  • This experiment was designed to evaluate the effects of the processing method of common vetch seed (CVS) (Vicia sativa) on laying performance, egg quality, metabolic parameters and liver histopatology during the peak production period in hens. Lohman layers, 46 wk of age in 6 replicate cages each containing 4 hens, were allocated randomly to one of four dietary treatments. Diets were control (C) diet containing no common vetch and experimental diets containing 25% raw common vetch (RCV), 25% soaked in water for 72 h with exchange of water every 24 h (SCV) and 25% soaked&boiled at $100^{\circ}C$ for 30 minute common vetch (SBCV). Inclusion of RCV into the diet deteriorated all laying performance variables. SCV did not alleviate the adverse effect of raw common vetch on feed intake, egg weight, feed conversion, final weight and weight change. SCV partially alleviated egg production (p<0.001). SBCV diminished the adverse effect on feed intake, egg weight, feed conversion, final weight and weight change compared to raw vicia sativa (p<0.001). No significant difference was detected between SBCV and the control group in terms of egg production, feed conversion, final weight and weight change. Regardless of the processing method, all the common vetch groups had lower shell strength compared to the control group. Haugh units did differ between all groups (p<0.001). Inclusion of RCV and SCV into the basal diet decreased triglyceride, cholesterol, total protein and serum glucose concentrations (p<0.001). Hovewer, inclusion of SBCV into the basal diet increased these parameters. Liver samples were stained with Hematoxylin and eosin (HE) and evaluated by light microscopy. A biopsy of native liver tissue was used as a control. No histopathologic finding was present in the control group. Raw V. sativa compared with the control caused lipid accumulations in hepatocytes, severe congestion of hepatic blood vessels, inflammation, increased numbers of Kupffer cells and sinusoidal dilatations. Whereas, the livers from groups given treated V. sativa showed only different degrees of sinusoidal dilatations. Findings from the present study point out the risk of increased hepatic damage due to use of raw Vicia sativa. Increasing treatment of V. sativa lead to a decrease of liver damages. Inclusion of raw and soaked vetch seeds in rations affected adversely all parameters examined in laying hens. But alleviation was observed when soaked and boiled vetch seeds (SBCV) were fed. The results of these experiments indicated that soaked&boiled Vicia sativa seeds may safely be used at a 25% level in rations of laying hens.

  • Background: There is a scarcity of information about the proportion of the adult Saudi population that meet the recommended guidelines of physical activity (PA) to reduce cancer risk. Moreover, their awareness about the role of PA in cancer prevention is unclear. Objectives: This cross-sectional study aimed at estimating the proportion of adult Saudis meeting the PA guidelines, specifically those recommended by American Cancer Society (ACS) for cancer prevention, and to assess the public awareness about the role of PA in cancer prevention. Materials and Methods: Using a multistage sampling method, 2,127 adult Saudis of both genders were recruited from 6 urban and 4 rural primary health care centers in Al Hassa, Saudi Arabia. Participants were personally interviewed to gather information about their sociodemographic characteristics, searching activity about PA and cancer, and the time spent in leisure time PA (moderate and vigorous)/week using the Global Physical Activity Questionnaire with show cards. Finally, items about the role of PA in cancer risk reduction were inquired. Results: Of the included participants, 11.6% met the recommendations for cancer prevention (${\geq}45$ minutes of moderate-vigorous PA activity/${\geq}5$ days/week or 225 minutes/week). Multivariate regression showed that being male (AOR=1.49, CI=1.09-2.06), <20 years of age (AOR=3.11, CI=2.03-4.76), and unemployed (AOR=2.22, CI=1.57-3.18) were significant predictors for meeting PA recommendations for cancer prevention. Only 11.4% of the sample indicated correctly the frequency and duration of PA required for an average adult to be physically active and while >70% of them indicated the role of PA in prevention of hypertension, coronary heart disease and lowering elevated blood cholesterol, only 18.6% and 21.7% correctly mentioned the role of PA in reducing colon and breast cancer risk, respectively. Poor knowledge was found among those with less than college education and aged ${\geq}50$ years. The level of knowledge was significantly positively correlated with total leisure time PA of the participants. Conclusions: A minority of adult Saudis in Al Hassa was aware about the role of PA in cancer prevention and engaged in sufficient LTPA for cancer risk reduction benefits, highlighting the need for public health actions to include policies and programs that address factors deterring their participation in LTPA and increasing their awareness with remedies to manage the prevalent misconceptions.

