We have utilized the PCR-RFLP method to detect the ryanodine receptor(RYR1) gene mutation and to estimate the genotype frequencies of the RYR1 gene in commercial crossbred pig population. The exon region(659bp) including point mutation(C ${\rightarrow}$T; Arg ${\rightarrow}$Cys) in the porcine ryanodine receptor gene, which is a causal mutation for PSS, was amplified by PCR and digested with Cfo I restriction enzyme. The RYR1 gene was classified into three genotypes by agarose gel electrophoresis. The normal homozygous(NN) individuals showed two DNA fragments consisted of 493 and 166bp. The mutant homozygous(nn) individuals showed only one DNA fragment of 659bp. Also, all three fragments(659, 493 and 166bp) were showed in heterozygous(Nn) carrier animals. The proportions of normal, carrier and PSS pigs within crossbred population of pigs were 81%, 15% and 4%, respectively. According to the results of analysis of variance for the association of genotypes of RYR1 of pigs at 30kg, day age at 90kg and average daily gains, the RYR1 nn genotype was very higher than RYR1 NN genotype for day age at 30kg with 5% level of significant difference, but no significant difference for association of any other genotypes with day age at 90kg and average daily gain in crossbred pigs. Therefore, DNA diagnosis by using PCR-RFLP analysis for the PSS gene was useful for large-scale screening of commercial pigs in the swine industry.
For gas hydrate exploration, long offset multichannel seismic data acquired using by the 4km streamer length in Ulleung basin of the East Sea. The dataset was processed to define the BSRs (Bottom Simulating Reflectors) and to estimate the amount of gas hydrates. Confirmation of the presence of Bottom Simulating reflectors (BSR) and investigation of its physical properties from seismic section are important for gas hydrate detection. Specially, faster interval velocity overlying slower interval velocity indicates the likely presences of gas hydrate above BSR and free gas underneath BSR. In consequence, estimation of correct interval velocities and analysis of their spatial variations are critical processes for gas hydrate detection using seismic reflection data. Using Dix's equation, Root Mean Square (RMS) velocities can be converted into interval velocities. However, it is not a proper way to investigate interval velocities above and below BSR considering the fact that RMS velocities have poor resolution and correctness and the assumption that interval velocities increase along the depth. Therefore, we incorporated Migration Velocity Analysis (MVA) software produced by Landmark CO. to estimate correct interval velocities in detail. MVA is a process to yield velocities of sediments between layers using Common Mid Point (CMP) gathered seismic data. The CMP gathered data for MVA should be produced after basic processing steps to enhance the signal to noise ratio of the first reflections. Prestack depth migrated section is produced using interval velocities and interval velocities are key parameters governing qualities of prestack depth migration section. Correctness of interval velocities can be examined by the presence of Residual Move Out (RMO) on CMP gathered data. If there is no RMO, peaks of primary reflection events are flat in horizontal direction for all offsets of Common Reflection Point (CRP) gathers and it proves that prestack depth migration is done with correct velocity field. Used method in this study, Tomographic inversion needs two initial input data. One is the dataset obtained from the results of preprocessing by removing multiples and noise and stacked partially. The other is the depth domain velocity model build by smoothing and editing the interval velocity converted from RMS velocity. After the three times iteration of tomography inversion, Optimum interval velocity field can be fixed. The conclusion of this study as follow, the final Interval velocity around the BSR decreased to 1400 m/s from 2500 m/s abruptly. BSR is showed about 200m depth under the seabottom

Strengthening vehicle emission regulation is one of important policies to improve air quality in urban area. Due to the limitation of specified driving cycles for certification test to reflect real driving conditions, additional off-cycle emission regulations have been adopted in US and being developed in Europe. The driving cycles of US or Europe have been used in emission certification for Korean light-duty vehicles, but it has not been known how well the driving cycles reflect various real driving patterns in Korea. In that point of view, it is required to estimate vehicle emission based on real road driving conditions to raise the effectiveness of vehicle emission regulation in Korea. In this study, real driving emission measurements have been conducted for three Korean light-duty vehicles with PEMS. The driving routes consisted of urban, rural and motorway in Seoul and Incheon. The data have been analyzed with various averaging methods including moving averaging windows method and compared to emission limits set with emission certification modes applied to tested vehicles. The results have shown that the real driving pollutant emissions of a gasoline and a LPG vehicles have been ranged quite lower than those of emission limits on CVS-75 driving cycle. But real driving NOx of a light duty diesel vehicle has been considerably higher than emission limit of NEDC driving cycle. The higher than expected NOx emission of a diesel vehicle might be caused by different strategy to control EGR in real driving condition from NEDC driving.
