Molecular characterization of a lectin, BPL-4, from the marine green alga Bryopsis plumosa (Chlorophyta)

  • A novel lectin specific to N-acetyl-D-galactosamine as well as N-acetyl-D-glucosamine was isolated from Bryopsis plumosa and named as BPL-4. Sodium dodecyl sulfate polyacrylamide gel electrophorese (SDS-PAGE) and matrix-assisted laser desorption / ionization-time of flight (MALDI-TOF) mass spectrometry data showed that this lectin was a monomeric protein with molecular weight 12.9 kDa. The N-terminal amino acid sequences of the lectin were determined by Edman degradation and the full cDNA sequence encoding this lectin was obtained using the degenerate primers designed from the amino acid sequence. The size of the cDNA was 414 bp containing single open reading frame (ORF) encoding the lectin precursor. The homology analysis showed that this lectin might belong to H lectin group. BPL-4 showed high sequence similarity (60.6%) to BPL-3, which is a previously reported lectin from the same species. The comparative analysis on the lectin's primary structure showed two conserved domains including one possible active domain of H lectin group.

  • Small peptides synthesized by nonribosomal peptide synthetases (NRPSs) genes are found in bacteria and fungi. While some microbial taxa have few, others make a large number and variety. However, biochemical characterization of the products synthesized by NPRS demands a great deal of efforts. Since the completion of genome projects of numerous microorganisms, the numbers of available NRPSs genes are being expanded. Prediction of the peptides encoded by NRPS could save time and efforts. We chose the NRPS gene from Bradyrhizobium japonicum as a model to predict the peptide structure encoded by NRPS genes. Using computational analyses, the domain structure of this gene was defined, and the structure of a peptide synthesized by this NRPS was deduced. It was found that it encoded a tripeptide consisting of proline-serine-phenylalanine. This method would be helpful to predict the structure of small peptides with various NPRS genes from the genome sequence.

Characterization of histone gene expression in sevenband grouper, Hyporthodus septemfasciatus against nervous necrosis virus infection

  • Recent studies revealed that histone proteins are involved in innate immune responses during pathogen invasion as well as DNA packing. This study characterized the histone genes (H2A.V) of sevenband groupers and analyzed gene expression in NNV-infected sevenband groupers. The open reading frame (ORF) of H2A.V is 387 bp which encoded 128 amino acid residues. The deduced amino acid sequence of H2A.V harbor a highly conserved domain for H2A/H2B/H3 and H2A_C binding domain. Quantitative real-time PCR analysis showed that H2A.V had a high gene expression level in the brain and blood after being NNV-infected. An increase in extracellular histone protein in the blood has been identified as a biomarker for vascular function in humans. More research is required to understand histone's immune response at the protein level or in aquatic animals.

Liquid Rubber를 이용한 Epoxy Resin의 개질 (The Modification of Epoxy Resins with Liquid Rubbers)

  • In order to Improve the fracture toughness of epoxy resin system, liquid chloroprene rubber(LCR) and liquid butadiene rubber (LBR) which have hydroxy group were used in the brittle epoxy system. Mechanical and toughness characterization of the modified epoxy resins were investigated as a function of liquid rubbers content. Epoxy resins modified with both of the liquid rubbers showed complete phase-separation microstructure : In all of the LCR content, it was observed that the domain size of LCR dispersed in the epoxy matrix ranging from $2{\mu}m\;to\;5{\mu}m$. It was found that fracture toughness, $K_{ic}$, of the modified LCR system was enhanced continuously as increasing LCR content. However around 10phr of LBR system showed maximum fracture toughness. Specifically, when BPA add to the modified LCR system, thermal and mechanical properties increased than neat epoxy. At the same time, fracture toughness was enhanced.

  • 감지부에 두 개의 간섭계형 센서가 있고 원격측정이 가능한 광섬유센서 어레이를 구성하였다. 각 센서의 신호를 분리 측정하기 위하여 짧은 광펄스를 광원으로 하는 시간분할방식의 신호처리를 도입하였다. 센서의 측정감도는 수 kHz 영역에서 $40{\mu}rad\sqrt{Hz}$였고, 센서간의 cross talk은 -50dB 이하였다.

