Phototransduction and Visual Cycle in the Ascidian Tadpole Larva

  • Ascidians are lower chordates, and their tadpole-like larvae share a basic body plan with vertebrates. To study photoreceptive systems in ascidians, we have isolated and characterized cDNA clones for three opsins, five G protein ${\alpha}$ subunits (G${\alpha}$), catalytic and regulatory subunits of cGMP phosphodiesterase (PDE), and arrestin from the ascidian Ciona intestinalis tadpole larva. Ci-opsin1 and Ci-opsin2 are vertebrate-type opsins, while Ci-opsin3 is a retinal photoisomerase similar to retinochrome and mammalian RGR. Both Ci-opsin1 and arrestin are specifically localized in the photoreceptor cells of the ocellus, whereas Ci -opsin2 is not expressed in the photoreceptors, but is co-localized in another population of neurons in the brain with PDE (Ci-PDE9 and Ci-PDE$\delta$). Ci-opsin3 is present in the entire region of the brain. Though five different cDNAs encoding Ga have been cloned, no transducin-type G protein has been found yet. Interestingly, one of G${\alpha}$i isoform is conspicuously expressed in the entire region of the brain. The Ci-opsin3 gene expression was observed in a broad area of the brain vesicle as well as in the visceral ganglion. Genes encoding ascidian homologs of CRALBP and ${\beta}$-CD, whose function is required for the mammalian visual cycle, are co-expressed with Ci-opsin3 in the brain vesicle and visceral ganglion. Localization of Ci-opsin3, CRALBP, and ${\beta}$-CD in a broad area of the brain suggests that the brain of the ascidian larva has a visual cycle system similar to that of the vertebrate RPE. Based on these data, we discuss the evolution of vertebrate visual systems.

  • A novel quantified method based on requirement analysis of image quality to improve display image quality was proposed. Nowadays, the image quality was limited by the poor understanding of the image quality requirement, which led to the critical factors of image quality could not be controlled during display development. Our method was set up to resolve this problem by clarifying the relationship between the image quality level and the effect factors in image processing. Moreover, the subjective factors were eliminated extremely by the image quality quantification. The method was applied in the RPTV development life cycle and its efficiency was demonstrated.

Comparative Interactomes of VRK1 and VRK3 with Their Distinct Roles in the Cell Cycle of Liver Cancer

  • Vaccinia-related kinase 1 (VRK1) and VRK3 are members of the VRK family of serine/threonine kinases and are principally localized in the nucleus. Despite the crucial roles of VRK1/VRK3 in physiology and disease, the molecular and functional interactions of VRK1/VRK3 are poorly understood. Here, we identified over 200 unreported VRK1/VRK3-interacting candidate proteins by affinity purification and LC-MS/MS. The networks of VRK1 and VRK3 interactomes were found to be associated with important biological processes such as the cell cycle, DNA repair, chromatin assembly, and RNA processing. Interactions of interacting proteins with VRK1/VRK3 were confirmed by biochemical assays. We also found that phosphorylations of XRCC5 were regulated by both VRK1/VRK3, and that of CCNB1 was regulated by VRK3. In liver cancer cells and tissues, VRK1/VRK3 were highly upregulated and its depletion affected cell cycle progression in the different phases. VRK3 seemed to affect S phase progression and G2 or M phase entry and exit, whereas VRK1 affects G1/S transition in the liver cancer, which could be explained by different interacting candidate proteins. Thus, this study not only provides a resource for investigating the unidentified functions of VRK1/VRK3, but also an insight into the regulatory roles of VRK1/VRK3 in biological processes.

STS 304 압연강의 저주기 피로특성에 관한 연구 (A Characteristic Study of Low Cycle Fatigue for Rolled STS 304 Steel)

  • In this study, low cyclic fatigue test was carried out at room temperature condition for rolled STS304 steel. The results of this study show that rolled STS304 steel has excellent static tensile strength and fatigue characteristics. The relationship between plastic strain range and fatigue life was examined using the triangular wave in order to predict the low cycle fatigue life of rolled STS304 steel by Coffin-Manson equation. Cyclic behavior of rolled STS304 steel was characterized by cyclic hardening with increasing number of cycle through the Hysteresis loop analysis and cyclic response of maximum stress versus number of cycles. It is found that the plastic deformation energy consumed per cycle is reduced by calculating the area of the hysteresis loop.

