OVERCOMING THE NUTRITIONAL LIMITATIONS OF RICE STRAW FOR RUMINANTS 1. UREA AMMONIA TREATMENT AND SUPPLEMENTATION WITH RICE BRAN AND GLIRICIDIA FOR LACTATING SURTI BUFFALOES

  • Fifty-six lactating Surti buffaloes, fed rice straw, were allocated to seven treatment groups as follows: 1. Straw supplemented with 2% urea (SS) + 1.5kg rice bran (RB) 2. Straw treated with 4% urea in an open stack (TS open) 3. TS open + 1.5 kg RB 4. TS open + 3.0 kg RB 5. TS open + 1.5 kg RB + 3.0 kg Gliricidia (Gl) 6. Straw treated with 4% urea in a closed pit (TS closed) 7. TS closed + 1.5 kg RB + 3.0 kg Gl Milk production, butterfat percentage and liveweight gain of cows and calves were measured and tested with analysis of variance. The results are: - The animals on urea treated straw (group 2) had a higher milk production (p<0.05), higher butterfat production (p<0.05) and less liveweight gain loss (p<0.05) than the animals on urea supplemented straw (group 1). Butterfat percentage also increased by treatment, although not significantly (p>0.05). - Increasing levels of rice bran (groups 3 and 4 compared to 2) increased total milk production and milked quantity of butterfat, while butterfat percentage decreased (p < 0.05). - Milk production increased (p <0.05) with extra rice bran added (group 4 compared to 3), but was not affected (p > 0.05) by Gliricidia addition (group 5 compared to 3). Butterfat percentage dropped with extra rice bran supplement (p <0.05). The lack of response to Gliricidia indicated that protein is not limiting in treated straw, or that Gliricidia protein is partly insoluble. - System of treatment had no effect on milk production (p >0.05), while supplementation with 1.5 kg RB and 3.0 kg Gliricidia increased production and caused a lower butterfat percentage (p <0.05) (groups 2, 5, 6 and 7 compared). A significant (p <0.05) interaction treatment system x supplementation was present. It was concluded, that both treatment and supplementation did affect milk production as well as milk composition. Gliricidia addition gave less effect than rice bran, indicating different requirements for starchy substances in the feed. Treatment of straw does not negatively affect butterfat production, it can increase butterfat production and even butterfat percentage.

Continuous Mesophilic-Dry Anaerobic Digestion of Organic Solid Waste (유기성고형폐기물의 연속 중온 건식혐기성소화)

  • Continuous dry anaerobic digestion of organic solid wastes (30% TS, Total Solids) comprised of food waste and paper was performed under mesophilic condition. During the operation, hydraulic retention time (HRT) was decreased as follows: 150 d, 100 d, 60 d, and 40 d, which corresponded to the solid loading rate of 2.0, 3.0, 5.0, and 7.5 kg TS/$m^3$/d, respectively. Volumetric biogas production rate ($m^3$/$m^3$/d) increased as HRT decreased, and the highest biogas production rate of 3.49${\pm}$0.31 $m^3$/$m^3$/d was achieved at 40 d of HRT. At this HRT, high volatile solids (VS) reduction of 76% was maintained, and methane production yield of 0.25 $m^3$/kg $TS_{added}$ was achieved, indicating 67.4% conversion of organic solid waste to bioenergy. The highest biogas production yield of 0.52 $m^3$/kg $TS_{added}$ was achieved at 100 d of HRT, but it did not change much with respect to HRT. For the ease feed pumping, some amount of digester sludge was recycled and mixed with fresh feed to decrease the solid content. Recirculation volume of 5Q was found to be the optimal in this experimental condition. Specific methanogenic activity (SMA) of microorganisms at mesophilic-dry condition was 2.66, 1.94, and 1.20 mL $CH_4$/g VS/d using acetate, butyrate, and propionate as a substrate, respectively.