Characterization of Fiber Direction Influence in CFRP Composites Using Advanced NDE Techniques

  • A nondestructive technique would be very useful. Advanced NDE T-ray (terahertz ray) techniques of technology and instrumentation has provided a probing field on the electromagnetic spectrum. However, the T-ray is limited in order to penetrate a conducting material to some degree. Here, the T-ray would not go through easily the CFRP composite laminates since carbon fibers are electrically conducting while the epoxy matrix is not. So, investigation of terahertz time domain spectroscopy (THz TDS) was made and reflection and transmission configurations were studied for a 48-ply thermoplastic PPS(poly-phenylene sulfide)-based CFRP solid laminate. It is found that the electrical conductivity of CFRP composites depends on the direction of unidirectional fibers.

Isolation and characterization of unrecorded yeast species from Korea in the families Debaryomycetaceae and Piskurozymaceae

  • The purpose of this study was to isolate and identify wild yeasts from soil of Gyeongju city, and Haemadipsa rjukjuana of Gageodo Island, characterizing unrecorded yeast strains from Korea. The molecular analysis of the D1/D2 domain of 26S rDNA of yeast was performed using the Basic Local Alignment Search Tool (BLAST). No official report exists describing these three species: one species in the genus Candida, one species in the genus Debaryomyces, and one species in the genus Solicoccozyma. Candida saitoana YL9, Debaryomyces fabryi YL1, and Solicoccozyma terrea 20g9-1 are recorded for the first time from Korea. All three strains were oval shaped and polar binding, while positive for glucose, ᴅ-xylose, and ᴅ-cellobiose. Morphological, physiological, and biochemical properties are described in the species descriptions.

프레스 금형 마모에 대한 음-초음파 신호 특성 분석 (Characterization of Acousto-ultrasonic Signals for Stamping Tool Wear)

  • This paper reports on the research which investigates acoustic signals acquired in progressive compressing, hole blanking, shearing and burr compacting process. The work piece is the head pin of the electric connector, whose raw material is the preformed steel bar. An acoustic sensor was set on the bed of hydraulic press. Because the acquired signals include the dynamic characteristics generated for all the processes, it is required to investigate signal characteristics corresponding to unit process. The corresponding dynamic characteristics to the respective process were first studied by analyzing the signals respectively acquired from compressing, blanking and compacting process. The combined signals were then periodically analyzed from the grinding to the grinding in the sound frequency domain and in the ultrasonic wave. The frequency of around 9 kHz in the sound frequency domain was much correlated to the tool wear. The characteristic frequency in the acoustic emission domain between 100 kHz and 500 kHz was not only clearly observed right after tool grinding but its amplitude was also related to the wear. The frequency amplitudes of 160 kHz and 320 kHz were big enough to be classified by the noise. The noise amplitudes are getting bigger, and their energy was much bigger as coming to the next regrinding. The signal analysis was based on the real time data and its frequency spectrum by Fourier Transform. As a result, the acousto-ultrasonic signals were much related to the tool wear progression.

Biocomputational Characterization and Evolutionary Analysis of Bubaline Dicer1 Enzyme

  • Dicer, an ribonuclease type III type endonuclease, is the key enzyme involved in biogenesis of microRNAs (miRNAs) and small interfering RNAs (siRNAs), and thus plays a critical role in RNA interference through post transcriptional regulation of gene expression. This enzyme has not been well studied in the Indian water buffalo, an important species known for disease resistance and high milk production. In this study, the primary coding sequence (5,778 bp) of bubaline dicer (GenBank: AB969677.1) was determined and the bubaline Dicer1 biocomputationally characterized to determine the phylogenetic signature among higher eukaryotes. The evolutionary tree revealed that all the transcript variants of Dicer1 belonging to a specific species were within the same node and the sequences belonging to primates, rodents and lagomorphs, avians and reptiles formed independent clusters. The bubaline dicer1 is closely related to that of cattle and other ruminants and significantly divergent from dicer of lower species such as tapeworm, sea urchin and fruit fly. Evolutionary divergence analysis conducted using MEGA6 software indicated that dicer has undergone purifying selection over the time. Seventeen divergent sequences, representing each of the families/taxa were selected to study the specific regions of positive vis-$\grave{a}$-vis negative selection using different models like single likelihood ancestor counting, fixed effects likelihood, and random effects likelihood of Datamonkey server. Comparative analysis of the domain structure revealed that Dicer1 is conserved across mammalian species while variation both in terms of length of Dicer enzyme and presence or absence of domain is evident in the lower organisms.
