Comparative analysis of water surface spectral characteristics based on hyperspectral images for chlorophyll-a estimation in Namyang estuarine reservoir and Baekje weir (남양호와 백제보의 Chlorophyll-a 산정을 위한 초분광 영상기반 수체분광특성 비교 분석)

  • In this study, we estimated the concentration of chlorophyll-a (Chl-a) using hyperspectral water surface reflectance in an inland weir (Baekjae weir) and estuarine reservoir (Namyang Reservoir) for monitoring the occurrence of algae in freshwater in South Korea. The hyperspectral reflectance was measured by aircraft in Baekjae Weir (BJW) from 2016 to 2017, and a drone in Namyang Reservoir (NYR) from 2020 to 2021. The 30 reflectance bands (BJW: 400-530, 620-680, 710-730, 760-790 nm, NYR: 400-430, 655-680, 740-800 nm) that were highly related to Chl-a concentration were selected using permutation importance. Artificial neural network based Chl-a estimation model was developed using the selected reflectance in both water bodies. And the performance of the model was evaluated with the coefficient of determination (R2), the root mean square error (RMSE), and the mean absolute error (MAE). The performance evaluation results of the Chl-a estimation model for each watershed was R2: 0.63, 0.82, RMSE: 9.67, 6.99, and MAE: 11.25, 8.48, respectively. The developed Chl-a model of this study may be used as foundation tool for the optimal management of freshwater algal blooms in the future.

Monitoring trafficking and expression of hemagglutinin-tagged transient receptor potential melastatin 4 channel in mammalian cells

  • The TRPM4 gene encodes a Ca2+-activated monovalent cation channel called transient receptor potential melastatin 4 (TRPM4) that is expressed in various tissues. Dysregulation or abnormal expression of TRPM4 has been linked to a range of diseases. We introduced the hemagglutinin (HA) tag into the extracellular S6 loop of TRPM4, resulting in an HA-tagged version called TRPM4-HA. This TRPM4-HA was developed to investigate the purification, localization, and function of TRPM4 in different physiological and pathological conditions. TRPM4-HA was successfully expressed in the intact cell membrane and exhibited similar electrophysiological properties, such as the current-voltage relationship, rapid desensitization, and current size, compared to the wild-type TRPM4. The presence of the TRPM4 inhibitor 9-phenanthrol did not affect these properties. Furthermore, a wound-healing assay showed that TRPM4-HA induced cell proliferation and migration, similar to the native TRPM4. Co-expression of protein tyrosine phosphatase, non-receptor type 6 (PTPN6 or SHP1) with TRPM4-HA led to the translocation of TRPM4-HA to the cytosol. To investigate the interaction between PTPN6 and tyrosine residues of TRPM4 in enhancing channel activity, we generated four mutants in which tyrosine (Y) residues were substituted with phenylalanine (F) at the N-terminus of TRPM4. The YF mutants displayed properties and functions similar to TRPM4-HA, except for the Y256F mutant, which showed resistance to 9-phenanthrol, suggesting that Y256 may be involved in the binding site for 9-phenanthrol. Overall, the creation of HA-tagged TRPM4 provides researchers with a valuable tool to study the role of TRPM4 in different conditions and its potential interactions with other proteins, such as PTPN6.

  • In recent years, ransomware attacks have become more organized and specialized, with the sophistication of attacks targeting specific individuals or organizations using tactics such as social engineering, spear phishing, and even machine learning, some operating as business models. In order to effectively respond to this, various researches and solutions are being developed and operated to detect and prevent attacks before they cause serious damage. In particular, honeypots can be used to minimize the risk of attack on IT systems and networks, as well as act as an early warning and advanced security monitoring tool, but in cases where ransomware does not have priority access to the decoy file, or bypasses it completely. has a disadvantage that effective ransomware response is limited. In this paper, this honeypot is optimized for the user environment to create a reliable real-time dynamic honeypot file, minimizing the possibility of an attacker bypassing the honeypot, and increasing the detection rate by preventing the attacker from recognizing that it is a honeypot file. To this end, four models, including a basic data collection model for dynamic honeypot generation, were designed (basic data collection model / user-defined model / sample statistical model / experience accumulation model), and their validity was verified.

Discussion on the Classification of Floristic Target Species - An Ecological Paradox of Floristic Regions - (식물구계학적 특정종 분류에 관한 고찰 - 식물구계의 역리 -)

  • The floristic target species (FTS) has been widely used as a important tool for environmental assessment in Korea. Here the floristic district is a key criterion for the selection and evaluation of FTS. The aim of this study is to clarify the intellectual meaning of the floristic district i.e. a sort of phytogeographical subunit. We reviewed 7 main literatures related to the floristic district. Nakai's 1919 Ulleungdo flora survey report was first to mention a region and/or province in terms of general geographical divisions. In 1977, the floristic district map of the Korea vascular plants was first described by Oh. In 1978, Lee & Yim proposed a new floristic district map dividing the central and southern regions, and the district boundaries have been adjusted by many other researchers. In the end, the floristic district map modified slightly from Lee & Yim's map was nationally recognized in 2018. However, the boundaries of floristic districts were inconsistent, both in their setting and adjustment, as well as the indicator species of the district. No basis for this inconsistency could be confirmed. We concluded that all district maps depend on arbitrary lines drawn a priori, and finally proposed the following two ecological strategies to ensure the effectiveness and practicality of the FTS as a scientific means for environmental impact assessment and national flora evaluation: (i) designing to achieve the flora list and the floristic district based on nativeness of plant distribution, (ii) constructing a habitat-based floral monitoring system.

