Qualitative Evaluation of Ligninolytic Enzymes in Xylariaceous Fungi

  • Sixty-one strains representing the main genera of wood-decaying xylariaceous fungi (mainly in Daldinia, Hypoxylon, Kretzschmaria, Rosellinia, Penzigia, and Xylaria) were tested for their ability to produce ligninolytic enzymes. The phenol oxidase activity and fungal growth of the xylariaceous fungi on gallic aicid and tannic acid media showed a variation in their ability to degrade lignocellulose. A number of species showed equal 개 betterligninolytic enzyme activities than Coriolus versicolor, a known basidiomycete wood-degrader. A large variation of the enzyme activity was observed by individual strains as well as a substantial variation between the isolates of the same species. The most frequent ligninolytic enzymes were peroxidase and general oxidase. With 19% of the strains tested, peroxidase showed the strongest ligninolytic enzyme activity, while tyrosinase activity was detected only in 7% of the strains. All strains of Kretzschmaria and Rosellinia tested was positive for laccase. Xylariaceous fungi were able to degrade the macromolecule, lignin, using each specific ligninolytic enzyme in the specfic lignin degradation pathway.

Antioxidant and Anticancer Activities of Methanol and Water Extracts from Leaves of Cirsium japonicum

  • Potential antioxidant activities of methanol and water extracts of Cirsium japonicum var. ussuriense (CJ) leaves were examined. The reducing power and hydroxyl raical-scavenging activity assays showed that the methanol extract had a significantly higher activity than the water extract. In addition, the methanol extract showed a concentration-dependent reducing power, ranging from 0.228 to 1.072($0.1{\sim}0.5\;mg/mL$), as well as a high DPPH free radical-scavenging activity($EC_{50}=40.25\;{\mu}/mL$). The total phenolic(as tannic acid) and flavonoid(as quercetin) contents of the extract were 62.41 mg/g and 13.48 mg/g, respectively. The cytotoxic activity indicated that the methanol extract has an inhibition activity in the stomach carcinoma cell (35.40%), suggesting that the methanol extract of CJ leaves could be used as a potential source of pharmaceutical material.

천연염료에 관한 연구(11) -코치닐 색소의 양모섬유 염색성- (Studies on the Natural Dyes(11) -Dyeing Properties of Cochineal Colors for Wool Fibers-)

  • In order to study the properties of cochineal colors, uv-visible spectra of cochineal colors solution, dyeing properties on the wool in several dyeing conditions and thermodynamic parameters were investigated. UV-visible spectra of cochineal colors solution showed hypochromic effect with the lapse of irradiation time but bathochromic shift with decreasing acidity of solution and addition of metallic ions. The concentration of cochineal colors in wool fiber increased with the increase of dyeing temperature, time, and acidity of initial dyebath. The value of apparent diffusion coefficients and standard affinities of dyeing decreased with the increase of dyeing temperature. The standard heats of dyeing$(\Delta{H}^\circ)$ and variation of entropy$(\Delta{S}^\circ)$ increased with the increase of concentration of initial dyebath. The activation energy$(E_a)$ were calculated to be 1.399~2.595kcal/mol in condition of 6~1%(o.w.f) dyebath. Wool fabrics were dyed reddish blue by iron sulfate, copper sulfate, aluminum acetate and tannic acid, and red by tin chloride, respectively. Lightfastness of wool fabrics dyed by cochineal colors were increased by mordant treatment, especially copper sulfate and iron sulfate treatment.

Antioxidant Activities of Various Extracts of Hovenia dulcis Thunb Fruits

  • Hovenia dulcis Thunb fruits were successively extracted with hot water, water, methanol, ethyl acetate, and chloroform. The crude extracts were investigated for potential antioxidant by measuring scavenging against DPPH free radicals, reducing power, superoxide radicals, and protection of protein damage and cultured cells from a lethal dose of hydrogen peroxide ($H_2O_2$). In all chemical assays used, the hot water extract of H. dulcis fruits, which contained $61.14{\pm}2.57$ (Tannic acid mg/g extract, n=3) of total phenolic compounds contents exhibited highest activity in in vitro models of DPPH free radical scavenging activity, reducing power assay, superoxide radical scavenging activity and protection of protein damage. In addition, the hot water extract protected cultured RAW 264.7 macrophages from a lethal dose of $H_2O_2$ and reduced reactive oxygen species level in RAW 264.7 cells.

환경변이원 및 항변이원에 관한 연구: 살균제 Captan 및 유기인계 살충제 (Diazinon, MEP, Malathion 및 EPN)의 혼합처리가 E. coli $WP_2 S$ (uvrA-, trp-) 균주의 돌연변이와 생잔율로 미치는 영향

  • 살균제 Captan, 살충제 Diazinon, MEP, Malathion 및 EPN 의 단독 처리 그리고 Captan 반복처리와 이들 살충제와 서로 다른 조합으로의 동시적, 시간적 복합처리가 E. coli WP2S(uvrA-, trp-) 균주의 돌연변이유발 빈도 및 생잔율에 미치는 영향을 검토하여 다음과 같은 결과를 얻었다. 1. Captan, Diazinon, MEP, Malathion, EPN 및 UV 의 동시적 복합처리는 이들 농약의 단독 처리에서 보다 E. coli WP2S (uvrA-, trp-)균주의 돌연변이유발빈도가 높았고 이들 농약의 단독처리효과를 합한 가산적 영향으로 나타났으며, 동일조건하에서 이들 농약의 처리농도를 배로 늘렸을 때는 배가효과를 보였으나 이 균주의 증가된 돌연변이 유발빈도는 cinnamaldehyde 30ng 및 60ng/ml 그리고 tannic acid 50ng/ml 배지로 각각 첨가했을 때 감소되었고 생잔율은 증가되었다.

