친환경 버섯배지에 녹차 잔류물의 처리가 새송이버섯의 생장 및 Catechin류 함량에 미치는 영향 (Effects of Green Tea Residue Treatment in Eco-Friendly Medium on Growth and Catechin Content of Pleurotus eryngii)

  • The present study was conducted to investigate the translocation of polyphenols, especially catechin derivatives, from mushroom medium mixed with green tea residues into fruiting body of Pleurotus eryngii. Pleurotus eryngii was grown on the media incorporated by mixing or surface-treated with dry materials including leaf petioles and young stems or leaves of green tea. The dry materials treated in medium did not affect plant height and fresh weight of Pleurotus eryngii body. From the samples of Pleurotus eryngii, the eight main catechin derivatives (-)-gallocatechin(GC), (+)-catechin (C), (-)-epicatechin (EC), (-)-epigallocatechin (EGC), (-)-epigallocatechin gallate (EGCG), (-)-gallocatechin gallate (GCG), (-)-epicatechin gallate (ECG), and (-)-catechin gallate (EGCG), and caffeine were analyzed quantitatively by HPLC. The results showed that EGC in Pleurotus eryngii was 45% more detected, when incorporated with the dry materials, than untreated control. Especially, content of EGCG was increased in surface-treated Pleurotus eryngii up to 3.2 ppm, while it was not detected or reduced in control and other treatments. Caffeine content was greatly increased regardless of treatment method, compared with control (0.1ppm), showing 44 fold-amount in Pleurotus eryngii at early growth stage when incorporated with the dry materials into medium. The results indicates that functional catechin derivatives of green tea would be partly translocated into Pleurotus eryngii throught incorporation and surface treatment with residues of green tea plants.

  • 돌외를 채취해서 잎과 줄기로 나누어 각자 실험실에서 차로 제조한 것과 제품으로 시판되고 있는 국산품과 일본산 제품 및 국산품 볶은것에 대해 무기성분의 함량을 측정하고 비교하였다. 돌외차에 함유된 무기성분은 잎에서 Mg, Mn, Ca 및 Cu의 함량이 많았고 줄기에서 K, Na 및 Fe이 많이 함유되어 있었으며 특히 줄기중 총회분의 K의 함량이 33.4%로서 가장 높았다. 제조된 돌외차와 시판제품에서 차 4g을 $1{\ell}$의 끓는물에 넣어 5분간 우려냈을 때 무기성분의 침출율은 K이 $83.2{\sim}96.3%$로 가장 높았으며 다음 Mg이$44.8{\sim}62.2%$ 이었고 Ca은 $23.1{\sim}64.4%$, Na은 $6.3{\sim}55.6%$ 순이었으나 Mn, Fe 및 Cu등은 극히 미량만 침출되었다.

The necrotroph Botrytis cinerea promotes disease development in Panax ginseng by manipulating plant defense signals and antifungal metabolites degradation

  • Background: Panax ginseng Meyer is one of the most valuable medicinal plants which is enriched in anti-microbe secondary metabolites and widely used in traditional medicine. Botrytis cinerea is a necrotrophic fungus that causes gray mold disease in a broad range of hosts. B. cinerea could overcome the ginseng defense and cause serious leaf and root diseases with unknown mechanism. Methods: We conducted simultaneous transcriptomic and metabolomic analysis of the host to investigate the defense response of ginseng affected by B. cinerea. The gene deletion and replacement were then performed to study the pathogenic gene in B. cinerea during ginseng - fungi interaction. Results: Upon B. cinerea infection, ginseng defense responses were switched from the activation to repression, thus the expression of many defense genes decreased and the biosynthesis of antifungal metabolites were reduced. Particularly, ginseng metabolites like kaempferol, quercetin and luteolin which could inhibit fungi growth were decreased after B. cinerea infection. B. cinerea quercetin dioxygenase (Qdo) involved in catalyzing flavonoids degradation and ∆BcQdo mutants showed increased substrates accumulation and reduced disease development. Conclusion: This work indicates the flavonoids play a role in ginseng defense and BcQdo involves in B. cinerea virulence towards the P. ginseng. B. cinerea promotes disease development in ginseng by suppressing of defense related genes expression and reduction of antifungal metabolites biosynthesis.

