Phenotypic Suppression of Rad53 Mutation by CYC8 (CYC8에 의한 rad53 돌연변이의 표현형 억제에 대한 연구)

  • RAD53 functions as an effector kinase of checkpoint pathways in Saccharomyces cerevisiae, which plays a central role to regulate many downstream cellular processes in response to DNA damage. It also involves in transcriptional activation of various genes including RNR genes which encode the key enzyme required for dNTP synthesis. In this study, we identified CYC8 as a suppressor for the hydroxyurea sensitivity of $rad53{\Delta}$ mutation. $Rad53{\Delta}$ mutant transformed with a multi-copy plasmid containing CYC8 showed increased hydroxyurea resistance. In contrast, TUP1 which forms a complex with CYC8 did not function as a suppressor. In the case of mutations, both $cyc8{\Delta}$ and $tup1{\Delta}$ suppressed hydroxyurea sensitivity of $rad53{\Delta}$. Since CYC8 can propagate as a prion in yeast, overexpression of CYC8 induced misfolding of the normal CYC8 proteins, resulting in dominant cyc8-phenotype. Therefore, it is suggested that CYC8 can act as a multi-copy suppressor due to its prion property. It was observed that the levels of RNR transcription were increased in the yeast strains containing either multi-copies of CYC8 gene or $cyc8{\Delta}$ mutation, suggesting that the increased level of RNR will elevate the intracellular pools of dNTPs, which, in turn, suppress the phenotype of $rad53{\Delta}$ mutation.

  • Pressure vessels by composite materials were damaged sometimes during manufacturing or assembling. The state and the size of damage by low-velocity tests were investigated in this paper. Impactors of various masses and various tup shapes were dropped freely in the range of 120mm height to 700mm height. Compared with hemispherical tup of 12.7mm diameter, for hemispherical tup of 25.4mm diameter the size of surface dent was smaller but the size of delamination was bigger.

  • Low-velocity impact damage of a thick pressure vessel by composite materials was studied using the modified Herzian contact radius theory. Impactors of various masses and various tup shapes were dropped freely in the range of 20m to 200mm height. With acceleration gage and strain gage installed on the impactor, impact force and acceleration and Contact radius were measured. After a test, the samples were radiographed to scan the state of damage. Compared with hemispherical tup of 12.7mm diameter, the contact radius of hemispherical tup of 25.4mm diameter was bigger. And the experimental data and the theoretical data was different due to the mechanical properties difference. The acceleration value was changed linearly according to the height.

  • Strain gage method is used to evaluate the mode I dynamic stress intensity factor of marging steel(18Ni) and titanium alloy(Ti-6A1-4V). To decide the best strain gage position on specimen, static fracture toughness test was performed. Then instrumented charpy impact test and dynamic tensile test was performed by using strain gage method for evlauating dynamic stress intensity factor. Strain gage signals on the crack tip region are used to calculate the stress intensity factors. It is found that strain gage method is more useful than method by using load which is obtained from impact tup to assess dynamic characteristics such as dynamic stress intensity factor.

  • In this study, the effect of reducing cement by proportional addition of waste powder rubber on the performance of concrete under impact three-point bending loading were investigated experimentally and numerically. Concrete specimens were prepared by adding 5%, 10% and 20 % of rubber powder as filler to the mix and decreasing the same percentage of cement. For each case, three beams of $50mm{\times}100mm{\times}500mm$ were loaded to failure in a drop-weight impact machine by subjecting them to 20 N weight from 300mm height, while another three similar beams were tested under static load. The bending load-displacement behavior was analyzed for the plain and rubberized specimens, under static and impact loads. A three dimensional finite-element method simulation was also performed by using LUSAS V.14 in order to study the impact load-displacement behavior, and the predictions were validated with the experimental results. It was observed that, despite decreasing the cement content, the proportional addition of powder rubber until 10% could yield enhancements in impact tup, inertial load and bending load.

  • Innovation and Scale-up of Start-up companies are becoming important national tasks. In the past, it was spread the start-up policy paradigm such as 'Start-up America', 'Start-up Chile', 'Start-up Britain' to overcome the recession globally. However as the economic recovery has become more visible recently in advanced economies, it is shifting from a start-up support policy to a scale-up oriented policy paradigm such as 'Scale-up America', Scale-up UK', 'Scale-up Denmark'. It is necessary to enter the scale-up phase beyond the start-up phase to increase the number of high-quality jobs and to continue economic growth. Therefore, it is necessary to grow the start-up into a strong medium-sized company and to lay the foundation for survival. Therefore, the purpose of this study is to consider the antecedent factors that influence the scale-up aspiration for the start-up firm to grow into a scale-up company, and empirically identifies the differences between the stages of economic development and entrepreneurs in the country. In order to accomplish the purpose, this study predicted scale-up by aspiration which is a predictor of scale-up behavior because it is difficult to achieve visible growth in a short period of time due to the characteristics of start-up companies. In order to empirically explore these relationships, the data were collected from nascent entrepreneurs who have less than 3.5 years of the Adult Population Survey(APS) among the subjects surveyed by the Global Entrepreneurship Monitor(GEM) and the national economic development stage are divided into Innovation-driven, Efficiency-driven, Factor-driven type economies. For the test hypotheses, this study adopted the multi-level model analysis for comparison between national economic development stages and using the R 3.5.0 program. The results of this study are as follows. There is difference between the national economic development and the entrepreneur in the relationship between innovation orientation of entrepreneurs and scale-up aspirations. As the economy of the country develops, the innovation activity of the entrepreneur becomes more active. Since start-ups are heavily influenced by entrepreneurs, there is a difference in the degree of aspiration depending on how innovative an entrepreneur is in the same environment. In terms of the relationship between innovation orientation and scale-up aspiration, the fear of failure was found to differ between national economic development and entrepreneurs. The fear of failure differ from country to country, and this is one of the important factors affecting entrepreneurial activities. It is expected that the factors influencing the growth of the start-up companies which are identified through the results of these studies, will be used to create a suitable scale-up ecosystem according to the national economic development stage.
