황기의 메탄올 추출물의 용량에 따른 면역생물학적 연구 (Immunobiological Studies on Doses of Methanol Extract of Astragali Radix)

  • Effects of methanol extract of Astragali Radix (AR) on the immune responses were studied using ICR mice. Mice were divided into 4 groups (10mice/group), and methanol extracts of AR at doses of 0.05, 0.25 and 1.25g/kg were orally administered to ICR mice once a day for 2 weeks. Mice were immunized and challenged with sheep red blood cells (SRBC). The results of this study were summarized as follows; (1) Methanol extract of AR at 0.05, 0.25 and 1.25g/kg didn't affect the weight ratios of thymus to body, as compared with those in controls, but significantly increased spleen weight ratio. (2)Methanol extract of AR at 0.05 and 0.25g/kg significantly increased hemagglutination titer and splenic plaque forming cells corresponding to humoral immunity, as compared with those in controls, but their enhancements were somewhat lowered at a high dose (1.25g/kg). (3) Methanol extract of AR at 0.05 and 0.25g/kg siginificantly increased delayed-type hypersensitivity reaction resulted from cell-mediated immunity, as compared with those in controls, but not so significant increases were observed at a high dose (1.25g/kg). (4) Methanol extract of AR at 0.05 and 0.25g/kg significantly increased phagocytic activity and the number of circulating leukocyte compared with those in controls, but their enhancements were lowered at a high dose (1.25g/kg). These results suggest that methanol extract of Astragali Radix increased humoral and cell-mediated immune responses, phagocytic activity and the number of circulating leukocyte, dependent upon dose, but inhibited their enhancement effects were decreased at a high dose (1.25g/kg).

대금음자(對金飮子)가 알코올 투여로 유발된 흰쥐의 간 지방 병증과 면역억제의 예방에 미치는 영향 (Preventive Effects of Daekumeumja on Fatty Degeneration of Liver and Immunosuppression Induced by Alcohol)

  • Objective : The present study has been undertaken to investigate the preventive effects of Daekumeumja on fatty degeneration of liver and immunosuppression induced by alcohol in rats. Method : Except for the normal group, we fed rat on 25% alcohol for 55 days. And Daekumeumja(DK) extract was administrated for the same period. We measured the serum component in rat's blood, weight of internal organs, liver triglyceride contents, histomorphometry and histopathological observation of internal organs. Results : 1. In the change measurement of serum components, DK group(50 mg/kg, 200 mg/kg) showed significant decrease of AST, ALT, albumin, ALP and triglyceride in comparison with those of the alcohol control group. 2. In the change measurement of internal organ's weight, DK group(50 mg/kg, 200 mg/kg) showed significant increase of relative body weights of liver, thymus and spleen in comparison with those of the alcohol control group. 3. DK group(50 mg/kg, 200 mg/kg) showed significant decrease of hepatic triglyceride contents in comparison with those of the alcohol control group. 4. In histomorphometrical changes of liver, DK group(50 mg/kg, 200 mg/kg) showed significant decrease of numbers of hepatocytes occupied by over 10% lipid droplets, percentages of regions occupied by lipid droplets and mean diameters of hepatocytes in comparison with those of the alcohol control group. In histomorphometrical changes of thymus, DK group(50 mg/kg, 200 mg/kg) showed significant increase of lobular thickness and cortex thickness in comparison with those of the alcohol control group. In histomorphometrical changes of spleen, DK group(50 mg/kg, 200 mg/kg) showed significant increase of splenic thicknesses, numbers of white pulps and mean diameters of white pulps in comparison with those of the alcohol control group. 5. In histopathological changes of liver, DK group(50 mg/kg, 200 mg/kg) showed effective inhibition of severe fatty changes in comparison with those of the alcohol control group. In histopathological changes of thymus and spleen, DK group(50 mg/kg, 200 mg/kg) showed effective inhibition of atrophic changes in comparison with those of the alcohol control group. Conclusions : Reviewing these experimental results, it appears that Daekumeumja have pharmaceutical preventive efficacy on fatty degeneration of liver and immunosuppression induced by alcohol in rats.

