Effect of Trichostatin A on Anti HepG2 Liver Carcinoma Cells: Inhibition of HDAC Activity and Activation of Wnt/β-Catenin Signaling

  • Purpose: To investigate the effect of deacetylase inhibitory trichostatin A (TSA) on anti HepG2 liver carcinoma cells and explore the underlying mechanisms. Materials and Methods: HepG2 cells exposed to different concentrations of TSA for 24, 48, or 72h were examined for cell growth inhibition using CCK8, changes in cell cycle distribution with flow cytometry, cell apoptosis with annexin V-FTIC/PI double staining, and cell morphology changes under an inverted microscope. Expression of ${\beta}$-catenin, HDAC1, HDAC3, H3K9, CyclinD1 and Bax proteins was tested by Western blotting. Gene expression for ${\beta}$-catenin, HDAC1and HDAC3 was tested by q-PCR. ${\beta}$-catenin and H3K9 proteins were also tested by immunofluorescence. Activity of Renilla luciferase (pTCF/LEF-luc) was assessed using the Luciferase Reporter Assay system reagent. The activity of total HDACs was detected with a HDACs colorimetric kit. Results: Exposure to TSA caused significant dose-and time-dependent inhibition of HepG2 cell proliferation (p<0.05) and resulted in increased cell percentages in G0/G1 and G2/M phases and decrease in the S phase. The apoptotic index in the control group was $6.22{\pm}0.25%$, which increased to $7.17{\pm}0.20%$ and $18.1{\pm}0.42%$ in the treatment group. Exposure to 250 and 500nmol/L TSA also caused cell morphology changes with numerous floating cells. Expression of ${\beta}$-catenin, H3K9and Bax proteins was significantly increased, expression levels of CyclinD1, HDAC1, HDAC3 were decreased. Expression of ${\beta}$-catenin at the genetic level was significantly increased, with no significant difference in HDAC1and HDAC3 genes. In the cytoplasm, expression of ${\beta}$-catenin fluorescence protein was not obvious changed and in the nucleus, small amounts of green fluorescence were observed. H3K9 fluorescence protein were increased. Expression levels of the transcription factor TCF werealso increased in HepG2 cells following induction by TSA, whikle the activity of total HDACs was decreased. Conclusions: TSA inhibits HDAC activity, promotes histone acetylation, and activates Wnt/${\beta}$-catenin signaling to inhibit proliferation of HepG2 cell, arrest cell cycling and induce apoptosis.

High-Performance Givens Rotation-based QR Decomposition Architecture Applicable for MIMO Receiver (MIMO 수신기에 적용 가능한 고성능 기븐스 회전 기반의 QR 분해 하드웨어 구조)

  • This paper presents an efficient hardware architecture to enable the high-speed Givens rotation-based QR decomposition. The proposed architecture achieves a highly parallel givens rotation process by maximizing the number of pivots selected for parallel zero-insertions. Sign-select lookahed (SSL)-CORDIC is also efficiently used for the high-speed givens rotation. The performance of QR decomposition hardware considerably increases compared to the conventional triangular systolic array (TSA) architecture. Moreover, the circuit area of QR decomposition hardware was reduced by decreasing the number of flip-flops for holding the pre-computed results during the decomposition process. The proposed QR decomposition hardware was implemented using TSMC $0.25{\mu}m$ technology. The experimental results show that the proposed architecture achieves up to 70 % speed-up over the TACR/TSA-based architecture for the $8{\times}8$ matrix decomposition.

