Effects of amino acid composition in pig diet on odorous compounds and microbial characteristics of swine excreta

  • Background: Major amino acids in pig diets are Lys, Met, Thr, and Trp, but little is known about the requirements for the other essential amino acids, especially on odorous compounds and microbial characteristics in feces of growing-finishing pigs. To this end, different levels of amino acid composition added to diets to investigate the effects of amino acid composition on microbial characteristics and odorous compounds concentration. Methods: A total eight (n = 8) barrows (Landrace ${\times}$ Yorkshire ${\times}$ Duroc) with an average bodyweight of $89.38{\pm}3.3kg$ were individually fed diets formulated by Korean Feeding Standards 2007 (old version) or 2012 (updated with ideal protein concept) in metabolism crates with two replication. After 15-day adaptation period, fresh faecal samples were collected directly from pigs every week for 4 weeks and analysed for total volatile fatty acids (VFA), phenols and indoles by using gas chromatography. The nitrogen was determined by Kjeldahl method. Bacterial communities were detected by using a 454 FLX titanium pyrosequencing system. Results: Level of VFA tended to be greater in 2012 than 2007 group. Among VFAs, 2012 group had greater (p < 0.05) level of short chain fatty acids (SCFA) than control.Concentration of odorous compounds in feces was also affected by amino acid composition in pig diet. Levels of ammonium and indoles tended to be higher in 2012 group when compared with 2007 group.Concentration of phenols, p-cresol, biochemical oxygen demand, and total Kjeldahl nitrogen, however, were lower (P < 0.05) in 2012 treatment group compare to 2007. The proportion of Firmicute phylum were decreased, while the Bacteriodetes phylum proportion increased and bacterial genera includingCoprococcus, Bacillus, and Bacteroides increased (p < 0.05) in 2012 compare to 2007 group. Conclusion: Results from our current study indicates that well balanced amino acid composition reduces odor by modulating the gut microbial community. Administration of pig diet formulated with the ideal protein concept may help improve gut fermentation as well as reduce the odor causing compounds in pig manure.

Effects of Volatile Fatty Acids on IGF-I, IGFBP-3, GH, Insulin and Glucagon in Plasma, and IGF-I and IGFBP-3 in Different Tissues of Growing Sheep Nourished by Total Intragastric Infusions

  • Twelve Suffolk${\times}$Small-tail-Han male sheep (body weight 21-26 kg), aged four months, were used to study the effects of volatile fatty acids (VFA) on IGF-I (insulin-like growth factor-I), IGFBP-3 (insulin-like growth factor binding protein-3), GH (growth hormone), insulin and glucagon in plasma, and IGF-I and IGFBP-3 in different tissues. The sheep were randomly divided into four groups with 3 sheep in each group. The sheep were sustained by total intragastric infusions and four levels of mixed VFA (the molar proportion of acetic acid, propionic acid and butyric acid was 65:25:10), which supplied 333, 378, 423 and 468 KJ energy/kg $W^{0.75}$/d, were infused into the rumen as experimental Treatments I, II, III and IV, respectively. The experiment lasted 12 days, of which the first 8 days were for pretreatment and the last 4 days for collection of samples. At the end of the experiment, blood samples were taken and then the sheep were slaughtered and tissue samples from the rumen ventral sac, rumen dorsal sac, liver, duodenum and Longissimus dorsi muscle were obtained. IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in different tissues were analysed. Results showed that the concentration of IGF-I, IGFBP-3, GH, insulin or glucagon in plasma and the content of IGF-I and IGFBP-3 in the rumen dorsal sac, rumen ventral sac, liver or Longissimus dorsi muscle were increased with VFA infusion level (p<0.05). No significant differences were found in duodenum IGF-I between Treatments I and II and in rumen dorsal sac IGFBP-3 between Treatments II and III (p>0.05). It was concluded that IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in rumen dorsal sac, rumen ventral sac, liver and Longissimus dorsi muscle were increased significantly with increasing level of ruminal infusion of mixed VFA.