  • The objective was to investigate the hypoglycemic action of catalpol in spontaneous diabetes db/db mice. 40 db/db mice were randomly divided into five groups: model control gourp; db/db plus catalpol 40, 80, 120 mg/kg body wt. groups and db/db plus metformin 250 mg/kg group. Age-matched db/m mice were selected as normal control group. The mice were administered with corresponding drugs or solvent by gavage for 4 weeks. The oral glucose tolerance test was carried out at the end of $3^{rd}$ week. After 4 weeks of treatment, the concentrations of fasting blood glucose (FBG), glycated serum protein (GSP), insulin (INS), triglyceride (TG), total cholesterol (TC) and adiponection (APN) in serum were detected. The protein expressions of phosphorylation-$AMPK{\alpha}$1/2 in liver, phosphorylation-$AMPK{\alpha}$1/2 and glucose transporter-4 (GLUT-4) in skeletal muscle and adipose tissues were detected by western blot. Real time RT-PCR was used to detect the mRNA expressions of acetyl-CoA carboxylase (ACC) and Hydroxymethyl glutaric acid acyl CoA reductase (HMGCR) in liver. Our results showed that catalpol could significantly improve the insulin resistance, decrease the serum concentrations of INS, GSP, TG, and TC. The concentrations of APN in serum, the protein expression of phosphorylation-$AMPK{\alpha}$1/2 in liver, phosphorylation-$AMPK{\alpha}$1/2 and GLUT-4 in peripheral tissue were increased. Catalpol could also down regulate the mRNA expressions of ACC and HMGCR in liver. In conclusion, catalpol ameliorates diabetes in db/db mice. It has benefit effects against lipid/glucose metabolism disorder and insulin resistance. The mechanism may be related to up-regulating the expression of phosphorylation-$AMPK{\alpha}$1/2.

  • For the elucidation of the metabolic effects due to alcoholic drink and moldy rice intake author designed two experiments as follows. In the first experiment, sixty male and female rats, divided into six groups, 10 rats each in both sexes, were given 7.5% sucrose-30% alcohol solution (Jinro-Soju) and diets varied of fat content (2%, 7%, 30% of diet) and protein quality (casein, bean, anchovy) for 6 weeks. In the Second experiment, six groups of rats, 10 male rats each, were fed the diets containing zero, 5%, 10%, 25%, 50%, and 100% moldy rice contaminated Penicillium Islandicum and the experimental periods were 4 weeks and 6 weeks. The results of these studies due to the toxicity of alcohol consumption and moldy rice ingestion of the fatty liver production and the other effects are as follows. 1. Food intake of alcohol consumed groups decreased to 50%-70% of that of standard group. In the second study, there was no significant differences on food intake due to the different contents of moldy rice and experimental period. 2. On the view paint of body weight gain, the body weight of alcohol consumed groups gained much less than that of standard group, because food consumption was decreased due to alcohol ingestion. 3. In comparison of F.E.R. and P.E.R. between standard and experimental groups, there was no significant differences in this study. 4. As a matter of fact, there was no significance on the nitrogen balance in both studies. 5. From the result of hematology, R.B.C. counts, W.B.C. counts, hemoglobin, and hematocrit showed normal values in all groups including in this study. So we might conclude that the toxicity of alcohol and moldy rice do not effect significantly on blood picture. 6. The larger organs shrank on the range of 20%-70% of that against standard group in this study. The major reason for the shrinkage of organs might be account of decreased food consumption due to alcoholic drink. There was no great changes on the organ weight due to the ingestion of moldy rice. 7. The nitrogen content in various organs in both experiments was revealed at the normal level for the worst condition in terms of the least food intake. In other word, it was noteworthy that the concentration of nitrogen in various organs was kept at the normal ratio as standard groups under the circumstances of this study. 8. The lipid content in the liver of rats fed alcohol and diets containing either various lipid contents or protein quality did not increase. Hepatic lipids accumulation due to the dietary fat content was observed, but there wvas no significances among the compared groups. In the second experiment, the difference of hepatic lipid content between the moldy rice groups and standard group was not showed. In addition to the result of total lipid, hepatic free cholesterol, free fatty acid, and triglyceride did not change in both studies, we might conclude that the toxicity of alcohol and moldy rice could not effect on the hepatic lipid contents. 9. There was no significant differences on the serum glucose level between alcohol groups and standard group. In the second experiment, serum glucose level increase in 6th week compared with in 4th week, but there was no significant differences.

