Five New Wood Decay Fungi (Polyporales and Hymenochaetales) in Korea

  • The wood decay fungi are a diverse taxonomic group that plays a pivotal role in forest carbon cycling. Wood decay fungi use various enzymatic pathways to digest dead or living wood in order to obtain carbon and other nutrients and these enzymatic systems have been exploited for both industrial and medical applications. Over 600 wood decay fungi species have been described in Korea; however, the recent application of molecular markers has dramatically altered the taxonomy of many of these wood decay fungi at both the genus and species levels. By combining molecular methods, specifically sequences of the internal transcribed spacer region, with traditional morphological characters, this study identified five new species records for Korea in five genera: Aurantiporus, Favolus, Neofavolus, Loweomyces, and Hymenochaetopsis. Three of these genera (Aurantiporus, Favolus, and Loweomyces) were previously unknown in Korea. The relatively simple morphology of the wood decay fungi often leads to ambiguous taxonomic assignment. Therefore, molecular markers are a necessary component of any taxonomic or evolutionary study of wood decay fungi. Our study highlights the need for a more robust and multifaceted approach in investigating new wood decay fungi in Korea.

A taxonomic note of the genus Lilium section Martagon in Korea (한국산 백합속(Lilium) 내 말나리절의 분류학적 검토)

  • The genus Lilium comprises about 100 species and is a major element of the family Liliaceae in temperate region of North Hemisphere. In the light of recent studies on the genus, we tried to examine the taxonomy of three Korean species (L. hansonii, L. tsingtauense, and L. distichum) belonging to section Martagon. The four available chloroplast regions (rbcL, matK, ndhF, and atpB) in Lilium species were selected and analyzed for a maximum likelihood phylogeny. As a results, three Korean species of section Martagon formed a monophyletic group. However, they nested within paraphyletic section Sinomartagon. Further, the discussions on taxonomic affinities of Korean species have made based on comparison between topology of phylogenetic tree and morphology.

Isolation, Characterization and Numerical Taxonomy of Novel Oxalate-oxidizing Bacteria

  • The present work is aimed at providing additional new pure cultures of oxalate utilizing bacteria and its preliminary characterization for further work in the field of oxalate-metabolism and taxonomic studies. The taxonomy of 14 mesophilic, aerobic oxalotrophic bacteria isolated by an enrichment culture technique from soils rhizosphers, and the juice of the petiole/stem tissue of plants was investigated. Isolates were characterized with 95 morphological, biochemical and physiological tests. Cellular lipid components and carotenoids of isolates were also studied as an aid to taxonomic characterization. All isolates were Gram-negative, oxidase and catalase positive and no growth factors were required. In addition to oxalates, some of the strains grow on methanol and/or formate. The taxonomic similarities among isolates, reference strains or previously reported oxalotrophic bacteria were analysed by using the Simple Matching (S/ sub SM/) and Jaccard (S$\_$J/) Coefficients. Clustering was performed by using the unweighted pair group method with arithmetic averages (UPGMA) algorithm. The oxalotrophic strains formed five major and two single-member clusters at the 70-86% similarity level. Based on the numerical taxonomy, isolates were separated into three phenotypic groups. Pink-pigmented strains belonged to Methylobacterium extorquens, yellow-pigmented strains were most similar to Pseudomonas sp. YOx and Xanthobacter autorophicus, and heterogeneous non-pigmented strains were closely related to genera Azospirillum, Ancylobacter, Burkholderia and Pseudomonas. New strains belonged to the genera Pseudomonas, Azospirillum and Ancylobacter that differ taxonomically from other known oxalate oxidizers were obtained. Numerical analysis indicated that some strains of the yellow-pigmented and nonpigmented clusters might represent new species.

