Tapioca주정증류 폐기물에서 Aspergillus niger 균주의 구연산 생산에 미치는 영향에 관한 연구 (Affection of Citric Acid Production from Tapioca Alcoholic Distillery Waste by Using the Cell of Aspergillus niger)

  • 주정공장에서 배출되는 타피오카 주정증류 폐기물을 발효기질로 하여 플라스크 진탕배양기에서 A. niger균주를 이용하여 폐기물 처리 및 구연산을 생산하는 실험에서 다음과 같은 결과를 얻었다. A. niger ATCC 9142균주를 사용하여 배양온도를 $25^{\circ}C\;30^{\circ}C$35^{\circ}C$로 변화하여 발효하였을 때 구연산 농도는 $30^{\circ}C$에서 6.09g/1로 최대가 되었으며, 플라스크 진탕배양기의 배양온도를 $30^{\circ}C$로 유지하면서 초기 pH를 조절하여 발효를 행한 결과, pH 4.3인 원폐수에서 구연산 생성량이 6.37g/l로 가장 높았다. 질소원, 인산원 및 금속이온이 구연산 생성에 미치는 영향을 알아보기 위하여 $NH_4NO_3,\;KH_2PO_4$$Mn^{2+}$를 첨가한 결과 이들 첨가물들이 구연산 생산을 감소시켰고, methanol, ethanol, isopropanol 및 n-propanol의 첨가가 구연산 발효에 미치는 영향을 알아보기 위한 실험에서는 methanol과 ethanol의 첨가가 구연산 생성을 촉진시켰다.

타피오카 전분을 첨가한 돈육 근원섬유 단백질의 물성 특성 및 돈육 모델소시지에 이용 (Evaluation of Rheological Properties of Pork Myofibrillar Protein with Tapioca Starch and its Utilization to the Pork Model Sausages)

  • 본 연구는 돈육 근원섬유 단백질에 식염의 농도와 타피오카 전분의 첨가량을 달리하여 점도, 겔 강도, 가열감량, 열량분석 및 전기영동의 변화를 관찰하여 근원섬유 단백질의 물성에 어떠한 영향을 주는지 알아보기 위해 실시하였다. 점도를 측정한 결과 식염의 농도가 증가함에 따라 혼합물의 점도가 유의적으로 증가하였으나, 타피오카 전분의 첨가에 따른 효과는 보이지 않았다(p<0.05). 겔강도는 식염의 농도와 타피오카 전분의 상호작용이 나타났고 식염의 농도와 타피오카 전분의 첨가량이 증가함에 따라 유의적으로 증가하였다(p<0.05). 가열감량에서도 타피오카 전분의 첨가로 인해 효과를 보여, 가열감량이 유의적으로 감소하였다(p<0.05). 열량분석을 통해 단백질의 변성을 관찰한 결과 $55^{\circ}C$에서 타피오카 전분이 겔을 형성하기 시작하는 피크가 보였으며, 그 이외의 피크에서는 식염의 농도와 타피오카 전분에 의한 피크 차이를 보이지 않았다. 전기영동의 결과 식염의 농도와 타피오카 전분의 첨가에 따른 단백질 분획 간의 큰 차이를 보이지 않았다. 따라서 근원섬유 단백질에 타피오카 전분을 첨가함으로써 겔 강도와 가열수율을 향상시키는 것으로 평가되었으며, 이러한 결과를 기초로 모델소시지를 제조하였다. 모델소시지의 결과로 2% 타피오카 첨가에 의해서는 유리수분이 낮아짐으로써 보수력을 증가하였으나 조직감의 증가는 나타나지 않은 반면 1%의 TGase의 첨가는 타피오카의 첨가유무에 상관없이 조직감을 증가시켰다. 따라서 TGase를 1% 첨가하고 타피오카 전분을 첨가할 경우 겔강도의 증가와 함께 보수력을 증진시킬 수 있을 것으로 판단된다.

Strain Improvement of Yeast for Ethanol Production Using a Combined Treatment of Electric Field and Chemical Mutagen N-Methyl-N'-nitro-N- nitrosoguanidine

  • The feasibility of using combined treatments of electric field and chemical mutagen N-methyl-N'-nitro-N-nitroso-guanidine (NTG) for the strain improvement of Saccharomyces sp. in ethanol production was examined. The treatment of electric field alone resulted in no effect on the lethality of yeast cells under the conditions of this study. However, when the electric field was applied together to the treatment of yeast cells with NTG, the electric field increased the lethal effect and auxotrophic mutation rate of NTG. The combined treatment of electric field and NTG also increased the chances of. obtaining superior yeast strains for the ethanol production from tapioca. A higher number of improved clones was obtained by the combined treatments of electric field and NTG than by the NTG treatment alone. The best clone, NF 30-9, which was also obtained by the combined treatment, produced $11.07\%$ (w/v) ethanol from tapioca slurry containing 25% (w/v) reducing sugar while the parental strain produced 9.77% (w/v).

