미생물 구조 및 집단 구성원의 변화를 배양에 의존하지 않고, 빠르게 평가할 수 있는 방법의 하나인 terminal-restriction fragment length polymorphism (I-RFLP) 분석을 김치 유산균 연구에 적용하여, $15^{\circ}C$ 및 $4^{\circ}C$ 발효 김치에 관여하는 유산균의 다양성과 역동성을 검토하였다. 두 발효온도에서 공통적으로 Leuconostoc mesenteroides, Lc, inhae, Lc. kimchii, Weissella koreensis, W. cibaria, Lactobacillus sakei, Lb. curvatus, Lb. plantarum, Lb. paraplantarum, Lb. pentosus, 및 Lb. brevis의 존재가 예상되었고, Lc. citreum과 Enterococcus faecalis는 각각 $15^{\circ}C$와 $4^{\circ}C$에서 검출되었다. W. koreensis는 중기발효에 우점을 점하였고, Lactobacillus 속은 발효 후기의 우점종으로 나타났다. Lb. sakei, Lb. curvatus는 발효온도에 관계 없이 우점종을 형성하였지만, Lb. plantarum, Lb. paraplantarum, Lb, pentosus와 Lb. brevis의 생육은 저온에서 잘 일어나지 않았다. 몇 종의 Leuconostoc 속 유산균은 발효 후기까지 생장이 유지되었다.
라임병의 원인균인 Borrelia burgdorferi에 대하여 각 균종의 표준균주와 진드기에서 추출한 DNA를 template로 PCR을 실시한 후 그 증폭산물을 Alu I으로 처리한 restriction fragment length polymorphism (RFLP) 방법으로 각 균종의 연관성을 조사하고자 하였다. 표준균주로 RFLP를 실시한 결과 B. burgdorferi sensu stricto와 B. garinii의 RFLP 형태 (50 bp, 70 bp, 150 bp)가 유사하였으며 B. afzelii에서는 다른 RFLP 형태 (50 bp,110 bp,150 bp)를 관찰하였다. 그 중 B. afzelii KK-1과 B. garinii HPI은 새로운 RFLP형태를 보여 B. afzelii자 B. garinii는 각각 2 type의 subgroup으로 분류할 수 있었다. 진드기 DNA에서는B. afzelii를 포함한 각 균종에 대하여 모두 유사한 RFLP형태를 보였는데, 진드기 DNA에서 확인된 B. afzelii는 KK-1과 같은 군에 속하는 것으로 사료되었다.
mRNA의 differential display방법에 의해 여름느타리 자실체의 상처 또는 자외선 처리시 발현되는 약 0.4kb의 cDNA 단편을 분리하였다. 이 cDNA 단편의 염기서열 분석결과 세포분열 촉진에 관여하는 cdc2-related protein kinase gene과 상당부분 유사성을 보였으며 RT-PCR 방법을 이용한 분리 유전자의 발현 실험을 통해 이 유전자가 정상 생장 환경에서는 갓, 대, 균사 등 모든 부위에서 기본적인 발현상태를 유지하고 있으며 기계적 상처 또는 자외선 처리에 의해 그 발현이 증폭됨을 확인하였다. 이러한 결과를 통해 향후 분리된 유전자의 연구를 통한 버섯 병 방어 관련 신호 전달 체계 분석에 대한 가능성을 검토해 보았다.
참게(Eriocheir sinensis)와 꽃게(Portunus trituberculatus)의 2종으로부터 genomic DNA를 분리 추출하였다. 선택된 7개의 OPA-05, OPA-13, OPA-16, OPB-06, OPB-15, OPB-17 and OPD-10의 RAPD primer를 이용하여 identical, polymorphic 그리고 specific fragment를 얻어냈다. 본 연구에서 부안산 참게 집단에서는 505개의 fragment가 나타났고, 꽃게 집단에서는 513개의 fragment가 확인되었다. 참게 집단에서는 165개의 identical fragment가 나타났으며, 이는 primer당 평균적으로 23.6개의 fragment로 확인되었다. 또한 꽃게 집단에서는 66개로서 평균해서 primer당 9.4개의 identical fragment가 나타났다. 참게 집단과 꽃게 집단의 polymorphic fragment는 각각 50개와 14개로 나타났고, 참게 집단과 꽃게 집단의 경우 OPB-17에서 identical fragment가 300 bp의 크기에서 확인되었다. 각각을 비교해 보았을 때 유전적 차이는 참게 집단에서보다 꽃게 집단에서 더 높은 수치를 나타내었고, 2종 사이에서 $0.726{\pm}0.004$의 수치를 나타내었다. 7개의 primer를 사용하여 얻어진 dendrogram은 cluster 1(FRESHWATER 01), cluster 2(FRESHWATER 02, 03, 04, 05 및 06), cluster 3(FRESHWATER 07, 08, 09, 10 및 11) 및 cluster 4(SWIMMING 12, 13, 14, 15, 16, 17, 18, 19, 20, 21 and 22)와 같이 4개의 유전적 클러스터로 나뉘어졌다. 꽃게 집단에서 18번째 개체(SWIMMING no. 18)와 17번째 개체 (SWIMMING no. 17) 사이가 가장 가까운 유전적 관계(genetic distance 0.096)를 나타내었다. 궁극적으로 볼 때 참게 집단의 2번째(FRESHWATER no. 02)와 참게 집단의 3번째(FRESHWATER no. 03) 개체 사이가 가장 먼 유전적 거리 (genetic distance=0.770)를 나타내었다. 위에서 언급했던 것처럼 RAPD-PCR 방법은 참게 및 꽃게 2종의 종 판별을 하기 위한 진단적 표지 (diagnostic marker)로 이용할 수 있는 잠재력을 가지고 있는 것으로 확인되었다.