The biodiversity representation assessment in South Korea's protected area network (보호지역 관리를 위한 생물다양성 평가)

  • National parks and other protected areas often do not adequately protect national biodiversity because they were originally created for socio-economic and/or aesthetic values. The Korean government has committed to expanding the extent of protected areas to fulfill its commitments to the Aichi Biodiversity Convention. To do so, it is necessary to quantify the current levels of biodiversity representation within existing protected areas and to identify additional conservation needs for vulnerable species and ecological systems. In this study, we assess the proportion of species ranges found in South Korea's protected areas, for the species documented in the 3rd National Ecosystem Survey. We modeled the range distribution of 3,645 species in the following taxonomic groups; plants (1,545 species), mammals (35), birds (132), herptiles (35), and insects (1,898) using the MaxEnt species distribution model and calculated how much of each species' range is within protected areas. On average, 17.4% of plant species' ranges are represented in protected areas, while for mammals and insects an average 12.0% is currently conserved. Conservation representation for herptiles averages 9.3%, while it is 8.6% for birds. Although large proportions of species that have restricted distributions should be represented in protected areas, 17 plant species, two insects (Parnassius bremeri and Lasioglossum occidens), and one bird species (Phylloscopus inornatus) with ranges smaller than 1,000 ㎢ have less than 10% of their ranges within protected areas. Establishing specific conservation goals such as the protection of endangered species or vulnerable taxonomic groups will increase the efficiency of the biodiversity conservation strategies. In addition, lowland coastal areas are critical for biodiversity conservation because the protected areas in South Korea are mainly composed of high mountainous areas.

A numerical taxonomic study on heterophyid trematodes (Heterophyidae에 관한 수리분류학적 연구)

  • A numerical taxonomy was studied on a group of heterophyid trematodes and analysis was made on the following species: Metagonimus yokogawai (3 OTU, Operational Taxonomic Unit) , Metagcnimus Miyata Type (3 OTU), Metagonimus takahashii (2 OTU), Heterophyes dispar (2 OTU), Heterephyes heterophyes (1 OTU), Heterophyes nocens (2 OTV), Heterophyopsis continua (1 OTU), Pygidiopsis summa (3 OTU), Stellantchasmus falcatus (2 OTU) and Stictodora sari (2 OTU). Twenty-six morphological characters were measured and their values were expressed as relative ratios. Similarity and correlation matrix among each individuals were calculated. Clustering analysis by Ward's method and factor analysis were performed using the SAS (Statistical Analysis System) package. As a results, the groups belonging to the genus of Metegenimus were divided into three phonons (Awetegonimus yokogawai, Metegcnimus Miyata Type, M. takahashii) , and Metagonimus Miyata Type was classified as the level of subspecies of M. takahashii. The groups belonging to the genus Heterophyes were clearly divided into three phonons (Heterophyes dispar, H. heterephyes, H. nocens), and H. nccens was classified as not a subspecies level of H. heterophyes but a distinct species. Other species were classified as distinct phonons. From these results, the appllcr lion of numerical taxonomy on trematode classification is considered to be a great aid to determine the limit of taxa.

The Taxonomic Study on the Genus Amanita in Korea (한국산(韓國産) 광대버섯속(屬)에 대(對)한 분류학적(分類學約) 연구(硏究))

  • For the purpose of investigating the distribution and flora of wild mushrooms in Korea, this study was conducted primarily on the genus Amanita. Through survey of Suweon, Kwangneung, Mt. Chiri, Taegwanryung, Yeongdong district and Mt. Hanta, 25 species of Amanita including 3 unrecorded species were discovered and are described in this paper. The author proposes to group 25 Korean species of Amanita into 3 subgenera according to morphological characteristics by the modern taxonomic system, and also designated the 3 unrecorded species of Amanita including one variety in Korean common name.

