Absorption, metabolism, and tissue concentrations of quercetin were examined and compared in mice and rats after oral administration of quercetin at 50 or 100 mg/kg. Quercetin was absorbed quickly in mice and reached maximum plasma concentration in I hr post-administration, and declined sharply after 4 hr. Plasma concentration of isorhamnetin, a major metabolite, also increased sharply, indicating rapid metabolic conversion, but elevated level was maintained longer than that of quercetin. Quercetin and isorhamnetin were found predominantly in glucuronide/sulfate-conjugate forms in both mice and rats. Tissue concentrations of quercetin and isorhamnetin in mice and rats were in the order of liver>kidney>spleen>plasma both 1 and 6 hr postadministration. These results show that quercetin is absorbed in mice after oral feeding and quickly metabolized into isorhamnetin as demonstrated in humans and other animal species. The results also can be used to explain various pharmacological activities reported in mouse models.
Prostaglandins (PGs) and Leukotrienes (LTs) have been implicated in the genesis of pulpal and periapical inflammation. In this study, the relationships among $PGE_2$, 6-keto-PG $F_1{\alpha}$ (a stable metabolite of $PGI_2$) and $LTB_4$ concentrations in inflamed pulp and periapical lesions were discussed. Pulp tissue were obtained in routine endodontic treatment and periapical lesions in periapical surgery after clinical diagnoses were made. These specimens were divided into four groups as normal pulp group (Control group), acute pulpitis group, chronic pulpitis group, and periapical lesion group. Pulp tissue and periapical lesions were stored in liquid nitrogen. The concentration of $PGE_2$, $PGI_2$ and $LTB_4$ were measured with ELISA. The data were analyzed by one-way ANOVA. Significantly higher levels of $PGE_2$, 6-keto-PG $F_1{\alpha}$ a and $LTB_4$ were found in acute pulpitis group than chronic pulpitis group and periapical lesion group(p<0.05). Periapical lesion group showed significantly higher mean concentrations of $PGE_2$ and $LTB_4$ than chronic pulpitis group. In control and chronic pulpitis group, significant higher levels of $PGI_2$ than $PGE_2$ and $LTB_4$ were found. These results suggested that the high levels of $PGE_2$ and $LTB_4$ in periapical lesions may be due to rich endothelium., fibroblast and lymphocyte known as the main producers of $PGE_2$ and $LTB_4$. $PGI_2$ may be thought to one of the most abundant PGs in normal pulp tissue.
In maize coleoptile segments where auxin transport capacity decreases with time following excision, susceptability of the tissue to transport inhibitors such as N-1-naphthylphthalamic acid (NPA), 3,4,5-triiodobenzoic acid (TIBA) or high concentrations of IAA was found to be rather increased. A time-dependent increase in the sensitivity to NPA can be postulated since the dose-response curve for NPA was shifted in the‘aged’tissue to the left (i.e. lower concentration). Preincubation of the tissue at a low temperature abolished the time-dependent sensitivity change, suggesting that cellular metabolism could be involved. The NPA-sensitive state was also brought about by calcium depletion of the tissue, which can be partially reversed by addition of calcium. Presence of exogenous IAA in the preincubation medium kept the auxin transport system from decay, implicating auxin as an endogenous controlling factor. Results of our experiments indicate a reversible, time-dependent changes of auxin transport system in which transport capacity and sensitivity to NPA are tightly coupled. Changes in the sensitivity to NPA were also seen in auxin action as well.
Insulin responsiveness to glucose and tissue responsiveness to insulin, using the hyperglycemic clamp and the hyperinsulinemic euglycemic clamp techniques, were compared among cows, sheep and pigs. The plasma insulin concentrations during the hyperglaycemic clamp period were highest (p < 0.05) in cows, followed by sheep and pigs. The glucose infusion rate in the hyperinsulinemic euglycemic clamp technique was greater (p < 0.01) in pigs than in cows and sheep. These results suggest responsiveness to insulin is higher in pigs than in cows and sheep.