Stable Maintenance of Recombinant Plasmid Containing trp $^+$ Operon in E. coli Cultures by the phe W$^+$ -pheS$^{t8}$ System (대장균 배양 중 phe W$^+$-pheS-$^{-ts}$ System에 의한 재조합 trp$^+$ 플라스미드의 안정적 유지)

  • To improve the stability of recombinant pBR322-trip$^+$ plasmid (pLTW24) in E. coli culture, a positive selection system was devised. A DNA fragment containing pheW$^+$ gene (a structural gene for tRNA$^{phe}$ was isolated and inserted into the pBR322-trip$^+$ plasmid(pLTP24). A temperature sensitive host strain. LC901-pheS$^{-ts}$, was constructed for this plasmid by transducing pheS$^{-ts}$ allele (phenylalanyl-tRNA synthetase) to E. coli LC901 using P1kc bacteriophage. The LC901-pheS$^{-ts}$ cells were unable to grow at a restrictive temperature when they had lost the pBR322 :: pheW$^+$ (pLTP24) plasmid. The effects of pheW$^+$ gene on the plasmid stability and the expression level of trip$^+$ gene in LC901-pheS$^{-ts}$ strain were investigated. The proportion of Trip$^+$ colonies among LC901-pheS$^{-ts}$ strain carrying plasmid pLTP24 was 99%, whereas that of LC901 strain carrying plasmid pLTW24 was 7% at the end of 20 generations. After 100 generations of growth, the strain LC901-pheS$^{-ts}$ carrying plasmid pLTP24 showed little loss of plasmids. While the majority of plasmid pLTW24 in LC901 strain were lost in the same period. The activities of tryptophan synthetase (T. Sase) and anthranilate synthetase (A. Sase) in LC901 strain carrying pLTW24 were about 1.2 times and 1.8 times respectively of those in LC901-pheS$^{-ts}$ strain carrying pLTP24.

Pig slurry treatment by the pilot scale hybrid multi-stage unit system (HMUS) followed by sequencing batch reactor (SBR) (HMUS와 SBR 반응조를 이용한 축분처리에 관한 연구)

  • Experiments in a pilot-scale hybrid multi-stage unit system (HMUS) combination of ATAD and EGSB followed by SBR process for pig slurry treatment were conducted to demonstrate the feasibility of using autothermal thermophilic aerobic digestion (ATAD) and expended granular sludge bed (EGSB) followed by sequencing batch reactor (SBR) system. Contaminants in pig slurry with high organic matter, nitrogen (N) and phosphorus (P) content were completely removed in the combined process. The highest removal rate for CODcr among contaminants in the feed pig slurry was attained by about 43.3% in ATAD unit process. Also TS removal rate of 96.5% was attained and the highest in the next coagulation unit process. The highest removal rate of CODcr under operating parameter conditions of OLR(organic loading rate), 3-6Kg $COD/m^3{\cdot}day$ and line velocity, 1.5-4m/h was earned at 3days of HRT. The disinfection of pathogens was effective at 50,000mg/L of TS in ATAD unit process. Biogas production per organic removal was $2.3{\sim}8.5m^3/kgTS{\cdot}d$ (average $5.2m^3/kgTS{\cdot}d$) in EGSB unit process. The average removal rates of CODcr 71.7%, TS 64.1%, TN 45.9%, and TP 50.4% were earned in the intermittent aeration SBR unit process.

