Species List of Aspergillus, Penicillium and Talaromyces in Korea, Based on 'One Fungus One Name' System (일균일명 체계에 의한 국내 보고 Aspergillus, Penicillium, Talaromyces 속의 종 목록 정리)

  • Aspergillus, Penicillium, and their teleomorphic genera have a worldwide distribution and large economic impacts on human life. The names of species in the genera that have been reported in Korea are listed in this study. Fourteen species of Aspergillus, 4 of Eurotium, 8 of Neosartorya, 47 of Penicillium, and 5 of Talaromyces were included in the National List of Species of Korea, Ascomycota in 2015. Based on the taxonomic system of single name nomenclature on ICN (International Code of Nomenclature for algae, fungi, and plants), Aspergillus and its teleomorphic genera such as Neosartorya, Eurotium, and Emericella were named as Aspergillus and Penicillium, and its teleomorphic genera such as Eupenicillium and Talaromyces were named as Penicillium (subgenera Aspergilloides, Furcatum, and Penicillium) and Talaromyces (subgenus Biverticillium) in this study. In total, 77 species were added and the revised list contains 55 spp. of Aspergillus, 82 of Penicillium, and 18 of Talaromyces.

New Records of Four Species Belonging to Eurotiales from Soil and Freshwater in Korea

  • Four strains of Penicillium and Talaromyces species are described and illustrated in an inventory of fungal species belonging to Eurotiales. The strains, CNUFC-DDS17-1, CNUFC-DDS27-1, CNUFC-PTM72-1, and CNUFC-YJW3-31, were isolated from soil and freshwater samples from South Korea. Based on their morphological characteristics and sequence analyses by the combined b-tubulin and calmodulin gene, the CNUFC-DDS17-1, CNUFC-DDS27-1, CNUFC-PTM72-1, and CNUFC-YJW3-31 isolates were identified as Penicillium pasqualense, Penicillium sanguifluum, Talaromyces apiculatus, and Talaromyces liani, respectively. The designated strains were found to represent a previously undescribed species of Korean fungal biota. In this study, detailed morphological descriptions and phylogenetic relationships of these species are provided.

A New Species and Five New Records of Talaromyces (Eurotiales, Aspergillaceae) Belonging to Section Talaromyces in Korea

  • Talaromyces is a genus within the phylum Ascomycota (class Eurotiomycetes, order Eurotiales, family Trichocomaceae). Many species in this genus are known to produce diverse secondary metabolites with great potential for agricultural, medical, and pharmaceutical applications. During a survey on fungal diversity in the genus Talaromyces in Korea, six strains were isolated from soil, indoor air, and freshwater environments. Based on morphological, physiological, and multi-locus (ITS, BenA, CaM, and RPB2) phylogenetic analyses, we identified five previously unrecorded species in Korea (T. brevis, T. fusiformis, T. muroii, T. ruber, and T. soli) and a new species (T. echinulatus sp. nov.) belonging to section Talaromyces. Herein, detailed descriptions, illustrations, and phylogenetic tree are provided.

  • Two new fungal species of the genus Talaromyces, Talaromyces purpurogenus and Talaromyces trachyspermus from the Trichocomaceae family, were recovered during an investigation of fungal communities in soil collected from the Gangwon-do and Jeollanam-do provinces of Korea. These two species have not been previously officially reported from Korea. In this study, detailed descriptions of internal transcribed spacer rDNA and beta-tubulin gene regions of these two fungi are presented. Morphological features of the two fungi in five agar media, potato dextrose, oatmeal, malt extract, czapek yeast extract, and yeast extract sucrose, are also reported. The species were identified on the basis of molecular and morphological analysis, and herein we present data with detailed descriptions and figures.

Molecular Docking Study of Novel Anti-Hepatitis B Virus Agents Isolated from Talaromyces Species

  • Hepatitis B virus is the leading source of liver disorders and is a global health problem and needs advancements in its treatment against increasing problems. Recently five vanitaracin derivatives were isolated from the fungus Talaromyces species which have anti-Hepatitis B virus activity. Hence, in the present study, molecular docking was carried out with five vanitaracin derivatives isolated from Talaromyces species and three known inhibitors.The objective of this work is to study the interaction of newly isolated compounds and compare its interaction with known inhibitors. The docking results revealed that vanitaracin derivatives have good interactions and has better docking score with the Hepatitis B virus and suggest SER2, SER4 and ASP30 are important residues involved in interaction with the inhibitors. These result authenticates vanitaracin derivatives contributes to inhibitory activity of Hepatitis B virus to treat liver disorders.

