Characterization of Cytosol Acid Protease from Dormant Cyst of Artemia franciscana (Artemia franciscana cyst의 세포내 산성 Protease)

  • A thiol protease has been isolated and partially purified from encysted brine shrimp Artemia franciscana using a four-step procedure(filtration, salting out, gel filtration and ion exchange chromatography). The optimum pH of the enzyme for caseinolytic activity was appeared to be 3.0, and the enzymematic activity was stable up to pH 6.0 but lost completely at the pH higher than 8.0. The optimal temperature of the enzyme was appeared to be 35$^{\circ}$C, and ninety percent of the enzyme activity was lost at 45$^{\circ}$C. Various metal ions, e.g., zinc, copper, iron, inhibited the enzyme activity; however, heavy metal chelator, e.g., EDTA, stimulated the enzyme activity. The protease was concluded to be a member of the thiol group protease, since it was inhibited by thiol protease inhibitors and iodoacetate. The protease was also concluded to be a acid protease based on optimum pH.

  • Streptomyces sp. KISl3 isolated from soil was found to produce low molecular weight thiol protease inhibitors. The protease inhibitor production was closely linked to the cell growth and regulated by growth condition. The inhibitor was purified from the culture broth through butanol extraction, silicagel 60 column chromatography, Sephadex LH-20 gel filtration and preparative HPLC. The inhibitor showed specific inhibitory activity to thiol protease such as papain, picin and bromelain. The mode of inhibition against papain to Hammersten casein as a substrate was non-competitive.

  • Chemotaxonomic and numerical identification were carried out for an isolate of Streptomyces strain SMF13 producing thiol protease inhibitor. Fifty taxonomic unit characters were tested and the data were analyzed numerically using the TAXON program. The isolate SMF13 was identified to be a member of the cluster 5 of Streptomyces and best matched to Streptomyces omiyaensis which is a synonym of Streptomyces exfoliatus. Therefore. it was concluded that the isolate was identified to be a strain of Streptomyces exfoliatus.

  • Protease related mold was isolated and selected as a starter culture for commercial production of meju. Isolated microorganism was identified as Syncephalastrum racemosum PDA 132 2. To obtain basic data about protease for production of soybean peptides and application of the strain in meju fermentation, we extrated and purified protease and charateristics of the enzyme were investigated. The optimum condition for the production of enzyme was pH 4.0, 30oC, 5 days. The protease was purified 19.7 folds by gel filtration and ion exchange chromatography and specific activity was 12.4unit/mg. The purified enzyme was 34kDa in size, thiol protease(100% inhibited by PCMB), and was acidic protease(stable between pH 2.0~5.0). Vmax of the enzyme was 2.14 g/min which was lower(1/50) than that of by Asp. wentti and B. subtilis.

Streptomyces griseus HH1, An A-factor Deficient Mutant Produces Diminished Level of Trypsin and Increased Level of Metalloproteases

  • A-factor I a microbial hormone that can positively control cell differentiation leading to spore formation and secondary metabolite formation in Streptomyces griseus. to identify a protease that is deeply involved in the morphological and physiological differentiation of Streptomyces, the proteases produced by Streptomyces griseus IFO 13350 and its A-factor deficient mutant strain, Streptomyces griseus HH1, as well as Streptomyces griseus HH1 transformed with the afsA gene were sturdied. In general Streptomyces griseus showed a higher degree of cell growth and protease activity in proportion to its ability to produce a higher amount of A-factor. In particular, the specific activity of the trypsin of Streptomyces griseus IFO 13350 was greatly enhanced more than twice compared with that of Streptomyces griseus HH1 in the later stage of growth. The specific activity of the metalloprotease of Streptomyces griseus HH1 was greatly enhanced more than twice compared with that of Streptomyces griseus IFO 13350, and this observation was reversed in the presence of thiostreptione, However, Streptomyces griseus HH1 transformed with the afsA gene showed a significantly decreased level of trypsin and metalloprotease activity compared with that of the HH1 strain. There was no significant difference between Streptomyces griseus IFO 13350 and HH1 strain in their chymotrypsin and thiol protease activity, yet the level of leu-amionpeptidase activity was 2 times higher in Streptomyces griseus HH1 than in strain IFO 13350 . Streptomyces griseus HH1 harboring afsA showed a similar level of enzyme activity , however, all the three protease activities sharply increased and the thiol protease activity was critically increased at the end of the fermentation. When a serine protease inhibitor, pefabloc SC, and metalloprotease inhibitor, EDTA, were applied to strain IFO 13350 to examine the in vivo effects of the protease inhibitors on the morpholofical differentiation, the formation of aerial meycelium and spores was delayed by two or three days.