소규모 국내생산 식물 발효식품의 바이오제닉아민 잔류특성 (Investigation on Biogenic Amines in Plant-based Minor Korean Fermented Foods)

  • 본 연구에서는 식물을 원료로 하여 제조된 발효식품들 중 지금까지 BAs에 대한 잔류분석연구가 진행되지 못하였던, 식품군들에 대해 BAs 잔류분석을 실시하였다. 조사대상 식품군으로는 식물 발효진액, 장아찌, 막걸리, 흑마늘, 발효차를 선정하였으며, 발효진액, 막걸리, 흑마늘, 발효차는 HIS, TYR, PUT, CAD, PHE 5종 BAs 의 총 함량이 1,000mg/kg을 초과하지 않았으나, 장아찌시료에서는 19.2-1,214 mg/kg으로 검출되었다. 이러한, 장아찌류의 고잔류 BAs는 간장과 된장 등 원료 장류에 의한 영향이 높을 것으로 사료되며, 장아찌에 대한 BAs 연구는 원료 및 제조 특성별 잔류 분석연구가 보완되어야 할 것이다.

송순차 제조를 위한 송순 및 솔잎의 화학적 조성 (Chemical Composition of Pine Sprouts and Pine Needles for the Production of Pine Sprout Tea)

  • As a basic study for preparing pine sprout tea, chemical components in pine sprouts and pine needles were analyzed as follows: In proximate composition the contents of most components except for crude fat were different between in pine sprouts and pine needles. Moisture content in pine sprouts was higher than that in pine needles. Calcium and potassium were major minerals contained in pine sprouts and needles, and their contents in pine needles picked in December were higher than those picked in June. Soluble tannin and vitamin C contents in leaf part of pine sprouts were much higher than those in stem part and their contents in pine needles were increased according to their growth. Free sugars like fructose, glucose and sucrose were contained in both pine sprouts and needles, and their contents in pine sprouts were higher in stem part as compared that in leaf part. Although fourteen kinds of amino acids were detected in pine sprouts and pine needles, their contents were extremely low. Amino acid composition between pine sprouts and pine needles was different each other, but major amino acids contained in them were same, those are acidic amino acids such as aspartic acid and glutamic acid. Amino acid contents in pine needles were increased according to the growth. In fatty acid composition in leaf part of pine sprouts, saturated fatty acid contents were higher than unsaturated fatty acid contents, but in stem part unsaturated fatty acid contents were higher. In pine needles the amount of saturated fatty acid was increased with the growth, but the amount of unsaturated fatty acid was rather decreased.

가루차용 차엽의 생육 및 성분 (Growth and Constituents of Tea Shoots for Powder Green Tea)

  • 차광한 차나무 잎의 엽위별 생육특성 및 차 품질 관련 주요 화학성분을 분석한 결과는 다음과 같다. 하위엽으로 내려 갈수록 엽장, 엽폭, 백아중은 컸으나 엽수분은 감소하였다. 하위엽으로 내려갈수록 total nitrogen, caffeine, vitamin C, saponin 함량은 감소하였고, total amino acid, chlorophyll 함량은 4번 엽과 3번엽에서 각각 최대함량을 나타냈다. tannin은 $9.53{\sim}11.24%$로 1엽이 11.24%로 함량이 제일 많았고 4엽이 9.53%로 가장 적었다. fatty acid는 2엽에서 3,594mg/100g으로 함량이 가장 많았고, 5엽에서 2,782mg/100g으로 함량이 가장 적었다. 포화지방산인 palmitic acid와 steatic acid는 5엽에서 함량이 많았고, 불포화 지방산인 oleic acid, linoleic acid, linolenic acid는 2엽에서 함량이 많았다. 이상 분석 결과로부터 15일간 차광한 후 채엽한 가루녹차 원료는 5엽까지 채취하여 제다해도 품질에 있어서 큰 문제가 없다고 생각된다.

다류원료 식물류 물 추출물의 항산화 효과 (The Antioxidative Effects of the Water-Soluble Extracts of Plants Used as Tea Materials)

  • 본 연구는 다류원료 식물류의 물 추출물에 대해 항산화성을 검토하고 그 항산화 물질(페놀성 화합물, 비타민 C, 셀레늄)을 분석하였다. 대두유에 식물류의 물 추출물을 0.02%(추출고형분 함량, w/w) 첨가하여 $55^{\circ}C$ 공기순환식 인큐베이터에서 36일 저장기간 동안 과산화물가 측정과 $110^{\circ}C$에서 Rancimat 방법에 의해 항산화성을 검토하였으며 첨가제가 들어있지 않은 대두유를 대조구에 사용했고 0.02% BHT 첨가한 대두유는 비교구로 사용하였다. 대조구와 BHT 첨가구에 비해 산화 유도기간이 $55^{\circ}C$에서는 구기자, 우롱차, 귤껍질(진피), 유자, 매실 물 추출물 등이 길었으며 $110^{\circ}C$에서는 우롱차, 인스탄트커피, 유자, 구기자, 오미자, 매실 물 추출물 등이 길었다. 추출고형분 g당 총페놀성 물질 함량은 우롱차, 녹차, 홍차, 커피, 계피, 결명자 물 추출물 등에서 높았고 비타민 C 함량은 커피, 두충, 홍차, 영지, 계피, 감잎, 치커리, 녹차, 우롱차 물 추출물 등에서 높았으며 셀레늄은 모든 물 추출물에서 검출되지 않았다. 이들의 항산화성은 단일 항산화성 물질의 효과에 기인하기 보다는 페놀성 화합물, 비타민 C 등 복합적 효과에 의한 것으로 사료된다.