Development of Simulation for Estimating Growth Changes of Locally Managed European Beech Forests in the Eifel Region of Germany (독일 아이펠의 지역적 관리에 따른 유럽너도밤나무 숲의 생장변화 추정을 위한 시뮬레이션 개발)

  • Forest management is known to beneficially influence stand structure and wood production, yet quantitative understanding as well as an illustrative depiction of the effects of different management approaches on tree growth and stand dynamics are still scarce. Long-term management of beech forests must balance public interests with ecological aspects. Efficient forest management requires the reliable prediction of tree growth change. We aimed to develop a novel hybrid simulation approach, which realistically simulates short- as well as long-term effects of different forest management regimes commonly applied, but not limited, to German low mountain ranges, including near-natural forest management based on single-tree selection harvesting. The model basically consists of three modules for (a) natural seedling regeneration, (b) mortality adjustment, and (c) tree growth simulation. In our approach, an existing validated growth model was used to calculate single year tree growth, and expanded on by including in a newly developed simulation process using calibrated modules based on practical experience in forest management and advice from the local forest. We included the following different beech forest-management scenarios that are representative for German low mountain ranges to our simulation tool: (1) plantation, (2) continuous cover forestry, and (3) reserved forest. The simulation results show a robust consistency with expert knowledge as well as a great comparability with mid-term monitoring data, indicating a strong model performance. We successfully developed a hybrid simulation that realistically reflects different management strategies and tree growth in low mountain range. This study represents a basis for a new model calibration method, which has translational potential for further studies to develop reliable tailor-made models adjusted to local situations in beech forest management.

The Use of the Internal Transcribed Spacer Region for Phylogenetic Analysis of the Microsporidian Parasite Enterocytozoon hepatopenaei Infecting Whiteleg Shrimp (Penaeus vannamei) and for the Development of a Nested PCR as Its Diagnostic Tool

  • The increasing economic losses associated with growth retardation caused by Enterocytozoon hepatopenaei (EHP), a microsporidian parasite infecting penaeid shrimp, require effective monitoring. The internal transcribed spacer (ITS)-1 region, the non-coding region of ribosomal clusters between 18S and 5.8S rRNA genes, is widely used in phylogenetic studies due to its high variability. In this study, the ITS-1 region sequence (~600-bp) of EHP was first identified, and primers for a polymerase chain reaction (PCR) assay targeting that sequence were designed. A newly developed nested-PCR method successfully detected the EHP in various shrimp (Penaeus vannamei and P. monodon) and related samples, including water and feces collected from Indonesia, Thailand, South Korea, India, and Malaysia. The primers did not cross-react with other hosts and pathogens, and this PCR assay is more sensitive than existing PCR detection methods targeting the small subunit ribosomal RNA (SSU rRNA) and spore wall protein (SWP) genes. Phylogenetic analysis based on the ITS-1 sequences indicated that the Indonesian strain was distinct (86.2% nucleotide sequence identity) from other strains collected from Thailand and South Korea, and also showed the internal diversity among Thailand (N = 7, divided into four branches) and South Korean (N = 5, divided into two branches) samples. The results revealed the ability of the ITS-1 region to determine the genetic diversity of EHP from different geographical origins.

Development of evaluation components and criteria for the Korean Healthy Diet and assessment of the adherence status among Korean adults (한국인을 위한 건강식단 평가 항목 및 기준 개발과 준수 현황)

  • Purpose: This study developed the evaluation components and criteria for the Korean Healthy Diet (KHD) and assessed the current compliance of Koreans. Methods: The study reviewed domestic and international dietary guidelines and literature and conducted an online survey of 514 Korean adults to understand their nutritional perceptions, specifically the perceived importance of health and incorporation into usual diet. Data from the Korea National Health and Nutrition Examination Survey (KNHANES) were used to investigate food and nutrient intake patterns and examine the relationship between intake and metabolic syndrome (MetS). Based on these data, the components and criteria for a KHD were established by sex and age, and adherence was assessed. Results: The KHD evaluation included 13 dietary components: carbohydrates, sugar, fiber, protein, total fat, saturated fat, sodium, calcium, mixed grains, meat·fish·eggs·beans, vegetables, fruits, and dairy products. Applying the selected components and criteria to data from the KNHANES (2019-2021), the average KHD adherence score for Korean adults was 5.465 ± 0.023 out of a maximum score of 13. The score significantly increased with age (4.766 ± 0.044 for 19-29 years; 5.276±0.032 for 30-49 years; 6.109 ± 0.033 for 50-64 years), and women (5.642 ± 0.028) had higher scores than men (5.284 ± 0.030) (p < 0.05). Furthermore, the total score significantly differed between those with MetS (5.518 ± 0.045) and those without (5.568 ± 0.026) after adjusted for sex and age (p < 0.05). When scoring the dietary components, sugar (0.852 ± 0.004) and proteins (0.881 ± 0.004) scored relatively higher in the association with MetS, whereas calcium (0.148 ± 0.004) and mixed grains (0.225 ± 0.005) scored relatively lower. Conclusions: The KHD evaluation criteria could be used as a tool for screening and monitoring the overall diet quality of Koreans.