  • 이차전지의 수명 평가는 크게 Cycle life test 와 Calendar life test로 나뉘어져서 평가 되고 있다. Cycle life test로 수명 검증을 위해서는 일반적으로 3000 사이클 이상 테스트를 진행 하여야 하지만 이 방법은 시간이 장기화 되어 신뢰성 검증 및 새로운 부품 적용에 한계가 따른다. 따라서 본 논문에서는 고온 챔버 및 사이클 시험기를 사용하여 빠른 시간 내에 평가를 할 수 있는 가속시험법을 적용하여 수명을 평가 한 연구결과를 발표하고자 한다.

  • 8200 Series Electric Locomotives are mostly imported from overseas due to aging and uncertainty of source technology, and it's the maintenance cost is increasing. We are analyzed life cycle costs based on international standards such as IEC 60300-3-3 and IEC62278. The main conversion system of the 8200 series electric locomotive is closely related to vehicle operation and is one of the subsystems requiring frequent maintenance. In this paper, the life cycle cost of the main conversion unit in 8200 series electric locomotive is analyzed based on the maintenance manual. As a result of analysis, maintenance cost of GTO module and control device is relatively high, and it is confirmed that the cost increases according to the useful life.

Life Cycle Impacts of Flexible-fiber Deep-bed Filter Compared to Sand-Filter including Coagulation and Sedimentation in Water Treatment Plant

  • Recently a new technology called the flexible-fiber deep-bed filter (FDF) claimed to replace the conventional sand filter including coagulation and sedimentation filter (CSF) processes in the water treatment plant. Therefore the life cycle assessment (LCA) approach was applied for evaluating the life cycle impacts of FDF compared with those of CSF. The used LCA softwares were the Simapro 6 and PASS and their life cycle impact assessment (LCIA) methodologies were the Eco-indicator 99 and the Korean Eco-indicator, respectively. The goal of this LCA was to identify environmental loads of CSF and FDF from raw material to disposal stages. The scopes of the systems have been determined based on the experiences of existing CSF and FDF. The function was to remove suspended solids by filtration and the functional unit was $1\;m^3$/day. Both systems showed that most environmental impacts were occurred during the operation stage. To reduce the environmental impacts the coagulants and electricity consumptions need to be cut down. If the CSF was replaced with the FDF, the environmental impacts would be reduced in most of the impact categories. The LCA results of Korean Eco-indicator and Eco- indicator99 were quite different from each other due to the indwelling differences such as category indicators, impact categories, characterization factors, normalization values and weighting factors. This study showed that the life cycle assessment could be a valuable tool for evaluating the environmental impact of the new technology which was introduced in water treatment process.

도로 종류와 도로생애주기별 탄소배출량, 에너지소모량 및 비용에 대한 거시적 분석방법 (Macro-level Methodology for Estimating Carbon Emissions, Energy Use, and Cost by Road Type and Road Life Cycle)

  • PURPOSES : The authors set out to estimate the related carbon emissions, energy use, and costs of the national freeways and highways in Korea. To achieve this goal, a macro-level methodology for estimating those amounts by road type, road structure type, and road life cycle was developed. METHODS : The carbon emissions, energy use, and costs associated with roads vary according to the road type, road structure type, and road life cycle. Therefore, in this study, the road type, road structure type, and road life cycle were classified into two or three categories based on criteria determined by the authors. The unit amounts of carbon emissions and energy use per unit road length by classification were estimated using data gathered from actual road samples. The unit amounts of cost per unit road length by classification were acquired from the standard cost values provided in the 2013 road business manual. The total carbon emissions, energy use, and cost of the national freeways and highways were calculated by multiplying the road length by the corresponding unit amounts. RESULTS: The total carbon emissions, energy use, and costs associated with the national freeways and highways in Korea were estimated by applying the estimated unit amounts and the developed method. CONCLUSIONS: The developed method can be employed in the road planning and design stage when decision makers need to consider the impact of road construction from an environmental and economic point of view.