Conformation of Soymilk Protein Treated by Pretense (단백분해효소 처리된 두유단백질의 구조적 특성)

  • Conformation of soymilk protein was examined to obtain basic information for improved calcium intolerence of soymilk protein partially hydrolyzed with protease. Surface hydrophobicities of three proteins showed the order of SMP(soymilk protein) < SPI(soy protein isolate) < PT-SMP(protease treated soymilk protein). Total thiol group contents of SMP and PT-SMP were similar but larger than that of SPI. Reducing rate of disulfide bond in PT-SMP after 2-mercaptoethanol treatment was laster than that in SMP. And so, this result indicates that PT-SMP may be less compacting due to protease treatement. From circular dichroism result, PT-SMP showed different pattern from SMP and SPI suggesting change of secondary structure by hydrolysis. And analysis of heat denaturating property by DSC showed that denaturation enthalpy of three proteins were all small. Especially enthalpy of PT-SMP was least, and this result suggested that PT-SMP was denatured easily by heating due to less compacting structure.

Cysteine Participates in Cell Proliferation by Inhibiting Caspase3-like Death Protease

  • Reduced thiols were important compounds for the maintenance of leukemia and lymphoma cell survival (and growth). In the course of examining the microenvirn-mental effects on lymphoma and leukemia cell growth, we found that cysteine suppressed apoptosis in these cells. In a present study, in order to investigate the role of cystein on the suppression of apoptotic cell death, we used CS21, P388, and L1210 cell lines. The addition of BSO, an inhibitor of glutathione synthase, induced apoptosis of these cells by blocking the cellular uptake of cysteine in CS21 cells. Although L1210 cells underwent apoptosis without thiol compounds, the addition of these compounds suppressed the apoptosis and promoted the growth or L1210 cells. When specific inhibitors of caspase3-like proteases, but not caspase1-like proteases, were activated during the L1210 cell apoptosis but the addition of thiol compounds suppressed the activation of caspase3-like proteases. These results suggest that reduced thiols including cysteine play an important role in the suppression of cell apoptosis by inhibiting the activation of caspase3-like proteases.

  • The extra-cellular hemolysin from Bacillus thuringiensis subsp. israelensis was purified in the process of suiting with (NH$_4$)$_2$SO$_4$, Sephadex G-200 gel filtration, and DEAE-cellulose column chromatography. The purified hemolysin had molecular weight of approximately 47,000 dalton on SDS-polyacrylamide gel electrophoresis. The activity of purified hemolysin on human red blood cells was increased by thiol agents, but that was Inhibited by cholesterol, protease treatment, and metal salts such as CuSO$_4$, and FeSO$_4$, respectively.

Alkaline Protease Production from Bacillus gibsonii 6BS15-4 Using Dairy Effluent and Its Characterization as a Laundry Detergent Additive

  • Protease is a widely used enzyme particularly in the detergent industry. In this research, we aimed to isolate alkaline protease-producing bacteria for characterization as a laundry detergent additive. The screening of alkaline protease production was investigated on basal medium agar plus 1% skim milk at pH 11, with incubation at 30℃. The highest alkaline protease-producing bacterium was 6BS15-4 strain, identified as Bacillus gibsonii by 16S rRNA gene sequencing. While the optimum pH was 12.0, the strain was stable at pH range 7.0-12.0 when incubated at 45℃ for 60 min. The alkaline protease produced by B. gibsonii 6BS15-4 using dairy effluent was characterized. The optimum temperature was 60℃ and the enzyme was stable at 55℃ when incubated at pH 11.0 for 60 min. Metal ions K+, Mg2+, Cu2+, Na+, and Zn2+ exhibited a slightly stimulatory effect on enzyme activity. The enzyme retained over 80% of its activity in the presence of Ca2+, Ba2+, and Mn2+. Thiol reagent and ethylenediaminetetraacetic acid did not inhibit the enzyme activity, whereas phenylmethylsulfonyl fluoride significantly inhibited the protease activity. The alkaline protease from B. gibsonii 6BS15-4 demonstrated efficiency in blood stain removal and could therefore be used as a detergent additive, with potential for various other industrial applications.

  • This report was investigate the biological characteristic of $\delta$-endotoxin of product by TH109 strain, one strain TH109 which has toxicity on Cockroach is isolate and identification. Generally the $\delta$-endotoxin of product by 3. thuringiensis strain was easily soluble in acid, alkaline and organic solvents but $\delta$-endotoxin of product by TH109 strain are insoluble in HCI, NaOH Thiol- reagent(25mM Dithiotheritol, 25mM Dithioeryritol, 25mM Nathioglycolate, 0.2M ESCN, 2% v/v $\beta$-mecaptethanol), organic solvents( acetone, $CCI_{4}$, ether, dioxin MeOH chloroforrh xylene ), Protease. Through this study of $\delta$-endotoxin produced by TH109 strain is insoluble in acid, alkaline, organic solvents and pretense etc. In the point of view, it is greater possibility that $\delta$-endotoxin will be transform into toxin by the reducible materials instead of the reaction of protease in the intestine.