Prosthetic shoulder arthroplasty in patients 40 years or younger: outcomes stratified by diagnosis and surgery

  • Background: The outcomes of patients 50-55 years old or younger undergoing prosthetic shoulder arthroplasty (PSA) may not generalize to younger patients. We report outcomes following PSA in a consecutive series of patients 40 years or younger. We hypothesize that total shoulder arthroplasty (TSA) provides better outcome and durability than resurfacing hemiarthroplasty (RHA). Methods: Patients were stratified by diagnosis and surgical procedure performed, RHA or TSA. Active range of motion and self-assessed outcome were evaluated preoperatively and at final follow-up. Results: Twenty-nine consecutive PSAs were identified in 26 patients, comprising 9 TSAs and 20 RHAs, with a minimum of 2-year follow-up. Twelve PSAs were performed for chondrolysis. Mean active forward elevation, abduction, external rotation, and internal rotation improved significantly (p<0.001 for all). Mean pain score improved from 6.3 to 2.1, Simple Shoulder Test from 4.0 to 9.0, and American Shoulder and Elbow Surgeons score from 38 to 75 (p<0.001 for all). Patients undergoing RHA and TSA had similar outcomes; but three RHAs required revision, two of these within 4 years of implantation. Four of five patients undergoing revision during the study period had an original diagnosis of chondrolysis. Conclusions: PSA in young patients provides substantial improvement in active range of motion and patient reported outcomes irrespective of diagnosis and glenoid management. However, patients undergoing RHA, especially for chondrolysis, frequently require subsequent revision surgery, so that RHA should be considered with caution in young patients and only after shared decision-making and counsel on the risk of early revision to TSA.

Intranasal Immunization With Nanoparticles Containing an Orientia tsutsugamushi Protein Vaccine Candidate and a Polysorbitol Transporter Adjuvant Enhances Both Humoral and Cellular Immune Responses

  • Scrub typhus, a mite-borne infectious disease, is caused by Orientia tsutsugamushi. Despite many attempts to develop a protective strategy, an effective preventive vaccine has not been developed. The identification of appropriate Ags that cover diverse antigenic strains and provide long-lasting immunity is a fundamental challenge in the development of a scrub typhus vaccine. We investigated whether this limitation could be overcome by harnessing the nanoparticle-forming polysorbitol transporter (PST) for an O. tsutsugamushi vaccine strategy. Two target proteins, 56-kDa type-specific Ag (TSA56) and surface cell Ag A (ScaA) were used as vaccine candidates. PST formed stable nano-size complexes with TSA56 (TSA56-PST) and ScaA (ScaA-PST); neither exhibited cytotoxicity. The formation of Ag-specific IgG2a, IgG2b, and IgA in mice was enhanced by intranasal vaccination with TSA56-PST or ScaA-PST. The vaccines containing PST induced Ag-specific proliferation of CD8+ and CD4+ T cells. Furthermore, the vaccines containing PST improved the mouse survival against O. tsutsugamushi infection. Collectively, the present study indicated that PST could enhance both Ag-specific humoral immunity and T cell response, which are essential to effectively confer protective immunity against O. tsutsugamushi infection. These findings suggest that PST has potential for use in an intranasal vaccination strategy.

p-Toluenesulfonic Acid 촉매를 이용한 1,4-Sorbitan 제조

  • This experiment was to determine the optimum conditions for D-sorbitol cyclization in the presence of p-toluenesulfonic acid(p-TSA) as acid catalyst before the esterification of sorbitol with fatty acids. The optimum conditions of hydration reaction to obtain maximum yield of 1,4-sorbitan were at $130^{\circ}C$, 200mmHg reduced pressure, after l50min and l%(w/w) p-TSA. In this condition the yield of 1,4-sorbitan was approximately 90%.

Characteristics of Carbon Dioxide Separation for Solid Absorbents According to Amine Order (아민 차수에 따른 고체 흡수제의 이산화탄소 분리 특성)

  • Primary and secondary amine-based sorbents were synthesized to investigate the operation capacity for the carbon dioxide separation TSA process. (3-Aminopropyl) triethoxysilane was used as a primary amine precursor as a crosslinking agent to synthesize a secondary amine precursor in which amine groups were crosslinked with a crosslinking agent. Carbon dioxide absorbed by primary amines is completely separated above 170 ℃. The working capacity of the primary amine absorbent was less than 2% when regenerated at 130℃. The secondary amine absorbent has a higher carbon dioxide separation capacity at a lower regeneration temperature than the primary amine absorbent. The secondary amine absorbent could predict process operation performance of about 6.5% with 2% carbon dioxide absorption and 100% carbon dioxide regeneration conditions. Therefore, it was found that the working capacity of the secondary amine absorbent was higher than that of the primary amine.