Acoustic Target Strength Characteristics of Two Species of Multiple Jellyfishes, Aurelia aurita and Cyanea nozakii, in the Southern Coast of Korea (남해 연안에 분포하는 해파리(Aurelia aurita, Cyanea nozakii)의 복수 개체에 의한 음향 표적강도 특성)

  • Understanding the abundance and distribution of massive jellyfish is necessary to forecast where or when their blooms will happen. The acoustic technique is one of the most useful methods of obtaining information if the acoustic characteristics of the targets are known. This study was conducted to determine the acoustic target strength (TS, dB) of two jellyfish species, Aurelia aurita and Cyanea nozakii, in the southern coast of Korea. For the ex situ measurements, 120, 200, and 420 kHz split beam transducers were used, and jellyfish with various bell lengths were arranged to prepare multiple jellyfish. Under 2 vertical individuals, the mean TS for multiple A. aurita at 120, 200, and 420 kHz was -72.7, -71.7, and -68.2 dB, respectively. In the case of 5 vertical individuals, the mean TS of the species was -71.3, -68.2, and -62.0 dB. Finally, the mean TS of C. nozakii at 120, 200, and 200 kHz was -62.0, -60.3, and -58.2 dB under 2 individuals and -58.1, -57.4, and -54.0 dB under 4 individuals, respectively. For both species, higher numbers of jellyfish resulted in a higher TS. In addition, higher frequencies were associated with a higher TS for the same jellyfish. These TS results for two species can be used as essential data for the acoustic detection of jellyfish in an open ocean or coastal area.

Therapeutic Profile of T11TS vs. T11TS+MiADMSA: A Hunt for a More Effective Therapeutic Regimen for Arsenic Exposure

  • Arsenic exposure is a serious health hazard worldwide. We have previously established that it may result in immune suppression by upregulating Th2 cytokines while downregulating Th1 cytokines and causing lymphocytic death. Treatment modalities for arsenic poisoning have mainly been restricted to the use of chelating agents in the past. Only recently have combination therapies using a chelating agent in conjunction with other compounds such as anti-oxidants, micronutrients and various plant products, been introduced. In the present study, we used T11TS, a novel immune potentiating glycopeptide alone and in combination with the sulfhydryl-containing chelator, mono-iso-amyl-dimarcaptosuccinic acid (MiADMSA) as a therapeutic regimen to combat arsenic toxicity in a mouse model. Results indicated that Th1 cytokines such as TNF-${\alpha}$, $IFN{\gamma}$, IL12 and the Th2 cytokines such as IL4, IL6, IL10 which were respectively downregulated and upregulated following arsenic induction were more efficiently restored to their near normal levels by T11TS alone in comparison with the combined regimen. Similar results were obtained with the apoptotic proteins studied, FasL, BAX, BCL2 and the caspases 3, 8 and 9, where again T11TS proved more potent than in combination with MiADMSA in preventing lymphocyte death. The results thus indicate that T11TS alone is more efficient in immune re-establishment after arsenic exposureas compared to combination therapy with T11TS+MiADMSA.

Impacts of Different Urban Surfaces on Summer Thermal Performance

  • This study measured temperatures and albedos of urban surfaces for different colors and materials during summer, and calculated the energy budget over different urban surfaces to find out the thermal performance affecting the heat built-up. The study selected six surface colors and 13 materials common in urban landscape. Their surface temperatures (Ts) and albedos were measured at a given time interval in the daytime from June to August. Average Ts over summer season for asphalt-colored brick was $4.0^{\circ}C$ higher than that for light red-colored one and $9.7^{\circ}C$ higher than that for white-colored one. The Ts for artificial surface materials of asphalt paving, brown brick wall, and green concrete wall was $6.0^{\circ}C$ higher than that for natural and semi-natural ones of grass, grassy block, and planted concrete wall. There was the greatest difference of $16.3^{\circ}C$ at midafternoon in the Ts between asphalt paving and planted concrete wall. Average albedo over summer season of surface materials ranged from 0.08 for asphalt paving to 0.67 for white concrete wall. This difference in the albedo was associated with a maximum of $15.7^{\circ}C$ difference at midafternoon in the Ts. Increasing the albedo by 0.1 (from 0.22 to 0.32) reduced the Ts by about $1.3^{\circ}C$. Average storage heat at midday by natural and semi-natural surfaces of grass and grassy block was about 10% lower than that by artificial ones of asphalt, light-red brick, and concrete. Reflected radiation, which ultimately contributes to heating the urban atmosphere, was 3.7 times greater for light-red brick and concrete surfaces than for asphalt surface. Thus, surfaces with in-between tone and color are more effective than dark- or white-colored ones, and natural or semi-natural surfaces are much greater than artificial ones in improving the urban thermal environment. This study provides new information on correlation between Ts and air temperature, relationship between albedo and Ts, and the energy budget.

