Synthesis and Characterization of Chelating Resins Containing Thiol Croups (티올기를 함유하는 킬레이트 수지의 합성 및 특성)

  • Three kinds of macro-reticular bead-typed chelating resins having thiol groups were obtained from basic resins like poly(strene-co-divinylbenzene) (PSD) and poly(styrene-co-methyl methacrylate-co-divinylbenzene) (PSMD): the chelating resin (I) was prepared by chloromethylation of phenyl rings of PSD followed by thiolation using thiourea. The chelating resin (ll) was designed to provide enough space to chelate heavy metal ions; one chloromethyl group was obtained by chlorination of hydroxymethyl group provided by reduction of carboxylic ester group of PSMD and another chloromethyl group was obtained by direct chloromethylation of pendent phenyl group using chloromethyl methyl ether. Both of chloromethyl groups were thiolated by using thiourea. The chelating resin (III) was prepared by chlorosulfonation of phenyl rings of PSD followed by thiolation using sodium hydrosulfide. The adsorbtivity toward heavy metal ions was evaluated. The hydrophobic chelating resin (I) with thiol groups showed highly selective adsorption capacity f3r mercury ions. However, the chelating resin (II) with thiol groups showed mere effective adsorption capacity toward mercury ions than chelating resin (I) with thiol groups, and showed some adsorption capacity for other heavy metal ions like Cu$\^$2+/, Pb$\^$2+/, Cd$\^$2+/ and Cr$\^$3+/. On the other hand, the chelating resin (III) which have hydrophilic thiosulfonic acid groups was found to be effective adsorbents for some heavy metal ions such as Hg$\^$2+/, Cu$\^$2+/, Ni$\^$2+/, Co$\^$2+/, Cr$\^$3+/ and especially Cd$\^$2+/ and Pb$\^$2+/.

  • A Streptomyces spp. strain SY 79-1 which was capable of producing metal protease was isolated from soil. The optimal pH and temperature of the protease were around pH 8.0 and $45^{\circ}C$, respectively. The stable pH range of the enzyme was between pH 6.0 to 8.0. The enzyme was stable at $45^{\circ}C$, but it lost the activity about 75 % for 5 min and completely for 30 min when it was treated at $60^{\circ}C$. The activity of the enzyme was inhibited by $Hg^{++},\;Cu^{++},\;Ag^{+}$ and activated by $Mg^{++},\;Mn^{++},\;Co^{++},\;but\;Fe^{++},\;Ca^{++},\;Pb^{++}\;and\;Al^{3+}$ did not affect enzyme activity. This enzyme was strongly inhibited by EDTA, but was not inhibited by 2, 4-DNP, ${\rho}$-CMB, ${\varepsilon}$-aminocaproic acid, cysteine, thiourea, citric acid, oxalic acid and sodium arsenate. When cobalt was added to the EDTA-denatured enzyme, the activity of the enzyme was restored.

Studies on Characteristics and Stability of Anthocyanin Pigment Extracted from Korean Purple - Fleshed Potatoes (한국산 유색감자 색소의 특성 및 안정성에 관한 연구)

  • Effects of pH, sugar, ascorbic acid, organic acids and light sources on the stability of anthocyanin pigment extracted from Korean purple-fleshed potatoes (PL-6, PL-28, PL-31 and Jasim) were studied. The pH had remarkable influence on the color stability of anthocyanin pigment. With increasing pH, the color gradually fades as colorless pseudobases are formed. In acidic pH, anthocyanin was stable, but with increasing pH the color gradually changed to colorless. The addition of sugar decrease in color stability of the pigment during storage period. The most of organic acids, such as a tartaric, citric and succinic acids, were found to improve the stability of the pigment, while malic and malonic acid reduced the stability of the pigment. The addition of ascorbic acid considerable decreased in anthocyanin pigment stability, but the effect was not decreased by adding thiourea. The effect of light sources such as a darkroom, a fluorescent light, and sunlight, reduced gradually the stability of anthocyanin pigment. Therefore the pigment degradation could be minimized by shielding the light from the pigment.