헛개나무잎차의 이화학적 특성 (Physiocochemical Properties of Hovenia dulcis Thunb. Leaf Tea)

  • In order to promote the utilization of Hovenia dulcis leaf as food, leaf tea was nanufactured and its physicochemical properties were examined. Aong proximate composiotns of Hovenia dulcis leaf tea were found in both fermented tea and roasted tea the higher contents of total sugar and crude protein. In minerals component of fermented tea and roasted tea, potassium (K) was the most high amounts of 11,560.6 ppm and 11,084.6 ppm, respectively. The highest contents of free sugar in both teas were mainly consisted of sucrose, the amounts of sucrose showed 1.47% and 1.94% . In case of organic acids, oxalic acid and citric acid were revealed 631.26mg% and 660.05mg%, respectively. The highest contents of total amino acid in fermented tea and roasted tea were 812.01mg% and 709.72mg% of glutamic acid. the fatty acids of fermented tea and roasted tea were mainly composed of 40.18% of linolenic acid and 26.92% of palmitic acid, respectively. The major volatile compounds of fermented tea and roasted tea were composed of 3, 7, 11, 15-tertramethyl-2-hexadecen-1-ol and methyl palmitate and the amounts of those showed 290.1ppm and 472.97 ppm, respectively. The tannins of fermented tea and roasted tea were 1.02% and 1.26% , as the extraction steps increased the tannin contents decreased. In Hunter's color values of tea extract L and b values decreased , while a value of those increased as the extraction steps were repeated.

Chemical Composition of Petals of Chrysanthemum spp.

  • Chemical compositions of petals of Chrysanthemum spp. were determined to renew its interest as a food material. The moisture contents of wild Chrysanthemum boreale and holticulturized Chrysanthemum morifolium were 11.7% and 11.0%. The proximate chemical ompositions were 6.1% and 11.7% in protein, 3.7% and 5.2% in ash, 11.1% and 12.8% in reducing sugar, of C. boreale and C. morifolium, respectively. Free sugars were mainly composed of fructose, glucose and sucrose. In fatty acid composition, the ratios of saturated to unsaturated fatty acids were 1.2:1 and 3.0:1, respectively. The amino acids determined were found 0.18 and 0.26 in the ratio of essential to total amino acids in each sample. Major elements of Chrysanthemum petals were Ca, K, Fe, Mg and Na, but the contents of Ca and Fe were more than twice higher in C. boreale than c. morifolium, Major volatile flavor components were 28.22% of epi-bicyclophellandrene and 24.55% of camphor in C. boreale, and 14.24% of 4-methyl-1-(10methylethyl)-3-cyclhexen-1-ol, 10.74% of camphor and 7.64% of 1,8-cineol in C. morifolium.

Effect of Panax ginseng Extract on Growth Responses of Human Intestinal Bacteria and Bacterial Metabolism

  • The growth responses of a variety of human Intestinal bacteria to extracts of Pun(1.vKy'n.ieny and five other oriental medicinal Araliaceae were evaluated in vitro and in vivo. The extracts enhanced the growth of Bifidobncterilim breve and B. longum in Media with or without carbon sources, suggesting the bifid factor (5) might be involved in the phenomenon. This effect was most pronounced with water extract of p. ginseng, the growth of 27 bifidobacteria strains belonging to B. ndolexcentium, H. longlrm, and 1. breve and B. iniuntis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and 5.fcherirhia coli had little or no ability to utilizes it (or growth. Methanol extracts of p, ginseng were found to selectively inhibit growth of various clostridia including C. perfringens and C. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake(600 mg/day for two weeks) on the fecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of fecal microflora including Bri'idobucterilim app. during the period of ginseng extract intake was significantly unaffected from the proceeding and sub sequent control periods. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The fecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobacterium spp. Other biochemical properties in faces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the pharmacological actions of P ginseng as an adaptogen.

Influence of Temperature and pH on Fermentation Pattern and Methane Production in the Rumen Simulating Fermenter (RUSITEC)

