Indonesian Lagocephalus wheeleri ("White-milbog") and Chinese Fugu obscurus ("Hwang-bong") puffers were dissected into liver, intestine, ovary or testis, skin and muscle for assay of toxicity, Also, the toxins from the puffer liver were partially purified and analyzed. The frequency of toxic specimens for Lagocephalus wheeleri was 20, 40, 0 and 10%, in terms of liver, intestine, skin and muscle, respectively; and average toxicity was 5.5$\pm$0.9(Mean$\pm$S.E.), 11.8$\pm$4.2, 2.2$\pm$0.1 and 3.0$\pm$0.8 MU/g, respectively. All of Fugu obscurus specimens had toxicity ranging between 2 and 210 MU/g. The frequency of toxicity in intestine and ovary, both 100% was to be higher percentage tan that of other tissues. The toxinsgave two spots tetrodotoxin(TTX) and anhydro-tetrodotoxin(anh-TTX) on TLC. Also, the toxin of each puffer species showed two spots in electrophoresis. In HPLC analysis of Fugu obscurus, the toxins showed TTX, 4-epi-TTX and anh-TTX.epi-TTX and anh-TTX.
Reproductive cycle, gonadal development and the spawning period of the Korean anchovy Coilia nasus were investigated by histologhical observations. Samples were collected at the coastal area of Geumgang dyke which is connected to Gunsan and Janghang, Korea, from February 2002 to January 2003. C. nasus is dioecious; the ovary consists of a pair of saccular structure with many ovarian lobules, and the testis consists of a pair of lobular structure with many testicular lobules and connected to the posterior seminal vesicle. Monthly changes in the gonadosomatic index (GSI) began to increase in April when seawater temperature increased and reached the maximum in June when the ovary was getting mature, the summer season of longer day length with higher water temperature. The reproductive cycle can be classified into five successive stage in females: early growing stage (February to March), late growing stage (March to April), mature stage (May to June), ripe and spent stage (June to July), and recovery and resting stage (July to January): in males, the cycle can be divided into four successive stages; growing stage (February to April), mature stage (May to July), ripe and spent (June to July), and recovery and resting stages (July to January). According to the frequency distributions of egg diameters in the spawning season. C. nasus is presumed to be summer spawning species and polycyclic species to spawn 2 times or more during the spawning season.
The possibility of inadvertent introduction of therapeutic gene expressing viral vectors has raised safety concerns about germ-line infection. Particularly, for indications such as prostate cancer and ovarian cancer, the proximity of the point of viral administration to organs of the reproductive system raises concerns regarding inadvertent germ-line transmission of genes carried by the virus vector. To evaluate the safety of in vivo adenovirus mediated gene transfer, we explored the biodistribution, persistance and potential germ-line transmission of p53-expressing adenovirus (Ad-CMV-p53). Both male and female Balb/c mice were injected with $1{\times}10^9$ PFU of Ad-CMV-p53. The PCR analysis showed that there were detectable vector sequences in liver, kidney, spleen, seminal vesicle, epididymis, prostate, ovary, and uterus. The RT-PCR analysis for detecting inserted gene, p53 showed that Ad-CMV-p53 viral RNA were present in spleen, prostate and ovary. Direct injected male and female mice of adenovirus vector into testis and ovary were mated and their of offspring were evaluated for germ-line transmission of the adenoviral vector. The PCR and RT-PCR analysis showed no evidence of germline transmission, although vector sequences were detected in DNA extracted from gonadal tissues. Real-time PCR result confirmed a significant decrease of adenovirus in gonad tissues 1 week after injection. We have also analysed the cell specific localization of viral DNA in gonad tissues by using in-situ PCR. Positive signals were detected in interstitial tissue but not in seminiferous tubule in sperm. In the case of ovary, adenovirus signal were localized to the stromal tissue, but no follicular signals were observed. Together, these data provide strong evidence that the risk of the Inadvertent germ-line transmission of vector sequences following intraperitoneal or direct injection into genito-urinary system of adenovirus is extremely low.
This study was carried out to find out specific proteins from different organs of Clonorchis sinensis. Crude extract, organ-specific and excretory-secretory (ES) proteins were analyzed by immunoblot with infected human sera. The bands of 7- and 17 -kDa were main component of intestinal fluid and ES protein and commonly found in all organ-specific proteins. The 17-kDa protein was observed from ES antigen, intestinal fluid, eggs and sperms, 26- and 28-kDa proteins were from the uterus, vitellaria, and ovary, and 34-, 37-, 43- and 50-kDa proteins were mainly from the testis and sperms. Serum of mice immunized with sperms reacted to the 50-kDa protein by immunoblotting and immunohistochemical staining showed a positive reaction at the seminal receptacle and seminiferous tubule. The present results show that the 7-kDa protein is a common antigen of every part or organ of C. sinensis, but different organs express their specific antigenic protein bands.
Gonadal development, gametogenesis, reproductive cycle, spawning, relative weight of flesh, and onset of sexual maturity of the murex shell, Ceratostoma rorifluum, collected from the rocky intertidal zone of Daehang-ri, Buan-gun, Jeollabuk-do, Korea were investigated monthly from January to December 2005 both cytologically and histologically. The gonads were widely placed on the digestive gland located in the posterior spiral fleshy part in the shell. C. rorifluum had separate sexes, and was an internal fertilizer. The sex ratio of females to males was approximately 1:1. The ovary and testis contained a great number of oogenic follicles and spermatogenic tubules, respectively. The oogonia and fully ripe oocytes were $15-19{\mu}m$ and $150-160{\mu}m$ in diameter, respectively, and the cytoplasm of the ripe oocytes contained a number of yolk granules. The relative weight of flesh reached a maximum in August($39.35{\pm}0.40%$), and then decreased rapidly in November($32.75{\pm}1.20%$). The percentages of female and male snails at first sexual maturity with shell heights ranging from 12.1-14.0 mm were 60.0% and 52.9%, respectively, while 100% of the snails of both sexes with shell heights over 18.1 mm were reproductively active. Based on the gonadal development and histological observations, the reproductive cycle of the snail could be categorized into five successive stages: early active(December to May), late active(March to July), ripe(June to September), spawning(July to October), and recovery(October to March). C. rorifluum spawned once a year between July and October, and the majority of spawning occurred in September when the seawater temperature exceeded $23.5^{\circ}C$.
