Sebum is secreted due to the effect of androgen, which start to be secreted at puberty. Androgens have profound effects on the physiology of the sebaceous gland, Using the human sebocyte cell line SZ95, we investigated the inhibitory effect of Chelidonii Harba (CH) on the subum production. Our results showed that numerous cytoplasmic lipid droplets were examined by Oil red staining and lipid droplets were increased markedly by testosterone. Cell viability was dose-dependently decreased by CH as compared with untreated cells, while total lipid content and cholesterol slightly were increased by CH. Testosterone significantly stimulated the synthesis of total lipid and the synthesis of specific sebaceous lipids such as cholesterol and triglyceride. Combined treatment with CH and testosterone resulted in a lower lipid synthesis than with testosterone alone. Especially cholesteol content was reduced by combined treatment with CH and testosterone. These results indicate that CH inhibits the testosterone-induced lipid synthesis in SZ95 cells and acts antagonistically to androgen at the cellular level.
Testosterone deficiency syndrome (TDS), also known as late-onset hypogonadism, is a clinical and biochemical syndrome associated with advanced age and characterized by deficient serum testosterone levels. The Elaeagnus multiflora fruit (EMF) and Cynanchum wilfordii (CW) have been used in traditional herbal medicine. This study aimed to investigate the therapeutic effects of EMF and CW mixtures (at the ratios of 3:7, 5:5, and 7:3) on TDS using TM3 cells and aging male rats. EMF, and mixtures of EMF and CW (at the ratios of 3:7, 5:5, and 7:3) significantly increased testosterone levels in TM3 cells (p <0.05). The rats were orally administered EMCW (EMF and CW mixed at the ratio of 3:7 50, 100 and 200 mg/kg/day) for 4 weeks consecutively. After 4 weeks of EMCW administration, latency time on the rotarod test, and serum testosterone and dehydroepiandrosterone sulfate levels were significantly increased (p <0.05 and p <0.01). Moreover, the levels of globulin-bound sex hormones were decreased in the EMCW-fed groups. However, prostate-specific antigen levels did not differ among the groups. These results suggest that EMCW can be effectively used to alleviate TDS.

BACKGROUND/OBJECTIVES: Male hypogonadism is a condition where the body does not produce enough testosterone and significantly impacts health. Age, obesity, genetics, and oxidative stress are some physiological factors that may contribute to testosterone deficiency. Previous studies have shown many pharmacological benefits of Schisandra chinensis (S. chinensis) Baillon as an anti-inflammatory and antioxidant. However, the molecular mechanism of attenuating hypogonadism is yet to be well established. This research was undertaken to study the effects of S. chinensis extract (SCE) on testosterone deficiency. MATERIALS/METHODS: S. chinensis fruit was pulverized and extracted using 60% aqueous ethanol. HPLC analysis was performed to analyze and quantify the lignans of the SCE. RESULTS: The 2,2-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) scavenging assays confirmed that the SCE and its major lignans (schisandrol A and gomisin N) inhibit oxidative stress. Effects of SCE analysis on the testosterone level under oxidative stress conditions revealed that both schisandrol A and gomisin N were able to recover the lowered testosterone levels. Through mRNA expression of TM3 Leydig cell, we observed that the SCE lignans were able to induce the enzymes involved in testosterone biosynthesis-related genes such as 3β-HSD4 (P < 0.01 for SCE, and P < 0.001 for schisandrol A and gomisin N), 17β-HSD3 (P < 0.001 for SCE, schisandrol A and gomisin N), and 17, 20-desmolase (P < 0.01 for schisandrol A, and P < 0.001 for SCE and gomisin N). CONCLUSIONS: These results support that SCE and its active components could be potential therapeutic agents for regulating and increasing testosterone production.