  • An experiment was conducted to study the effect of temperature and pH on in vitro nutrient degradability, volatile fatty acid profile and methane production. The fermenter used was the semi-continuous system, known as the rumen simulation technique (RUSITEC). Sixteen cylinders were used at one time with a volume of 800 ml, the dilution rate was set at 3.5%/hour, the infused buffer being McDougall's artificial saliva. Basal diet (9.6 g DM) used in RUSITEC consisted of (DM) 6.40 g Timothy hay, 1.86 g crushed corn and 1.34 g soybean meal. The food for the fermentation vessel was provided in nylon bags, which were gently agitated in the liquid phase. The experiment lasted for 17 d with all the samples taken during the last 5 d. Treatments were allocated at random to four vessels each and were (1) two temperature levels of $39^{\circ}C$ and $41^{\circ}C$ (2) two pH levels of 6.0 and 7.0. The total diet contained ($g\;kg^{-1}$ DM) 957 OM, 115 CP and $167MJ\;kg^{-1}$ (DM) GE. Although increase in temperature from $39^{\circ}C$ to $41^{\circ}C$ reduced degradation of major nutrients in vitro, it was non-significant. Interaction effect of temperature with pH also reflected a similar trend. However, pH showed a significant (p<0.05) negative effect on the degradability of all the nutrients in vitro. Altering the in vitro pH from 7 to 6 caused marked reduction in DMD from 60.2 to 41.8, CPD from 76.3 to 55.3 and GED from 55.3 to 35.1, respectively. Low pH (6) depressed total VFA production (61.9 vs. 34.9 mM) as well as acetate to propionate ratio in vitro (from 2.0 to 1.5) when compared to pH 7. Compared to pH 7, total gas production decreased from 1,841 ml to 1,148 ml at pH 6, $CO_2$ and $CH_4$ production also reduced from 639 to 260 ml and 138 to 45 ml, respectively. This study supported the premise that pH is one of the principal factors affecting the microbial production of volatile fatty acids and gas. Regulating the ruminal pH to increase bacterial activity may be one of the methods to optimize VFA production, reduce methane and, possibly, improve animal performance.

Emissions of Volatile Organic Compounds from a Swine Shed

  • The concentrations and chemical compositions of volatile organic compounds (VOCs), including volatile fatty acids, phenols, indoles, aldehydes, and ketones, which are the main organic compounds generated by swine, were investigated in July and October 2016 and January 2017. In addition, the emission rates and annual emissions of these components from the swine shed were estimated. The concentrations of VOCs in the swine shed averaged $511.3{\mu}g\;m^{-3}$ in summer, $315.5{\mu}g\;m^{-3}$ in fall and $218.6{\mu}g\;m^{-3}$ in winter. Acetone, acetic acid, propionic acid, and butyric acid were the predominant components of the VOCs, accounting for 80-88% of the total VOCs. The hourly variations of VOC concentrations in the swine shed in fall and winter suggest that the VOC concentrations were related to the ventilation rate of the swine shed, the activity of the swine, and the temperature in the swine shed. Accordingly, the emission rates of VOCs from the swine shed were $1-2{\times}10^3{\mu}g(h\;kg-swine)^{-1}$.

Effect of Feeding Bypass Protein on Rumen Fermentation Profile of Crossbred Cows

  • The effect of three varying ratios (high, medium and low) of Rumen Degradable Protein (RDP) to Undegradable Dietary Protein (UDP) of 37:63, 52:48 and 70:30 in iso-nitrogenous and iso-caloric concentrate mixtures on rumen fermentation profile was studied using rumen fistulated Jersey crossbred cows. Rumen pH and ammonia nitrogen concentration were found to be lower with a concentrate mixture containing a higher UDP level of 63.38% when compared with those having medium and low UDP levels of 47.55 and 29.75%, respectively, at all post feeding intervals. Total volatile fatty acid concentration as well as concentrations of individual fatty acids viz., acetate, propionate and butyrate were also found higher in animals fed concentrate mixture with the highest UDP level.

볏짚의 가수분해과정중 유기산 생성 특성 (Characteristics of Volatile Fatty Acids Release During the Hydrolysis of Rice Straw)

  • 농업 바이오매스를 혐기소화 시스템에 투입 가능성 여부를 구명하기 위해, 생산량이 가장 많은 볏짚을 원료로 하여 전처리에 따른 볏짚의 가수분해과정에서 휘발성유기산(VFAs)의 변화 특성을 조사하였다. 전처리는 3 cm 절단과 분쇄, $35^{\circ}C$$55^{\circ}C$, 알카리 처리, 교반속도 등을 조사하였다. 총 휘발성유기산(TVFAs)의 농도는 가수분해 시간이 경과하면서 증가하였다. 볏짚의 크기에서는 분쇄한 시료가 3cm 절단 시료에 비해 분쇄한 시료에서 TVFAs의 농도가 높게 나타났으며, 알칼리를 처리한 시료는 처리전에 비해 3배 증가하는 것으로 나타났다. 교반속도별로는 온도에 따라 다르게 나타났는데 $35^{\circ}C$에서는 150 rpm이, $55^{\circ}C$에서는 80rpm과 200 rpm에서 TVFAs의 농도가 높게 관측되었다. 전체적으로는 TVFAs중에서 acetic acid가 대부분이었으며, TVFAs와 유사한 증감경향을 보였다. 섬유소의 변화는 셀룰로스와 헤미셀룰로스가 약간 감소하였으나, 리그닌은 변화가 없는 것으로 나타났다.
