동결기간과 해동 후 냉장이 우육의 기호성에 미치는 영향 (Effects of Freezing Period and Chilling Process after Tawing on the Palatability of Beet Loin)

  • 동결기간과 해동 후 재냉장이 우육의 기호성에 미치는 영향을 검토하기 위하여 도축 후 1일 냉장한 홀 스타인 등심육을- 2$0^{\circ}C$에 동결하여 20, 40 및 60일째에 해동하는것과 이것을 1$^{\circ}C$에 재냉장한 것의 전단력가, 부수성, 액토미 오신의 추출성과 ATPase 활성에 대한 실험과 함께 관능평가를 실시하였다. 우육의 전단력가능 동결 중 변화가 없었으며 해동 후 재냉장에 의하여 낮아졌다. 부수성은 동결기간이 길수록 낮아지고 재냉장에 의한 유의적인 차이를 보이지 않았다. 액토미오산의 추출성과 Ma-ATP-ase 활성은 동결기간이 길수록 낮아지고 재냉장에 의하여 높아지고 경향을 보였다. 재냉장에 의한 기홍성 향상 효과는 동결기간이 짧을수록 크게 나타났다. 재냉장에 의하여 기호성이 우수하게 된 것은 조직감과 가열육향의 향상이 크게 관련되었다.

포유류 초기 배아의 효율적인 동결 보존 방법에 관한 연구 (Development of Effective Cryopreservation Method for Mammalian Embryo)

  • 본 연구에서 생쥐 초기 배아를 이용하여 동결 보존된 배아의 생존율 또는 발달율에 영향을 미치는 요인들의 상관관계를 알아본 결과, 배아의 발달 단계에 따른 동결 - 해동 후 생존율에 있어서는 2세포기 배아가 4~8세포기 배아보다 유의하게 높았으나(p<0.01), 배 발달율에 있어서는 4~8세포기 배아가 유의하게(p<0.01), 높아 생존율과 배 발달율과는 상관관계가 없는 것으로 사료된다. 또한, 동결 보호제에 따른 배아의 생존율에 있어서는 2, 4~8세포기 배아 모두 DMSO에서 유의하게 높았으나(p<0.01), 배 발달율에 있어서는 EG가 DMSO에 비해 더 유의하게 높은 성적을 나타내어(p<0.01, p<0.05) 동해로 인한 상해가 적은 것으로 생각되어 2, 4~8세포기 배아에서 EG가 더 효과적인 동결 보호제로 사료되며, 동결 프로그램으로는 완만 동결 - 급속 해동법이 더 우수한 프로그램으로 보인다.

Delayed Deproteinization Causes Methodological Errors in Amino Acid Levels in Plasma Stored at Room Temperature or -20℃

  • Deproteinization has been recognized as a prerequisite for amino acid analysis of plasma samples. For plasma stored at room temperature, delaying deproteinization for 30, 60 or 120 minutes did not result in significant changes in the mean CV (coefficient of variation), which ranged from 4.4 to 5.6%. However the mean CV of aspartic acid, ${\alpha}$-aminoadipic acid, alanine and lysine was about 10%. When the plasma was stored frozen at -20${^{\circ}C}$, the CV was increased at 0 and 120 minutes after thawing, to 12.4% (range, 4.1 to 35.3%) and 8.0% (2.5 to 30.7%), respectively. The concentrations in plasma during storage at room temperature of all the amino acids analyzed showed significant changes. In plasma stored for 30 minutes at room temperature, 17 amino acids increased in concentrations and two decreased. Extending this period to 60 or 120 minutes increased the instability as compare to the reference group. Storing plasma at -20${^{\circ}C}$ for 2 weeks resulted in significantly greater changes in the amino acid concentrations than at room temperature. On extending the storage time at room temperature, after thawing, to 30, 60, and 120 minutes, 21, 20, and all 22 amino acids respectively changed significantly (p<0.01). The present study indicates that methodological errors occur in the concentrations determined for all amino acids when plasma is left at room temperature. The storage of frozen non-deproteinized plasma accompanied more significant changes in most amino acid concentrations and thus should be avoided. Deproteinization should be performed as soon as possible after plasma collection.