A comparative study on immune-stimulatory and antioxidant activities of various types of ginseng extracts in murine and rodent models

  • Background: Ginseng (Panax ginseng) is a widely used traditional herbal supplement that possesses various health-enhancing efficacies. Various ginseng products are available in market, especially in the Korean peninsula, in the form of drinks, tablets, and capsules. The different ginseng types include the traditional red ginseng extract (RGE), white ginseng, and black red ginseng extract (BRGE). Their fermented and enzyme-treated products are also available. Different treatment regimens alter the bioavailability of certain compounds present in the respective ginseng extracts. Therefore, in this study, we aimed to compare the antioxidant and immune-stimulating activities of RGE, BRGE, and fermented red ginseng extract (FRGE). Methods: We used an acetaminophen-induced oxidative stress model for investigating the reduction of oxidative stress by RGE, BRGE, and FRGE in Sprague Dawley rats. A cyclophosphamide-induced immunosuppression model was used to evaluate the immune-stimulating activities of these ginseng extracts in BALB/c mice. Results: Our results showed that most prominently, RGE (in almost all experiments) exhibited excellent antioxidant effects via increasing superoxide dismutase, catalase, and glutathione peroxidase activities in the liver and decreasing serum 8-hydroxy-2'-deoxyguanosine, aspartate aminotransferase, and lactate dehydrogenase levels compared with the groups treated with FRGE and BRGE. Moreover, RGE significantly increased the number of white blood cells, especially T and B lymphocytes, and antibody-forming cells in the spleen and thymus, and it also activated a number of immune cell subtypes. Conclusion: Taken together, these results indicate that RGE is the best supplement for consumption in everyday life for overall health-enhancing properties.

The Inhibitory Effect of Premature Citrus unshiu Extract on Atopic Dermatitis In Vitro and In Vivo

  • Atopic dermatitis (AD) is a chronic, recurrent inflammatory skin disease that is associated with Th2 cell-mediated allergy. The process that leads to infiltration of inflammatory cells into an AD lesion is remarkably dependent on various chemokines, especially TARC (thymus and activation-regulated chemokine/CCL17) and MDC (macrophage-derived chemokine/CCL22). Serum levels of these chemokines are over-expressed in AD patients. Citrus unshiu, which is known as Satsuma mandarin, has anti-oxidative, anti-inflammation, and anti-microviral activity. Therefore, we investigated the effect of EtOH extract of premature C. unshiu on AD. We did this using a DNCB-induced AD mouse model. We also tried to confirm an inhibitory effect for premature C. unshiu on the expression of inflammatory chemokines in IFN-${\gamma}$ and TNF-${\alpha}$ stimulated HaCaT human keratinocytes. We found that extract of premature C. unshiu reduced DNCB-induced symptoms such as hyperkeratosis, increased skin thickness, and infiltrated mast cells, in our AD-like animal model. The extract decreased levels of IFN-${\gamma}$ and IL-4 in ConA-stimulated splenocytes isolated from DNCB-treated mice. Also, extract of premature C. unshiu inhibited mRNA expression and protein production of TARC and MDC through the inhibition of STAT1 phosphorylation. These results suggest that C. unshiu has anti-atopic activity by regulating inflammatory chemokines such as TARC and MDC.

