This study aimed to investigate the effect of treating environmental purification insect larvae to pig manure on crude ash contents and ammonia production. The experiment set up consisted go two groups: 1 kg of each 3rd instar TM (Tenebrio molitor) and 3rd instar PBS (Protaetia brevitarsis seulensis) larvae in Experiment 1 or 3rd and 4th instar of HI (Hermetia illucens L.) larvae in Experiment 2 were treated with 5 kg of pig manure. In Experiment 1, the crude ash content was higher in TM larvae-treated pig manure at days 0 and 5 (p>0.05), but was similar to that in PBS larvae-treated pig manure over (p>0.05). Ammonia production was observed at day 0 of TM and PBS larvae-treated pig manure (p<0.05), but did not occur thereafter. For Experiment 2, there was significant difference in crude ash content of 3th and 4th instar HI larvae-treated pig manure on day 15. Additionally, ammonia production was found in 3th and 4th instar HI larvae-treated pig manure at days 0 and 5, but did not continue over time. In conclusion, treating TM, PBS and HI to pig manure changed the crude ash contents and reduced ammonia through the ability to decompose pig manure. Thus, environmental impact can be minimized using environmental purification insect larvae.
Edible insects are attracting growing interest as a sustainable source of protein for addition to processed meat and dairy products. The current study investigated the optimal method for protein extraction from mealworm larvae (Tenebrio molitor), cricket adults (Gryllus bimaculatus), and silkworm pupae (Bombyx mori), for use in further applications. After defatting with n-hexane for up to 48 h, sonication was applied for 1-20 min and the protein yield was measured. All samples showed a total residual fat percentage below 1.36%, and a 35% to 94% improvement in protein yield (%). In conclusion, defatting with n-hexane combined with sonication improves the protein yield from insect samples.
최근 들어 곤충을 식품 및 바이오 소재로 이용한 연구가 활발히 진행되고 있다. 그러나 곤충을 이용한 모발 성장 효과에 대한 연구는 아직 미흡한 실정이다. 따라서 본 연구에서는 탈모 예방 및 모발 성장 효과를 가진 새로운 천연물 소재 개발을 위해 갈색거저리 유충 추출물의 항산화 활성 및 모발 성장 촉진 효과를 연구하였다. 갈색거저리 유충 추출물의 항산화 활성 평가를 위해서 DPPH 라디칼 및 아질산염 소거능을 측정하였다. 모발 성장촉진 효과를 측정하기 위해서는 인간 모유두세포(human dermal papilla cell)와 섬유아세포(fibroblast, NIH3T3 cell)를 이용하였으며 MTS assay를 통해 세포생존율 및 세포증식률을 측정하였다. 모유두세포에서 dihydrotesteone (DHT)에 의한 세포사 억제 효과를 확인하였으며, 섬유아세포에서는 tolbutamide (TBM)의 potassium channel blocker 역할에 의한 세포사 억제 효과를 확인하였다. DPPH radical 및 아질산염 소거능 측정 결과 갈색거저리 유충 추출물은 항산화 역할이 뛰어난 것으로 보고된 블루베리와 유사하거나 높은 정도의 항산화능을 가지는 것으로 확인되었다. In vitro 상에서 갈색거저리 유충 추출물을 48시간 동안 처리한 경우, 모유두세포와 섬유아세포의 세포증식을 218% 및 116%까지 증가시켰다. 또한, 모유두세포에서 DHT 처리에 의한 세포사가 갈색거저리 유충 추출물에 의해 억제되는 것을 확인하였으며, 섬유아세포에서는 potassium channel blocker인 TBM에 의해 세포생존율이 감소하였으나 갈색거저리 유충 추출물 처리 시 세포생존율이 정상군과 비슷한 정도로 회복되는 것을 확인하였다. 이상의 결과로부터 갈색거저리 유충 추출물을 이용한 모발성장 및 탈모방지 기능성 소재 개발 가능성을 확인하였다.
