THE EFFECTS OF UREA NITROGEN ON THE METABOLISM OF PLANTS (II) The response of some nitrogen components of barley to urea and other nitrogen in water culture.

  • For the comparison with the previous paper (4) the present report deals with the absorption and metabolism of urea and other nitrogen ions in barley seedling absorbed through root. 1. The amount of nitrate in barley treated with urea reach it peak on the 8th day. NO3 on the 4th, NH4 on the 6th or 8th, respectively. 2. The ammonia content in urea group reaches its peak on the 6th day but other groups on the 4th day. The present data in the urea group show to shorten 4 days compared with that of the previous paper(4). 3. the content of total amide from the present data aare gradully increased on all of the groups during this experiment. These are agreement with the result of the previous paper (4). 4. the alcohol solution nitrogen in the urea gorup shows the similar tendency to the NaNO3 group but reaches it peak 2 days later than in the (NH4)2SO4 group. 5. The content of total nitrogen in the urea series has the lowest amount at the beginning while the richest from the 4th day after. These would be explained on that the absorption of urea is delayed and the PH in the urea solution does not change, so called "physiological neutrality". LP-BM5 Leukemia Retrovirus 감염으로 유발된 Murine AIDS에서 Dehydroepiandrosterone Sulfate의 Th1/Th2 Cytokines의 발현 조절 및 산화적 스트레스 억제 효과 연구 (Dehydroepiandrosterone Sulfate Inhibited Immune Dysfunction Induced by LP-BM5 Leukemia Retrovirus Infection through Regulating Th1/Th2 Type Cytokine mRNA Expression and Oxidative Stress in Murine AIDS Model)

  • AIDS로의 진행과정은 단순히 몇몇 기작에 의한 것이 아니라 여러 경로를 거쳐 발생하며 그 진행속도도 개개인에 따라 다양하게 나타나고 있다. 본 연구에서는 쥐 AIDS 모델을 이용하여 항산화호르몬으로 알려진 DHEAS의 면역조절 효과를 확인하고자 하였다. DHEAS의 투여는 LP-BM5 retrovirus 감염으로 인한 T와 B 임파구의 mitogenesis를 증가시켰으며 Th1/Th2 type cytokines의 발현에도 영향을 미쳤는데 주로 전사수준에서 작용한 것으로 생각된다/ 또한 retrovirus 감염으로 인한 간조직의 지질과산화 유발을 억제하여 조직 내의 항산화제인 vitamin E의 함량을 유지시킴으로 NF-kB의 활성화를 통한 retrovirus의 복제를 억제시켜 Th1/Th2 type cytokines의 불균형적인 발현을 저해하였을 것으로 여겨진다. 따라서 쥐 AIDS모델에 있어서 DHEAS의 면역조절 효과를 확인한 본 연구는 향후 HIV감염 이후 AIDS로의 진행 과정 시 항산화제의 역할과 기작 규명에 대한 가능성을 제시할 수 있을 것으로 생각된다.

Ochnaflavone의 면역보조제 효과 (Effect of Ochnaflavone as An Immunoadjuvant)

  • In this present study, we determined whether or not there is an immunoadjuvant effect of ochnaflavone, a biflavone isolated from Lonicera japonica. As an antigenic source, the cell wall (CACW) of Candida albicans, a fungal pathogen, was used. CACW consists of 95% carbohydrate (mannan). In the experiments, BALB/c mice were immunized with emersion forms of CACW combined with or without ochnaflavone (Och) in the presence of IFA containing mineral oil or CACW alone. Then, the amounts of antisera collected from these mice groups were measured by the ELISA method. Data from these experiments showed that CACW combined with Och (CACW/Och/IFA) provoked the production of antisera app. 2.2 or 5 times more than the corresponding CACW/IFA or CACW alone (CACW/DPBS), respectively, in mice (P<0.05). We further examined the immune response type induced by Och. Analysis of the values of the IgG1/IgG2a ratios obtained from IgG isotyping revealed that Och induced Th2-immunity more dominantly than Th1. This finding was confirmed by cytokine profile. CACW/Och/IFA formulation induced IL-4 (Th2-type cytokine) more than IFN${\gamma}$ (Th1-type cytokine) as compared with CACW/IFA and CACW/DPBS formulations (P<0.05). All data combined, Och appears to have an immunoadjuvant activity that may convert Th1 immunity into Th2 immunity.