Anti-Inflammatory Effect of Quercetagetin, an Active Component of Immature Citrus unshiu, in HaCaT Human Keratinocytes

  • Citrus fruit contain various flavonoids that have multiple biological activities. However, the content of these flavonoids are changed during maturation and immature Citrus is known to contain larger amounts than mature. Chemokines are significant mediators for cell migration, while thymus and activation-regulated chemokine (TARC/CCL17) and macrophage-derived chemokine (MDC/CCL22) are well known as the typical inflammatory chemokines in atopic dermatitis (AD), a pruritic and chronic inflammatory skin disease. We reported recently that the EtOH extract of immature Citrus unshiu inhibits TARC and MDC production. Therefore, we investigated the activity of flavonoids contained in immature Citrus on TARC and MDC levels. As a result, among the various flavonoids, quercetagetin has stronger inhibitory effects on the protein and mRNA expression of TARC and MDC than other flavonoids. Quercetagetin particularly has better activity on TARC and MDC level than quercetin. In HPLC analysis, the standard peak of quercetagetin matches the peaks of extract of immature C. unshiu. This suggests that quercetagetin is an anti-inflammatory component in immature C. unshiu.

  • Phellinus linteus (PL)-hot water extract (PL-W) or- methanol extract (PL-M) was orally administered alone (single dose of 400, 800, 1600 mg/kg; 800 mg/kg/day for 5 days) or with cyclophosphamide (CY, 20 mg/kg, i.p.) to female ICR mice. Within PL alone-treated group, WBC and plaque forming cells (PFC) to SRBC were slightly and significantly enhanced when compared with control group. The relative thymus and spleen weights, WBC and PFC numbers were significantly decreased by the treatment of CY, whereas those values were markedly increased by the concomitant treatment of CY and PL when compared with CY administration alone. To assess the effects of PL and/or CY on the mitogen response of splenocytes io LPS, mouse splenocytes were stimulated with or without LPS in the presence of various concentration of PL and/or CY in vitro and splenocytes proliferation (SP) was measured by MTT assay. PL alone increased both SP and LPS- stimulated SP. Moreover, SP and LPS-induced SP suppressed by the treatment of CY alone were significantly restored by PL-treatment. These activities were higher by PL-M than by PL-W, These results indicated that PL was able to increase humoral immunity and to inhibit immunotoxicity induced by CY.

수종 생약 수침 엑기스의 면역증강 작용에 관한 연구 (Immunopotentiating Activity of Water Extracts of Some Crude Drugs)

  • Immunopotentiating activity of water extract of Lycii Fructus, Rubus Fructus, Torilis Fructus, Schizandrae Fructus and Cuscutae Semen were investigated using mouse fed with normal diet and protein deficient diet. Body weight, ratio of spleen or thymus to body weight, number of total peritoneal exudate cell, content of total serum protein and albumin content were decreased in the mouse fed with protein deficient diets as compared to the mouse fed with normal diet. Regardless of the nutritional condition, the supplement of the water extract of Lycii, Rubus Torilis, Schizandrae Fructus or Cuscutae Semen increased the ratio of spleen to body weight. The increment of this ratio was significant by the supplement of water extracts of Lycii and Torilis Fructus in mouse fed with protein deficient diet. The supplement of water extracts of Torilis Fructus to the mouse fed with protein deficient diet increased the content of total serum protein and albumin content significantly. Similar results were obtained with Lycii, Rubus, Schizandrae Fructus and Cuscutae Similar. The electrophorogram shows the difference in the concentration of some serum protein constituents between the mouse fed with normal diet and protein deficient diet.

두충 추출물이 DNCB로 유발된 알레르기성 접촉피부염과 산화적 손상에 미치는 영향 (Effect of Eucommia ulmoides Extracts on Allergic Contact Dermatitis and Oxidative Damage Induced by Repeat Elicitation of DNCB)