영양물질 주입에 의한 메탄 발효 주정폐액의 효율증진에 관한 연구 (A study on Enhanced Efficiencies of Methane Fermented Alcohol Wastewater Treatment by Supplement of Nutrients)

  • In Korea, naked barley and tapioca are main raw materials for the production of fermentation ethyl alcohol, and one million drums bf 95% fermentation ethyl alcohol is produced per year by use of them. Stillage of alcoholic fermentation is mostly digested by methane fermentation process, and methane gas occured if methane fermentation process is recovered and mixed with fuel to decrease 25-30% for total fuel used in factories. In the anaerobic digestion process of naked barley stillage, supplement of nutrients is necessary to slove the problems caused by inhibitory materials contained if stillage and deficiency of nutrients. Therefore, the objective of this study was to examine why the anaerobic digesters using the naked barley distillery wastewater have shown the poor digestability frequently and how to control it. As the poor digestion was supposed to be occurred by the lack of iron as trace nutrient, the experiments were carried out to find out the optimum dosage and the way of addition of iron and to assess the quantitative evaluation of the type of iron in digesters. Initially, bottle test as batch digesters and lab-scaled continuous flow digesters were used in order to determine the digestion characteristics with tapioca and naked barley distillery wastewater. According to the results of batch tests, the poor digestion was caused by volatile fatty acids and could be improved by adding of calcium. The activity of the methanogenic bacteria were increased remarkably when the iron was added to the digester in the form of mixture with substrates.

혐기성 소화 및 막분리에 의한 Tapioca 전분의 폐수처리 (Treatment of Tapioca Starch Wastewater By Anaerobic Digestion Coupled With Membrane Separation Process)

  • This study thus looks into two treatment processess : i) Anaerobic digester coupled with hollow fibre membrane unit. Treatment of starch waste with anaerobic digester-membrane system was studied. $0.17\m^2$ area of hollow fibre membrane unit of known pore size was immersed into laboratory-scale anaerobic digestion system. The pore size of membrane was varied from 0.03 to $\0.15mu$m. The hydraulic retention time of anaerobic digester was varied from 1.5 to 10 days. The effect of hydraulic retention time on treatment efficiency was significant while effect of membrane size was not significant. The gas production was about 0.74㎥/kg COD treated. The COD removal efficient was about 80-95% depending on the hydraulic retention time. ii ) Crossflow ultrafiltration as post treatment to anaerobic filter. The effluent from anaerobic filter, which had a total COD in the range of 4,500-5,200 mg/L was treated by crossflow ultrafiltration units. The study conducted with different membrane pore size indicated that membrace with 1,000,000 molecular weight cut-off size gave a higher COD removal efficiency in the range of 83-87% while giving a study flux of $120-130 L/\m^2$.h. A study was conducted to see the long term clogging effect of membrane also.

알코올 발효 부산물(타피오카 · 겉보리 주정박) 활용한 생리활성 및 기능성 소재연구 (Study on Bioactive Materials Using By-Products of Alcohol Fermentation (Tapioca-Unhulled Barley Dried Distiller's Grains))

  • In this study, the bioactive activities (antioxidant activity, whitening activity, and anti-wrinkle effect) of dried tapioca-unhulled barley grain residue (TUDDG) obtained after alcohol fermentation, were measured. In the case of DPPH radical scavenging activity, the 50% ethanol extract of TUDDG showed the highest level of about 83% at the concentration of 100 mg/ml, and the ABTS radical scavenging activity was also about 98% of the 50% ethanol extract of TUDDG even at the concentration of 10 mg/mL. In the case of mushroom tyrosinase inhibitory activity, the 50% ethanol extract of TUDDG showed the highest activity at 100 mg/ml concentration of 37%. As a result of collagenage activity inhibition and elastase analysis, the 50% ethanol extract of TUDDG showed high activity with 4.2 mg/mL (IC50) and 26.1 mg/mL (IC50), respectively. Therefore, considering the physiological activity as well as the extraction efficiency of physiologically active substances, the 50% ethanol extract of TUDDG is judged to be highly effective.

변성 전분을 첨가한 식빵의 품질 특성 (Quality Characteristics of White Bread added Modified Starch)

  • 밀가루에 3종의 변성 전분(초산 감자 전분, 알파화 찰옥수수 전분, 히드록시프로필화 타피오카 전분)의 첨가량(3, 6, 9, 12, 15%)을 달리하여 제조하고, 이의 텍스쳐 특성과 기호도를 분석하였다. 첨가한 변성 전분 종류별 기호도는 대조구에 비하여 알파화 찰옥수수 전분은 3% 이상 6% 이하 사용 시, 초산 감자 전분의 경우 사용량 6%까지, 히드록시프로필화 타피오카 전분은 9%까지 높은 결과를 보였다. 이와 같은 결과로 볼 때 밀가루에 변성 전분을 첨가할 때 사용량의 증가에 따라 식빵의 firmness가 낮아지며, 변성 전분 종류 별로 일정 사용량의 범위에서 기호도가 상승되었다. 최근 식빵 텍스쳐에 대한 기호도가 기존의 일반 식빵보다 찰지고 보습성이 있는 제품을 선호하는 경향으로 변하고 있어 밀가루에 변성 전분을 첨가하여 소비자들이 요구하는 텍스쳐의 제품을 제조할 수 있으며, 변성 전분 첨가 식빵이 대조군에 비해 firmness가 낮아짐으로서 변성 전분의 첨가가 식빵의 노화 지연에도 도움을 주는 것으로 알 수 있었다. 또한 변성 전분 첨가 시에 식빵의 텍스쳐는 향상될 수 있으며, 부피가 작아지는 특성이 수반됨을 알 수 있었으나, 이들 전분류의 특성을 파악하여 제빵에 적정량을 첨가할 경우 다양한 텍스쳐의 빵 제품들을 만들 수 있을 것으로 기대된다.