Sulforaphane inhibits the Th2 immune response in ovalbumin-induced asthma

  • Sulforaphane (1-isothiocyanato-4-(methylsulfinyl)-butane), belonging to a family of natural compounds that are abundant in broccoli, has received significant therapeutic interest in recent years. However, the molecular basis of its effects remains to be elucidated. In this study, we attempt to determine whether sulforaphane regulates the inflammatory response in an ovalbumin (OVA)-induced murine asthma model. Mice were sensitized with OVA, treated with sulforaphane, and then challenged with OVA. Sulforaphane administration significantly alleviated the OVA-induced airway hyperresponsiveness to inhaled methacholine. Additionally, sulforaphane suppressed the increase in the levels of SOCS-3 and GATA-3 and IL-4 expression in the OVA-challenged mice. Collectively, our results demonstrate that sulforaphane regulates Th2 immune responses. This sutdy provides novel insights into the regulatory role of sulforaphane in allergen-induced Th2 inflammation and airway responses, which indicates its therapeutic potential for asthma and other allergic diseases.

성견에서 치아회분말의 하악골체부 매식시 치유과정에 관한 실험적 연구 (AN EXPERIMENTAL STUDY ON THE TISSUE REACTION OF TOOTHASH IMPLANTED IN MANDIBLE BODY OF THE MATURE DOG)

  • The purpose of this study is to evaluate whether the ashed tooth powder is utilized as an alternative material of the implant. For this purpose the author performed the experimental study to investigate the tissue response of sintered toothash and its histocompatibility. Bony defects to expose the body of marrow, $1{\times}1cm$ in size, were created in the right and left mandibular body of mature dog, and then the ashed tooth powders were filled in right side and the blood clot was filled in the left side as an control. The dogs were sacrificed at 4th, 7th, and 16th week after implantation and histologic examination was performed. The results of this study were obtained as follows : 1. Any inflammatory response was not noted after implanting of the ashed tooth powder during the whole experimental period. 2. At 4th week, ashed tooth powders were surrounded by mature connective tissue. And we could observe hydroxyapatite crystal structure within the ashed tooth powder. 3. At 7th week, we could observe that macrophage phagocyted the small granules of ashed tooth powders. 4. At 16th week, the union with host bone by growth of new trabeculae was observed. And there were remnants of ashed tooth power within some of new trabeculae.

Modulation of Immune Response Induced by Co-Administration of DNA Vaccine Encoding HBV Surface Antigen and HCV Envelope Antigen in BALB/c Mice

  • Plasmid DNA vaccines encoding the hepatitis B virus (HBV) surface and hepatitis C virus (HCV) envelope antigens, respectively, were constructed, and attempt were made to find the possibility of a divalent vaccine against HBV and HCV. The expression of each plasmid in Cos-1 cells was confirmed using immunocytochemistry. To measure the induced immune response by these plasmids in vivo, female BALB/c mice were immunized intramuscularly with $100\;{\mu}g$ of either both or just one of the plasmids. Anti-HBV and HCV-specific antibodies and related cytokines were evaluated to investigate the generation of both humoral and cellular immune responses. As a result, specific anti-HBV and anti-HCV serum antibodies from mice immunized with these plasmids were observed using immunoblot. The levels of IL-2 and RANTES showing a $Th_{1}$ immune response were significantly increased, but there was no change in the level of IL-4 ($Th_{1}$ immune response) in any of the immunized groups. Compared with each plasmid DNA vaccine, the combined vaccine elicited similar immune responses in both humoral and cell-mediated immunities. These results suggest that the combined DNA vaccine can induce not only comparable immunity experimentally without antigenic interference, but also humoral and $Th_{1}$ dominant cellular immune responses. Therefore, they could serve as candidates for a simultaneous bivalent vaccine against HBV and HCV infections.

  • 고체 로켓 추진기관 노즐의 내열재로 사용되는 탄소/페놀릭 복합재료의 열반응 수치해석을 수행하였다. 본 논문에서 탄소/페놀릭 재료의 열반응 해석은 (1) 로켓 노즐벽에서 대류열전달계수를 구하기 위한 연소가스의 경계층 적분방정식 수치해석과 (2) 삭마두께, 숯깊이 및 온도를 계산하기 위한 탄소/페놀릭의 열반응(열분해, 삭마)을 고려한 1차원 열전도 해석으로 구성된다. 시험결과와 해석결과를 비교 분석하였으며, 목삽입재 좌우 인접 부위를 제외하고 잘 일치하는 것을 확인 할 수 있었다.