  • Obesity, the world's leading metabolic disease, is a serious health problem in both industrialized and developing countries. Natural substances are of great interest in preventative medicine, especially in the field of metabolic syndromes-from insulin resistance to obesity and diabetes. In the present study, we investigated the effect of A. pullulans SM-2001 Extract (Polycan®) on the adipocyte differentiation of 3T3-L1 preadipocytes and the anti-obesity effect of 3T3-L1 adipocytes. Although β-glucan has been found to have health benefits in the regulation of the immune system and blood cholesterol levels, its role in obesity has not been fully investigated. Polycan® suppressed lipid accumulation and glycerol-3-phosphate dehydrogenase (GPDH) activity without affecting cell viability in 3T3-L1 preadipocytes and adipocytes. Polycan® also inhibited cellular lipid accumulation through down-regulation of transcription factors, such as PPARγ and C/EBPα, and induced dose-dependent phosphorylation of AMP-activated protein kinase (AMPK)-a cellular energy sensor-while the total AMPK protein content remained unchanged. Taken together, this shows that the activation of AMPK by Polycan® in adipocytes plays a critical role in Polycan®-induced inhibition of adipocyte differentiation. Our results show that Polycan® has an anti-obesity action in vitro, suggesting a potential novel preventative agent for obesity and other metabolic diseases.

  • The purpose of this study was to examine the effect of 8 weeks of aerobic exercise on serum lipids and kidney function in middle-aged T2DM patients. Subjects participated in aerobic exercise training during 8 wk. They started with an exercise intensity of $HR_{max}$ 60-75%, 20~45 minutes per day, 3~5 times a week. The results were compared to the baseline after 8 weeks. After the 8-week aerobic exercise routine, the body fat percentage and fasting glucose were significantly decreased, and the total cholesterol (TC) andtriglycerides (TG) were significantly decreased. Additionally, kidney function (Blood Urea Nitrogen (BUN], uric acids, and creatinine) was not significantly changed. In conclusion, a regular 8-week aerobic exercise training routine may improve serum lipids; however, kidney function (BUN, uric acids, and creatinine) did not change in middle-age type 2 diabetes mellitus (T2DM) and T2DM in diabetic nephropathy patients.

  • Forty-eight individually fed Angus steers (body weight $220kg{\pm}9.1$) were utilized to investigate the effects of copper (Cu) source and concentration on lipid metabolism and carcass quality. Steers were stratified by body weight and initial liver Cu concentration and randomly assigned to one of five groups. Groups were then randomly assigned to treatments. Treatments consisted of: 1) control (no supplemental Cu); 2) 10 mg Cu/kg DM from $CuSO_4$; 3) 10 mg Cu/kg DM from a Cu amino acid complex (Availa Cu) 4) 20 mg Cu/kg DM from $CuSO_4$; and 5) 20 mg Cu/kg DM from Availa Cu. Steers were fed a corn-alfalfa-based growing diet for 56 d. Steers were then switched to a high concentrate finishing diet for 145 d. On day 74 of the finishing phase subcutaneous adipose tissue biopsies were obtained from three steers/treatment to determine basal and stimulated lipolytic rates in vitro. Steers were then slaughtered after receiving the finishing diet for 145 d. Control steers tended (p<0.12) to have lower ceruloplasmin (Cp) activity than Cu supplemented steers. Steers receiving 20 mg Cu/kg DM from Availa Cu had higher (p<0.03) Cp activity than steers receiving 20 mg Cu/kg DM from $CuSO_4$. Plasma non-esterified fatty acids were similar across treatments. Steers receiving 10 mg Cu/kg DM from Availa Cu had higher (p<0.02) total plasma cholesterol concentrations relative to steers receiving 10 mg Cu/kg DM from $CuSO_4$. Steers receiving 20 mg Cu/kg DM from Availa Cu had lower (p<0.03) plasma triglyceride concentrations than steers supplemented with 20 mg Cu/kg DM from $CuSO_4$. Fatty acid profile of longissimus muscle was similar across treatments. Backfat depth tended (p<0.18) to be lower in Cu supplemented steers relative to controls. Steers supplemented with 20 mg Cu/kg DM from Availa Cu had heavier (p<0.03) hot carcass weights and a greater (p<0.02) dressing percentage than steers supplemented with 20 mg Cu/kg DM from $CuSO_4$. Furthermore, in vitro basal (p<0.06) and epinephrine stimulated (p<0.04) lipolytic rates of subcutaneous adipose tissue were higher in Cu supplemented steers relative to controls. The results of this study suggest that Cu supplementation has minimal effects on blood and lean tissue lipid profile. However, it appears that Cu may play a role in lipid metabolism in subcutaneous adipose tissue.