파골세포에 의한 뼈의 재흡수와 조골세포에 의한 뼈 형성의 균형은 뼈 건강에 매우 중요한 요소이다. 따라서 이들의 불균형은 골다공증, 골연화증 및 골석화증과 같은 다양한 골 질환을 유발할 수 있다. 특히 파골세포의 기능이 비정상적으로 항진된 경우 골 파괴가 증가되어 골다공증이 야기되며 이런 현상은 류마티스 관절염 같은 염증성 질환의 골 소실과 밀접한 연관이 있다. 그러나 현재 사용중인 골 흡수 억제제는 장기간 사용시 부작용이 보고되고 있어 천연물 소재를 기반으로한 새로운 골 흡수 억제제 개발이 요구되고 있다. 따라서 본 연구에서는 갈색거저리 유충 에탄올 추출물이 RANKL에 의해 유도되는 파골세포 분화에 미치는 영향을 확인하고 그 작용기작을 구명하고자 하였다. 갈색거저리 유충 에탄올 추출물이 파골세포 분화에 미치는 영향을 조사하기 위하여 RAW264.7 세포에 RANKL을 단독 처리 및 추출물과 함께 5일간 처리한 후 TRAP 활성을 비교하였다. 그 결과 RANKL에 의해 증가한 파골세포 분화는 갈색거저리 유충 추출물 2 mg/ml 농도까지 세포독성 없이 농도 의존적으로 감소하는 것을 확인할 수 있었다. 또한 갈색거저리 추출물이 TRAP, NFATc1 및 CtsK와 같은 파골세포 분화마커 유전자 및 단백질 발현량에 미치는 변화를 확인한 결과, RANKL 처리에 의해 현저히 증가한 이들 유전자가 갈색거저리 추출물에 의해 현저하게 감소하는 것을 확인하였다. 또한 갈색거저리 추출물의 파골세포 분화 억제작용기작을 확인한 결과 mitogen activated protein kinases (MAPKs)중 p38의 신호 전달 억제 기작을 통하여 파골세포 분화가 억제됨을 확인 할 수 있었다. 이러한 결과로 보아 갈색거저리 유충 에탄올 추출물 및 그 생리활성 물질들은 골다공증과 같은 골 질환 치료 및 예방을 위한 잠재적 기능성 소재로 사용될 수 있음을 확인하였다.
현재 우리나라는 고령화 사회에 접어들었으며, 이와 함께 골 대사 질환이 큰 문제로 대두되고 있다. 그 중 가장 흔하게 발생하는 골다공증은 여러 요인에 의해 발생하고 있으며 이의 예방 및 치료를 위한 소재 개발 연구가 활발하게 진행되고 있다. 그러나 곤충을 소재로 한 골다공증 예방 및 치료 연구는 미흡한 실정이다. 따라서 본 연구에서는 골 형성 촉진 효능을 가진 새로운 소재 개발을 위해 갈색거저리 유충 오일의 MG-63 조골세포의 분화 촉진 효과를 연구하였다. 조골세포에서 갈색거저리 유충 오일의 독성 및 증식 효과를 평가하기 위하여 MTS assay를 진행한 결과 $80{\mu}g/ml$ 농도까지 세포 독성이 나타나지 않았으며, 48시간 배양했을 때 $40-80{\mu}g/ml$ 농도에서 120% 정도의 세포 증식 효능을 확인 하였다. 갈색거저리 유충 오일이 조골세포 분화에 미치는 영향을 확인하기 위하여 5일간 갈색거저리 유충 오일을 MG-63 조골세포에 처리한 후 ALP 활성을 측정하였다. 그 결과 $80{\mu}g/ml$ 농도에서 무처리 군에 비해 약 180%의 조골세포 분화 촉진 효능을 관찰 하였다. 이 결과는 ALP staining에서도 유사하게 나타났다. mRNA 발현량의 변화를 측정한 결과, Alpl과 Runx2 유전자 발현량이 증가하였고, 단백질 발현량을 측정했을 때에도 유사한 결과를 확인하였다. 이를 통해 ALP와 Runx2 유전자 및 단백질 발현에 의해서 ALP 활성이 증가하고 조골세포 분화가 촉진되었을 것으로 판단되며, 갈색거저리 유충 오일을 이용한 골 형성 촉진에 따른 골다공증 예방 및 치료 기능성 소재 개발에 대한 가능성을 확인하였다.

