The expressed sequence tag (EST) effort in Toxoplasma gondii has generated a substantial amount of gene information. To exploit this valuable resource, we chose to study tgd057, a novel gene identified by a large number of ESTs that otherwise show no significant match to known sequences in the database. Northern analysis showed that tgd057 is transcribed in this tachyzoite. The complete cDNA sequence of tgd057 is 1169 bp in length. Sequence analysis revealed that tgd057 possibly adopts two polyadenylation sites, utilizes the fourth in-frame ATG for translation initiation, and codes for a secretory protein. The longest open reading frame for the tgd057 gene was cloned and expressed as a recombinant protein (rd57) in Escherichia coli. Western analysis revealed that serum against rd57 recognized a molecule of ~21 kDa in the tachyzoite protein extract. This suggests that the tgd057 gene is expressed in vivo in the parasite.
Toxoplasmn gondii이 강독주인 RH주와 조직내 cyst 형성주인 Fukaya주의 세포막 단배 성분을 SDS 존재하에 서 전기영동하여 분석하였다. 먼저 RH tachyzoite와 Fukaya의 cyst를 각각 마우스의 복강액과 뇌조직으로부터 분리하였는데, 불연속 Percoll density-gradient서 원심분리하여 tachygoite는 50 U와 605 Percoll용액 경계면에서, cyst는 40%와 50%의 경계면 및 50%와 60 % 경계면에서 얻었으며, cyst는 저장액으로 처리하여 bradyzoite를 얻었다. Lactoperoxidase를 촉매로 세포막에 방사성 요오드를 표지시킨 후 자가방사표지그림을 얻었을 때, bradyzoite 는 15 KDa와 14 KDa의 분자량을 가진 단백질이 주요 단백질로 나타났으며, tachyzoite에서는 30 KDa 단백질이 주요 단백질로 나타났다. 또, 당단백질의 존재를 파악하기 위해서 lectin blotting을 시행하였는데, concanavalin A는 bradyzoite에서 200K∼50KDa의 여러 단백질을, .그리고 tachyzoite에서는 52KDa 단백질을 주로 하는 33K∼20 KDa단백질을 검출하였으며, phytohemagglutinin은 두·유형에서 아무런 단백질도 검출하지 못하였다. 한편, 이들을 효소면역이적법으로 항 Fukfya항체와 항 RH항체로 반응시켰을 때, 많은 교차 반응을 보였으나, bradyzoite에서는 15 KDa 단백질이, 그리고 tachyzoite에서는 52 KDa, 30 KDa 및 25 KDa 단백 짙이 각각 유형 특이 항원 단백으로 나타났다. 위의 결과들로, bradyzoite에서는 15 KDa 단백질이 당단백질은 아니지만 특이 항원성을 갖는 주요 백으로 나타났으며, tachyzoite에서는 지금까지 주요 세포막 단백으로. 알려진 P3O외에 당단백질이며 성을 갖는 세포막 단백으로 SaKDa 단백 (gps2)을 확인할 수 있었다.

Toxoplasmic encephalitis is caused by reactivation of bradyzoites to rapidly dividing tachyzoites of the apicomplexan parasite Toxoplasma gondii in immunocompromised hosts. Diagnosis of this life-threatening disease is problematic, because it is difficult to discriminate between these 2 stages. Toxoplasma PCR assays using gDNA as a template have been unable to discriminate between an increase or decrease in SAG1 and BAG1 expression between the active tachyzoite stage and the latent bradyzoite stage. In the present study, real-time RT-PCR assay was used to detect the expression of bradyzoite (BAG1)- and tachyzoite-specific genes (SAG1) during bradyzoite/tachyzoite stage conversion in mice infected with T. gondii Tehran strain after dexamethasone sodium phosphate (DXM) administration. The conversion reaction was observed in the lungs and brain tissues of experimental mice, indicated by SAG1 expression at day 6 after DXM administration, and continued until day 14. Bradyzoites were also detected in both organs throughout the study; however, it decreased at day 14 significantly. It is suggested that during the reactivation period, bradyzoites not only escape from the cysts and reinvade neighboring cells as tachyzoites, but also converted to new bradyzoites. In summary, the real-time RT-PCR assay provided a reliable, fast, and quantitative way of detecting T. gondii reactivation in an animal model. Thus, this method may be useful for diagnosing stage conversion in clinical specimens of immunocompromised patients (HIV or transplant patients) for early identification of tachyzoite-bradyzoite stage conversion.