The Leaf of Diospyros kaki Thumb Ameliorates Renal Oxidative Damage in Mice with Type 2 Diabetes

  • Diabetic kidney disease is the most common and severe chronic complication of diabetes. The leaf of Diospyros kaki Thumb (persimmon) has been commonly used for herbal tea and medicinal purposes to treat a variety of conditions, including hypertension and atherosclerosis. However, the effect of persimmon leaf on kidney failure has not been investigated. This study aimed to examine the role of persimmon leaf in protecting the diabetes-associated kidney damage in a mouse model of type 2 diabetes. Mice were fed either a normal chow diet with or without powered persimmon leaf (5%, w/w) for 5 weeks. In addition to kidney morphology and blood markers of kidney function, we assessed levels of oxidative stress markers as well as antioxidant enzymes activities and mRNA expression in the kidney. Supplementation of the diet with powered persimmon leaf not only decreased the concentration of blood urea nitrogen in the plasma but also improved glomerular hypertrophy. Furthermore, the persimmon leaf significantly decreased the levels of hydrogen peroxide and lipid peroxide in the kidney. The activities of superoxide dismutase, catalase, and glutathione peroxidase and the mRNA expression of their respective genes were also increased in the kidney of persimmon leaf-supplemented db/db mice. Taken together, these results suggest that supplementation with the persimmon leaf may have protective effects against type 2 diabetes-induced kidney dysfunction and oxidative stress.

  • Green tea seed was extracted with absolute ethanol,and the green tea seed extract(GTSE)was subjected to assays for toxicity, antioxidant ability, angiogenesis inhibitory effects and cell adhesion, as well as western blotting, and an in vivo experiment against 4 high-ranking adult cancers in Korea. Our series of experimental data demonstrated that GTSE has an antioxidant ability superior to that of EGCG in the green tea leaf, and also exhibits a profound high tumor growth inhibitory activity on a variety of cancer cell lines, as well as nude mice infected with cancer cells. GTSE was identified as a natural anticancer compound showing excellent angiogenesis inhibition and cancer cell suppression abilities. Our preliminary observations also indicate that GTSE may be another potential source of natural dietary antioxidants and also may be applicable as a novel natural anticancer agent.

Gas chromatography-Thermal Energy Analyzer에 의한 휘발성 니트로소아민과 담배 특유의 니트로소아민들의 동시 분석연구 (Studies on the Simultaneous Determination of VNA and TSNA by GC - TEA)

  • This is to investigate the methodology for the simultaneous determination of Wk, mk and TSNA using gas chromatography(GC) in combination with chemiluminescence detector, thermal energy analyzer(TEA) . The simultaneous analysis has been estimated by evaluating tobacco. The TEA was linked to GC equipped with non -polar SPB -5 fused silica capillary column which was introduced into the ceramic pyrolysis tube by the point of 16cm from the end of TEA. Quantification was carried out by internal standardization with WDPA after calibration of retention times and response factors with authentic nitrosoamines. It was demonstrated that WDPA was most preferable as internal standard for the simultaneous analysis. The recoveries of the internal standard were in the range of 83∼96% . Nitrosoamines in this method were detected with determination limit of 0.1ng and was made by a straight line in calibration curve by TEA response. The suitability of nitrosoamines extraction in tobacco leaf was investigated. It was most suitable to extract nitrosoamines from tobacco leaves with 0.01 M NaOH within a period of 8 hours. Thimerosal as an antibacterial agent was added to NaOH solution to prevent artifactual formation. The fractionation and the purification of nitrosoamines form alkaline extracts were conveniently performed using Extrelut multilayer column and dichloromethane. Reproducible and reliable results were obtained for the determination of nitrosamines in a relatively short time compared to previous known method. TSNA contents in burley were about 4 times higher as those in the fluecured tobacco.