Distillers dried grain (DDG) and makgeolli spent grain (MSG) are agricultural byproducts to produce alcoholic beverage. However, they are known to contain enough nutrients. Mealworm is a promising insect resource for an animal feed ingredient as well as alternative human food. With low cost, DDG and MSG were investigated as a feed ingredient for rearing high quality mealworms. DDG and MSG were mixed with wheat bran and compared to control feed (only wheat bran) for its effects on larval survivorship, larval weight, duration for developmental period, pupation rate, and pupal weight. When DDG added, larval survivorship was reduced to 50~70% compared to the control group. Larvae fed on DDG were heavier from third to sixth week. Especially, larvae with 50% DDG were 28% heavier than the control group at the third week. For the larval period, the 50% DDG group was 11% less than that for the control. The pupal weight for the 30% DDG group was 7% heavier than that for the control group. Pupation rates for all the DDG groups were higher than 90%. When compared to the control, larval survivorship for the 70% MSG group was low, but the 50% and 70% MSG groups were high during the seventh and eighth weeks because of delayed development. After the eighth week, larvae with 70% MSG showed the highest larval weight increase as 9~18% compared to the control group. Except 70% MSG group, all of MSG groups showed more than 90% pupation rates. We confirmed that adding 30~50% of DDG or MSG to conventional wheat bran have a strong potential to replace the conventional wheat bran insect feed for quality insect production.
Among edible insects, black soldier fly (Hermetia illucens), yellow mealworm (Tenebrio molitor), and common housefly (Musca domestica) have been considered as an alternative protein source for pigs. Because they are easy to breed and grow in the organic wastes, and they have well-balanced nutritional value as a protein source for pigs. The black soldier fly larvae and mealworm could replace the fish meal in the diets for weaned pigs without adverse effects on growth performance and nutrient digestibility. Black soldier fly could also be included in the finishing pig's diet without any negative effects on the growth performance and pork quality of the market pigs. Insect products showed a greater standardized ileal digestibility value of amino acids than conventional animal proteins in growing pigs. Due to the limited amount of insect products used for pig feeding study, most previous pig studies have been conducted in weaned pigs. Thus, further study is needed about the optimal inclusion level of insect products in every phase diet from weaned pigs to sows. The use of insect products in swine diets has some challenges in terms of cost, supply, and safety. Lastly, intrinsic differences among insect species, processing method, and feeding phase should be taken into consideration for the use of insect products in the swine diets.
난방제 해충인 파밤나방의 친환경적 방제를 위하여 토양에서 곤충병원성 곰팡이를 수집한 다음, 파밤나방 유충에 대해 우수한 방제효과가 있는 곰팡이 Metarhizium anisopliae FT83을 분리하여 파밤나방의 생육단계별 살충율을 검정하였다. M. anisopliae FT83의 파밤나방 1령~3령 유충에 대한 살충율은 $1{\times}10^8conidia/ml$ 수준에서 100%이었고 반수치사시간은 0.5일, 2.6일, 2.5일이었으며 4령과 5령에 대한 살충율은 각각 $83.3{\pm}6.2%$, $86.0{\pm}5.7%$였으며 반수치사시간은 4.2일, 3.6일이었다. 번데기와 알에 대한 살충율도 100%로 파밤나방의 전 생육단계에 대해 높은 살충활성을 나타내었다.
Among different types of spoilage, microbial contamination can cause feed decomposition, which results in decreases in feed intake and productivity, infection, and breeding disorder. During the storage time, various microbes have a chance to inoculate with depreciation of feed and to infect the animals. We investigated bacteria that inhabit diverse feed ingredients and complete feed which have been stored for a few months. We isolated and identified 30 genera and 62 species of bacteria. Among these 62 species, 21 species were of non-pathogenic bacteria, 18 species were of pathogenic bacteria, 9 species were of opportunistic pathogens, and 14 species were of unknown bacteria. Pantoea allii and 24 species showed proteolytic enzyme activity. We also confirmed that 6 species including Pseudomonas psychrotolerans showed ${\alpha}$-amylase activity, and 29 species including Burkholderia vietnamiensis showed cellulase activity. Microbacterium testaceum and 3 species showed resistance to Ampicillin, Kanamycin, Streptomycin, Gentamicin, Carbenicillin, and Erythromycin ($50{\mu}g/mL$). Using mealworm larvae (Tenebrio molitor L.) as a model for pathogenicity, we confirmed that 8 species including Staphylococcus xylosus had pathogenicity for mealworm larvae. Especially, Enterobacter hormaechei, Staphylococcus xylosus, and Staphylococcus hominis were reported as being pathogenic for humans. This research suggests that hygienic management of animal feed is essential because beneficial and harmful bacteria can inhabit animal feed differently during storage and distribution.