톡소포자충(Tomplasmngondii)은 세포내에 기생하는 구포자충(Cuidia)의 일종으로 항원은 여러 가지 단백질로 구성되어 있으며 이들 항원의 분석은 면역학적 생화학적인 면에서 시도 해 볼 필요가 있다. 톡소포자충(RH주) tachyzoite의 용해물 및 분비항원을 SDS-PAGE(sodium dodecyl sulfate polyacrylamide gel electrophoresis)로 단백질 분획을 관찰하고 면역 전기영동이적법 (EITB, enzyme-linked immunelectrohansfer blot)을 시행하여 반응대를 관찰하였다. 토끼 (New Zealand산) 또는 BALB/c마우스를 톡소포자충으로 면역 또는 감염시키고 3주 또는 7주 후에 채혈 하였다. 면역 후 토끼나 마우스 혈청은 간접형광항체법(IFA)으로 항체가 1:1024 또는 1 : 128임을 확인하였다. 톡소포자충 용해물 및 분비항원으로 SDS-PAGE를 시행하였을 때 10 kDa에서 220 kDa까지 광범위한 단백질 분포대를 보였다. 톡소포자충 용해물 항원을 IgG 항혈청과 반응시켰을 때 주요 항원의 분자량은 23에서 35 kDa까지 5개의 반응대를 관찰하였으며 IgG와 IgM의 공동반응 대는 24. 27, 30. 35 kDa에서 관찰되었다. 분비항원을 IgG 항혈청과 반응시켰을 때 감염 마우스 의 복강액을 항원으로 한 경우 33(P30), 45 kDa의 반응대가 주요 항원임을 알 수 있었고 톡소포자 충을 CHL 세포로 in vitro에서 배양시킨 후 배양액의 상청액을 항원으로 EITB를 시행하였을 때 20 kDa. 30 kDa 이하의 분획에서 반응대가 관찰되었다. 이상으로 30 kDa 항원이 톡소포자충 tachyzoite의 용해물 뿐만 아니라 분비물에서도 관찰되는 중요한 항원임을 알 수 있었다.
톡소포자충(RH strain) tachyzoltes를 감마선에 조사하여 인체 백혈병 세포인 HL-60 세포와 정상 마우스의 복강 대식세포에 감염시킨 다음 충체 증식 능력에 어떠한 변화가 오는지 $^3H-uracil$ 흡수시험을 통하여 알아보았다 감마선 조사량은 30, 70, 100및 300 Gy로 하였고 tachyzoite감염은 시켰으나 감마선을 조사하지 않은 비조사군(NI군)과 tachyzoites를 넣지 않고 숙주세포만을 배양한 비감염군을 각각 두었다. 3H-uracil 흡수시험 결과 12-24시간 후 대식세포의 NI 군은 2,190-4,787(countperminute. 이하 같음), HL-60세포의 NI 군은 2,967-8,254의 높은 흡수량을 보였으나. 감마선 조사군은 대식세포 감염시 381-703(100 Gy조사군) 및 218-408(300 Gy조사군), HL-60 세포 감염시 1,911-2,618(30 Gy), 1,253-1,384(70 Gy), 1,013-1,090(100 Gy) 및 483-588(300 Gy)의 흡수량을 각각 보였다. 충체 증식에 대한 억제율은 300 Gy 조사시 12-24시간에 대식세포의 경우 90-92%. HL-60 세포의 경우 80-94%이었다. 이상과 같이 톡소포자충 RH tachyzoites가 감마선 조사 후 세포내 분열증식 능력에 치명적인 영향을 받는다는 것을 uracil 흡수 시험법으로 확인하였다.
