Improvement of Productivity by Forest Tree Breeding Work in Korea (우리나라에서의 임목육종(林木育種)에 의(依)한 생산성(生産性) 증가(增加))

  • Improvement of productivity by forest tree breeding work in Korea was estimated for a few important tree species. Progenies of 17 plus trees of red pine (Pines densiflora) outgrew by 57 percentage compared with progenies of unselected trees at age 15. If best three families are selected among the 17, more than double in volume grow-th is expected. The hybrid Pinus rigida ${\times}$ P. taeda showed more than double volume growth compare to P. rigida at a southern plantation at age 15. However, the superiority of the hybrid decreased at northern plantations, mainly because of low coldhardiness of the hybrid. At a northern plantation, the hybrid grew less than the P. rigida on upper hill, while the hybrid grew much better than the P. rigida on flat area. Another hybrid Populus alba ${\times}$ P. glandulosa grew faster than both parents by two to two and half times according to planting sites at age 10. Introduction of Pinus rigida also showed increased volume growth. Volume increase by selection of best five provenances among 45 at age 12 was estimated as 53 percent compare to progenies of plus trees in Korea, Additional 19 percent of volume increase was expected by selection of the best families within the best provenances. Annual production of chestnuts reached about 70,000 M/T by planting resistant clones to chestnut gall wasp (Dryocosmus kuriphilus), which killed almost all susceptible trees. Although polyploid trees and mutants have been produced by colchicine treatments in over 10 tree species, none of them is economically important Remarkable improvement of productivity is expected by biotechnology in future through selection, hybridization, introduction of foreign genes at cell, cell organelle and gene level, and gene transformation. At present, mass propagation of superior planting materials by tissue culture will increase the productivity.

Survival Results and Prognostic Factors in T4 N0-3 Non-small Cell Lung Cancer Patients According to the AJCC 7th Edition Staging System

  • Background: The American Joint Committee on Cancer (AJCC) published a new staging system ($7^{th}$ edition) in 2009. In our study, we evaluated the survival results and prognostic factors among T4 local advanced non-small cell lung cancer (LA-NSCLC) patients in a large heterogeneous group, in accordance with this new system. Materials and Methods: We retrospectively evaluated the files of 122 T4 N0-3 M0 LA-NSCLC patients, identified according to the new staging system, treated at two centers between November 2003 and June 2012. Variables correlating with univariate survival at p<0.20 were later included in multivariate Cox regression analysis. Here, selection of relevant predictors of survival was carried out in accordance with the likelihood ratio formula with p<0.05 regarded as significant. Results: The median age was 60 and the median follow-up period was 17.4 months. Median overall survival (OS) was 18.3 months, the 1 year overall survival (OS) rate was 72%, and the 5 year OS rate was 28%. Statistically significant predictors of survival were (p<0.20) ECOG-PS (Eastern Cooperative Oncology Group Performance Status), age, T4 factor subgroup, stage and primary treatment in OS univariate analysis. On multivariate analysis for OS ECOG-PS (p=0.001), diagnostic stage (p=0.021), and primary treatment (p=0.004) were significant. In the group receiving non-curative treatment, the median OS was 11.0 months, while it was 19.0 months in the definitive RT group and 26.6 months in the curative treatment group. There was a significant difference between the non-curative group and the groups which had definitive RT and curative operations (respectively p<0.001 and p=0.001) in terms of OS, but not between the groups which had definitive RT and curative operations. The median event free survival (EFS) rate was 9.9 months, with rates of 46% and 19% at 3 and 5 years, respectively. On univariate analysis of EFS rate with ECOG-PS, weight loss and staging, statistical significance was found only for thorax computerized tomography (CT)+18F-fluorodeoxy-glucose positron emission tomography-CT (PET-CT) use, stage and primary treatment (p<0.20). In multivariate analysis with EFS, only the primary treatment was statistically significant (p=0.001). In the group receiving non-curative treatment, the median EFS was 10.5 months while in the curative operation group it was 14.7 months. When all the primary treatment groups were taken into consideration, grade III/IV side effect swas observed in 57 patients (46.6%). Esophagitis was most prominent among those that received definitive radiotherapy. Conclusions: Independent prognostic factors among these 122 heterogeneous LA-NSCLC T4 N0-3 M0 patients were age at diagnosis, ECOG-PS, stage and primary treatment, the last also being a significant prognostic indicator of EFS. Our findings point to the importance of appropriate staging and a multidisciplinary approach with modern imaging methods in this patient group. In those with T4 lesions, treatment selection and the effective use of curative potential should be the most important goal of clinical care.