  • 두충(잎, 껍질, 줄기)의 추출물이 DNCB로 감작된 4주령 BALB/c mouse 암컷에 유도된 접촉성 피부염의 억제효과를 조사하였다. 림프절, 비장 및 흉선의 무게는 두충 투여군이 DNCB 대조군보다는 낮게 나타났다. 두충 부위별 추출물을 투여한 군에서 귀의 무게는 두충 추출물의 농도가 높을수록 감소하는 경향을 보였으며, 대조군과 비교하여 무게에 함량 변화가 있었다. 두충의 부위별 추출물을 1,000 mg/kg 농도로 투여하면 귀의 무게는 대조군과 유사한 수준까지 낮아졌으며, 귀의 두께는 두충 추출물을 투여한 군에서 시간이 경과할수록 두께가 감소하는 경향을 나타내었다. MDA 함량은 DNCB 대조군과 두충 투여군을 비교하였을 경우에, 간 조직에서 차이가 나타나지 않았으나, 염증이 발생한 귀 조직에서는 차이가 나타났으며(p<0.05), NO 함량은 SW, ST 및 BT 그룹에서 염증이 억제되어 대조군과 마찬가지로 측정되지 않았다.

Origanum vulgare and Thymus vulgaris Extract Usability to Improve Silage Hygienic Quality and Reduce Mycotoxin Concentrations

  • Silage is one of the main feed components of ruminants around the world and can make up about 50-80% of the rations of dairy cows during the winter. The aim of this study was to evaluate the usability of oregano (Origanum vulgare L.) and thyme (Thymus vulgaris L.) aqueous and ethanol extracts to improve the hygienic quality of perennial ryegrass, red clover and blue alfalfa silage samples and estimate their effect on mycotoxins concentrations. Under laboratory conditions, 63 silage samples (21 perennial ryegrass, 21 blue alfalfa, 21 red clover) were fermented with inserted aqueous and ethanol extracts of oregano and thyme and two commercial inoculants with mesophilic lactic acid bacteria. After 96 days of fermentation, in silage samples were established fermentation parameters, microbiological status and mycotoxins concentrations. It was determined that the best results for achieving hygienic quality of perennial ryegrass and red clover silage samples was by insertion of aqueous and ethanol extracts of oregano and thyme. In blue alfalfa samples, the best results of silage hygienic indicators were determined by inserting aqueous and ethanol extracts of oregano. Aflatoxin B1 (AFB1), deoxynivalenol (DON), zearalenone (ZEA) and T-2 toxin concentrations in perennial ryegrass, red clover and blue alfalfa silage samples were best reduced by inserting aqueous and ethanol extracts of oregano and thyme. The present study shows that these extracts can be used to improve silage hygienic quality, reduce mycotoxins concentrations and thus ensure the wellness of cattle.

In vivo와 in vitro에서 김치가 혈액성상과 면역세포배양에 미치는 영향 (The Effects of Kimchi on Hematological and Immunological Parameters in vivo and in vitro)

  • This study was designed to know the effect of kimchi on the hematological and immunological parameters in vivo and in vitro, respectively. To study the effects of kimchi on the hematological parameters, rats(S.D., male) were divided into 4 groups and fed diets containing of 3%, 5% and 10% kimchi or kimchi free diet(control) for 6 weeks. The results of CBC(complete blood cell) tests obtained from the bloods of rates were as follows ; In 10% kimchi group, the level of WBC(white blood cells), RBC(red blood cells), Hgb(hemoglobin), Hct(hematocrit) were increased significantly than those of control group(p<0.05). MCV(mean corpuscular volume), one of the red cell indices, was also increased significantly in the animals fed 10% kimchi(p<0.05). RDW(Red cell distritution width) and PCT(plateletcrit) was lowest in 10% kimchi group(p<0.05). To examine the effects of kimchi on immune cell growth in vitro, three types of mouse immune cells-spleen cells, bone marrow cells, thymus cells-were cultured with extracts of salted Chinese cabbage, fresh kimchi and fermented kimchi(for 1 week) for 12 or 20 days. Control was supplemented with PBS(phosphate buffer saline) excluding kimchi extract. The results of spleen cell, bone marrow cell, and thymus cell cultures showed similar tendency: control medium accelerated death of cells, extracts of salted Chinese cabbage reduced the rate of cell death, and extracts of fresh kimchi and fermented kimchi promoted cell growth. From these results, it could be suggested that kimchi possibly has an effect on the hematopoietic ability and increases immune cell development and growth in vivo.