The subacute oral toxicity of 1-(4-methylpiperazinyl)-3-phenylisoquinoline (CWJ- a-5) was investigated in Sprague-Dawley (SD) rats. Five groups of 5 males and 5 females were orally administered at doses of 0, 37.5, 75, 150 and 200 mg/kg with CWJ-a-5 for 2 weeks. In clinical signs, Salivation was observed in the 75, 150 and 500 mg/kg male and female groups. Loss of fur was observed in the 500 mg/kg male and female group. Body weight were significantly decreased in the 150 and 500 mg/kg male groups and in the 500 mg/kg female group. Food consumption was significantly decreased in the 300 mg/kg male group. In serum biochemistry, total cholesterol and phospholipid were significantly increased in 500 mg/kg male and female group. Aspartate aminotransferase was significantly increased in the 500 mg/kg female group. In histopathological examination, vacuolar degeneration of renal tubules in the kidney, vacuolar degeneration of hepatocytes in the liver vacuolar degeneration of myocytes in the heart, vacuolar degeneration of histiocytes in the spleen and thymus, atrophy of seminiferous tubule and degeneration of germinal epithelium in the testis, vacuolar degeneration of corpus luteum, granulosa cell and theca cell in the ovary were observed in the 150 and 500 mg/kg male and female groups. Based on these results, the no observed adverse effect level (NOAEL) with CWJ-a-5 was considered to be 75 mg/kg and the absolute toxic dose was considered to be 150 mg/kg in this study

Sex differentiation and gonad development were investigated in a marine medaka species, Oryzias dancena (Beloniformes; Teleostei). The average time to hatch was 11 days post-fertilization (dpf) at $25^{\circ}C$. Primordial germ cell (PGC) was first observed at 5 dpf and migrated to presumptive gonadal area between the gut and pronephric duct at 9 dpf. Male and female gonads were morphologically differentiated at 12 days post-hatching (dph). Early oocytes at perinucleolus stage as well as the formation of spermatid and efferent duct were observed at 28 dph. At 6 weeks of age, the ovary exhibited yolk granulation in many oocytes, while testis possessed a considerable number of spermatogonia and spermatids. The first ovulation was observed in 9-week-old females, and at the same age, males contained fully-matured spermatozoa. Data obtained in this study indicate that the gonad differentiation of O. dancena is the typical type of differentiated gonochorism.
Localization of isoenzyme of glutathione transferase Pi class was compared in different human tissues by immunohistochemical analysis. Strong enrich-ment of GST-Pi in the epithelial tissues was observed in the granular layer of skin, nipple and esophagus which are vulnerable to exogenous chemicals and in the duct epithelium such as pancreatic, biliary, salibvary, renal tubules as well as in the steroid biosynthesis organs such as theca and granulosa of ovary, leydig cell of testis and zona reticularis of adrenal glands.
SOHLH2 is a novel germ cell-specific transcription factor that is crucial for folliculogenesis in the ovary and spermatogenesis in the testis. SOHLH2 represents a candidate gene for infertility with premature ovarian failure. We analyzed whether mutations in the SOHLH2 gene in 98 Korean women with premature ovarian failure. The sequence analysis identified six novel SNPs (c.431-41G>C, c.656A>T, c.1000+27C>T, c.1000+33G>T, c1258-106G>A, and c.2094+ 11T>C) from Korean patients with premature ovarian failure. The c.656A>T found in exon 7 results in change of an amino acid, tyrosine to phenylalanine. Functional mutations in SOHLH2 gene are rare in Korean women with premature ovarian failure.
An experiment was conducted to investigate the annual reproductive cycle of a Korean endemic species, Acheilognathus majusculus, from Jeokseong-myeon located in Seomjin River. The reproductive cycle is examined histologically regarding water temperature and day length of the habitat, the gonadosomatic index (GSI), the female ovipositor length index (OLI), monthly variation in egg diameter distribution, and developmental characteristics of female and male gonads. The maximum GSI was found in $19.21{\pm}2.32$ and $6.90{\pm}0.53$ for female and male respectively when water temperature ($14^{\circ}C$) and day length (11.1hr) began to rise. On the other hand, the minimum level was reached during August ($1.87{\pm}0.67$ for female and $0.88{\pm}0.50$ for male). No samples represent with measurable ovipositor between September and November, while the longest ovipositor length index was in April ($79.68{\pm}4.69%$). We compared and calculated the stages of testis and ovary development process in order to determine the germ cell development characteristics and the reproductive cycle. According to the result, we classified the female Acheilognathus majusculus reproductive cycle into four stages: Ripe (April) and spawning phase (May to June), degenerative phase (July), growing phase (August to December), and mature phase (January to March). The annual reproductive cycle of male Acheilognathus majusculus was categorized into five stages viz. Ripe and spawning phase (May to June), degenerative phase (July to August), resting phase (September to November), growing phase (December to February), and mature phase (March to April).