Effects of Various Kinds of Salt on the Quality and Storage Characteristics of Tteokgalbi

  • This study was carried out to evaluate the effects of various kinds of salt on the quality and storage characteristics of tteokgalbi. The tteokgalbi was prepared using four types of salt: 1.5% purified salt (control, C), 1.5% five-year-old solar salt (FS), 1.5% Topan solar salt (TS), and 1.5% French Gu$\acute{e}$rande solar salt (GS). The moisture, crude lipid, crude ash, crude protein and calorie contents, water holding capacity, and cooking loss were not significantly different between control and all other treatments. As for the textural characteristics, the use of GS increased the hardness of the tteokgalbi. According to the sensory evaluation, the use of TS had the best score in springiness. Tteokgalbi made with TS and GS had the two highest scores in flavor and total acceptability. During 15 d of storage, the contents of 2-thiobarbituric acid (TBA) value, volatile basic nitrogen (VBN) and the total microbial counts increased, while the pH decreased. The TBA values of the tteokgalbi containing TS and GS were lower than that of C. Lightness (L) and yellowness (b) values decreased during storage, but redness (a) displayed no significant difference during storage. Overall, the best results, in terms of TBA value and sensory attributes, were obtained for the tteokgalbi containing TS and GS.

Determining the target strength bambood wrasse (Pseudolabrus japonicus) using Kirchhoff-ray mode

  • Although ex situ target strength (TS) measurements using dual- and split-beam systems have become the primary approach of estimating fish abundance, theoretical model estimation is a powerful tool for verifying the measurements, as well as for providing values when making direct measurements is difficult. TS values for 20 samples of live bambooleaf wrasse (Pseudolabrus japonicus) whose target length (TL) ranged between 13.7 and 21.3 cm were estimated theoretically using the Kirchhoff-ray mode model, and the TS values for 18 live fish samples were additionally measured at ${\sim}0^{\circ}$ tilt angle to the swimming aspect using a tethered method at a frequency of 120 kHz to verify the theoretical values. The digitizing intervals used to extract the fish body and swim bladder morphology in the X-ray photographs significantly affected the calculated TS patterns, but variations based on the speed of sound and density ratio values for the general range of fish flesh were relatively small (within 1 dB). Close agreement was observed between the measured and theoretical TS values, and the correlation between the average TS and body length of the fish could be calculated accurately as <$TS_{120kHz}$>= 20logTL (cm) -71.6 using the theoretical method.

  • Black porgy and fat greenling are commercially important fish species due to the continuously increasing demand in Korea. When estimating acoustically the fish length by a fish sizing echo sounder, it is of crucial importance to know the target strength (TS) to length dependence. In relation to these needs, the target strength experiments for live fishes were conducted in an acrylic salt water tank using two split-beam echo sounders operating at 70 and 120kHz. The target strength under well-controlled laboratory conditions was simultaneously measured with the swimming movement by digital video recording (DVR) system and analyzed as a function of fish length (L) and frequency (or wavelength ${\lambda}$). Equations of the form TS-alog (L)+blog (1)+c were derived for their TS-length dependence. The best fit regression of TS on fork length for black porgy was TS=20.62 log (L, m)-0.62 log (${\lambda}$, m)-30.68 ($r^2$=0.77). The best fit regression of TS on fork length for fat greenling was TS=12.06 log (L, m)-5.85 log (${\lambda}$, m)-22.15 ($r^2$=0.44).