  • Inflammatory bowel disease, known as Crohn's disease and ulcerative colitis, is an unexplained disease characterized by chronic inflammation that repeats a cycle of relapse, improvement, and complications. The cause of inflammatory bowel disease is not clearly known, but it is predicted that a complex of various factors precipitate its occurrence. In particular, inflammatory mediators, such as cytokine, induce an increase in cell-mediated inflammatory responses. Focal tissue damage then occurs in the intestinal mucosa because of the weakening of the immune-modulating functions of cotton. Immune and inflammatory responses do not decrease appropriately but continue until they lead to chronic inflammation. Current research has focused on the cytokine genes, which have important roles in these inflammatory responses. Cytokine is a glycoprotein that is produced mostly in activated immune cells. It connects the activation, multiplication, and differentiation between immune cells, which causes focal tissue damage and inflammatory response. Moreover, butyrate, which originates in dietary fiber and plays an important role in the structure and function of the intestinal area, shows control functions in the intestinal immune system by decreasing the proinflammatory cytokine and increasing the anti-inflammatory cytokine. Therefore, this research investigated the molecular mechanism of the anti-inflammatory effects of butyrate to comprehend the cytokine controlling abilities of butyrate in the immune cells. Butyrate is expected to have potential in new treatment strategies for inflammatory bowel disease.

Effects of platelet-derived growth factor and epidermal growth factor on the characteristics of beagle dog's periodontal ligament and bone marrow cells (혈소판유래성장인자와 상피성장인자가 치주인대세포와 골수세포의 성상에 미치는 영향)

  • This study was performed to evaluate the effects of platelet-derived growth factor (PDGF) and epidermal growth factor (EGF) on the characteristics of beagle dog's periodontal ligament (BPD) cells and bone marrow (BBM) cells which have the important role on the early stage of periodontal tissue regeneration in vitro. In control group, the cells ($1.5{\times}10^5$cells/ml) were cultured alone with Dulbecco's Modified Eagle's Medium contained with 10% fetal bovine serum, $50{\mu]g/ml$ ascorbic acid, and 10mM/ml ${\beta}-glycerophosphate$. In experimental groups, growth factors, PDGF or EGF(10ng/ml), were added into the above culture condition. And then each group was characterized by examining the cell proliferation rate, amount of total protein synthesis, alkaline phosphatase activity at 1, 5, 9, 13, 17th day after seeding of cells into the culture wells. The results were as follows: 1. Both BPD and BBM cells in PDGF-treated group proliferated more rapidly than non-treated cells. This finding also was observed in EGF-treated group but it was not as prominent as that of PDGF-treated group. The proliferation rates of both cells showed the time-dependent pattern during experimental periods in all three groups. 2. Amount of total protein synthesis was more increased in PDGF-treated group than in control group. But no significant difference between EGF-treated group and control group was observed throughout experimental periods even though the tendency of amount of protein synthesis was time-dependent pattern. 3. Alkaline phosphatase activity also more increased in PDGF-treated group than control group. But slight decrease tendency was seen in both cells of EGF-treated group. From the above results, PDGF appeared to enhance the proliferation and cellular activities including amount of total protein synthesis and alkaline phosphatase activity of BPD and BBM cell, but EGF did not show notable effects. The optimal application of these growth factors was thought to be useful as the adjunctive means in periodontal regeneration procedures.

Syphacia obvelata: A New Hope to Induction of Intestinal Immunological Tolerance in C57BL/6 Mice

  • The ability of nematodes to manipulate the immune system of their host towards a Th2 and T regulatory responses has been proposed to suppress the inflammatory response. Clinical trials have proposed a useful effect of helminth infections on improvement of inflammatory disorders. In this study, we investigated the immunomodulatory effect of Syphacia obvelata infection to induce intestinal tolerance in C57BL/6 mice. Mice were infected through the cagemates with self-infected BALB/c mice. Four weeks post-infection, expression levels of $IFN-{\gamma}$, $TNF-{\alpha}$, IL-17, and IL-10 were assessed in the supernatant of mesenteric lymph node (MLN) culture. $Foxp3^+Treg$ were measured in MLN cells by flow cytometry. In the S. obvelata-infected group, the percentage of Tregs ($5.2{\pm}0.4$) was significantly higher than the control ($3.6{\pm}0.5$) (P<0.05). The levels of IL-10 ($55.3{\pm}2.2$ vs $35.2{\pm}3.2$), IL-17 ($52.9{\pm}3.8$ vs $41{\pm}1.8$), $IFN-{\gamma}$ ($44.8{\pm}4.8$ vs $22.3{\pm}2.3$) and $TNF-{\alpha}$ ($71.1{\pm}5.8$ vs $60.1{\pm}3.3$) were significantly increased in infected mice compared to the control group (P<0.05). The above results showed the potential effects of S. obvelata to induce intestinal tolerance. Therefore, it seems that S. obvelata may increase the immunological suppressive function in the intestinal tract.

