Measurement Technique for Single Phase Local Heat Transfer Coefficients of Subchannels in a Rod Bundle using a Copper Sensor (봉다발 부수로의 단상 국부열전달 계수 측정기법에 관한 연구)

  • This paper presents the measuring technique for local heat transfer coefficients using a copper sensor in a rod bundle with mixing vanes. A copper sensor consists of a cartridge heater and four pieces of thermocouple. The Heater is located at the center of the copper sensor and thermocouples measure the surface temperature of the copper sensor. Unheated copper sensor and heated copper sensor are able to measure the local heat transfer coefficient at the position where the heated copper sensor is installed. However the entire region of a rod bundle is actually not heated, the decay of local heat transfer coefficients measured represents overestimated value rather than an actual value. The calibration curve for local heat transfer coefficients is presented using the correction factor calculated by CFD.

Voltammetric Determination of Cobalt(Ⅱ) Using Carbon Paste Electrodes Modified with 1-(2-Pyridylazo)-2-naphthol

  • A method for the determination of cobalt(Ⅱ) by differential pulse voltammetry using a carbon paste electrode constructed by incorporating 1-(2-pyridylazo)-2-naphthol(PAN) into a conventional carbon paste mixture composed of graphite powder and Nujol oil has been developed. Several influencing factors for the determination of cobalt(Ⅱ) were studied in detail and the optimum analytical conditions were found to be as follows: pH, 4.6; composition of electrode, 20%; temperature of deposition, 43 ℃; time of preconcentration, 15 min. Regeneration of the electrode surface for the continuous uses of the electrode was achieved by exposing the carbon paste electrode to an acidic solution. Response of the electrode was reproducible for the uses of five times and the relative standard deviations were 6.7 and 4.6% for 2.0×10-5 M and 4.0×10-6 M cobalt(Ⅱ), respectively. The calibration curve for cobalt(Ⅱ) obtained by differential pulse voltammetry was divided into two linear ranges of 1.7× 10-6-1.3×10-4 M and 2.0×10-7-8.0×10-7 M. The detection limit was estimated to be 1.3×10-7 M. The effects of coexisting ions were also investigated to test the applicability of the proposed method to the determination of cobalt(Ⅱ) in real samples.

Cyanide detection based on natural dyes reaction from blue butterfly pea flowers (Clitoria Ternatea)

  • A green spectrophotometric method for the determination of cyanide has been proposed using, a green reagent, aqueous extract of blue butterfly pea. The test tube was filled with anthocyanin rich extract (pH 6) and cyanide solution. The reaction was kept constant for 10 minutes at room temperature. The reaction mixture changed color from blue to green as the amount of CN-ions increased. The 620 nm peak intensity increased with CN concentration. Therefore, this wavelength was used for all cyanide analyses. The cyanide calibration curve had a linear range of 0.25-1.00, 1.00-4.00, and 4.00-10.00 mg/L, with a satisfactory correlation coefficient of 0.99 and a LOD of 0.57 mg/L. The recovery ranged from 8.33 to 76.94 percent, indicating that this method is inaccurate at low cyanide concentrations. The intra-day and intermediate precision relative deviations were 0.391-0.871 % and 1.112-1.583 %. An H-bond forms between the C-4 group of the B-carbonyl ring and the HCN molecule according to the B3LYP/TZVP calculation. The method is convenient for cyanide concentrations above the LOQ of 1.09 mg/L, cost-effective, and capable of reducing toxic solvents with acceptable precision. The method could also be used to detect total cyanide in biological, environmental, and industrial waste samples.

Residue by elapsed time of non-enzymatic antioxidants in dentifrice (세치제에 함유된 비효소계 항산화제의 경시변화에 따른 잔류량)

  • Objectives: The purpose of this study is to evaluate the non-enzymatic antioxidants stabilities in dentifrices by ascorbic acid and tocopherol according to the chemical condition. Methods: For the analysis of two antioxidants, HPLC UV detector system was used. HPLC was performed using sodium sulfate, acetonitrile(ACN), methanol(MeOH) and measuring absorbance at 240-295 nm. To confirm general pH reaction of two compounds, buffer solution was prepared for the analysis. The dentifrice was titrated by pH so as to examine the change of elapsed time in dentifrice. Linearity of calibration curve of two antioxidants was measured. Results: Each compound showed good linearity at optimized wavelength as well as showing good precision. General pH reaction of two antioxidants was examined. Ascorbic acid showed the highest residue(63.23%) at pH 10 and the lowest residue(2.77%) at pH 4. Tocopherol showed the highest residue(55.70%) at pH 7 and the lowest residue(3.31%) at pH 4. As a result of changing elapsed time of antioxidants in dentifrice by pH, components were remained stably at low temperature($39.2^{\circ}F$) and pH 7. Conclusions: It is necessary to keep dentifrice including ascorbic acid and tocopherol, and non-enzymatic antioxidants at pH 7 and low temperature for improving chemical stability.

