Identification study of rare and high-priced natural products used for oriental medicine by RAPD analysis (RAPD 분석법을 이용한 산삼, 웅담, 녹용 등의 한약재 판별연구)

  • Natural products used for oriental medicine often come from various geographical sources, after several different distribution channels. Therefore some form of quality control procedure is required to safeguard naturl products for prescriptions purposes. To achieve this, systematic apprroaches such as morphological examination, microscopic analysis of powdered herbs and chemical analysis can be carried out. However, to ensure absolute criteria for quality assurance of natural products, DNA fingerprinting method such as RAPD(Random amplified polymorphism DNA) analysis can be used for authentication of natural products for authenticatin of natural products. In this study, warious oligonucleotide primers will be synthesized for the detection of RAPD markers and also parameters of affecting PCR(Polymerase Chain Reaction) in the detection of RAPD markers of rare and high-priced natural products will be studied with genomic DNA of chosen samples.

Study on The Anti-HSV(Herpes Simplex Virus) Activity of Natural complex Products (한약 탕제를 이용한 항 Herpes virus 제제의 개발 연구)

  • In order to search for anti-HSV agents from natural complex products, we extended the number of specimens. Both methanol extract and boiling water extract of the natural complex products were screened to detect anti-HSV activity by MTT assay. Anti-HSV activities of thirteen natural complex products extracted by methanol and boiling water were screened. Three of 13 natural complex products extracted by methanol showed efficacy against HSV. Natural complex products showing anti-HSV activities as methanol extracts were No.3, 6, 11 and their Sl were 323.809, 2811.041 and 708.20. As water boiling extracts, No.8 and No.11 have displayed Sl of 16.45 and 60.39 respectively. Especially anti-HSV activities of natural complex products extracted by methanol No.6 was stronger than other ones.

A Survey for Some Asbestos Containing Products in Korea (우리나라 일부 석면 함유제품에 대한 실태조사)

  • This study was performed to investigate the current status, in Korea, of the production of asbestos containing products (ACPs) such as asbestos cement products, asbestos friction materials, asbestos joint sheets, asbestos textile products, and other asbestos-containing products from May 2 to July 30, 2007. The information on ACPs was obtained through written questionnaires from 16 of the 27 companies that produce ACPs in Korea. The production amounts of asbestos containing gaskets were 70 tons in 2004, 90 tons in 2005, and 55 tons in 2006 in 4 companies and that of asbestos friction materials were 435.5 tons in 2004, 540.4 tons in 2005, and 454.3s ton in 2006 in 10 companies. The type and number of ACPs were:- 19 asbestos cement products, produced by 2 companies; 47 asbestos friction materials produced by 18 companies; 12 asbestos joint sheets productes by 4 companies; 18 asbestos textile products from 3 companies, and 6 other asbestos products from 5 companies. The database of ACPs was constructed to include the products name, identification number, name of company, production year, composition, asbestos content (%), usage, specification, and a picture. The database will be used to efficiently identify ACPs and to avoid asbestos exposure in workers and the general population.

  • Recent studies in molecular biology and proteomics have identified a significant number of novel diagnostic, prognostic, and therapeutic disease markers. However, validation of these markers in clinical specimens with traditional histopathological techniques involves low throughput and is time consuming and labor intensive. Tissue microarrays (TMAs) offer a means of combining tens to hundreds of specimens of tissue onto a single slide for simultaneous analysis. This capability is particularly pertinent in the field of cancer for target verification of data obtained from cDNA micro arrays and protein expression profiling of tissues, as well as in epidemiology-based investigations using histochemical/immunohistochemical staining or in situ hybridization. In combination with automated image analysis, TMA technology can be used in the global cellular network analysis of tissues. In particular, this potential has generated much excitement in cardiovascular disease research. The following review discusses recent advances in the construction and application of TMAs and the opportunity for developing novel, highly sensitive diagnostic tools for the early detection of cardiovascular disease.