This study was carried out to investigate the influence of molybden (Mo) concentrations in fertilizer solution on the growth of and nutrient uptake by domestically bred strawberries. Tissue analysis based on the dry weight was also conducted to determine the threshold levels in plants when Mo toxicity developed in strawberries. The leaf chlorophyll contents decreased lineally as Mo concentrations in the fertilizer solution were elevated. The differences among treatments in chlorophyll contents were statistically significant. The fresh and dry weights decreased significantly when the Mo concentrations in fertilizer solution were higher than 3.0 mM in 'Keumhyang' and 'Maehyang' strawberries and 1.0 mM in 'Seolhyang' strawberry. The elevation of Mo concentrations in fertilizer solution resulted in severe toxicity and crops developed the unique symptoms. The margin of older leaves became yellow and desiccated. Then, the margin of leaf blade rapidly became bronze colored and died as the symptoms spread up the plants. The interveinal area of the young leaves became yellowing. The elevation of Mo concentrations in fertilizer solution did not influence the tissue P, K, and Mg contents based on the dry weight. The tissue Ca contents were higher in 1.0 mM treatment than other treatments of 'Keumhyang' and 'Seolhyang' strawberries. The tissue Mo contents based on the dry weight of 'Keumhyang', 'Maehyang', and 'Seolyahng' strawberries were 76.5, 104.0, and 187.3 $mg{\cdot}kg^{-1}$, respectively, in the 0.25 mM treatments and 4,155, 5,367, and 2,190 $mg{\cdot}kg^{-1}$, respectively, in the 4.0 mM treatments. The contents increased lineally as Mo concentrations in fertilizer solution were elevated. When the concentration of Mo at which growth of crops were retarded by 10% is regarded as threshold level, the Mo contents based on dry weight of above ground plant tissue should be lower than 653.4, 686.2, and 589.7 $mg{\cdot}kg^{-1}$, in 'Keumhyang', 'Maehyang', and 'Seolyahng' strawberries, respectively.
A laboratory microcosm experiment was conducted to evaluate a major metal uptake route as well as chronic toxic effects of the clam, Macoma balthica exposed to Ag and Cu contaminated sediments. Twenty five clams were exposed to the sediments contaminated with four levels of Ag $Ag(0.01-0.87\mu{mol}\;g^{-1})$ and $Cu(0.75-5.55\mu{mol\;g^{-1})$ for 90 days. AVS (acid volatile sulfide) concentration in the sediments, considered as major factor controlling metal geochemistry and bioavailability, was manipulated to evaluate its effects on Ag and Cu bioaccumulation in M. balthica. Following 90-d exposure, the tissue Ag and Cu in M. balthica increased linearly with the Ag and Cu concentrations in sediments extracted with 1 N HCI (SEM, simultaneously extracted metals with AVS). The bioaccumulation of Ag and Cu in M. balthica was little influenced by difference in [SEM] - [AVS] values, suggesting a minor contribution of pore water metals to bioaccumulation. Tissue Ag and Cu concentrations directly influenced on the clearance rate and glycogen content of the clams. The clams with highest tissue Ag $(1.0\pm{0.2}\mu{mol}\;g^{-1})$ and Cu concentrations $(2.7\pm{0.3}\;\mu{mol}\;g^{-1})$ had only 18-43% of clearance of the clams exposed to uncontaminated sediments. Similarly, glycogen content of the exposed clams had a inverse relationship with tissue Ag and Cu concentrations. These results suggest that M. balthica exposed to Ag and Cu contaminated sediments accumulates metals largely by ingestion of contaminated sediments and can display chronic effects as reduced clearance rate and glycogen content.
A method for induction of multiple shoots using cotyledonary nodes and shoot tips of Macrotyloma uniflorum (Lam.) Verdc. was described. The experiment was conducted in which shoot induction was noticed on Murashige and Skoog (MS) medium supplemented with different concentrations and combinations of four cytokinins (KIN, 2iP, Ads, BAP). These multiple shoots were later developed into normal shoots. The highest rate of shoot proliferation came from MS medium added with BAP 1.5 mg/L. The multiple shoot buds were subcultured into MS medium with BAP (0.5-1.5 mg/L) along with Ads (1.0 mg/L) and GA$_3$ (0.5 mg/L), which gave rise to the highest frequency of shoot proliferation and elongation. The shoots were rooted on MS medium supplemented with 1.75 mg/L IBA.