방사선 피폭선량의 예측을 위한 방사선 민감 지표 모델 개발을 위하여 코발트-60 감마선과 의료용 싸이크로트론 50 MeV($p{\to}RBe^+$) fast neutron을 0.25 Gy에서 1 Gy의 선량을 랫드에 각각 전신 조사한 후 말초혈액내 임파구와 소장에 음와세포의 형태학적 변화를 apoptotic fragment assay법을 이용하여 관찰하였다. 모든 방사선조사군에서 음와세포와 말초 임파구에 아포토시스의 유도가 증가된 것이 관찰되었으며, 이것은 방사선이 방사선 민감세포의 아포토시스 유도를 자극한 것으로 보인다. 상기의 결과는 아포토시스가 손상된 세포를 제거하므로 손상된 방사선 민감 표적 장기의 항상성 유지에 중요한 역할을 하는 것으로 판단되었다. Apoptotic fragments의 발생빈도에 대한 선량-반응곡선에 있어서 음와세포는 중선자 조사군이 $y=0.3+(6.512{\pm}0.279)D(r^2=0.975)$으로, 반면에 감마선 조사군은 $y=0.3+(4.435{\pm}0.473)D+(-1.300{\pm}0.551)D^2(r^2=0.988)$의 식을 얻었다. 그리고 말초 임파구에서는 감마조사군이 $y=3.5+(118.410{\pm}10.325)D+(-33.548{\pm}12.023)D^2(r^2=0.992)$의 식으로 나타났다. 이와 같이 감마선조사군은 공히 linear quadratic model 이였으나 중성자조사군은 linear model 로 관찰되었다. 조사된 세포의 종류와 상관없이 apoptotic fragments의 발생빈도와 조사 선량간의 유의한 효과가 있는 것으로 확인되었다. 이상의 결과에서 조사선량의 증가와 비례하여 방사선 민감 세포의 apoptotic fragments가 수적으로 증가하였으며, 고준위 방사선이 저준위 방사선보다 선량 반응 곡선과 시간 경과에 따른 영향이 보다 강한 것으로 인지되었으며, 음와세포의 apoptosis 유도에 대한 중성자선의 방사선 생물학적 효과비(RBE)는 1.919 였다. 그리고 모든 방사선조사군에서 방사선피폭 후 4시간과 6시간에 apoptosis 유도가 가장 많았으며, 음와세포의 형태학적 소견은 정상 대조군에서 관찰되지 않는 전형적인 apoptotic fragments가 나타났다. 따라서 음와 세포와 말초 임파구에서의 아포토시스 유도는 방사선 조사에 의한 세포 손상의 생물학적 영향 평가를 위한 검색 및 방사선 피폭선량 예측의 지표로 이용 가능할 것으로 사료됨.