PURPOSES : The authors set out to estimate the related carbon emissions, energy use, and costs of the national freeways and highways in Korea. To achieve this goal, a macro-level methodology for estimating those amounts by road type, road structure type, and road life cycle was developed. METHODS : The carbon emissions, energy use, and costs associated with roads vary according to the road type, road structure type, and road life cycle. Therefore, in this study, the road type, road structure type, and road life cycle were classified into two or three categories based on criteria determined by the authors. The unit amounts of carbon emissions and energy use per unit road length by classification were estimated using data gathered from actual road samples. The unit amounts of cost per unit road length by classification were acquired from the standard cost values provided in the 2013 road business manual. The total carbon emissions, energy use, and cost of the national freeways and highways were calculated by multiplying the road length by the corresponding unit amounts. RESULTS: The total carbon emissions, energy use, and costs associated with the national freeways and highways in Korea were estimated by applying the estimated unit amounts and the developed method. CONCLUSIONS: The developed method can be employed in the road planning and design stage when decision makers need to consider the impact of road construction from an environmental and economic point of view.

Assessment of a storm is accomplished by the duration and storm area rather than a simple analysis given by the ground rain gauge stations. One of the best method for assessing storm is Depth-Area-Duration(DAD) of rainfall analysis. but existing DAD analysis method is likely to possible errors. Therefore, DAD analysis and its applicability were examined using a grid-based DAD analysis program that can reduce the possibility of errors in this study. Three spatial distribution techniques were used to analyze the applicability. Then, DAD analysis was performed using the converted grid-based rainfall data. As a result, it was possible to estimate the MAAR values by area for each duration, and showed high applicability in the rainfall data using ordinary kriging technique.
This study was conducted to estimate the dimensions of horticultural products and the mean plant height of plug seedlings using three-dimensional (3D) images. Two types of camera, a ToF camera and a stereo-vision camera, were used to acquire 3D images for horticultural products and plug seedlings. The errors calculated from the ToF images for dimensions of horticultural products and mean height of plug seedlings were lower than those predicted from stereo-vision images. A new indicator was defined for determining the mean plant height of plug seedlings. Except for watermelon with tap, the errors of circumference and height of horticultural products were 0.0-3.0% and 0.0-4.7%, respectively. Also, the error of mean plant height for plug seedlings was 0.0-5.5%. The results revealed that 3D images can be utilized to estimate accurately the dimensions of horticultural products and the plant height of plug seedlings. Moreover, our method is potentially applicable for segmenting objects and for removing outliers from the point cloud data based on the 3D images of horticultural crops.

This study aimed to estimate and evaluate the thermal mitigation of the urban tree canopy on the summer outdoor environment by quantitative use of mean radiant temperature. This study applied the SOLWEIG model based $T_{mrt}$ comparison method by using both (1) urban tree canopy presence examples and (2) urban tree canopy absence examples as constructed from airborne LiDAR system based three-dimensional point cloud data. As a result, it was found that an urban tree canopy can provide a decrease in the entire domain averaged daily mean $T_{mrt}$ about $5^{\circ}C$ and that the difference can increase up to $33^{\circ}C$ depending both on sun position and site conditions. These results will enhance urban microclimate studies such as indices (e.g., wind speed, humidity, air temperature) and biometeorology (e.g., perceived temperature) and will be used to support forest based public green policy development.