Deep Learning for Herbal Medicine Image Recognition: Case Study on Four-herb Product

  • The consumption of herbal medicine and related products (herbal products) have increased in South Korea. At the same time the quality, safety, and efficacy of herbal products is being raised. Currently, the herbal products are standardized and controlled according to the requirements of the Korean Pharmacopoeia, the National Institute of Health and the Ministry of Public Health and Social Affairs. The validation of herbal products and their medicinal component is important, since many of these herbal products are composed of two or more medicinal plants. However, there are no tools to support the validation process. Interest in deep learning has exploded over the past decade, for herbal medicine using algorithms to achieve herb recognition, symptom related target prediction, and drug repositioning have been reported. In this study, individual images of four herbs (Panax ginseng C.A. Meyer, Atractylodes macrocephala Koidz, Poria cocos Wolf, Glycyrrhiza uralensis Fischer), actually sold in the market, were achieved. Certain image preprocessing steps such as noise reduction and resize were formatted. After the features are optimized, we applied GoogLeNet_Inception v4 model for herb image recognition. Experimental results show that our method achieved test accuracy of 95%. However, there are two limitations in the current study. Firstly, due to the relatively small data collection (100 images), the training loss is much lower than validation loss which possess overfitting problem. Secondly, herbal products are mostly in a mixture, the applied method cannot be reliable to detect a single herb from a mixture. Thus, further large data collection and improved object detection is needed for better classification.

An analysis of natural products patents (천연물 신약 특허 동향 분석)

  • Objectives : This study aims to investigate the current situation of domestic patents in Korea on natural products and to provide directions for developing and applying herbal medicinal products by in-depth analyses. Methods : Combinations of words, such as "A61K" or "A61P", "herb" or "Korean medicinal herb" or "Korean medicine" or "traditional" or "natural" or "$botanic^*$" or "China", and "medicine" or "treatment" or "prevention" or "improvement", were used to search patents in the World Intellectual Property Service (WIPS) database. Three methods were applied to analyze natural products patents. First, the number of patent registrations was sorted by year. Second, the assignees were analyzed by count and country of origin. Finally, cluster map analysis was conducted to explore frequently emerging words in natural products and the relationship amongst those words to treat corresponding diseases/technologies. Results : Results showed that, first, the total number of patents had been increasing. Among the patents, 76.4% were registered by domestic institutes/companies, and 23.6% by foreign institutes/companies. Second, USA, Japan, and China possess a considerable number of Korean patents and, therefore, domestic institutes/companies can seek joint technological development opportunities with their counterparts from those countries in the future. Finally, a total of four clusters were identified by cluster map analysis. Each of the clusters includes natural products related to diseases involving skin, aging, and blood sugar, as well as adult diseases. Conclusions : In this study, natural products patents registered in the Korean Intellectual Property Office were analyzed. The analyses results showed the kinds of natural products that had been employed for the treatment of certain diseases. However, natural products included in existing patents were minimal given that 4,174 species of indigenous plants are found in Korea. Thus, Korean institutes/companies should utilize unexplored plants to develop more value-added drugs.

Modulation of dendritic cell function by Trichomonas vaginalis-derived secretory products

  • Trichomoniasis caused by the parasitic protozoan Trichomonas vaginalis is the most common sexually transmitted disease in the world. Dendritic cells are antigen presenting cells that initiate immune responses by directing the activation and differentiation of naive T cells. In this study, we analyzed the effect of Trichomonas vaginalis-derived Secretory Products on the differentiation and function of dendritic cells. Differentiation of bone marrow-derived dendritic cells in the presence of T. vaginalis-derived Secretory Products resulted in inhibition of lipopolysaccharide-induced maturation of dendritic cells, down-regulation of IL-12, and up-regulation of IL-10. The protein components of T. vaginalis-derived Secretory Products were shown to be responsible for altered function of bone marrow-derived dendritic cells. Chromatin immunoprecipitation assay demonstrated that IL-12 expression was regulated at the chromatin level in T. vaginalis-derived Secretory Products-treated dendritic cells. Our results demonstrated that T. vaginalis- derived Secretory Products modulate the maturation and cytokine production of dendritic cells leading to immune tolerance.