The structural stability of brain pyrydoxine-5'-phosphate (PNP) oxidase and the catalytic properties of the monomeric species were investigated. The unfolding of brain pyridoxine-5'-phosphate (PNP) oxidase by guanidine hydrochloride (GuHCl) was monitored by means of fluorescence and circular dichroism spectroscopy Reversible dissociation of the dimeric enzyme into subunits was attained by the addition of 2 M GuHCl. The perturbation of the secondary structure under the denaturation condition resulted in the release of the cofactor FMN. Separation of the processes of refolding and reassociation of the monomeric species was achieved by the immobilization method. Dimeric PNP oxidase was immobilized by the covalent attachment to Affi-gel 15 without any significant lass of its catalytic activity. Matrix-bound monomeric species were obtained from the reversible refolding processes. The matrix bound-monomer was found to be catalytically active, possessing only a slightly decreased specific activity when compared to the refolded dimeric enzyme. In addition, limited chymotrypsin digestion of the oxidase yields two fragments of 12 and 161 kDa with a concomitant increase of catalytic activity The catalytically active fragment was isolated by ion exchange chromatography and analyzed for association of two subunits using the FPLC gel filtration analysis. The retention time indicated that the catalytic fragment of 16 kDa behaves as a compact monomer. Taken together, these results are consistent with the hypothesis that the native quaternary structure of PNP oxidase is not a prerequisite for catalytic function, but it could play a role in the regulation.

Objective: This study was conducted to screen scrotal hernia in domesticated swine from selected breeders in the Philippines. This defect is associated with a cytosine to thymine mutation in the BCL-2 associated X protein (BAX) gene of swine. Methods: Genetic screening was done by DNA extraction followed by amplification and digestion using polymerase chain reaction-restriction fragment length polymorphism, amplifying the 416 bp region of the BAX gene that was subjected to digestion using the Ear I enzyme. Sequencing was also conducted to validate the results. Results: Results revealed that out of 538 samples tested, 411 (76.4%) of the samples were found to be normal whereas the remaining were carriers of the mutation in which 80 (14.9%) were heterozygous mutants and 47 (8.7%) were homozygous mutants. Pietrain breed was found to have the highest incidence. Conclusion: Having a scrotal hernia eliminates the chances of using the boar as a breeder stock because the following generations arising from it would most likely exhibit herniation. It is therefore advised to establish a genetic screening method for Scrotal Hernia in the Philippines to eliminate the negative gene from the herd.
In the present paper, the impact damage behavior of steel plate reinforced concrete panels exposed to shock impulsive loading and fragment impact loading is investigated. To evaluate the retrofit performance of a steel-strengthened concrete panels, a numerical experiment using a numerical simulation with AUTODYN, an explicit analysis program is introduced because a real explosion experiment requires the vast investment and expense for facilities as well as the deformation mechanisms are too complicated to be reproduced with a conventional closed-form analyses. The model for the analysis is simplified and idealized as a two-dimensional and axisymmetric case controled with geometry, boundary condition and material properties in order to obtain a resonable computation time. As a result of the analysis, panels subject to either shock loading or fragment loading without the steel plate reinforcement experience the perforation with spalled fragments. In addition, the panels reinforced with steel plate can prevent the perforation and provide the good mechanical effect such as the increase of global stiffness and strength through the composite action between the concrete slab and the steel plate.
The genus Streptomyces is intensively studied due to its excellent ability to produce secondary metabolites with diverse bioactivities. In particular, adequate precursors of secondary metabolites as well as sophisticated post modification systems make some high-yield industrial strains of Streptomyces the promising chassis for the heterologous production of natural products. However, lack of efficient genetic tools for the manipulation of industrial strains, especially the episomal vector independent tools suitable for large DNA fragment deletion, makes it difficult to remold the metabolic pathways and streamline the genomes in these strains. In this respect, we developed an efficient deletion system independent of the episomal vector for large DNA fragment deletion. Based on this system, four large segments of DNA, ranging in length from 10 kb to 200 kb, were knocked out successfully from three industrial Streptomyces strains without any marker left. Notably, compared to the classical deletion system used in Streptomyces, this deletion system takes about 25% less time in our cases. This work provides a very effective tool for further genetic engineering of the industrial Streptomyces.
The objective of this study was to develop a simplified, efficient, and accurate protocol for sexing goat embryos. Based on the amelogenin gene located on the conservation region of X- and Y- chromosomes, a pair of primers was utilized and the system of PCR was established to amplify a 262 bp fragment from the X- chromosome in female goats, and a 262 bp fragment from X- chromosome and 202 bp fragment from the Y- chromosome in male goats, respectively. The accuracy and specificity of the primers were assessed using DNA template extracted from goat whole blood sample of known sex. 100% (10/10) concordance was obtained by using the PCR assay. Fifty-one biopsied embryos were transferred into 25 recipient goats on the same day that the embryos were collected and sex of the kid was confirmed after parturition. Eighteen kids of predicted sex were born. The biopsied samples from 51 goat embryos were amplified with 100% efficiency and 94.7% accuracy. In conclusion, our results indicated that PCR sexing protocols based on the amelogenin gene is highly reliable and suitable for sex determination of goats.
