Cell Type-Specific and Inducible PTEN Gene Silencing by a Tetracycline Transcriptional Activator-Regulated Short Hairpin RNA

  • Inducible and reversible gene silencing in desired types of cells is instrumental for deciphering gene functions using cultured cells or in vivo models. However, efficient conditional gene knockdown systems remain to be established. Here, we report the generation of an inducible expression system for short hairpin RNA (shRNA) targeted to PTEN, a well-documented dual-specificity phosphatase involved in tumor suppression and ontogenesis. Upon induction by doxycycline (DOX), the reverse tetracycline transcriptional activator (rtTA) switched on the concomitant expression of GFP and a miR-30 precursor, the subsequent processing of which released the embedded PTEN-targeted shRNA. The efficacy and reversibility of PTEN knockdown by this construct was validated in normal and neoplastic cells, in which PTEN deficiency resulted in accelerated cell proliferation, suppressed apoptosis, and increased invasiveness. Transgenic mice harboring the conditional shRNA-expression cassette were obtained; GFP expression and concurrent PTEN silencing were observed upon ectopic expression of rtTA and induction with Dox. Therefore, this study provides novel tools for the precise dissection of PTEN functions and the generation of PTEN loss of function models in specific subsets of cells during carcinogenesis and ontogenesis.

Understanding the Nuclear Technological Innovation: Focussing on the Transition Issue of 4th Generation Nuclear Energy Technology (원자력 기술혁신에 대한 고찰: 4세대 원자력 에너지기술 전환 이슈를 중심으로)

  • In this study, we conducted an in-depth literature review of the innovation characteristics of nuclear technology. We understood that the main issue may be a transition of fourth generation nuclear energy technology. We also analyzed a present status of Korean policy on nuclear energy technology and identified some implications for a successful settlement of the fourth generation nuclear energy technology. This study could provide some policy implications for maintaining the sustainable competitiveness of the fourth generation nuclear energy technology in Korea. According to our study, the factors that influence on a transition of nuclear energy technology are a policy-induced market for technical application and demonstration, stable and long-term resource allocation, constant interaction among stakeholders of innovation, accumulation of skill and know-how for an entire system. In addition, we conducted a case analysis on policy for Korean fourth generation nuclear energy technology. According to this case study, it would be necessary for Korea to pursue systematic training of human resources, active response to a social acceptance and reaction, establishment of specific plan for technical demonstration, long-term policy suggestion, and active promotion of constant interaction between stakeholders.

  • With technology rapidly changing our economic, cultural and social realities, the question of how to educate the generation for the fourth industrial revolution has been a pressing issue. The purpose of this study is to review teachers' perception regarding the educational change in the fourth industrial revolution. For this purpose, we surveyed 420 school teachers and administrators. Most teachers highly understand The Fourth Industrial Revolution and they think AI(Artificial Intelligence) is a core technology. They recognize The Fourth Industrial Revolution is very important and great influence in the education field. For this, they need educational innovation in the education field. Elementary school teacher perceive understanding Elementary school teachers think that understand The Fourth Industrial Revolution and change of curriculum is highly important. And, Secondary school teachers perceive that the change of education system and class is very necessary. Lastly, STEAM education is the most appropriate to prepare for a changing society.

Wind energy into the future: The challenge of deep-water wind farms

  • In 2019, 5.6% of the total energy produced worldwide came from wind. Offshore wind generation is still a small portion of the total wind generation, yet its growth is exponential. Higher availability of sites, larger producibility and potentially lower environmental impacts make offshore wind generation attractive. On the other hand, as the water depth increases, fixed foundations are no more viable, and the new frontier is that of floating foundations. This paper brings an overview of why and how offshore wind energy should move deep water; it contains material from the Keynote Lecture given by the first author at the ACEM20/Structures20 Conference, held in Seoul in August 2020. The paper is organized into four sections: the first giving general concepts about wind generation especially offshore, the second and the third considering economic and technical aspects, respectively, of offshore deep-water wind generation, in the fourth, some challenges of floating offshore wind generation are presented and some conclusions are drawn.