Diversity of Aspergillus, Penicillium, and Talaromyces Species Isolated from Freshwater Environments in Korea

  • In order to elucidate the fungal diversity and community structure in freshwater environments, numerous fungal strains were isolated from freshwater, submerged soils, twigs, dead insects, etc. Among them, the present study has focused specifically on Aspergillus, Penicillium, and Talaromyces species, which produce diverse useful metabolites in general. Twelve strains of Aspergillus isolated were identified as A. japonicus (n = 5), A. tubingensis (3), A. niger (2), and A. flavus (2), 10 strains of which belong to Aspergillus section Nigri, named black Aspergillus. Eight strains of Penicillium were identified as P. brasilianim (n = 3), P. oxalicum (2), P. crustosum (1), P. expansum (1), and P. piscarium (1). Two different strains of Talaromyces were identified as T. pinophilus and T. versatilis. Thus far, Penicillium piscarium and Talaromyces versatilis have been unrecorded in Korea, for which we provide detailed morphological and molecular characteristics.

Talaromyces halophytorum sp. nov. Isolated from Roots of Limonium tetragonum in Korea

  • Talaromyces halophytorum sp. nov. was isolated from the roots of halophyte Limonium tetragonum collected from Seocheon-gun, Korea in November 2015. It showed a slow growth on yeast extract sucrose agar at 25 ℃, no growth at 4 ℃ or 37 ℃ and produced smooth-walled and globose to sub-globose conidia. T. halophytorum is phylogenetically distinct from the other reported Talaromyces species of section Trachyspermi based on multi-locus sequence typing results using partial fragments of β-tubulin, calmodulin, ITS, and RNA polymerase II genes.

  • During fungal diversity surveys of the order Eurotiales in Korea, two fungal strains, EML-DG33-1 and EML-NCP50, were isolated from samples of rat dung and fig tree leaf collected at a garden located in Gwangju in 2014. To complete the National Species List of Korea, it is a prerequisite to verify whether many questionable species, which were previously recorded but not confirmed, indeed present in Korea. Herein, the isolates were confirmed as undescribed species, Paecilomyces variotii and Talaromyces amestolkiae based on the combination of morphological and phylogenetic analyses of multigenes including the rDNA internal transcribed spacer, ${\beta}-tubulin$, and RNA polymerase II subunit 2.

Eleven previously unrecorded fungal species isolated from hyphosphere soil supporting wild mushrooms in Jeju Island (제주도 야생버섯 hyphosphere 토양에서 분리된 국내 미기록 진균 11종 보고)

  • To understand microorganism effects on wild mushroom fruiting bodies, we investigated the fungi in hyphosphere soil supporting wild mushroom species Cortinarius violaceus, Amanita hemibapha, Laccaria vinacelavellanea, and Amanita verna found in the Gotjawal area of Jeju Island. Fungal species identification based on morphological traits and molecular analysis of ITS, LSU rDNA, and β-tubulin gene sequences resulted in isolation and identification of eleven fungal species previously unrecorded in Korea. These newly-recorded species are: Arthrinium kogelbergensis, Kalmusia longisporum, Keithomyces carneum, Neopyrenochaeta cercidis, Penicillium ranomafanaense, Phomatodes nebulosa, Pyrenochaeta nobilis, Tolypocladium album, Talaromyces kendrickii, Talaromyces qii, and Umbelopsis gibberispora, and their morphological characteristics and phylogenetic positions are described.

  • Two kinds of heat treatment method for the selective isolation of soil fungi to eliminate the commonest fungi and also to examine the vertical and seasonal distributions of the fungal population were applied to soil samples from two plots around Seoul area. The incubation method at $42^{\circ}C$ and heat treatment at $70^{\circ}C$ were used in this experiment. In the incubation method, the almost all the fungi isolated from two plots were mesophile, while the thermotolerant fungi was Aspergillus fumigatus and thermophilic fungi were Sporotrichum thermophile and Malbranchea pulchella var. sulfrea. The most dominant species isolated by this method was A. fumigatus. Nine genera and fourteen species were isolated from the two plots, and S. thermophile, Talaromyces ucrainicus,Malbranchea pulchella var. sulfrea were new to Korea. From the selection method by heat treatment at $70^{\circ}C$, ten genera and twenty species were isolated. Among these, the most fungi were also mesophile and thermotolerant fungus was A. fumigatus. The most dominant species isolated by this method was T. stipitatus, Talaromyces helicus var. major, Emericella nidulans var. nidulans, Chaetomium subspirale and Neosartorya fisheri var. fisheri were new to Korea. From the two isolation methods, it was found that the total number of soil fungi and frequency of species appeared including dominant ones were the highest at the soils of upper layer while the lowest at the soils of lower layer in its vertical distribution.