EFFECTS OF CIMATEROL (CL 263,780) ON GROWTH PERFORMANCE AND CARCASS QUALITY OF BROILERS FED ON DIFFERENT LEVELS OF DIETARY PROTEIN AND ENERGY

  • The present study was carried out to investigate the effect of cimaterol on growth performance, carcass quality and cellular functional activity of broilers as affected by the various protein and energy levels. In starter period (0-21 days) all chicks were fed the basal diet which contained approximately 23 % crude protein and 3200 kcal of metabolizable energy per kg of diet. The cimaterol was added during 22-49 days and during the period of 8th week the cimaterol was withdrawn. In finisher period (22-49 days), a $2{\times}2{\times}3$ factorial arrangement consisting of 2 levels of cimaterol (0 mg/kg, 0.25 mg/kg), 2 levels of protein (19%, 17%) and 3 levels of energy (3200, 2900, 2600 kcal/kg) was used. In the finisher period, the body weight gain and feed efficiency was improved by the supplementation of cimaterol. The high protein and high energy level with supplementation of cimaterol had showed the highest body weight gain and feed efficiency, without significant difference. The administration of cimaterol had no effects on percentage of abdominal fat content, giblet and neck. Eventhough the difference was not significant (p>0.05), carcass yield was improved slightly by the administration of cimaterol. The effect of cimaterol on carcass composition was clearly demonstrated that protein content of broilers was not increased (p>0.05) but fat content decreased significantly (p<0.05). The ultilization of nutrients in experimental diets was not significantly affected by feeding cimaterol compared to control group. The results of in vitro studies with liver and adipose tissue showed that cimaterol increased the lipolytic activities at 19% protein level whereas at 17% protein level this effect was variable. Lipogenic activities in liver and adipose tissue were not affected with the administration of cimaterol but the activities increased as energy decreased, particularly in liver tissue. In cell studies with acinar culture of liver tissues, cimaterol had no effect on protein synthetic activity but the parameter was increased at higher level of dietary protein and energy. Protein secretion in liver was increased by the supplementation of cimaterol. In addition, at high protein level the protein secretion was increased and has shown the highest values at medium energy level.

Increased Frequency of Sister Chromatid Exchanges After $^{131}I$ Therapy in Lymphocytes of Thyroid Cancer Patients (갑상선암 환자에서 방사성옥소($^{131}I$) 치료로 인한 림프구의 자매염색분체교환(SCE) 빈도증가)

  • To evaluate the genotoxic effect of $^{131}I$, lymphocytes from 9 patients who underwent large dose (150 mCi) $^{131}I$ therapy after total thyroidectomy were studied for sister chromatid exchanges (SCE) before and after their first radioiodine treatments. Frequency of SCE (FSCE) was counted in chromosomes of 30 lymphocytes in each patients, and was expressed as numbers of SCE per cell. Numbers of leukocytes were also observed during $^{131}I$ therapy. Pretreatment FSCE ($4.2{\pm}0.7$) was not different from the control ($3.8{\pm}0.4$, p=0.17). However, the frequency was significantly elevated after $^{131}I$ administration (at the second day, $7.9{\pm}0.8$, p<0.001) and was diminished but still significantly elevated after a week (at 9th day, $6.4{\pm}0.6$, p<0.001). While counts of leukocytes in the peripheral blood showed no change (p> 0.05). In conclusion, chromatids of human lymphocytes were significantly damaged after $^{131}I$ treatment without any bone marrow supression. And the repair of SCE was not complete within one week.