This study aimed at finding a fast, sensitive, and efficient protocol for molecular identification of intracellular protozoa Toxoplasma (T.) gondii. For molecular detection of T. gondii, we developed a polymerase chain reaction coupled with dot blot hybridization assay (PCR/DBH). For DBH analysis, the amplified DNA of T. gondii tachyzoite was labeled by incorporation of digoxigenin. The DBH assay alone was capable of detecting down to $1{\times}10^4$ pg of T. gondii genomic DNA. The PCR alone was capable of detecting down to $1{\times}10^3$ pg of T. gondii genomic DNA, whereas the PCR/DBH assay was capable of detecting down to $1{\times}10^2$ pg of T. gondii genomic DNA, indicating that sensitivity of the PCR/DBH method was approximately 10 to 100 times higher than PCR or DBH alone. Our PCR/DBH assay will be useful for confirming the presence of T. gondii on the samples and differentiating T. gondii infection from other intracellular protozoa infections.
톡소포자충의 숙주-기생충 상호관계 연구의 일환으로 마우스 비장에서 분리한 T 림프구. B 림프 구 및 과립구(대부분 호중구로 구성)에 톡소포자층의 tachyzoites를 감염시킨 후 감염된 림프구와 호중구의 미세형태 변화를 관찰하는 한편 각 세포의 충체 감염에 대한 감수성을 동위원소 흡수시 험법을 이용하여 정량화하였다. 충체는 병원성이 강한 RH 주를 샤용하였고 각 세포는 BALB/c와 CBA 마우스의 비장에서 분리하여 사용하였다. 감염 후 24시간에 관찰한 결과, T 림프구, B 림프 구 및 호중구는 마우스 주에 상관없이 세포질 내에 tachyzoites가 한 개, 두 개 또는 7-8개까지 관찰되었다. 감염된 T 림프구는 충체 주변에 형성죈 parasitophorous vacuole로 인해 핵이 한 쪽으로 밀리며. 미토콘드리아의 수가 증가하였다 감염된 B 림프구는 조내형질세망(RER)이 대조군에 비해 발달하지 않았으며 감염된 호중구는 과립의 수가 현저히 감소하였다 림프구와 호중구의 톡소포자충 감염에 대한 감수성을 3H-uracil 흡수량으로 정량화한 결과. 마우스 주에 따른 차이는 없었고 모든 종류의 세포 내에서 충체가 활발히 증식함이 확인되었다. 이상의 결과로 볼 때, BALB/c와 CBA 마우스의 비장 T 림프구, B 림프구 및 호중구는 모두 톡소포자충의 tachyzoites 감염에 대해 감수성이 높음을 알 수 있었고, 감염된 면역세포는 그 기능이 저하될 것으로 추측된다.
A preservation test of Toxoplasma gondii tachyzoites for considerable time was tried to obtain simple and economical methods using various suspending media at $4^{\circ}C$ rather than serial passage of the parasite in mice. The preservation period was a term that the tachyzoites were detected from the peritoneal fluid of mice after inoculation of 2$\times$$10^5$ organisms preserved according to the lapse of time. The numbers of tachyzoite per 1\textrm{mm}^3$ of the peritoneal fluid with 2$m\ell$ of the saline solution taken in 4days after inoculation were presented as percentage in proportion to the control. The numbers. of tachyzoite per 1\textrm{mm}^3$ of the peritoneal fluid of the control were consisted of the average number of the tachyzoites of 10 mice inoculated with 2$\times$$10^5$ organisms by serial mouse passage. The tachyzoites could be preserved for 26 days in the suspending medium of saline solution at $4^{\circ}C$ Ringer's solution for 18 days, Hank's solution for 28 days, and egg-glycerine solution for 50 days.