Objective: Testicular fat deposition has been reported to affect animal reproduction. However, the underlying mechanism remains poorly understood. The present study explored whether sperm meiosis and testosterone synthesis contribute to mouse testicular fat deposition-induced reproductive performance. Methods: High fat diet (HFD)-induced obesity CD1 mice (DIO) were used as a testicular fat deposition model. The serum hormone test was performed by agent kit. The quality of sperm was assessed using a Sperm Class Analyzer. Testicular tissue morphology was analyzed by histochemical methods. The expression of spermatocyte marker molecules was monitored by an immuno-fluorescence microscope during meiosis. Analysis of the synthesis of testosterone was performed by real-time polymerase chain reaction and reagent kit. Results: It was found that there was a significant increase in body weight among DIO mice, however, the food intake showed no difference compared to control mice fed a normal diet (CTR). The number of offspring in DIO mice decreased, but there was no significant difference from the CTR group. The levels of follicle-stimulating hormone were lower in DIO mice and their luteinizing hormone levels were similar. The results showed a remarkable decrease in sperm density and motility among DIO mice. We also found that fat accumulation affected the meiosis process, mainly reflected in the cross-exchange of homologous chromosomes. In addition, overweight increased fat deposition in the testis and reduced the expression of testosterone synthesis-related enzymes, thereby affecting the synthesis and secretion of testosterone by testicular Leydig cells. Conclusion: Fat accumulation in the testes causes testicular cell dysfunction, which affects testosterone hormone synthesis and ultimately affects sperm formation.
본 연구에서는 넙치(Paralichthys olivaceus) 정소로부터 DMRT-1 partial cDNA를 분리하였다. 넙치 DMRT-1은 317개의 염기로 구성되어 있으며, Atlantic halibut, 감성돔 및 무지개 송어와 각각 94%, 85%, 82%의 높은 상동성을 보였다. RT-PCR을 이용하여 DMRT-1 mRNA 발현은 난소보다 정소에서 높게 나타난 것을 관찰할 수 있었다. 또한, 암컷 넙치에 웅성호르몬인 testosterone 처리시 처리 농도에 따라 난소에서 DMRT-1 mRNA 발현이 증가함을 볼 수 있었다. 따라서, DMRT-1은 수컷에서 특이적으로 발현되는 유전자임을 알 수 있으며, 본 연구의 결과는 넙치의 성 전환 및 유도에 대한 기초자료를 제공할 것이다.
마카, 홍삼, 남가새 추출물을 일정한 무게 비율로 혼합하여 발효시켜 얻어진 식물추출복합발효물(MP119)이 성기능에 미치는 영향을 조사하였다. ACE(angiotensin converting enzyme)과 PDE(phosphodiesterase)에 대한 저해효과를 통해 혈액순환부진 개선 및 cGMP 농도에 의한 발기능 개선에 미치는 효과를 측정하였다. MP119의 ACE 저해효과를 측정한 결과 $IC_{50}$값이 241.3${\pm}$35.5 ppm, PDE 저해효과를 측정한 결과 $IC_{50}$값이 372.2${\pm}$33.8 ppm으로 나타나 혈관확장으로 인한 혈류량 증가 및 cGMP농도 증가에 영향을 줄 수 있을 것으로 예상된다. MP119는 마우스 정소세포인 TM3 cell에 유의성 있는 독성이 나타나지 않았으며, TM3 cell에 농도별로 처리했을 때 세포배양액으로부터 대조군과 비교하여 최대 20.11% 증가한 testosterone 농도를 확인하였다. 또한 HUVEC에 농도별로 처리했을 때 NO생산량이 MP119 처리 농도가 증가할수록 유의성 있게 증가하였다. 웅성마우스에 MP119를 7일간 경구투여 후 생식장기의 무게와 정자수를 측정하고, MP119를 경구투여한 후 염화카드뮴을 투여하여 생식장기의 무게와 정자수를 측정하였다. 또한 실험쥐로부터 혈청을 분리하여 혈청 내 testosterone과 cGMP를 측정하였다. 그 결과 MP119의 투여를 통해 생식장기의 무게 변화 없이 정자수 증가에 도움을 준다는 것을 확인하였으며, 더불어 혈청 내 testosterone 및 cGMP의 농도 역시 증가하였다. MP119를 투여한 후 염화카드뮴을 투여한 실험군에서 역시 정자수 및 혈청 내 testosterone, cGMP 농도 증가를 나타내었으므로 결과적으로 MP119가 염화카드뮴의 독성예방 및 치료에 효과적임을 확인하였다.