Respiratory Protective Effect of Salvia plebeia R. Br. Extracts against Ambient Particulate Matter-induced Airway Inflammation (미세먼지 유도 기도염증에 대한 배암차즈기 추출물의 호흡기 보호 효과)

  • Background: Small particles increase airway inflammation upon reaching the alveoli. Here, we investigated the protective or therapeutic effects of Salvia plebeia R. Br. (SP_R) extracts on airway inflammation. Methods and Results: To investigate the anti-inflammatory activity of SP_R extracts, we measured their inhibitory effect on the production of reactive oxygen species (ROS) expression of inflammatory mediators, and immune cell infiltration in MH-S alveolar macrophage cells and in the ambient particulate matter (APM)-exposed airway inflammation mice model. The SP_R extracts inhibited the production of ROS and expression of IL-4, IL-10, IL-15, and IL-17A mRNA in APM-stimulated MH-S cells. Oral administration of SP_R extracts suppressed APM-induced inflammatory symptoms, such as high alveolar wall thickness, excess collagen fibers, decreased mRNA expression of chemokines (Ccr9, Ccl5, Ccr3), inflammatory cytokines (IL-15, TNF-${\alpha}$), and IL-4 Th2 cytokine in the lung. The SP_R extracts also inhibited ROS production, granulocyte ($CD11b^+Gr-1^+$) infiltration, IL-17A, TNF-${\alpha}$, macrophage inflammatory protein (Mip-2), and chemokine (C-X-C motif) ligand 1 (Cxcl-1) production in the airway. The specific compounds in the SR-R extracts that mediate the anti-inflammatory effects were identified. Conclusions: In this study, SP_R extracts effectively inhibited airway inflammatory responses, such as ROS production and granulocyte infiltration into the airway, by regulating the expression of chemokines and inflammatory cytokines.

The Effect of the Unsaturated Oil on the Normal Liver and Lipid Metabolism of Rats Fed Several Plant Oils (불포화도가 다른 식물성 유지를 섭취시킨 흰쥐에서 정상적 간 및 지질대사변화의 고찰)

  • To investigate the effect of the unsaturated plant oil on the normal liver and lipid metabalism rats were fed the daily dose of 1.57ml/kg body weight of perilla oil (Iodine value 190~207), corn oil (Iodine value 115~130) and olive oil (Iodine value 80~85) respectively for 28 days. The results were as follows. For the 14 days the test groups showed normal weight gain with 7.86~20.89% increase rate. In the period of the 3rd and the 4th week the increase rate of the perilla oil group was decreased significantly under 17.53~13.8% of control level, but the remainders(corn oil, olive oil) keep normal. The feeding of corn and olive oil for 28 days did show any harmful effect on normal GOT, GPT activity, ALK-P, serum cholesterol and serum triglyceride value of rat. The perilla oil feeding for 21~28 days slightly increased the GOT, GPT activity as 174.87, 93.46u but decreased the cholesterol and triglycerids value as 54.6~0.36mg/dl compared to control. In the pathological finding of test group liver some rats in 28 days feeding group showed reactive vesicula nuclei in corn oil group and mild fatty metamorphosis in olive oil group. But most subjects did not show any characterized sign of acute or subacute liver damage.

Mucosal Immunity Related to CD8+ T Lymphocytes in Children with Helicobacter pylori Gastritis

  • Purpose: We investigated the role of CD8+T cells as host immune factors in pediatric patients with Helicobacter pylori gastritis. Methods: Gastric mucosal tissue and blood samples were collected from 39 children, including 11 children with H. pylori infection and 28 children as controls. Anti-CD8 and anti-T-bet antibodies were used for immunohistochemistry of the gastric mucosa. For the cell surface and intracellular staining, peripheral blood mononuclear cells were stained with anti-IL7Rα, anti-CX3CR1, anti-CD8, anti-T-bet, and anti-IFN-γ antibodies. Cytokines of sera such as tumor necrosis factor alpha (TNF-α) and CX3CL1 were analyzed using enzyme- linked immunosorbent assay (ELISA). Results: In the immunohistochemistry of gastric mucosa, the frequency of CD8+ and T-bet+ T cells cells was higher in the H. pylori-positive group than in the control group (26.9± 7.8% vs. 16.9±3.3%, p<0.001; 5.0±2.5% vs. 2.2±0.7%, p=0.001). Between the control and H. pylori-positive groups, the frequency of IL-7RαlowCX3CR1+ CD8+ and T-bet+ INF-γ+ CD8+ T cells were not significantly different between surface and intracellular staining, respectively (40.4±24.0% vs. 38.2±17.8%, p=0.914; 40.4±24.0% vs. 38.2±17.8%, p=0.914). In the ELISA, no significant differences in TNF-α and CX3CL1 concentrations were observed between the control and H. pylori-positive groups (34.3±12.1 pg/mL vs. 47.0±22.6 pg/mL, p=0.114/0.5± 0.1 pg/mL vs. 0.5±0.1 pg/mL, p=0.188). Conclusion: CD8+ T and Th1 cells, which secrete IFN-γ, might play important roles in the mucosal immunity of the stomach in children with H. pylori infection.