키토산과 탄닌 처리가 황토 염색에 미치는 영향 (The Effect of Chitosan and Tannin Treatment on the Natural Dyeing using Loess)

  • Tannin treatment has been used for improving the color fastness of dyed materials. In natural dyeing, the tannin treatment is highly effective in improving the fastness. The chitosan treatment also improves the fastness and depth of shade in natural dyeing. In this study, the effects of tannin and chitosan pre-treatment on the color and fastness in loess dyeing were investigated. Cotton woven fabric specimens and cotton knit fabric specimens were pre-treated with chitosan solution in acetic acid, and the specimens were then treated with or without tannin. The specimens were finally dyed with loess. The tannin treatment decreased the K/S values, while the chitosan treatment increased the K/S. Both the tannin treatment and the chitosan treatment increased the wash fastness and light fastness. In tannin treatment, tannin component and Fe component of loess may react together to decrease the lightness and develop dark color. For maintaining inherent color of the loess, it is much preferable to employ chitosan treatment rather than tannin treatment.

Antioxidant and Skin Anti-Aging Effects of Marigold Methanol Extract

  • The objective of this study was to evaluate the antioxidant and anti-aging effects of marigold methanol extract (MGME) in human dermal fibroblasts. Total polyphenolic and flavonoid contents in MGME were 74.8 mg TAE (tannic acid equivalent)/g and 85.6 mg RE (rutin equivalent)/g, respectively. MGME ($500{\mu}g/mL$) increased 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-azino-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical-scavenging, and superoxide dismutase (SOD)-like antioxidant activities by 36.5, 54.7, and 14.8%, respectively, compared with the control. At $1,000{\mu}g/mL$, these activities increased by 63.7, 70.6, and 20.6%, respectively. MGME ($100{\mu}g/mL$) significantly increased the synthesis of type 1 procollagen by 83.7% compared with control treatment. It also significantly decreased Matrix Metalloproteinase-2 (MMP-2) activity and MMP-1 mRNA expression by 36.5% and 69.5%, respectively; however, it significantly increased laminin-5 mRNA expression by 181.2%. These findings suggest that MGME could protect human skin against photo-aging by attenuating oxidative damage, suppressing MMP expression and/or activity as well as by stimulating collagen synthesis.

Antioxidant and Antidiabetic Activities of Ulmus davidiana Extracts

  • The antioxidant activities of water, ethanol, methanol, and chloroform extracts of Ulmus davidiana were evaluated using various antioxidant assays: DPPH (1,1-diphenyl-2-pricrylhydrazyl) free radical scavenging; hydroxyl radical scavenging; lipid peroxidation scavenging; and reducing power assays. All extracts, except the chloroform extract, demonstrated strong antioxidant activity in all assays. The chloroform extract had the highest hydroxyl radical scavenging activity, and its activity was equivalent to $\alpha$-tocopherol at a concentration of 0.5 mg/mL. Additionally, the antidiabetic activity of their extracts was also evaluated using a rat intestinal $\alpha$-glucosidase inhibition assay. Among all extracts investigated, the methanol extract had the highest $\alpha$-glucosidase inhibitory activity, although its activity was less than acarbose at $0.5\;{\mu}g/mL$. This result suggested that U. davidiana extracts may have antidiabetic activity. Total phenolic compounds and flavonoids were also measured. Phenolic compounds such as tannic acid, p-coumatric acid, and kaempferol were detected by high-performance liquid chromatography (HPLC). These results suggest that U. davidiana extracts may be useful as a potential source of antioxidant and antidiabetic materials.

Antioxidant Activity of Fractions from 70% Methanolic Extract of Sonchus oleraceus L.

  • The potential antioxidant activities of different fractions from a 70% methanolic (MeOH) extract of Sonchus oleraceus were assayed in vitro. All of the fractions exception of n-hexane showed a strong antioxidant activity, especially the ethyl acetate (EtOAc) fraction, which showed the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity ($IC_{50}=19.25{\mu}g/mL$). The results of hydroxyl radical scavenging activity and a reducing power assay showed concentration dependence, the EtOAc fraction demonstrating a better result than the other fractions at the same concentration in the studies. Additionally, the fractions' total phenolic (TP) contents was measured, phenolic compounds such as tannic acid, p-coumatric acid, quercetin, epicathchin, and kaempferol being detected by high performance liquid chromatography (HPLC). Meanwhile, a regression analysis revealed a moderate-to-high correlation coefficient between the antiradical activity and the TP contents, suggesting that fractions obtained from the 70% MeOH extract of S. oleraceus are of potential use as sources of antioxidant material.
