Stroke occurs when local thrombosis, embolic particle or the rupture of blood vessele interrupts the blood floe to the brain. $\beta$-estradiol 17-valerate has been reported to exert neuroprotective effects when administered before an ischemic insult. Recently, the pathophysiology of cerebral ischemia has been studied extensively in rat with various methods. In the present study, we investigates whether $\beta$-estrodiol 17-valerate can protect against brain injury. RNA sample were extracted from the hippocampus of female rat, reverse-transcription in the presence of [$\alpha$32p] dATP. Differential gene express-ion profiles were revealed (Bone morphogenetic protein type 1A receptor, Protein disulphide isomerase, Leukemia inhibitor factor receptor, cytochrome bc- 1 complex-x core P, thiol-specific antioxidant protein). RT-PCR was used to validate the relative expression pattern obtained by the cDNA array. The precise relationship between the early expression of recovery genes and stroke is a matter of luther investigation. 효모의 티올특이성 항산화단백과 아미노산 서열상 상동성을 보이는 50종류의 단백은 새로운 항산화 단백군을 형성하며 또한 병원성 미생물에도 널리 분포하고 있으나 이들 단백의 생화학적 및 생리적인 기능은 거의 알려져 있지 않은 실정이다. 본 연구는 병원성 미생물의 티올특이성 항산화단백의 기능에 관한 연구로서 Saccharomyces cerevisiae의 TSA 및 Salmonella typhimurium alkcyl hydroperoxide reductase의 AhpC subunit와 상동성을 나타내는 Corynebacterium diphtheriae의 DirA 유전자를 PCR 방법으로 클로닝하고 대장균에 발현시킨 후 정제하여 항산화 특성을 조사하였다. 정제된 DirA는 티올을 함유하는 금속촉매 산화계인 DTT/Fe$^{3+}$를 선택적으로 억제하였으며 티오레독신 의존성 과산화물 분해활성을 나타내었다. DTT/Fe$^{3+}$ 금속촉매 산화계에 의한 효소의 불활성화를 50% 억제 하는 DirA의 농도는 0.12 mg/ml로 효모 TSA 항산화활성의 약1/4 수준이었으며, 효모의 티 오레 독신계와 반응시켰을때 과산화물 분해활성은 0.02 unit/mg로서 효모 TSA의 티오레독신 의존성 과산화물 분해활성의 1/20수준이었다. 정제된 단백질을 이용하여 항체를 제조하였으며 이항체를 이용하여 Corynebacterium diphtheriae에서 발현됨을 확인하였다. 이러한 결과를 통하여 Corynebacterium diphtheriae의 병원성은 숙주세포의 방어기전인 백혈구에 의하여 생성되는 과산화수소 또는 다른 활성산소종을 제거하는 DirA작용과 연관이 있는 것으로 사료된다.
Thioredoxin (TRX) protein is an antioxidant responsible for reducing other proteins by exchanging cysteine thiol-disulfide and is also known for its anti-allergic and anti-aging properties. On the other hand, epidermal growth factor (EGF) is an important material used in the cosmetics industry and an essential protein necessary for dermal wound healing facilitated by the proliferation and migration of keratinocytes. EGF also assists in the formation of granulation tissues and stimulates the motility of fibroblasts. Hence, genetically modified soybeans were developed to overexpress these industrially important proteins for mass production. A single-dose oral-toxicity-based study was conducted to evaluate the potential toxic effects of TRX and EGF proteins, as safety assessments are necessary for the commercial use of seed-specific protein-expressing transgenic soybeans. To achieve this rationale, TRX and EGF proteins were mass purified from recombinant E. coli. The single-dose oral-toxicity tests of the TRX and EGF proteins were carried out in six-week old male and female Institute of Cancer Research (ICR) mice. The initial evaluation of the single-dose TRF and EGF treatments was based on monitoring the toxicity signatures and mortality rates among the mice, and the resultant mortality rates did not show any specific clinical symptoms related to the proteins. Furthermore, no significant differences were observed in the weights between the treatment and control groups of male and female ICR mice. After 14 days of treatment, no differences were observed in the autopsy reports between the various treatment and control groups. These results suggest that the minimum lethal dose of TRX and EGF proteins is higher than the allowed 2,000 mg·kg-1 limit.
Mitogen-activated protein kinases (MAPK) affect the activation of activator protein-1 (AP-1), which plays an important role in regulating a range of cellular processes. However, the roles of these signaling factors on hydrogen peroxide ($H_2O_2$)-induced cell death are unclear. This study examined the effects of $H_2O_2$ on the activation of MAPK and AP-1 by exposing the cells to $H_2O_2$ generated by either glucose oxidase or a bolus addition. Exposing BJAB or Jurkat cells to $H_2O_2$ affected the activities of MAPK differently according to the method of $H_2O_2$ exposure. $H_2O_2$ increased the AP-1-DNA binding activity in these cells, where continuously generated $H_2O_2$ led to an increase in mainly the c-Fos, FosB and c-Jun proteins. The c-Jun-$NH_2$-terminal kinase (JNK)-mediated activation of c-Jun was shown to be related to the $H_2O_2$-induced cell death. However, the suppression of $H_2O_2$-induced oxidative stress by either JNK inhibitor or c-Jun specific antisense transfection was temporary in the cells exposed to glucose oxidase but not to a bolus $H_2O_2$. This was associated with the disruption of death signaling according to the severe and prolonged depletion of reduced glutathione. Overall, these results suggest that $H_2O_2$ may decide differently the mode of cell death by affecting the intracellular redox state of thiol-containing antioxidants, and this depends more closely on the duration exposed to $H_2O_2$ than the concentration of this agent.
