Oleanane Triterpenoids from Gordonia ceylanica

  • Chemical investigation of hot hexane extract of the stem bark of Gordonia ceylanica afforded a new triterpenoid, $3{\beta}-acetoxy-28-hydroxyolean-12-ene$ (1) and three other oleanane triterpenoids, $3{\beta}-hydroxyolean-12-ene$ (2), $3{\beta}-acetoxyolean-12-ene$ (3), and $3{\beta}-acetoxyolean-12-en-11-one$ (4) which are new to the species. Structure of compound 1 was suggested by $^1H\;NMR,\;^{13}C\;NMR$ and MS spectral data and confirmed by converting to previously reported compound, erythrodiol diacetate (5). Structures of 2, 3 and 4 were established by comparison of the spectral data with the previously reported compounds. Further the acid hydrolysate of 4 was identical with $3{\beta}-hydroxyolean-12-en-11-one$ (6).

  • Three dihydrochalcone compounds were isolated from the twigs of Stewartia koreana (Theaceae) through repeated column chromatography. Their chemical structures were elucidated as phloridizin, 2',4,4'-trihydroxy-6'-methoxydihydrochalcone and confusoside, respectively, by spectroscopic analysis. These compounds were isolated for the first time from this plant.

A review on Camellia oleifera Abel.: A valuable material in food and medicine

  • This study aims to provide an overview of the research on the chemical composition, nutritional value, biological activities, and potential applications of Camellia oleifera seeds. Camellia oleifera Abel. (Theaceae) is a type of woody plant found in various regions, including China, Japan, India, and Southeast Asia. This plant is highly valued for its cooking oil, as the oil extracted from its seeds contains many unsaturated fatty acids (90%), mainly oleic acid (80%), and various biologically active compounds. Oil derived from C. oleifera seeds has been shown to possess numerous health benefits, such as reducing low-density lipoproteins cholesterol levels, preventing cardiovascular diseases and cancer, and regulating blood pressure. Apart from its oil, the seeds of C. oleifera also contain remarkable biological compounds that offer additional health advantages. Despite these promising attributes, C. oleifera has yet to be widely recognized as a potential source of raw materials for pharmaceutical purposes. This lack of popularity and awareness has hindered further exploration of its pharmaceutical benefits and other uses. Through this article, we hope everyone can better understand this plant and have more practical applications in the future.

동백(冬柏)을 위시한 차나무과(科) 식물(植物) 종실(種實)에 함유(含有)된 항산화제(抗酸化劑)에 관한 연구(硏究) (Studies on the Antioxidative Substances in the Seeds of Some Theaceae Family)

  • Dried seeds of Camellia japonica and Thea sinensis were investigated to determine the nature of their antioxidative activity. Activity was measured by the induction period in the coupled oxidation of a substrate lard and extracts or isolates to be tested. 70% methanol and dichloromethane extracts were found to be antioxidative abilities. Their unsaponifiables revealed weak antioxidative activity, although hexane extracts did not show antioxidative effect on lard. Column chromatography for dlchloromethane extracts gave 4 fractions(only 2 fractions were potent). HPLC was used in isolating potent antioxidative components from the column fractions and the precolumn-passed methanol extracts. They were separated into 7 and 8 components, respectively. The column fractions obtained from both seeds comprised trans-p-coumaric acid. trans-p-ferulic acid and an unknown component with minor components such as chlorogenic acid and catechin. On the other hand, the most prominent components in the methanol extracts were an unidentified component. trans-pcoumaric acid, trans-p-ferulic acid, catechin and chlorogenic acid. The unknown compound isolated from the column fractions and methanol extracts was identified as epicatechin by $^1H-and\;^{13}C-NMR$. The antioxidative activities of these components were epicatechin > catechin > chlorogenic acid > trans-p-ferulic acid > trans-p-coumaric acid.