  • Objective: The experiments were carried out to measure the effect of multivitamin, acidifier and Azolla on growth performance, profitability and lipid profiles of blood of broiler chickens to produce safe and cost effective broilers. Methods: In experiment 1, 240 day-old Cobb-500 broiler chicks were fed diets; $D_1$ (control), $D_2$ ($D_1$ with 1 mL multivitamin/liter water), $D_3$ ($D_1$ with 1 mL acidifier/liter water), $D_4$ ($D_1$ with 1 mL multivitamin and 2 mL acidifier/liter water) having 3 replications in each, and 20 chicks/replication. In experiment 2, 150 day-old Cobb-500 broiler chicks were fed diets; $T_1$ (control), $T_2$ (5% Azolla in the diet), $T_3$ (7% Azolla in the diet) and $T_4$ ($T_1$ with 1 mL multivitamin and 1 mL acidifier/liter water) having 3 replications in each, and 20 chicks/replication in control, and 10 chicks/replication in the remaining dietary treatment groups for 35 days. Results: In experiment 1, the highest live weight was observed in $D_4$ (p<0.05), however, feed intake was statistically similar between diets (p>0.05). The lowest feed conversion ratio (FCR) (p<0.001) and mortality (p<0.05) were observed in $D_2$ followed by $D_4$, $D_1$, and $D_3$, respectively. There were no significant differences between diets for feed cost and net profit (p>0.05). However, evidently but not significantly, the highest net profit was obtained in $D_2$ followed by $D_4$, $D_1$, and $D_3$, respectively. In experiment 2, the highest live weight (p<0.05) and feed intake (p<0.001) were observed in $T_4$. Mortality (p<0.01), FCR (p<0.01), feed cost (p<0.05) and net profit (p<0.05) were significantly different among diets. Considering net profit, $T_2$ was the best performing dietary group followed by $T_3$, $T_1$, and $T_4$, respectively. The lowest lipid profiles were observed in $D_3$ followed by $D_1$, $D_4$, and $D_2$, respectively (p<0.05). In experiment 2, the lowest total cholesterol, TG, and the highest amount of high density lipoprotein were observed in $T_2$, followed by $T_3$, $T_1$, and $T_4$, respectively (p<0.05). Evidently but not significantly, low density lipoprotein was the highest in $T_2$ followed by $T_3$, $T_4$, and $T_1$, respectively (p>0.05). Conclusion: In conclusion, Azolla and acidifier reduced lipid profiles of broiler chickens. Considering net profit and lipid profiles, 5% Azolla may be the suitable dietary group for producing safe and profitable broilers. However, more studies are needed to confirm this study prior to suggesting using Azolla in the poultry industry.

  • A total of 80 piglets ($5.85{\pm}0.62kg$ BW; 21 d of age) were used to study the effect of carnitine addition to extruded full-fat soybean (EFS) diets on the growth of weaned pigs. Pigs were allotted into five treatments based on body weight, in a completely randomized block design. Each treatment has 4 replicates of 4 heads each. Treatments were 1) SBM (positive control), 2) EFS without carnitine (negative control), 3) EFS with 50 ppm carnitine, 4) EFS with 100 ppm carnitine and 5) EFS with 150 ppm carnitine. During d 0 to 14, piglets were fed diets containing 3,400 kcal ME, 23% crude protein, 1.65% lysine, 0.9% Ca and 0.8% P and for the period of d 15 to 28, piglets were fed diets supplying 3,300 kcal ME, 20% crude protein, 1.55% lysine, 0.9% Ca and 0.8% P. The urease activity of EFS (0.18) were three times higher than SBM (0.07). During d 0-14, pigs fed SBM had greater ADG and ADFI compared to pigs fed extruded full-fat soybean diets (p<0.05). Feed conversion ratio was not different among treatments. No linear or quadratic effect of carnitine addition was found in growth performance. During d 15-28, piglets fed SBM diet also showed better ADG and FCR with no significant differences among treatments. Feed intake tended to increase as carnitine addition level was increased (p=0.10). For overall period (d o to 28), the best performance was observed in pigs fed SBM diet. CP digestibility was higher in pigs fed SBM diet than piglets fed EFS diet at d 14, and DM and CP digestibility tended to be higher in pigs fed SBM diet at d 28. Blood metabolites (BUN, glucose and cholesterol)were not affected by treatments. In conclusion, based on the results of this study piglets at 21 d of age appeared to be not ready for extruded full-fat soybean (FFSB) in their diets. Piglets fed extruded FFSB showed decreased growth rate compared to piglets fed SBM diet. Nutrient utilization was also poor in piglets fed extruded FFSB diets. L-carnitine addition at the level of 50 to 150 ppm was not effective in improving the growth performance of pigs fed EFS diets.