경남지역 위탁급식업체 조리종사자의 위생교육 및 근무환경에 따른 위생 직무수행도 평가 (A Study on the Hygiene Practices of Foodservice Employees by Hygiene Education and Work Environment in the Gyeongnam Area)

  • This study examined the hygiene practices of contract foodservice employees and investigated the influence of education and work environment on these hygienes practices. A questionnaire was distributed to 250 contract foodservice employees and a total of 232 responses were received and analyzed. The overall score for hygiene practices of contract foodservice employees was 3.89 based on a 5-point scale. Cross-contamination was prominent (highest score at 4.46) and the heating temperature was less prominent (lowest score at 3.49). The factors most affecting contract foodservice employees were their work period, the size of the contract foodservice management company, the number of meals served daily, the frequency of meal service per day and the frequency of hygiene education. Compared to small and medium-sized contract foodservice management companies, the major contract foodservice management companies showed higher scores for refrigerator/freezer control (P<0.001), vegetable/fruit disinfection (P<0.001), thawing (P<0.001), heating temperature (P<0.001), cleaning/disinfection (P<0.01), and personal hygiene (P<0.05). The frequency of hygiene education had a significant effect on the performance levels for refrigerator/freezer control (P<0.001), vegetable/fruit disinfection (P<0.001), thawing (P<0.001), heating temperature (P<0.001), cleaning/disinfection control (P<0.001), food supply control (P<0.05), and personal hygiene (P<0.05). From these results, to increase the sanitation quality of contract foodservice operations, hygiene practice levels need to increase and hygiene education systematically should be enforced for foodservice employees.

Chromosomal Modification in Human Embryonic Stem Cells Cultured in a Feeder-Free Condition after Single Cell Dissociation using Accutase

  • Human embryonic stem (ES) cells are a potential source of cells for developmental studies and for a variety of applications in transplantation therapies and drug discovery. However, human ES cells are difficult to culture and maintain at a large scale, which is one of the most serious obstacles in human ES cell research. Culture of human ES cells on MEF cells after disassociation with accutase has previously been demonstrated by other research groups. Here, we confirmed that human ES cells (H9) can maintain stem cell properties when the cells are passaged as single cells under a feeder-free culture condition. Accutase-dissociated human ES cells showed normal karyotype, stem cell marker expression, and morphology. We prepared frozen stocks during the culture period, thawed two of the human ES cell stocks, and analyzed the cells after culture with the same method. Although the cells revealed normal expression of stem cell marker genes, they had abnormal karyotypes. Therefore, we suggest that accutase-dissociated single cells can be usefully expanded in a feeder-free condition but chromosomal modification should be considered in the culture after freeze-thawing.

동결/융해에 따른 폐기물 매립지 복토층 연구 (A Study of Landfill Coyer Liners by Freezing/Thawing)

  • 일반 및 특정폐기물 매립지에서 복토층의 중요성은 매립지의 바닥층 만큼 강조되지는 않는 것 같다. 그러나 실제로 매립지의 파괴 원인중에 가장 커다란 영향을 미치는 것은 복토층 설치의 실패에서 온다고 볼 수 있다. 특히 복토층 기능은 우수를 지표면으로 유출증진하여 매립장 안으로 침투 억제시키며, 폐기물의 노출시 자연환경 위생에 대하여 완충작용을 하며, 매립지의 침하 및 침강을 억제하는데 있다. 본 연구는 겨울철 동결/융해에 따른 폐기물 매립지 최종복토의 거동을 수행하였으며, 폐기물 매립지에서와 같은 조건을 부여하기 위해 거대한 Lysimeter를 설치하여 실제로 최종복토에 쓰여지는 물질로 세가지 실험을 수행하였다. 실험결과는 동결/융해에 따른 점토층의 변화를 묘사하고 있으며 또한, 매립지에서의 동결깊이에 따른 복토층의 파괴는 점토의 물리 적, 공학적인 측면에 영향을 주며 이러한 영향은 매립지 설계시 고려되어야 한다고 본다. 본문은 실험에 사용되어진 복토층의 물질, 복토층의 묘사와 그들의 실험결과에 대한 결과분석 및 결론을 설명하고 있다.

Ethylene Glycol을 이용한 유리화 동결시 평형시간과 배 발달단계별 생쥐 배의 생존성 (Effect of Equilibration Tine and Developmental Stages on the Survival of Mouse Embryos Cryopreserved by Vitrification in EFS Solution)

  • The present experirnents on cryopreservation were carried out to investigate effect of solution toxicity, equilibration time and cell stages on the post-thaw survival of mouse morulae and blastocyst embryos cryopreserved by vitrification in EFS solution. The mouse embryos were exposed to the EFS solution in one step at room temperature, kept in the EFS solution during different period for toxicity test, vitrified in liquid nitrogen and thawed rapidly. After the mouse morulae embryos were exposed to EFS solution for 2 and 5 ruin. at room temperature and then they were washed in 0.5 M sucrose solution and basal mediurn(D-PBS + 10% FCS), they were cultured to examined cryoprotectant toxicity induced injury during exposure, most of embryos developed to expanded blastocysts(100 and 90.0%). However, when the exposure time was extended to 10 and 20 min, these development rates dropped dramatically in 10 ruin. (75.0%) and 20 ruin. (4.5%), respectively. When the compacted morulae were vitrified in EFS solution after equilibration for 2 and 5 min, the embryos have developed to normal blastocyst following thawing, washing and culture processes was 89.3 and 89.6%. However, when the exposure time was expanded to 10 ruin, this survival rate dropped to 68.8%. When the blastocyst were vitrified in EFS solution after equilibration for 2, 5 and 10 minutes, the survival rate of embryos which developed to normal blastocyst following thawing and culture processing were 58.5, 46.7 and 22.4%, respectively. The optimal time of equilibration of mouse morula and blastocysts in EFS solution seemed o be 2 and 5 ruin.