Effect of Extraction Conditions of Green Tea on Antioxidant Activity and EGCG Content: Optimization using Response Surface Methodology

  • Green tea, the leaves of Camellia sinsneis (Theaceae), is generally acknowledged as the most consumed beverage with multiple pharmacological functions including antioxidant activity. This study was performed to analyze the effect of extraction conditions of green tea on its antioxidant effects using DPPH assay. Three extraction factors such as extraction solvent (EtOH, 0 - 100%), extraction time (3 - 15 min) and extraction temperature ($10-70^{\circ}C$) were analyzed and optimized extraction condition for antioxidant activity of green tea extract (GTE) was determined using response surface methodology with three-level-three-factor Box-Behnken design (BBD). Regression analysis showed a good fit of data and the optimal conditions of extraction were found to be 57.7% EtOH, 15 min and $70^{\circ}C$. Under this condition, antioxidant activity of experimental data was 88.4% which was almost fit to the ideal value of 88.6%. As epigallocatechin gallate (EGCG) is known for the major ingredient for antioxidant activity of green tea, we investigated the effect of EGCG on antioxidant activity of GTE. EGCG showed antioxidant activity with the $IC_{50}$ value of $4.2{\mu}g/ml$ and a positive correlation was observed between EGCG content and the antioxidant activity of GTE with $R^2=0.7134$. Interestingly, however, GTE with 50 - 70% antioxidant activity contain less than $1.0{\mu}g/ml$ of EGCG, which is much lower than $IC_{50}$ value of EGCG. Therefore, we suppose that EGCG together with other constituents contribute to antioxidant activity of GTE. Taken together, these results suggest that green tea is more beneficial than EGCG alone for antioxidant ability and optimal extraction condition of green tea will be useful for the development of food and pharmaceutical applications

Anti-Cancer Effects of Green Tea by Either Anti- or Pro-Oxidative Mechanisms

  • Tea derived from the leaves and buds of Camellia sinensis (Theaceae) is consumed worldwide. Green tea contains various components with specific health-promoting effects, and is believed to exert protective effects against diseases including cancer, diabetes and hepatitis, as well as obesity. Of the various tea components, the polyphenol catechins have been the subject of extensive investigation and among the catechins, (-)-epigallocatechin gallate has the strongest bioactivity in most cases. Our research group has postulated that hepatocyte nuclear factor-$4{\alpha}$, sterol regulatory element-binding proteins, and tumor necrosis factor-${\alpha}$ are targets of green tea constituents including (-)-epigallocatechin gallate for their anti-diabetes, anti-obesity, and anti-hepatitis effects, respectively. Published papers were reviewed to determine whether the observed changes in these factors can be correlated with anti-cancer effects of green tea. Two major action mechanisms of (-)-epigallocatechin gallate have been proposed; one associated with its anti-oxidative properties and the other with its pro-oxidative activity. When reactive oxygen species are assumed to be involved, our findings that (-)-epigallocatechin gallate downregulated hepatocyte nuclear factor-$4{\alpha}$, sterol regulatory element-binding proteins, and tumor necrosis factor-${\alpha}$ may explain the anti-cancer effect of green tea as well. However, further studies are required to elucidate which determinant directs (-)-epigallocatechin gallate action as an anti-oxidant or a pro-oxidant for favorable activity.

  • Eurya emarginata (Thunb.) Makino (Theaceae) is distributed in coastal areas of island. The leaves of Eurya are used in the traditional medicine of the coastal areas of jeju island with the aim of diuresis or to treat ulcers. Nevertheless, there are few reports on the biological activity and constituents of E. emarginata. In this study, we investigated the pharmacological activity of the solvent extracts of E. emarginata on the several inflammatory markers (TNF-$\alpha$, IL-1$\beta$, IL-6, NO, iNOS and COX-2). Also we examined the antioxidizing effect of the solvent extracts by determination of DPPH radical-scavenging activity. Among the solvent fractions, EtOAc and BuOH extracts showed potent radical scavenging activity (RC$_{50}$=10.9 and 12.7 respectively). The subtractions of EF 5-4-6-3-2 and BF 1 potentially inhibited the mRNA expression of pro-inflammatory cytokines (IL-1$\beta$, IL-6 and TNF-$\alpha$) at the concentration of 100 $\mu\textrm{g}$/mι. Also the fractions inhibited the mRNA expression of pro-inflammatory cytokines (IL-1$\beta$, IL-6 and TNF-$\alpha$) and protein expression of iNOS and COX-2 at the concentration of 100 $\mu\textrm{g}$/mι. And then, the inhibition of iNOS was correlated with the decrease of nitrite level. These results suggest that E. emarginata may have anti-inflammatory activity through the inhibition of pro-inflammatory cytokines, iNOS and COX-2.2.