This study was carried out to investigate PSS (Porcine Stress Syndrome) with the PSE (Pale, Soft, Exudative) in 319 different pigs(Yorkshire 150; Landrace 89 and Duroc 80). The PCR-RFLP method was adapted to detect the ryanodine receptor (RYR 1) gene mutation and to estimate the genotype frequency of the RYR1 gene in breeding pig population. The DNA samples were collected from hair follicles of pigs of Yorkshire, Landrace and Duroc. After DNA amplification by PCR, the PCR products were digested by restriction enzyme, Cfo I. Primary PCR products of ryanodine receptor gene were length of 659 bp in hair follicle and their second PCR products were length of 522 bp in hair follicle. The exon region (522 bp) including point mutation ($C \arrow T; Arg \arrow Cys$) in the porcine ryanodine receptor gene, which is a causal mutation for PSS, was digested with Cfo I restriction enzyme. The RYR1 gene was classifed into three genotypes by agarose gel electrophoresis. The normal homozygous (NN) individuals showed two DNA fragments consisted of 439 and 83 bp. The mutant homozygous (nn) individuals showed only one DNA fragment 522 bp. In addition, all three fragments (522, 439 and 83 bp) were showed in heterozygous (Nn) carrier animals. The normal homozygous (NN), heterozygous (Nn) and mutant homozygous (nn) were 98.00, 2.00 and 0.00% in Yorkshire pigs, 87.64, 11.24 and 1.12% in Landrace, 100.00, 0.00 and 0.00% in Duroc, respectively. The gene frequencies of N and n were 0.990 and 0.010 in Yorkshire pigs, 0.933 and 0.067 in Landrace, 1.000 and 0.000 in Duroc, respectively.
Objectives: This study evaluated the antibacterial effect and mechanical properties of composite resins ($L_{CR}$, $M_{CR}$, $H_{CR}$) incorporating chitosan with three different molecular weights (L, Low; M, Medium; H, High). Materials and Methods: Streptococcus (S). mutans 100 mL and each chitosan powder were inoculated in sterilized 10 mL Brain-Heart Infusion (BHI) solution, and was centrifuged for 12 hr. Absorbance of the supernatent was measured at $OD_{660}$ to estimate the antibacterial activities of chitosan. After S. mutans was inoculated in the disc shaped chitosan-containing composite resins, the disc was cleansed with BHI and diluted with serial dilution method. S. mutans was spread on Mitis-salivarius bacitracin agar. After then, colony forming unit (CFU) was measured to verify the inhibitory effect on S. mutans biofilm. To ascertain the effect on the mechanical properties of composite resin, 3-point bending and Vickers hardness tests were done after 1 and 3 wk water storage, respectively. Using 2-way analysis of variance (ANOVA) and Scheffe test, statistical analysis was done with 95% significance level. Results: All chitosan powder showed inhibition effect against S. mutans. CFU number in chitosan-containing composite resins was smaller than that of control resin without chitosan. The chitosan containing composite resins did not show any significant difference in flexural strength and Vickers hardness in comparison with the control resin. However, the composite resin, $M_{CR}$ showed a slightly decreased flexural strength and the maximum load than those of control and the other composite resins $H_{CR}$ and $L_{CR}$. Conclusions: $L_{CR}$ and $H_{CR}$ would be recommended as a feasible antibacterial restorative due to its antibacterial nature and mechanical properties.

This study aimed to estimate accuracy of GPS/Leveling and to present availability of GPS/Leveling in public surveying. For this purpose, we carried out GPS survey for bench marks and control points of Hongsung area. Orthometric heights calculated by two GPS/Leveling methods were compared to reference height. The one is calculated by base of geoid models such as EGM96, OSU91A, KGEOID99, and the other is calculated by network adjustment using fixed point. The results of GPS/Leveling by geoid models show that RMSE of EGM96 is ${\pm}0.061m,\;OSU91A\;{\pm}0.725m,\;KGEOID99\;{\pm}0.598m$. The results of GPS/Leveling by network adjustment show that the best RMSE is ${\pm}0.043m$ in case of using three fixed bench mark, and this method can be used for leveling effectively. GPS/Leveling would be able to apply in forth order public leveling and height determination of public control points.