냉동 및 레토르트식품에서 상업적 변성전분의 적용에 관한 연구 (The Study for Application of Commercial Modified Starch to Frozen and Retort Foods)

  • For the application of the commercially modified starch in frozen and retort foods, apparent viscosity and water loss were measured at each stages of heating, sterilization and freezing-thawing stages. Apparent viscosity showed the exponential increase with concentration and the slopes of apparent viscosity against concentration in the Firm-Tex(hydroxypropyl distarch phosphate), Hi-Flo(acetylated distarch adipate) and Colflo 67(acetylated distarch adipate) from waxy maize starch were higher than those of Amyloacetate M20(starch acetate) and X-amylo 250(distarch phosphare) from potato starch. In the presence of 1 or 2% NaCl, X-amylo 250 among modified starches showed the increase in water loss and the decrease in apparent viscosity, whereas Colflo 67, Hi-Flo and Firm-Tex were little affected by NaCl. In the presence of 1 or 2% sucrose, water loss and apparent viscosity of the modified starches were not affected. In the range of pH 4~8, water loss and apparent viscosity of the modified starches had no change but the differences were detected to some extent between each of heating, sterilization and freezing-thawing stages. In the apparent viscosity and water loss of the modified starches after 3 week storage from heating and sterilization, Hi-Flo, Amyloacetate M20 and X-amylo 250 were not changed at the storage period, and the overall acceptability of retort food containing the modified starches such as Firm-Tex and Amyloacetate M20 were favored more than others. In the apparent viscosity and water loss of the modified starches which have been frozen and thawed three times repeatedly, Colflo 67, Hi-Flo and Firm-Tex were not changed in freezingthawing, and the overall acceptability of frozen food containing Firm-Tex was most favored.

Quality Evaluation of Mackerel Fillets Stored under Different Conditions by Hyperspectral Imaging Analysis

  • This study was designed to compare the quality changes in mackerel fillets stored under different conditions by using hyperspectral imaging (HSI) techniques. Fillets packaged in vacuum were stored for six days under five different conditions: refrigerated at 4℃ (R group); iced at 5±3℃ (I group); kept at an ambient of 17±2℃ (A group); frozen at -18℃ for 24 h and thawed in a refrigerator at 4℃ for 5 h on the sampling day (FTR group); FTR thawed in tap water instead of thawing in a refrigerator (FTW group). The FTR group had the lowest total bacterial count, drip loss, 2-thiobarbituric acid reactive substances, volatile basic nitrogen, and texture profile analysis values among groups during the entire storage period (p<0.05). Scanning electron microscopy revealed that the FTR group had less damage, while the other groups had shrunken muscle tissues. HIS integrated with the partial least squares model yielded reliable and efficient results, with high R2cv values, for several quality parameters of the mackerel fillets. Overall, the FTR group, involving freezing and thawing in a refrigerator, appears to be the most favorable option for maintaining the quality of mackerel fillets, which could be practically implemented in the industry. HSI is a suitable and effective technique for determining the quality of mackerel fillets stored under different conditions.

Cryopreservation of mesenchymal stem cells derived from dental pulp: a systematic review

  • Objectives: The aim of the present systematic review was to investigate the cryopreservation process of dental pulp mesenchymal stromal cells and whether cryopreservation is effective in promoting cell viability and recovery. Materials and Methods: This systematic review was developed in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement and the research question was determined using the population, exposure, comparison, and outcomes strategy. Electronic searches were conducted in the PubMed, Cochrane Library, Science Direct, LILACS, and SciELO databases and in the gray literature (dissertations and thesis databases and Google Scholar) for relevant articles published up to March 2019. Clinical trial studies performed with dental pulp of human permanent or primary teeth, containing concrete information regarding the cryopreservation stages, and with cryopreservation performed for a period of at least 1 week were included in this study. Results: The search strategy resulted in the retrieval of 185 publications. After the application of the eligibility criteria, 21 articles were selected for a qualitative analysis. Conclusions: The cryopreservation process must be carried out in 6 stages: tooth disinfection, pulp extraction, cell isolation, cell proliferation, cryopreservation, and thawing. In addition, it can be inferred that the use of dimethyl sulfoxide, programmable freezing, and storage in liquid nitrogen are associated with a high rate of cell viability after thawing and a high rate of cell proliferation in both primary and permanent teeth.