Two Cases of Androgenetic Alopecia Patients Treated with Traditional Oriental Hair Care Products and Yukmijihwang-huan (한방모발관리 제품과 육미지황환(六味地黃丸)을 병용한 남성형 탈모 환자 치험 2례)

  • Objectives : The purpose of this study is to report the effect of traditional oriental hair care products and Yukmijihwang-huan on two androgenetic alopecia patients. Methods : Two androgenetic alopecia patients were treated by traditional oriental hair care products and Yukmijihwang-huan was prescribed to all two patients. Improvements of patients were evaluated through photographs. Results : As a result of examining photographs, symptoms of alopecia in two cases were improved. Conclusions : This study shows us that traditional oriental hair care products and Yukmijihwang-huan effect on two androgenetic alopecia patients. However, prospective study with big sample size control would be required.

Contamination Investigation and Risk Characterization on the Polycyclic Aromatic Hydrocarbon of Agricultural Products Used for Food and Medicine (식·약공용 농·임산물의 다환방향족탄화수소 오염도 조사 및 위해도 결정)

  • Polycyclic aromatic hydrocarbons(PAHs) contents were analyzed by measuring benz(a)anthracene(BaA), chrysene(Chr), benzo(b)fluoranthene(BbF) and benzo(a)pyrene(BaP), and the related risk characterization was conducted for 113 samples out of 14 different agricultural products used for food and medicine. Detection rate of PAHs was 90.3% as a whole, and the highest one was 80.5% for BaP. The detection rate of BaP exceeding the maximum permitted concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, 5.0 ㎍/kg was 1.8%, and the detection rates of BaA, Chr and BbF were within the range of 2.7~10.6%. The highest average concentration of BaA was 3.41 ㎍/kg detected from Lycii Fructus, while those of Chr, BbF, BaP and PAH4(sum of detected BaA, Chr, BbF and BaP) were 5.00, 1.79, 2.36, 12.36 ㎍/kg, respectively, detected from Rehmanniae Radix Preparata. As for the risk characterization on PAHs, the overall MOE(Margin of Exposure) values were measured within the range of 105~107, which is unlikely to cause direct health concerns, but the worring values of MOE were measured 6.57×104 for BaP and 6.10×104 for PAH4 from Rehmanniae Radix Preparata, which may require an improvement plan to reduce BaP contents.

Changes of Cytosolic $Ca^{2+}$ under Metabolic Inhibition in Isolated Rat Ventricular Myocytes

  • To characterize cytosolic $Ca^{2+}$ fluctuations under metabolic inhibition, rat ventricular myocytes were exposed to $200{\mu}M$ 2,4-dinitrophenol (DNP), and mitochondrial $Ca^{2+}$, mitochondrial membrane potential (${\Delta}{\Psi}m$), and cytosolic $Ca^{2+}$ were measured, using Rhod-2 AM, TMRE, and Fluo-4 AM fluorescent dyes, respectively, by Laser Scanning Confocal Microscopy (LSCM). Furthermore, the role of sarcolemmal $Na^+$/$Ca^{2+}$ exchange (NCX) in cytosolic $Ca^{2+}$ efflux was studied in KB-R7943 and $Na^+$-free normal Tyrode's solution (143 mM LiCl ). When DNP was applied to cells loaded with Fluo-4 AM, Fluo-4 AM fluorescence intensity initially increased by $70{\pm}10$% within $70{\pm}10$ s, and later by $400{\pm}200$% at $850{\pm}45$ s. Fluorescence intensity of both Rhod-2 AM and TMRE were initially decreased by DNP, coincident with the initial increase of Fluo-4 AM fluorescence intensity. When sarcoplasmic reticulum (SR) $Ca^{2+}$ was depleted by $1{\mu}M thapsigargin plus $10{\mu}M ryanodine, the initial increase of Fluo-4 AM fluorescence intensity was unaffected, however, the subsequent progressive increase was abolished. KB-R7943 delayed both the first and the second phases of cytosolic $Ca^{2+}$ overload, while $Na^+$-free solution accelerated the second. The above results suggest that: 1) the initial rise in cytosolic $Ca^{2+}$ under DNP results from mitochondrial depolarization; 2) the secondary increase is caused by progressive $Ca^{2+}$ release from SR; 3) NCX plays an important role in transient cytosolic $Ca^{2+}$ shifts under metabolic inhibition with DNP.