  • 한국연안도 해상교통량 증가, 선원자질저하 및 노후선박증가로 수만톤을 초과하는 기름유출사고의 위험이 상존하고 있다. 이러한 대형 오염사고 발생에 효과적으로 대응하기 위한 선진 방제체제의 구축방안은 다음과 같다. 1. 범국가적 차원의 국가긴급계획을 효율적으로 실행하기 위한 과학적인 지역긴급계획의 수립 2. 2만톤의 축가방제능력 조기 확보 3. 해양오염방제 전문교육ㆍ훈련의 강화 4. NOWPAP사업 추진등 인접국가간 방제협력체제의 구축 21세기 방제정책은 선진 방제체제를 조기에 구축하여 방제작업의 과학적ㆍ전문화ㆍ국제화를 동한 실행적 방제능력 향상에 주력하여야 할 것이다.

비만 유발 생쥐에서 Th2분화조절을 통한 황련-감초 하태독법의 알레르기성 비염 발현 억제효과 (Effect of Hataedock Method with Coptidis Rhizoma and Glycyrrhiza Uralensis in Allergic Rhinitis-induced Obese Mice)

  • Objective This study is to learn the effects of Hataedock method using Coptidis rhizoma and Glycyrrhiza uralensis mixed extract on inflammatory response in allergic rhinitis-induced obese NC/Nga mice. Materials and Methods The mice were fed with high fat-diet to be obese, and were divided into 3 groups as follows; allergic rhinitis-induced obese mice group with Hataedock method (CGT, n=10), no treatment group (Ctrl), allergic rhinitis elicited obese mice group (ARE). To induce allergic rhinitis, NC/Nga mice of 3 weeks age were sensitized on 7, 8 and 9 weeks by ovalbumin antigen in intraperitoneal space. After 7 days of final sensitization, allergic rhinitis was initially induced in mice through nasal cavities for 5 days. After 1-week, allergic rhinitis was induced again by the same method. Histological examination was used to identify distribution of IL-4, CD40, STAT6, $Fc{\varepsilon}RI$, substance P, MMP-9, NF-${\kappa}B$ p65, iNOS and COX-2. Results Hataedock method significantly reduced IL-4, STAT6 and CD40 response (p<0.05). In CGT, the inhibition of Th2 differentiation decreased inflammatory mediators such as $Fc{\varepsilon}RI$, substance P, MMP-9, NF-${\kappa}B$ p65, iNOS and COX-2 (all p<0.05). The immunological improvement led reduction of respiratory epithelial damage and mucin secretion in goblet cell. Conclusion The results of this study show that the Hataedock method suppresses the expression of allergic rhinitis by decreasing the inflammatory mediators through the regulation of Th2 differentiation even when the inflammation reaction is increased by obesity. Therefore, Hataedock may have potential preventive measure of allergic rhinitis accompanied by obese.

시호(柴胡)가 MC 및 Mouse의 염증관련 cytokine 분비에 미치는 영향 (Effect of Bupleuri Radix on Inflammatory Cytokine Secretion to HMC and Mouse Immmune Cells)

  • This research was performed in order to investigate the anti-inflammatory effects of Bupleuri Radix(BR) on the Immune response in vitro. Cellular proliferation and cytokine production were measured in mast cells or mouse B cells or CD4 Th cells. BR water extract inhibited the secretions of TNF-$\alpha$ and IL-6 in PMA/A23187 stimulated HMC-1 cells. It increased proliferation but did not affect the expressions of CD69 or CD23 in rIL-4/anti-CD40 activated S cells. BR reduced surface IgE expression and secreted IgE but increased the production of IL-4, IFN-$\gamma$ and IgG1 in the same cells. BR caused an increase in proliferation in anti-CD3/anti-CD28 stimulated CD4 Th cells but it did not affect the differentiation of Th1 or Th2 cells. However, IL-2 was increased in BR treated Th2 cells. Considering the above-mentioned results, BR can be applied to a broad range of anti-inflammatory reactions, but our data suggest that it will not be likely to exert any effects on type 1 allergic response.