  • A facile and simple procedure for the synthesis of novel and known calix[4]resorcinarene derivatives were developed via a reaction of arylaldehydes with resorcinol in the presence of catalytic amounts of tungstate sulfuric acid (TSA) under solvent-free conditions. This eco-friendly method has many appealing attributes, such as excellent yields, short reactions times, use of safe and recoverable catalyst, and simple work-up procedures. TSA was characterized by powdered X-ray diffraction (XRD), X-ray fluorescence (XRF) and FTIR spectroscopy.

  • Purpose: The ability to noninvasively track the migration of neural progenitor cells would have significant clinical and research implications. We generated stably transfected F3 human neural progenitor cells with human sodium/iodide symporter (hNIS) for noninvasively tracking F3. In this study, the expression patterns of hNIS gene in F3-NIS were examined according to the cultured time and the epigenetic modulation. Materials and Methods: F3 human neural stem cells had been obtained from Dr. Seung U. Kim (Ajou University, Suwon, Korea). hNIS and hygromycin resistance gene were linked with IRES (Internal Ribosome Entry Site) under control of CMV promoter. This construct was transfected to F3 with Liposome. To investigate the restoration of hNIS gene expression in F3-NIS, cells were treated with demethylating agent (5-Azacytidine) and Histone deacetylase inhibitor (Trichostatin A: TSA). The expression of hNIS was measured by I-125 uptake assay and RT-PCR analysis. Results: The iodide uptake of the F3-NIS was higher 12.86 times than F3 cell line. According to the cell passage number, hNIS expression in F3-NIS gradually diminished. After treatment of 5-Azacytidine and TSA with serial doses (up to $20{\mu}M$, up to 62.5nM, respectively) for 24 hours, I-125 uptake and mRNA of hNIS in F3-NIS were increased. Conclusion: These results suggest that hNIS transfected F3 might undergo a change in its biological characters by cell passage. Therefore, the gene ex[ressopm of exogenous gene transferred human stem cell might be affected to the epigenetic modulation such as promoter methylation and Histone deacetylation and to the cell culture conditions.

  • Quality and distribution of the TEA(Total Enclosed Area), TSA(Twine Surface Area), solidity ratio and resistance according to the partial nets of the midwater trawl net were examined through experiment at the sea to standardize a design of the midwater trawl net.The results can be summarized as follows;1. The TEA, TSA and resistance of the trawl net were increased definitely according to increasing the scale of the trawl net, and this is the most important factor to design the trawl net.2. As the result of analysis of the distributions of the bag net was smaller than the others. Because TEA for rear end of the wing net is decreased suddenly, we can judge that the trawl net was towed irregularly. 3. The resistance distribution of the partial net was increased suddenly from central part of the bag net and was showed large value in the codend.4. The resistance of the net from the position one of three to gab net and codened charged 75% at all resistance of net.

Purification and Characterization of Thiol-Specific Antioxidant Protein from Human Liver: A Mer5-Like Human Isoenzyme

  • A 23-kDa molecular mass of antioxidant protein was purified from human liver. This protein exhibited the preventive effect against the inactivation of glutamine synthetase by a metal-catalyzed oxidation system. This antioxidant activity was supported by a thiol-reducing equivalent such as dithiothreitol in a similar manner to that of the 25-kDa thiol-specific antioxidant protein (TSA) from human red blood cells (HR). However, a thioredoxin-linked peroxidase activity of thiol-specific antioxidant protein of human liver (HLTSA) (0.91 ${\mu}mol/min/nmol$ of HLTSA) was much lower than that of thiol-specific antioxidant protein of human red blood cells (HRTSA) (16.4 ${\mu}mol/min/nmol$ of HRTSA). This HLTSA is also immnologically distinct from HRTSA Amino acid sequences of the three tryptic peptides (P1, P2, P3) of HLTSA were found to be completely homologous to segments of the known Mer5-like protein, which belongs to the known TSA family.