Taxonomic assessment of North American species of the genera Cumathamnion, Delesseria, Membranoptera and Pantoneura (Delesseriaceae, Rhodophyta) using molecular data

  • Evidence from molecular data supports the close taxonomic relationship of the two North Pacific species Delesseria decipiens and D. serrulata with Cumathamnion, up to now a monotypic genus known only from northern California, rather than with D. sanguinea, the type of the genus Delesseria and known only from the northeastern North Atlantic. The transfers of D. decipiens and D. serrulata into Cumathamnion are effected. Molecular data also reveal that what has passed as Membranoptera alata in the northwestern North Atlantic is distinct at the species level from northeastern North Atlantic (European) material; M. alata has a type locality in England. Multiple collections of Membranoptera and Pantoneura fabriciana on the North American coast of the North Atlantic prove to be identical for the three markers that have been sequenced, and the name Membranoptera fabriciana (Lyngbye) comb. nov. is proposed for them. Many collections of Membranoptera from the northeastern North Pacific (predominantly British Columbia), although representing the morphologies of several species that have been previously recognized, are genetically assignable to a single group for which the oldest name applicable is M. platyphylla.

Taxonomic Revision of the genus Chelon (Pisces, Mugilidae) from Korea (한국산 Chelon속 (Pisces, Mugilidae) 어류의 분류학적 재검토)

  • This study was carried out to reveal the taxonomic status of the two mullets, Chelon affinis and Chelon haematocheilus from Korea by comparison of their morphological characteristics. Chelan affinis, occurred in the southern coastal area, is a distinct species characterized by having a keel on the middorsal line in front of the spinous dorsal fin. However, Chelon haematocheilus, found in the all coastal areas of Korea excluding Cheju island is easily distinguished from the former in having the deep flat head. By comparison of five groups of Chelan haematocheilus, Kunsan group is slightly differed from others in the height of anal fin and caudal peduncle length. Although Chelon affinis is similar with Chelan haematocheilus in the view of meristic characters, they are distinguished from each other in the number of pyloric caeca and lateral line scales.

Phylogenetic rind Taxonomic Status of the Phytoplasmas Associated with Water Dropwort (Oenanthe javanica DC) Disease in Korea and Japan

  • To evaluate the phylogenetic and taxonomic status of the phytoplasmas associated with water dropwort (Oenanthe javanica DC) disease in Korea and Japan, their 16S rDNA was analyzed. DNAs extracted from water dropworts collected in Korea (Kyongnam province) and Japan (Chiba prefecture) affected by witches' broom and yellows were subjected to PCR using phytoplasma-specific primers, which amplified a 1.4-kbp fragment that included the 16S rDNA. Phytoplasmas were characterized by RFLP analysis using AluI, HaeIII, HhaI, KpnI, MseI, and RsaI restriction enzymes and by sequence analysis of the PCR products. The mater dropwort witches'broom (WDWB) and water dropwort yellows (WDY) 16S rDNA sequences were identical and closely related to onion yellows (OY, 99.9% identity), which belong to the aster yellows (AY) 16S-subgroup. However, the KpnI RFLP analyses clearly distinguished the WDY and WDWB phytoplasmas from the OY phytoplasma. The phylogenetic analysis based on 16S rDNA showed that WDWE and WDY phytoplasmas are members of a relatively homogeneous group that evolved from a common ancestor.

Molecular and morphological evidence for the taxonomic status of a newly reported species of Albula (Albuliformes: Albulidae) from Korea and Taiwan

  • To clarify the taxonomic status of five specimens of Albula collected from Korea and Taiwan (Albula sp.), genetic differences among them and other congeneric species were investigated based on mitochondrial DNA cytochrome b sequences. All five specimens of Albula sp. showed identical haplotypes and strongly supported a monophyletic group on phylogenetic analysis. The genetic differences between Albula sp. and ten other species of Albula ranged from 0.100 to 0.164. These values were almost equal to or exceeded the interspecific differences among the latter ten species (0.038-0.206). Morphological comparisons of Albula sp. and another four Indo-Pacific species, A. argentea, A. glossodonta, A. oligolepis and A. virgata, are also presented. Albula sp. could be distinguished from the latter four species in their numbers of pored lateral-line scales, anal fin rays and vertebrae, as well as in several proportional measurements.