Tritium is an important nuclide that must be monitored for radiation safety management. In this study, HTO was orally administered to rats at the level of 37 kBq ($1{\mu}Ci$) or 370 kBq ($10{\mu}Ci$) to examine tissue distribution and excretion levels. After sacrifice, wet and dry tissue samples were weighed and analyzed for tissue free-water tritium (TFWT) and organically bound tritium (OBT). The mean tissue concentrations of TFWT (OBT) were 30.9 (17.8) and 4.4 (8.1) Bq/g on days 7 and 13 at the 37 kBq level and 30.8 (64.6) Bq/g on day 17 at the 370 kBq level. To assess the cytogenetic damage due to tritium exposure, a cytokinesis-blocked micronucleus (MN) assay was performed in blood samples from rats exposed to HTO for 14 and 21 days after oral administration. There was no significant difference in the MN frequencies between the control and exposed rats.
An experiment was conducted to investigate correlations of litter size and average serum progesterone concentrations during pregnancy with mammary gland growth and development at parturition. Twenty ewes (5, 9, 4, and 2 ewes carrying 0, 1, 2, and 3 lambs, respectively) were used to measure weekly serum progesterone concentration during pregnancy. At parturition, the experimental ewes were slaughtered for determination of mammary gland growth and development at parturition (mammary dry fat-free tissue [DFFT], DNA, RNA, collagen, protein, and glycogen). Correlation of mammary DFFT with litter size and averages serum progesterone concentrations were 0.75 and 0.72, respectively. Litter size or maternal serum progesterone concentrations did not correlate with the mammary DNA concentration. However, litter size or maternal serum progesterone concentrations positively correlated (p < 0.01) with the mammary RNA and protein concentrations, but negatively correlated with the mammary collagen (p < 0.01) and. glycogen (p < 0.05) concentrations. Litter size or maternal serum progesterone positively correlated (p < 0.01) with the total mammary DNA, RNA, collagen, protein and glycogen contents. These results implied that the increased concentrations of progesterone with the increased litter size during pregnancy improved mammary gland growth and development at parturition.
Purpose: Obesity is often associated with disturbances in the mineral metabolism. The purpose of this study was to investigate the effects of high-fat diet-induced obesity on tissue zinc concentrations and zinc transporter expressions in mice. Methods: C57BL/6J male mice were fed either a control diet (10% energy from fat, control group) or a high-fat diet (45% energy from fat, obese group) for 15 weeks. The zinc concentrations in the serum, stool, and various tissues were measured by inductively coupled plasma (ICP)-atomic emission spectrophotometry or ICP-mass spectrophotometry. The levels of zinc transporter mRNAs in the liver, duodenum, and pancreas were measured by real-time RT-PCR. The levels of serum adipokines, such as leptin and IL-6, were determined. Results: The total body weight, adipose tissue weight, and hepatic TG and cholesterol concentrations were significantly higher in the obese group, as compared to the control group. The obese group had significantly higher levels of serum leptin and pro-inflammatory IL-6 concentrations, and had significantly lower levels of serum alkaline phosphatase activity. The zinc concentrations of the liver, kidney, duodenum, and pancreas were all significantly lower in the obese group than in the control group. On the other hand, the fecal zinc concentrations were significantly higher in the obese group than in the control group. The serum zinc concentrations were not significantly different between the two groups. The ZnT1 mRNA levels of the liver and the pancreas were significantly higher in the obese group, as compared to the control group. Hepatic Zip10 mRNA was also increased in the obese group. Conclusion: Our study findings suggest that obesity increases fecal zinc excretion and lowers the tissue zinc concentrations, which may be associated with alterations in the zinc transporter expressions.