In order to get insight into the mechanism of cadmium (Cd)-induced brain injury, we investigated the effects of Cd on the induction of COX-2 in bEnd.3 mouse brain endothelial cells. Cd induced COX-2 expression and $PGE_2$ release, which were attenuated by thiol-reducing antioxidant N-acetylcysteine (NAC) indicating oxidative components might contribute to these events. Indeed, Cd increased cellular reactive oxygen species (ROS) level and DNA binding activity of nuclear factor-kB (NF-kB), an oxidative stress sensitive transcription factor. Cd-induced $PGE_2$ production and COX-2 expression were significantly attenuated by Bay 11 7082, a specific inhibitor of NF-kB and by SB203580, a specific inhibitor of p38 mitogen activated protein kinase (MAPK). These data suggest that Cd induces COX-2 expression through activation of NF-kB and p38 MAPK, the oxidative stress-sensitive signaling molecules, in brain endothelial cells.
P. pastoris는 대장균에 비해 정확한 접힘, 당화, 효율적인 분비기작등의 장점을 가지고 있어 재조합 단백질의 생산을 위한 균주로서 관심을 받고 있다. 또한 재조합 단백질의 효율적인 생산공정 개발을 위하여 배양공정 중 특정 시간이나 조건에서 발현될 수 있는 효율적인 유도성 프로모터의 개발도 중요 관심 분야이다. 본 연구에서는 연속배양을 이용하여 P. pastoris의 배양 중 특정 기질이 소모되었을 때 발현되는 자동 유도성 프로모터를 개발하기 위하여, 인산이 고갈되었을 때 과발현 되는 유전자들을 탐색하였다. 인산 제한 연속배양의 정상상태에서 얻어진 균체로 부터 total RNA와 mRNA를 분리하였고, 이로부터 cDN를 합성하여 인산제한조건에서 과발현되는 유전자들을 확보하였다. 그 중 빈도수가 높은 8종의 유전자 3-phosphoglycerate kinase, glyceraldehyde 3-phosphate dehydrogenase(GAPDH), glucokinase, thiol-specific antioxidant protein, triosephosphate isomerase, sodium/phosphate symporter(NPS) 그리고 pyruvate decarboxylase를 선별하였고, Northern blot analysis를 수행한 결과 인산 섭취에 관련된 NPS 유전자가 인산제한조건에서 과발현 됨을 확인하였다. 본 연구실에서는 자동유도성 프로모터로서 NPS유래의 프로모터의 잠재성을 알아보기 위하여, 관련 유전자를 확보하여 외래 유전자를 이용한 발현연구를 진행 중이다.
Peroxiredoxin 6 (Prx 6)는 티올-특이적 항산화 단백질에 속하는 효소로서 산화적 스트레스로부터 세포를 보호할 뿐만 아니라 과산화물의 환원작용을 촉매한다. 본 연구에서는 인간 Prx 6의 promoter를 이용하여 항산화반응을 유발하는 추출물을 효과적으로 스크리닝하는 새로운 형질전환마우스를 개발하는 최종목적을 달성하기 위한 중간단계로서, hPrx 6/Luc 벡터를 개발하고, 이들 벡터의 안정적 발현과 성공적 반응성을 세포주를 이용하여 확인하고자 하였다. 이를 위해, 인간 Prx 6 promoter를 증폭하여 luciferase cDNA와 결합한 hPrx 6/Luc 벡터를 제조하였으며, 제조된 벡터를 제한효소 절단과 염기서열분석을 통해 확인하였다. hPrx 6/Luc 벡터는 NCI-H460 세포에 transfection한 후 인삼(KWG), 홍삼(KRG), 맥문동(LP), 홍문동(RLP)의 4가지 추출물을 처리하여 luciferase activity를 측정하였다. 그 결과, luciferase activity는 4가지 추출물에 의해 효과적으로 증가하였고, 특히 KRG과 LP를 처리한 그룹이 KWG과 RLP를 처리한 그룹보다 높았다. 또한, luciferase activity는 RLP 농도에 의존적으로 증가하였다. hPrx 6/Luc 벡터와 hPrx 6 mRNA반응의 차이를 비교하기 위해, 4가지 추출물을 처리한 후 hPrx 6 mRNA의 양을 RT-PCR로 분석하였다. 그러나, hPrx 6 mRNA의 양은 비록 고농도의 RLP 추출물에서는 약간의 증가가 관찰되었지만, 대조군에 비하여 4가지 추출물에서 유의적인 차이가 없었다. 한편, 4가지 추출물에 의한 superoxide dismutase (SOD) 활성의 변화는 비록 일부 차이는 있었지만 hPrx 6/Luc 벡터와 유사한 반응을 나타내었다. 따라서, 이러한 결과는 hPrx 6/Luc 벡터는 성공적으로 제조되었고, 세포내에서 안정적으로 발현하면서 항상화물질에 민감하고 정량적으로 반응할 수 있음을 제시하고 있다. 더불어, 이러한 세포주에서 확인결과를 바탕으로 형질전환마우스가 개발된다면, 항산화물질을 정량적으로 스크리닝하는 시스템으로 적용가능성이 매우 높음을 보여주고 있다.