이 연구의 목적은 시험관내 TCD (테스토스테론 화합물 검출) 시스템으로 8개 한약재의 테스토스테론 유사 활성을 검색하는 것이었다. 녹각상, 백수오, 야관문, 홍삼, 토령, 민들레, 남가세, 호로파의 열수 추출물과 80% 에탄올 추출물을 5, 50, 500 ㎍/ml의 농도로 in vitro TCD 시스템에 적용하였다. 80% 에탄올 추출물의 테스토스테론 유사 활성은 일반적으로 농도에 비례하였고, 음성대조군보다 0.9 내지 5.0 배 더 높았다. 호로파와 야관문은 동일한 농도에서 우수한 테스토스테론 유사 활성을 보였으며 민들레와 홍삼이 그 뒤를 이었다. 토령과 남가세는 낮은 테스토스테론 유사 활성을 보였으며 녹각상과 백수오는 매우 낮은 활성을 나타냈다. 양성대조군 5α-DHT의 활성과 비교하면, 호로파 및 야관문의 80% 에탄올 추출물은 500 ㎍/ml에서 10-8 M 5α-DHT보다 높은 테스토스테론 유사 활성을 나타냈다. 열수 추출물은 80% 에탄올 추출물보다 낮은 테스토스테론 유사 활성을 보였고, 농도에 따른 활성의 증가도 에탄올 추출물보다 낮았다. 호로파, 야관문 및 민들레 등의 80% 에탄올 추출물이 테스토스테론 관련 유전자의 전사 효율을 향상시킬 수 있음이 확인되었다. 따라서, 본 연구는 남성 갱년기 증상의 개선에 필요한 건강 기능성 식품 재료 또는 의약 재료를 개발하는 데 참고가 될 수 있을 것이다.
5${\alpha}$-dihydrotestosterone(DHT)과 testosterone(T)은 남성 생식기관의 주 생성 호르몬들로 그 구조가 매우 비슷하여 이들 각 개를 특이하게 측정(specific determination)하는 방법이 개발되지 않고 있다. 본 연구는 고속액체 크로마토그라피(HPLC)를 이용하여 이들을 분리한 후 섬광면역측 정법(Luminescence immunoassay, LIA)으로 정량하는 방법을 개발하여 이들의 응용 가능성을 검토하고져 하였다. DHT와 T의 retention time은 각각 10.3min, 17.6min이었다. DHT-LIA와 T-LIA에 서 다른 스테로이드들과의 교차반응도는 방사면역측정법(RIA)과 대동소이하였다. 정도관리(quality control) 시료의 intra-assay variation은 DHT-LIA가 8.7%, T-LIA가 6.0%의 변이계수를 나타내었고, inter-assay variation의 변이계수는 각각 12.0% 및 15.3%이었다. 실측치(y)와 기대치(x)간의 관계를 보면, DHT-LIA경우는 Y=0.94X+0.9(r=0.989), T-LIA는 Y=1.01X+0.06(r =0.988)로 두 측정치 사이에는 통계적으로 유의한 차이가 없었다. 위의-측정방법을 이용하여 DHT-enanthate와 T-enanthate 처리후 혈청내 DHT 및 T의 농도변화를 조사한 실험결과 LIA와 RIA의 값사이에 유의한 차이가 없었다. 위의 결과로 보아 본 실험에서 개발된 DHT와 T의 섬광면역측정법은 정립되었다고 사려된다.
We screened the pharmacological effects of a 50% ethanol extract of Yacon tubers and leaves on spermatogenesis in rats. As a result, we found that Yacon tuber extracts increased sperm number and serum testosterone level in rats. It has been reported that the crude extract of Yacon tubers and leaves contain phenolic acids, such as, chlorogenic acid, ferulic acid and caffeic acid by HPLC/MS analysis. We were interested in the contributions made by phenolic acid, particularly chlorogenic acid of Yacon tuber extract to the spermatogenic activity. After administering Yacon tuber extract or chlorogenic acid to rats for 5 weeks, numbers of sperm in epididymis were increased by 34% and 20%, respectively. We also administered ferulic acid, which has been reported to be a metabolite of chlorogenic acid and a constituent of Yacon tuber extract to investigate its spermatogenic activity in rats. Yacon tuber extract and ferulic acid increased sperm numbers by 43% and 37%, respectively. And, Yacon tuber extract, and chlorogenic acid showed significantly inhibition effect of testoeterone degradation in rat liver homogenate. We considered that the spermatogenic effect of Yacon tuber extract might be related to phenolic compounds and their inhibitory effect of testosterone degradation. Yacon showed the possibility as ameliorable agents of infertility by sperm deficiency and late onset hypogonadism syndrome with low level of testosterone.