  • A field experiment was carried out to investigate the sulfur effects on yield, growth and nutritive qualities of soybean in the different fertilizer application methods. Ammonium sulfate and super-phosphate were applied to the soil with levels of 0, 2, 4, and 6Kg S/10a and potassium sulfate, ammonium sulfate and thiourea were applied by foliar application with 2KgS/10a. The results obtained were as follows; 1. Grain and dry matter yield of soybean were increased by the increase of sulfur application and sulfur application showed positive effect on yield components. 2. Sulfur application increased the sulfur content and decreased N/S ratio in grain of soybean. 3. Protein content was increased by raising sulfur application up to 4Kg S/10a. Sulfur application influenced on amino acid composition of soybean protein. Cysteine and methionine contents were increased by sulfur application. 4. Lipid content of soybean grain and inorganic ion absorption by soybean plant were not influenced significantly by sulfur application. 5. In application effects, there was no significant difference among sulfur fertilizers, but foliar application was more effective than soil application.

Solubilization of Proteins from Human Lymph Node Tissue and Two-Dimensional Gel Storage

  • In the present study, we compared six different solubilization buffers and optimized two-dimensional electrophoresis (2-DE) conditions for human lymph node proteins. In addition, we developed a simple protocol for 2-D gel storage. Efficient solubilization was obtained with lysis buffers containing (a) 8M urea, 4% CHAPS (3-[(3-cholamidopropyl) dimethylammonio]-1-propanesulfonate), 40 mM Tris base, 65 mM DTT(dithiothreitol) and 0.2% carrier ampholytes; (b) 5M urea, 2M thiourea, 2% CHAPS, 2% SB 3-10 (N-decyl-N, N-dimethyl-3-ammonio-1-propanesulfonate), 40mM Tris base, 65 mM DTT and 0.2% carrier ampholytes or (c) 7M urea, 2M thiourea, 4% CHAPS, 65 mM DTT and 0.2% carrier ampholytes. The optimal protocol for isoelectric focusing (IEF) was accumulated voltage of 16,500 Vh and 0.6% DTT in the rehydration solution. In the experiments conducted for the sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), best results were obtained with a doubled concentration (50 mM Tris, 384 mM glycine, 0.2% SDS) of the SDS electrophoresis buffer in the cathodic reservoir as compared to the concentration in the anodic reservoir (25 mM Tris, 192 mM glycine, 0.1% SDS). Among the five protocols tested for gel storing, success was attained when the gels were stored in plastic bags with 50% glycerol. This is the first report describing the successful solubilization and 2D-electrophoresis of proteins from human lymph node tissue and a 2-D gel storage protocol for easy gel handling before mass spectrometry (MS) analysis.

Effects of Nitrofurantoin on Lipid Peroxidation and Reactive Oxygen Radical Generation in Porcine Lung Microsome (Nitrofurantion이 폐장 미크로솜 지질과산화와 반응성 산소 라디칼 생성에 미치는 영향)

  • In vitro effects of nitrofurantoin, an antimicrobial agent for acute and chronic urinary tract infection, on the lung microsomal lipid peroxidation and the generation of reactive oxygen radicals were investigated to elucidate the biochemical mechanisms of its in vivopulmonary toxicity. The interaction of nitrofurantoin with porcine lung microsome resulted in significant lipid peroxidation. In addition, nitrofurantoin stimulated the generation of reactive oxygen radicals, $O^{-}_{2}{\cdot},\;H_2O_2$ as well as a highly reactive secondary oxygen species, $OH{\cdot}$. The stimulation of lipid peroxidation was inhibited not only by superoxide dismutase and catalase, but also by hydroxyl radical scavengers, mannitol and thiourea. Neither singlet oxygen $({^1}O_{2})$ was detected during the incubation of microsome with nitrofurantoin, nor lipid peroxidation was inhibited by singlet oxygen scavengers. When incubated anaerobically under the nitrogen atmosphere, the ability of nitrofurantoin to stimulatle lipid peroxidation was abolished. It appears that NADPH-dependent metaboliam of nitrofurantoin in pulmonary microsome under aerobic condition is accompanied by the stimulation of lipid peroxidation through the mediation of reactive oxygen radicals, particularly hydroxyl radical. It is strongly suggested from these results that the stimulation of pulmonary microsomal lipid peroxidation by the reactive oxygen radical may be a in vivo mechanism of pulmonary toxicity caused by nitrofurantoin.