Blockchain-Based Access Control Audit System for Next Generation Learning Management (차세대학습관리를 위한 블록체인 기반의 접근제어 감사시스템)

  • With the spread of COVID-19 infections, the need for next-generation learning management system for undact education is rapidly increasing, and the Ministry of Education is planning future education through the establishment of fourth-generation NEIS. If the fourth-generation NEIS System is well utilized, there are advantages such as providing personalized education services and activating the use of educational data, but a solution to the illegal access problem in an access control environment where strict authorization is difficult due to various user rights. In this paper, we propose a blockchain-based access control audit system for next-generation learning management. Sensitive personal information is encrypted and stored using the proposed system, and when the auditor performs an audit later, a secret key for decryption is issued to ensure auditing. In addition, in order to prevent modification and deletion of stored log information, log information was stored in the blockchain to ensure stability. In this paper, a hierarchical ID-based encryption and a private blockchain are used so that higher-level institutions such as the Ministry of Education can hierarchically manage the access rights of each institution.

  • The introduction of new technologies is a result of the advent of the Fourth Industrial Revolution and the change from management environment to cloud environment. Changes in the production and management environments are expected because of changes in the external environment. However, responding flexibly to changes is not enough. The problem of the records management process, system, and infrastructure is intertwined, and partial reorganization is difficult to cope with changes in the external environment. Therefore, we need to redesign the whole process and study the major issues to be considered so we could redesign the process and produce an improved next-generation electronic records management.

Towards Sustainable Environmental Policy and Management in the Fourth Industrial Revolution: Evidence from Big Data Analytics

  • This study is to explore the relationship between the Fourth Industrial Revolution and the environment using the big data methodology. We scrutinize the trend of the Fourth Industrial revolution, in association with the environment, and provide implications for a more desirable future environmental policy. The results show that the Industrial Revolution has been generally perceived as negative to environment before the 2010s, while it has been widely regarded as positive after the period. It is highly expected that the Fourth Industrial Revolution will be capable of functioning as a new alternative to enhance the quality of the biophysical and social environment. This study justifies that the new wave of technological development may serve as a cure for the enhancement of the environmental quality. The positive linkage between the new technological development and the environment from this study clearly indicates that the environmental industry and environmental technologies will be key economic factors in the next-generation society. They should be of critical importance in shaping our cities into clearer and greener spaces, and people will continuously depend on the development of new environmental technologies in order to correct environmental damages.

The Spirit of the March First Movement and the Role of a Missional Church in the Era of the Fourth Industrial Revolution (3·1 운동 정신과 제4차 산업혁명 시대에서의 선교적 교회 역할)

  • In commemoration of the centennial anniversary of the March 1 movement in 2019, the research looked at the significance and influence of the March 1 movement on Korean history and highlighted what the church of the 4th Industrial Revolution era should play. The meaning of the March 1 movements to modern society has been identified as follows. First, the Church needs a clear sense of responsibility for society. Second, there is a need for attention to the next generation, the existing future. Third, practical education is needed for young people. Fourth, what is as important as a dreams is the value in it. Focusing on these facts, the study suggested what modern church roles should be like in the era of the Fourth Industrial Revolution. First, the church should attempt a missional approach that emphasizes the sense of belonging. Second, education for the next generation, called the Z generation, is paramount. Third, the church of the new era should be the leader of the civil society movement. Fourth, efforts are needed to realize the identity of the church. So the Church of the Fourth Industrial Revolution era should be a more missional community that reveals the kingdom of God in everyday life.

Survey of Service Industry Policy and Big Data Analysis of Core Technology in Preparation of the Fourth Industrial Revolution (4차 산업혁명에 대비한 서비스산업 정책 고찰과 핵심기술의 빅데이터 분석)

  • Countries around the world are preparing policies to promote service economy. Recently, as the fourth industrial revolution is accelerating, interest in the service industry is increasing. Korea's service industry is among the lowest among OECD countries in terms of employment, value-added and productivity, and it is time to explore new development strategies. The Korean government is establishing a service economic development strategy to promote employment and economic vitality. However, in the era of the 4th industrial revolution, the service industry is very important in that it has to be fused with the manufacturing industry. This study examines the service industry policy related to the 4th industrial revolution which the central government, local governments, and countries around the world are pursuing through literature review. The Big data analysis is used to determine the interest rate of the seven major service industries and core technologies for the fourth generation industrial revolution.