전분의 종류에 따른 생선연육 스낵의 물리화학적 품질 특성 (Physicochemical Characteristics of Fried-fish Snacks with Different Types of Starch)

  • Fried-fish snacks containing surimi are a popular seafood product in Asia. To prepare fried-fish snacks, various ingredients are added to surimi. Among them, starch is important for producing snacks of desirable texture and taste. We investigated the physicochemical properties of fried-fish snacks containing arrowroot, potato, sweet potato, corn, and tapioca starches. The fried-fish snack with arrowroot starch showed the highest drying rate; after drying for 3 hours, the water content was unchanged. None of the starches affected the total volatile basic nitrogen values of the fried-fish snacks, which were approximately 1.5 mg/100 g, indicating freshness. The fried-fish snacks with corn and arrowroot starches had the highest brightness (L) values and the fried-fish snack with tapioca starch, the lowest L value. The hardness value of the fried-fish snack with tapioca starch was significantly higher than that of the other fried-fish snacks. In the sensory evaluation, the fried-fish snacks with arrowroot starch showed the best overall acceptance.

타피오카와 겉보리 혼합원료의 당화 및 알코올 발효의 특성 연구 (Saccharification and alcohol fermentation characteristics of a mixture of tapioca and hulled barley)

  • 알코올 발효에 이용되는 타피오카와 겉보리 혼합원료의 효소당화액을 제조하여 알코올 발효 특성을 조사하였다. 타피오카와 겉보리 혼합원료를 내열성 액화효소인 Spezyme Fred를 0.04% 사용하여 액화하고 글루코아밀레이스(G), 단백질가수분해효소(P), 베타글루칸 가수분해효소(B)를 혼합한 당화효소제(GPB)를 사용하여 당화하였다. 타피오카와 겉보리 혼합원료(7:3, w/w)를 $50^{\circ}C$에서 150분간 당화하였을 때 당화액의 포도당 함량은 11.9%였고 점도는 26 cp로 나타났다. 베타글루칸 가수분해효소(B)의 첨가는 당화액의 포도당 수율의 증가와 점성의 저하에 도움을 주었다. 타피오카와 겉보리의 혼합비율을 7:3(w/w)으로 하였을 때 다른 비율의 당화액에 비하여 점도가 낮으며 포도당 함량이 높게 나타났다. 타피오카와 겉보리 혼합원료에 300%의 가수량으로 확립된 조건에서 당화하고 $30^{\circ}C$에서 72시간 알코올 발효를 수행한 결과 타피오카와 겉보리 혼합원료를 이용한 알코올 발효에 적합한 발효액의 알코올 및 포도당 함량은 각각 9.0과 0.02%였고, pH와 산도는 각각 pH 4.3과 0.3%이었다.

Effect of Nonstarch Polysaccharide-Rich By-Product Diets on Nitrogen Excretion and Nitrogen Losses from Slurry of Growing-Finishing Pigs

  • An experiment was conducted to investigate the effect of diet for growing-finishing pigs with high level of non-starch polysaccharides (NSP) from by-products on nitrogen excretion and nitrogen losses from slurry during storage. Sixteen commercial crossbred barrows of about 68 kg BW were randomly allotted to one of four diets. The control diet was formulated using tapioca and rice as basal energy sources. In the other diets, tapioca was replaced by either coconut expellar, rice bran or beer by-product. The diets differed mainly in the amount and compostition of NSP. After a 12-day adaptation period, urine and faeces were collected separately in metabolism cages for 9 days. Urine and faeces from the first four days were used to analyse the nitrogen partitioning. Urine and faeces from the last 5 days were mixed as slurry. The slurry was sampled at the end of the collection period and again after 30 days storage, to analyse for nitrogen to calculate the losses. Increasing dietary NSP reduced urinary nitrogen and nitrogen losses from the slurry during storage. The pigs fed the diet based on beer by-product excreted the most nitrogen via faeces and the least nitrogen via urine. Nitrogen losses from slurry of pigs fed the beer by-product were from 34 to 65% lower than from the other three diets. It is concluded that including NSP-rich by-products in the diet of growing-finishing pigs reduces urinary nitrogen excretion and nitrogen losses from slurry during storage.