  • Local extravasation during intravenous administration of adriamycin (doxorubicin HCl) can cause severe skin ulceration and necrosis. To investigate the mechanism of adriamycin-induced skin toxicity, effects of adriamycin on reactive oxygen radical metabolism using cultured skin cells of fetal rat. Adriamycin produced significant release of lactic dehydrogenase from cultured skin cell preparations dose- and time-dependently. The production of superoxide anion in sonicated suspensions of cultured skin cells was significantly increased by adriamycin under the presence of NADPH and NADH. The drug also stimulated malondialdehyde (MDA) production, an index of lipid peroxidation, in NADPH- and NADH-supported cell preparations. The increased production of MDA was significantly inhibited by oxygen radical scavengers (superoxide dismutase, catalase, thiourea) and antioxidants (butylated hydroxytoluene, ${\alpha}-tocopherol$). Treatment of cultured skin cells with 1, 3,-bis (2-chloroethyl)-1-nitrosourea (BCNU), an inhibitor of glutathione reductase, enhanced the lipid peroxidation induced by adriamycin. The present study suggests that lipid peroxidation which is resulted from the stimulated production of reactive oxygen radical causes cellular damage in adriamycin-treated skin cells of rat.

  • This study was conducted to understand browning characteristics of Chinese Chestnut during processing and storage. For this, the isolation and identification of polyphenolic compounds and the biochemical characteristics of polyphenol oxidase(PPO) were investigated. The content of total phenol was $6.5{\mu}g/g$ and it was consisted of ferulic acid, caffeic acid, synapic acid, p-coumaric acid, gallic acid and salicylic acid in order. PPO was purified 11.7 fold through ammonium sulfate fractionation, DEAE-cellulose column chromatography and Sephadex G-200 column chromatography. Purified enzyme showed single protein and activity band by polyacrylamide gel electrophoresis. The optimum pH and temperature of PPO were 5.9 and $45^{\circ}C$, respectively. The activity of PPO was lost 93% by exposing at $80^{\circ}C$ for 15minutes. $Mg^{{+}{+}}$, $Ca^{{+}{+}}$, $Zn^{{+}{+}}$ increased the activity of PPO, but $Fe^{{+}{+}}$, $K^+$, $Hg^{{+}{+}}$ inhibited PPO at 10mM concentration. L-ascorbic acid, thiourea, sodium chloride and L-cysteine were effective inhibitors of PPO. The activity of PPO was higher for o-diphenols than other polyphenols. The Km value of PPO for catechol was 5mM.

  • This research was carried out to study the seed dormancy of radishes which were used for summer vegetation. The differences of dormancy of cultivars, maternal effects, the effect of full day length to mother plants, and the effects of chemical treatments for breaking the seed dormancy were considered for this study. These results sere summarized as follows; 1. The germination percentage of 7 cultlvars sown just after harvest were as follows;SP70-:0, PS11-:37.5, GJ75-:75.5, JD127-:77.5, YH85-:81.0, M55-:96.5, and JC73-:98.0%. 2. The dormany of $F_1$ seed of radish was severe when SP70-was used for mother plant. 3. Germination behavior was different by the maturity of plants grown for seed production. The dormancy of seed harvested from immature field-grown plants sown Nov. 10 was deeper than mature plants sown Mar. 22. 4. The germination ratio of tire seed of SP70-radish was increased by treatment of full day length to tire mother plant during full period of growing or since first flowering stage, while that was 0% by treatment since yellow ripening stage. 5. GA 200 ppm+thiourea 0.2% treatment was most effective for breaking of radish seed dormancy.

  • In this study, ZnS: Cu nano-crystals are synthesized by solution synthesis technique (SST). The structural properties such as crystal structure and particle morphology, and the optical properties such as light absorption/transmittance, energy bandgap, and photoluminescence (PL) excitation/emission are investigated. In an attempt to realize the Cu-doping easiness, the synthesis temperature (~$80^{\circ}C$) is applied to the synthesis bath, and the thiourea is used as sulfur precursor, unlike other general chemical synthesis route. Both undoped ZnS and ZnS : Cu nano-crystals have the cubic crystal structure and have the spherical particle shape. The position of light absorption edge is ~305 nm, indicating the occurrence of quantum size effect. The PL emission intensity and line-width are maximum and minimum, respectively, for Cu-doping concentration 0.03M. In particular, the dependence of PL intensity and line-width on the Cu-doping concentration for ZnS : Cu nano-crystals synthesized by SST is reported for the first time in this study. Experimental results of the absorption edge and the PL excitation show that the main emission peak of ZnS : Cu nano-crystals (~510 nm) in this study is due to the radiative recombination center in the energy bandgap induced by Cu dopant.