한국산 비단풀목에 관한 본초학적 연구 (A Herbological Study on the Rhodophyta in Korean)

  • Objective : The purpose of this dissertation is to make a list of the Ceramiales growing naturally in the Korean coastal waters, and to carry out a survey on the current distribution status to investigate the Ceramiales which can be used as medicinal herbs. Methods : References and research papers about herbology published at home and abroad were examined. Results : The results of this study were as follows: 1. A total of 60 genera and 157 species of Ceramiales was found in the Korean coastal waters among which 5 genera and 6 species(approximately 4%) were medicinal plants. 2. Out of the 157 species of Ceramiales 67 species belonged to Rhodomelaceae, and out of the 6 species of medicinal plants 3 species belonged to Ceramiacera. 3. Among the medicinal parts 6 species belonged to algae. 2 species had flatness property, and 3 species had salty flavors. 4. As for the domestic distribution of medicinal plants, four species are distributed on the coast of the country, and Charmakji Caloglossa leprieuriii is distributed at the mouth of the Nakdonggang River and Goheung, Jeollanam-do. 5. 6 species had the efficacy of Spleen, stomach and large intestine meridian 2 species had the potency of remove phlegm, which helps to remove quench one's phlegm in human body. 6. None of the total medicinal Ceramiales were toxic. Conclusion : There were totaled to 60 genera and 157 species in Theaceae in Korea and among them medicinal plants are 5 genera, 9 species, some 4% in total.

Schima wallichii subsp. liukiuensis의 Candida종에 대한 항균효과 및 항균물질의 분리정제 (Purification of Antimicrobial Compounds and Antimicrobial Effects of Schima wallichii subsp. liukiuensis against Candida sp.)

  • 218종의 목본식물 자원으포부터 높은 항율력올 보인 Schima wallichii subsp. liukiuellsis로부터 항균활성물질을 추출, 정제하였다. Schim$\alpha$의 향균물질 추출에 가장 적합한 용매는 70% 에탄올이었으며, 계젤에 따른 항균활성은 차이를 보이지 않았고, 부위별로는 가을에 채취한 수피가 가장 좋았다. 조추출물을 유가용매로 정제하여 최종적으혹 buthanol 분획올 얻었고, 이를 silica gel과 sephadex LH-20 및 HPLC분석을 통 하여 흰색결정의 항균물질 Compound I을 얻었다. 항균물질은 UV, IR, MS분석 결과 aglycone으로 ${\alpha}$-sitosterol에 rhamnose, galactose, glucose가 1:1:1로 결합된 물질로 추정되었다. Compound I의 미생물에 대한 MIC는 3종의 bacteria에 L 1.25 g/L, 2종의 fungi에 5.0 g/L로 나타났으며, 효모에 대한 MIC는 0.04 g/L로 매우 높게 나타났다. Compound I은 천연 보존제, 의약품 등으로의 향후 개발이 기대된다.

  • Objectives : Camellia sinensis (Theaceae) possesses a various beneficial effects such as free radical-scavenging, inactivation of urokinase in cancer cell proliferation, antibacterial, and hypotensive. Dental caries is one of the most common oral infectious disease in a human. Oral microorganisms play a significant role in the etiology of dental caries. An aberration to this ecology due to dietary habits, improper oral hygiene or systemic factors lead to an increase in cariogenic microorganisms. Cariogenic microorganisms like Streptococcus mutans and Streptococcus sobrinus encourage the accumulation and adherence of plaque biofilm by metabolizing sucrose into glucans. The purpose of this study was to investigate the antimicrobial activity of phenolic compounds of Camellia sinensis and R-carvone, monoterpenes, is can be found naturally in numerous essential oils, on Streptococcus mutans and Streptococcus sobrinus .Methods : The antimicrobial activity of these compounds was determined by the broth microdilution method and checkerboard dilution assay to investigate the potential synergistic effects of each five compounds of Camellia sinensis (C. sinensis) and R-carvone.Results : C. sinensis-isolated compounds and R-carvone were determined with MIC of more than 1,000 ㎍/mL. However, the combination test showed significant synergism against S. mutans and S. sobrinus, implicated in the lowered MICs.Conclusions : These results suggest that combinatory application of phenolic five compounds (theophyllin, l-theanine, epicatechin, epicatechin gallate, and caffeine) from C. sinensis and R-carvone has a potential synergistic effect and thus may be useful as a mouthrinse in helping control cariogenic microorganism.