A Case Study of Software Architecture Design by Applying the Quality Attribute-Driven Design Method (품질속성 기반 설계방법을 적용한 소프트웨어 아키텍처 설계 사례연구)

  • in a software development, the design or architecture prior to implementing the software is essential for the success. This paper presents a case that we successfully designed a software architecture of radiation monitoring system (RMS) for HANARO research reactor currently operating in KAERI by applying the quality attribute-driven design method which is modified from the attribute-driven design (ADD) introduced by Bass[1]. The quality attribute-driven design method consists of following procedures: eliciting functionality and quality requirements of system as architecture drivers, selecting tactics to satisfy the drivers, determining architectures based on the tactics, and implementing and validating the architectures. The availability, maintainability, and interchangeability were elicited as duality requirements, hot-standby dual servers and weak-coupled modulization were selected as tactics, and client-server structure and object-oriented data processing structure were determined at architectures for the RMS. The architecture was implemented using Adroit which is a commercial off-the-shelf software tool and was validated based on performing the function-oriented testing. We found that the design method in this paper is an efficient method for a project which has constraints such as low budget and short period of development time. The architecture will be reused for the development of other RMS in KAERI. Further works are necessary to quantitatively evaluate the architecture.

On-line Monitoring of the Flocs in Mixing Zone using iPDA in the Drinking Water Treatment Plant (정수장 응집혼화공정에서의 응집플럭 연속 모니터링)

  • This study evaluated the flocs forming characteristics in the mixing zone to increase the coagulation effect in the drinking water plant. As a measuring tool of formed flocs, on-line particle dispersion analyzer (iPDA) was used in Y drinking water plant. To evaluate the forming flocs, many parameters such as poly amine, coagulant dosing amount, raw water turbidity, and pH was applied in this study. During the periods of field test, poly aluminium chloride (PACl) as a coagulant was used. With the increase of the raw water turbidities, poly amine was also added as one of aids for increasing in coagulation efficiency. The turbidity and pH of raw water was ranged from 7 to 9 and from 25 to 140 NTU, respectively. The increasing of raw water turbidity brought the bigger floc sizes accordingly. From a regression analysis, $R^2$ value was 0.8040 as a function of T, raw water turbidity. Floc size index (FSI) was obtained from a correlation equation as follows; FSI = 0.9388logT - 0.3214 Also, polyamine gave the bigger flocs the moment it is added to the coagulated water in the rapid mixing zone. One of parameters influencing the floc sizes was the addition of powdered active carbon(PAC) in the mixing zone. In case of higher turbidity of raw water, $R^2$ value was 0.9050 in the parameters of [PACl] and [PAC]; FSI = $0.0407[T]^{0.324}[PACI]^{0.769}[PAC]^{0.178}$ On-line floc monitor was beneficial to evaluate the flocs sizes depending on the many parameters consisting raw water properties, bring the profitable basic data to control the mixing zone more effectively.

A Longitudinal Trend Analysis in Scientific Knowledge Achievement Progress (초.중.고 학생들의 과학 지식 성취 수준 추이 분석을 위한 종단적 연구)

  • The long term trend of studensts' science achievement is a very important factor to check the effectiveness of science educational policy. However, up to date no such effort to understand the trend of Korean students' science achievement has been put into action. Recently, the Science Education Center in Korea National University of Education has been attempted to collect nation wide data for students' science achievement. The first part of the effort was to develop item pools. This study was the second part to collect nation wide data and to check any change during the two year time interval. In this study, the item pools developed by Kwon et. al.(1998) were used with some modification. The data were collected two times; February 1997 and March 1999. The subjects collected nationally were 8,766 students in 1997 and were 4,398 in 1999. The subjects were collected randomly but stratified by region and sex. As the results, the trends of achievement change during the two years were different from elementary to high school. The achievement scores were decreased in elementary schools and increased in high school. In case of middle schools, the change was not significant. However, even in elementary schools the knowledge on theory was increased significantly while knowledge on facts and principles were decreased. In contrast, the knowledge on fact showed the most increase in high schools. In this study, the data were analysed in light of region, sex, behavioral objective levels(ability) and context of test items. The science achievement monitoring system developed by the Science Education Center in Korea National University of Education can be an effective tool for monitoring students' achievement on the national level.
