The ethanolic extract of the heartwood of Taxus baccata L. afforded six taxoids, belonging to the skeletally three different groups, $2{\alpha},\;5{\alpha},\;10{\beta}-triacetoxy-14{\beta}-(2-methyl)-butryloxy-4(20)$,11-taxadiene (1), taxusin (2), baccatin VI (3), baccatin III (4), $1{\beta}-hydroxybaccatin$ I (5) and taxol (6). The isolation and structure elucidation of three taxoids (3-5) have been presented in this study. This is the first report for the presence of these taxoids in Taxus baccata L. growing in Turkey.

Chemical investigation on a sample of the needles of Himalayan Taxus baccata has resulted in the isolation of several taxoids including taxol (1) 10-deacetyl-baccatin III (2) and 2-deacetoxytaxinine J (3) along with different lignans (6 and 7) and simple phenolics (8, 9, 10, 11 and 12). The occurrence of 4-(4'-hydroxyphenyl)-butane-2-one and 4-(4'-hydroxyphenyl)-trans-but 3-ene-2-one (8) in Taxus species is reported for the first time. The $^{13}C-NMR$ spectral data of two rearranged taxiod constituents, brevifoliol (4) and 13-decinnamoyltaxchinin B (5) are presented. The acid-catalyzed decomposition of taxol has been discussed. The synthesis of other two constituents, rhododendrol (10) and hibalactone (7) has been described.
We have isolated endophytic fungi from the Indian yew tree, Taxus baccata, and then screened for taxol production. Out of the 40 fungal cultures screened, one fungus Gliocladium sp. was found to produce taxol and 10-DABIII (10-deacetyl baccatin III). These compounds were purified by TLC and HPLC and characterized using UV-spectroscopy, ESI-MS, MS/MS, and proton NMR. One liter of Gliocladium sp. culture yielded $10\;{\mu}g$ of taxol and $65\;{\mu}g$ of 10-DABIII. The purified taxol from the fungus showed cytotoxicity towards cancer lines HL-60 (leukemia), A431 (epidermal carcinoma), and MCF-7 (breast cancer).
A large number of chemical constituents have been reported from the Himalayan yew [Taxus baccata (Linn) or T. wallichiana (Zucc)]. These constituents are mainly taxoids and phenolics. Taxol, a lead anticancer agent, is the most important constituent. Other compounds have also been found to possess interesting biological properties. The literature concerning the chemistry and bioactivity of the constituents of the Himalayan yew has been briefly reviewed.
Influence of fructose addition to the cultivation medium on the production of taxanes and the growth of callus culture of Taxus baccata was studied. The cultures showed an ability to adjust to the substitution of some of the sucrose in the media by fructose and the fresh biomass accumulation was higher on the media containing different concentrations of fructose during the second cultivation period.
Anticancer agent taxol was synthesized from baccatin III which was extracted from cell cultures of European yew, Taxus baccata Pendula. Callus and suspension cultures of T. baccata Pendula showed the formation baccatin III. The content of baccatin III in cell cultures reached 0.015% (w/w) on dry weight basis. The semisynthetic approaches were made with baccatin III and taxol side chain. To prepare taxol side chain, (-)-N-((S)-2-hydroxy-1-phenylethyl) benzamide was synthesized first from (S)-(+)-phenylglycine. Then (-)-N-((IS,2S)-2-hydroxy-1-phenyl-3-butenyl) benzamide was synthesized with vinyl magnesium bromide. The synthesis of (2R, 3S)-(-)-2-(1-ethoxyethoxy)-3-phenyl-3-(phenylmethanamido) propanoic acid with RuCl3, catalyst was the final step to prepare taxol side chain. The semisynthetic reactions yielded 0.0002% taxol, 0.0005% 7-epi-10-deacetyltaxol, and unidentified taxanes on dry weight basis. It is suggested that the semisynthesis of taxol from baccatin III could be an alternative source of taxol and related taxanes.