Estimation of shelf-life by long-term storage test of Pyungwi-san (평위산 전탕팩의 장기보존 시험에 따른 유통기한 설정)

  • Objectives : To estimate the shelf-life by long-term storage test of Pyungwi-san. Methods : Experiments were conducted to evaluate the stability such as the selected physicochemical, heavy metal, microbilogical experiment under an acceleration test and long-term storage test of Pyungwi-san in different storage under room temperature, refrigeration and freezing. Futhermore, HPLC analysis was performed for the determinations of glycyrrhizin in the Pyungwi-san on an Inertsil ODS-3 column(250 mm ${\times}$ 4.6 mm, 5 um) using solvent 35% acetonitrile include 0.05% phosphoric acid at 254 nm. The flow rate was 1.0 mL/min. Results : The significant change was not showed in pH, heavy metal, microbiological, identification test and quantitative analysis based on acceleration test and long-term storage test. Retention time of glycyrrhizin in HPLC chromatogram was about 16.065 min and calibration curve showed good linearity($R^2$ = 0.9999). The contents of glycyrrhizin in acceleration test and long-term storage test were 0.068~0.076 mg/mL and 0.066~0.077 mg/mL, respectively. Shelf-lifes of room temperature, refrigeration and freezing by long-term storage test were predicted 41, 24 and 34 months, respectively. Conclusions : The suggested shelf-life would be helpful on the storage and distribution of herbal medicine.

Determination of Non-Steroidal Anti-Inflammatory Drugs in Human Urine Sample using HPLC/UV and Three Phase Hollow Fiber-Liquid Phase Microextraction (HF-LPME)

  • Three phase hollow fiber-liquid phase microextraction (HF-LPME), which is faster, simpler and uses a more environmentally friendly sample-preparation technique, was developed for the analysis of Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) in human urine. For the effective simultaneous extraction/concentration of NSAIDs by three phase HF-LPME, parameters (such as extraction organic solvent, pH of donor/acceptor phase, stirring speed, salting-out effect, sample temperature, and extraction time) which influence the extraction efficiency were optimized. NSAIDs were extracted and concentrated from 4 mL of aqueous solution at pH 3 (donor phase) into dihexyl ether immobilized in the wall pores of a porous hollow fiber, and then extracted into the acceptor phase at pH 13 located in the lumen of the hollow fiber. After the extraction, 5 ${\mu}L$ of the acceptor phase was directly injected into the HPLC/UV system. Simultaneous chromatographic separation of seven NSAIDs was achieved on an Eclipse XDB-C18 (4.6 mm i.d. ${\times}$ 150 mm length, 5 ${\mu}m$ particle size) column using isocratic elution with 0.1% formic acid and methanol (30:70) at a HPLC-UV/Vis system. Under optimized conditions (extraction solvent, dihexyl ether; $pH_{donor}$, 3; $pH_{acceptor}$, 13; stirring speed, 1500 rpm; NaCl salt, 10%; sample temperature, $60^{\circ}C$; and extraction time, 45 min), enrichment factors (EF) were between 59 and 260. The limit of detection (LOD) and limit of quantitation (LOQ) in the spiked urine matrix were in the concentration range of 5-15 ng/mL and 15-45 ng/mL, respectively. The relative recovery and precision obtained were between 58 and 136% and below 15.7% RSD, respectively. The calibration curve was linear within the range of 0.015-0.96 ng/mL with the square of the correlation coefficient being more than 0.997. The established method can be used to analyse of NSAIDs of low concentration (ng/mL) in urine.

