginsenoside Rd among the saponin fraction were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation. This comprehensive antioxidant effects of red ginseng saponin seems to be by a certain action of saponin other than a direct antioxidant action.

Ginsenoside Changes in Red Ginseng Manufactured by Acid Impregnation Treatment

  • To enhance the functionalities of ginseng, an acid impregnation pre-treatment was applied during red ginseng processing. Acetic, ascorbic, citric, malic, lactic, and oxalic acid were used for the acid impregnation treatment, and total and crude saponin concentrations and ginsenoside patterns were evaluated. Total and crude saponin contents of red ginseng pre-treated by acetic, ascorbic, and citric acid were similar to those of red ginseng without pre-treatment, whereas lactic, malic, and oxalic acid pre-treatment caused a reduction of total and crude saponin in red ginseng. From the high performance liquid chromatography analysis of ginsenosides, increased $Rg_3$ density was shown in red ginseng pre-treated by acetic, ascorbic, and citric acid impregnation. In the case of lactic, malic, and oxalic acid pre-treatment, increased $Rg_1$ density was observed in red ginseng. Increased $Rg_1$ and $Rg_3$ contents due to acid impregnation during red ginseng processing may contribute to improving bioactive functionalities of red ginseng.