The endophytic fungal populations of different tissues of Taxus baccata grown at high altitudes in West Bengal, India were explored. These isolated fungal populations represented different genera, which were screened for taxol production using immunoassay technique. The culture AAT-TS-$4_1$ that produced taxol was identified as Gliocladium sp. based on its cultural, morphological characteristics, internal transcribed spacer, and 18S rRNA sequence analysis. Kinetics of taxol production as a function of culture growth were investigated.
Enhanced production of (10-deacetyl) baccatin III and related taxanes was observed in suspension cultures of Taxus baccata Pendula. six % of initial glucose and sucrose concentration increased 10-deacetyl baccatin III production 3.5 and 2.5 times, respectively. Methyl jasmonate, as an elicitor, increased taxane production. Time course changes of taxane production after methyl jasmonate addition showed that baccatin II and 10-deacetyl baccatin III were detected first and paclitaxel, 10-deacetyl taxol and cephalomanine were produced in sequence. Feeding experiments with $500{\mu}M$ of benzoic acid increased 10-deacetyl baccatin III production 10 times. Baccatin III production was also increased 8 times by feeding of $500{\mu}M$ of lysine as a precursor.
This study was carried out for the purpose of supplying the basic data for artificial forestation, natural regeneration and ecological conservation etc., and obtaining information on alpine vegetation, by establishing vegetation units on the basis of phytosociological classification of community and studying growth pattern on the basis of species composition, hierarchy structure and population dynamics, for Taxes cuspidata naturally growing at the alpine districts in Korea. The importance value of Taxus cuspidata by districts mostly showed above 100 in the upper story but at Mt. Chiri and Mt. Hanra communities its value was comparatively low. In the middle story it showed lower value than that of upper story and not quite showed at Mt. Chiri. Taxus cuspidata communities in the lower story were only in Mt. Hanra, Mt. Odae and Mt. Sobaek, and their importance values were about 10 which were relatively low values. The communities were classified into five groups as Taxus cuspidata-Males baccata var. mandshurica, Taxus cuspidata-Abies nephrolepis, Taxus cuspidata-Abies koreana, Taxus cuspidata-Acer mono and Taxus cuspidata-Euonymus quelpaertensis by TWINSPAN analysis. Taxus cuspidata-Abies koreana community was distributed at the northern aspect of the mountain ridges and at higher elevation than other communities and distributed. Taxus cuspidata-Acer mono community was relatively low than the others and distributed at the hillsides of mountain. And Taxus cuspidata-Euonymus quelpaertensis communities were distributed at the relatively high elevation and northern and eastern aspect of the mountain tap, and Taxus cuspidata-Malus baccata var. mandshurica communities were distributed at the medium elevation, and southern and eastern aspect of the mountain ridge. In the relation between communities and environmental factors, it was correlated with aspect, elevation and topography at the first axis, and elevation, slope at the second axis.

The purpose of this study is to review the taxonomic position of Taxus cuspidata var. latifolia endemic to Ulleung Island with related taxa T. cuspidata var. cuspidata, T. caespitosa, and T. cuspidata var. nana based on external morphological characters and DNA barcoding study. T. cuspidata var. latifolia was similar to T. cuspidata var. cuspidata in the arbor, straight trunk, and symmetric arrangement of leaf. But the unique differences between T. cuspidata var. latifolia and T. cuspidata var. cuspidata were leaf size and the exposure of seed from aril. Additionally, sequences of four chloroplast DNA regions including matK, rbcL, trnL intron and trnL-trnF spacer regions were analyzed. Korean Taxus species and their related taxon T. cuspidata var. nana were strongly supported as a monophyletic group in neighbor-joining analysis. Taxus cuspidata var. latifolia showed 100% sequence identity to related taxa. Korean endemic T. caespitosa is also distinguishable from related taxa by prostrate stems and spiral arrangement of leaf. The examinations of external morphology and DNA barcoding study suggest that the taxonomic position of T. cuspidata var. latifolia should be maintained as a variety of T. cuspidata.