  • A matrix modification was studied for the determination of trace germanium in mineral waters by electrothermal atomic absorption spectrophotometry (ET-AAS). For this, the type and quantity of modifier as well as the use of auxiliary modifier were investigated to realize the efficient modification. Germanium suffers from low sensitivity and poor reproducibility in ET-AAS determination because of the premature loss of germanium via volatile germanium monoxide formation when heated in the presence of carbon. Therefore, the addition of a matrix modifier is necessary to stablize the germanium, thermally and chemically. By the addition of palladium (10 ${\mu}g/mL)$ as a single modifier to the sample containing 500 ng/mL germanium, the charring temperature could be raised from 800 to $1000^{\circ}C$, and its absorbance was also increased, but the atomization temperature was not raised. In this case, the absorbance of germanium was not changed in the range of 10∼70 ${\mu}g/mL$ of palladium added. On the other hand, it was considered that the use of a mixed modifier could modifiy the matrix more effectively than with a single modifier. The best results were obtained by using 1% ammonium hydroxide as an auxiliary modifier together with 10 ${\mu}g/mL$ palladium. The charring temperature could be raised from 800 to $1100^{\circ}C$, without any change of the atomization temperature. With above optimum conditions, the trace amount of germanium in several mineral waters were determined by a calibration curve method, and good recoveries of more than 95% were also obtained in the samples in which a given amount of germanium was spiked. The detection limit of this method was about 6.9 ng/mL.

Anodic Stripping Voltammetric Determination of Cadmium(Ⅱ) Using Alga-Modified Carbon Paste Electrodes (Alga변성전극을 이용한 Cadmium(Ⅱ)의 양극벗김 전압-전류법적 정량)

  • Microorganisms such as alga are able to uptake toxic and heavy metal ions. After Cd(Ⅱ) was preconcentrated on the carbon paste electrode constructed by incorporating alga (Anabaena), it was determined with differential pulse anodic stripping voltammetry. A well-defined oxidation peak of Cd(Ⅱ) was obtained at - 0.75 V vs. SCE. We investigated the optimum conditions using the peak, which are the effect of the amount of alga, pH, ionic strength, temperature, and preconcentration time on the preconcentration of Cd(Ⅱ) and that of the reduction time and potential on the reduction of Cd(Ⅱ) preconcentrated. Calibration curve for the determination of Cd(Ⅱ) was linear over the range of $1.0{\times}10^6\;M\;to\;8.0{\times}10^6$\;M (R=0.9978) and the detection limit was $5.0{\times}10^{-7}$\;M. The relative standard deviation was 3.1% (n=6) for $7.0{\times}10^{-6}\;M Cd(Ⅱ). In regeneration of the electrode surface with 0.1 M HCl, the response was reproducible continuously by 10 times.

Simultaneous Determination of Six Bioactive Components in Guibi-tang by HPLC-DAD (HPLC-DAD를 이용한 귀비탕 중 6종 생리활성 물질의 동시분석법 확립)

  • Guibi-tang, a traditional herbal medicine, is used for anti-oxidant, anti-osteoporosis, hemostasis and gastroprotection. To develop an analysis method of simultaneous determination of six compounds, swertisin, decursinol, glycyrrhizin, 6-gingerol, costunolide and decursin in Guibi-tang, a high performance liquid chromatography was used with diode array detector. Six bioactive components were separated on a SHISEIDO $C_{18}$ column ($5\;{\mu}m$, 4.6 mm I.D.${\times}$250 mm) with column temperature $30^{\circ}C$. The gradient elution was composed of water with 0.1% trifluoroacetic acid (TFA) and acetonitrile. UV wavelength was set at 230 nm, 254 nm and 330 nm, respectively. Calibration curve showed good linear regression ($R^2$ > 0.9999). The limits of detection (LOD) and the limits of quantification (LOQ) ranged in 0.03 - 0.23 ${\mu}g/ml$ and 0.08 - $0.70\;{\mu}g/ml$, respectively. The RSD values of intra- and inter-day test were in the range of 0.03 ~ 0.96% and 0.01 ~ 1.46%, respectively. The evaluated results of accuracy test were varied from 92.28% ~ 105.14% with RSD < 1.60%. In conclusion, this developed simultaneous determination method was accuracy and sensitive to the quality evaluation of Guibi-tang.

Determination of Wood Flour Content in WPC Through Thermogravimetic Analysis and Accelerator Mass Spectrometry (열중량 분석기와 질량가속기를 이용한 목재·플라스틱 복합재의 목질섬유함량 분석)

  • Determination of the wood content in wood plastic composite (WPC) is crucial to form reliable WPC market. WPC with simple formulation consisting of only two components (wood flour and polypropylene) was examined using thermogravimetric analysis (TGA) and accelerator mass spectrometry (AMS) for determining wood content in the WPC. TGA method using derivative peak temperature (DTp) of polypropylene under low heating rate ($5^{\circ}C/min$) showed more reliable calibration curve and lower error factor compared to method of using the percentage of weight loss of wood flour. In addition, AMS using bio-based carbon content showed greater reliability for the determination of wood content in the WPC in comparison with the TGA method.
