  • Korea Railroad Research Institute(KRRI) has developed the rubber-tired AGT system from 1999 to 2004. The rubber-tired AGT vehicle is now on test for its performance and function in the Gyeong-San test line. Tn this paper, we provide the reliability management plan to assure required the RAMS(reliability, availability, maintainability & safety) of the AGT vehicle system.

  • Tn this paper. a numerical method for the system analysis of the slab track is presented. In the model rail is considered to have a distributed mass and to be supported by sleepers discretely on slab. The slab supported by discrete slab pads- isolators Put on floor is modelled by finite beam elements The system analysis for the same type floating slab laid in Puchon station is carried out using the developed program. Then the numerical results are compared with system requirements for slab track.

  • Low Earth Orbit(LEO) Non-Terrestrial-Network(NTN) 은 위성을 사용하여 통신 서비스를 제공하지 못하는 도시 이외의 산간, 바다, 항공기 또는 외진 지역에 통신 서비스를 제공하는 모델이다. Terrestrial Network(TN) 핸드오버에서 사용되는 Measurement-Based Triggering(MHT)의 경우 User Equipment(UE)에서 측정된 값을 기반으로 Measurement Report를 전달한다. 그러나 NTN 환경에서의 HO triggering은 TN 과는 달리 장거리 통신을 요구하고 위성이 빠른 속도로 이동함에 따라 MHT에 사용되는 지표들을 대체할 방법이 필요하다. 이 논문에서는 측정 유효성을 대체할 수 있는 지표로 UE와 셀 중심 간의 거리 및 고도 각을 활용하여 HO triggering을 예측하는 시스템을 제안한다. 제안한 예측 시스템은 핸드오버 triggering 예측에 대해 우수한 성능을 보였으며 좋은 측정 결과를 얻을 수 있었다.

Stability Evaluation of Phased Isolation Intra-Clarifier Ditch Process on Short-Term Hydraulic Shock Loading (단기 수리학적 충격부하시 침전지 내장형 상분리 산화구공정의 처리 안정성 평가)

  • The phased isolation intra-clarifier ditch system used in this study is a simplified novel process enhancing simultaneous removal of biological nitrogen and phosphorus in municipal wastewater in terms of elimination of additional pre-anaerobic reactor, external clarifier, recycle of sludge, and nitrified effluent recirculation by employing intrachannel clarifier. Laboratory-scale phased isolation ditch system was used to assess the treatability on municipal wastewater. When the system was operated at the HRTs of 6~12hours, SRTs of 9~31days, and cycle times of 2~8hours, the system showed removals of BOD, TN, and TP as high as 88~97%, 70~84%, and 65~90%, respectively. The rainfall in Korea is generally concentrated in summer because of site-specific characteristics. Especially, the wet season has set in on June to August. In combined sewers, seasonal variations are primarily a function of the amount of stormwater that enters the system. In order to investigate the effect of hydraulic shock loading on system performance, the laboratory-scale system was operated at an HRT of 6hours (two times of influent flowrate) during two cycles (8hours). The system performance slightly decreased by increasing of influent flowrate and decreasing of system HRT. Nitrification efficiency and TN removal were slightly decreased by increasing of influent flowrate (decreasing of system HRT), whereas, the denitrification was not affected by hydraulic shock loading. However, the higher system performance could be achieved again after four cycles. Thus, the phased isolation technology for enhanced biological nutrient removal in medium- and small-scale wastewater treatment plants suffering fluctuation of influent quality and flowrate.

The Limnological Survey of Lagoons in the Eastern Coast of Korea (1): Lake Chungcho (동해안 석호의 육수학적 조사(1): 청초호)

  • Water quality and Pollution state of Lake Chungcho were evaluated during three years from 1998 to 2000. We surveyed physicochemical parameters, and TSI (trophic state index) was calculated using TP, Chl. a, and SD (secchi disc transparency) data of growing season average. Water samples were collected bimonthly except freezing season. During the study period, total annual precipitation in 1998, 1999 and 2000 year was 1,797,1,722 and 1,345 mm, respectively. Salinity and conductivity were high($29.3{\pm}5.5\;ppt$, and $45,105{\pm}7,585\;{\mu}S/cm$) then other lagoons in the Eastern Coast of Korea. Chemocline was formed by salinity at $0.5{\sim}1.5\;m$ water depth. As a result of this, DO concentration of hypolimnion was below $3.0\;mgO_2/L$. Especially, when intense chemocline was formed, temperature of hypolimnion was higher than epilimnion. Secchi disc transparency, chlorophyll a, and COD were $1.8{\pm}0.3\;m$, $15.7{\pm}20.7\;mg/m^3$, and $3.1{\pm}0.8\;mgO_2/L$, respectively. Most of TN/TP ratios below 20, but concentration of TN and TP was high. Values of TSI ranged between 59 and 77, indicating a eutrophic condition in this system.

Establishment of an In Vivo Report System for the Evaluation of Amber Suppression Activity in Escherichia coli (대장균에서 비천연 아미노산의 위치특이적 삽입을 위한 Amber Suppressor tRNA와 Aminoacyl-tRNA Synthetase의 Amber Suppression 활성측정시스템 개발)

  • Site-specific incorporation of unnatural amino acids into proteins in vivo can be achieved by co-expression of an orthogonal pair of suppressor tRNA and engineered aminoacyl-tRNA synthetase (ARS) that specifically ligates an unnatural amino acid to the suppressor tRNA. As a step to establish this technique, here we generated an Escherichia coli reporter strain DH10B(Tn:lacZam) by integrating amber mutated lacZ gene into the chromosome of E. coli DH10B strain. In vivo expression of E. coli amber suppressor $tRNA^{Gln}$ produced blue colonies in culture plates containing X-Gal as well as dramatically increased $\beta$-galactosidase activity. In addition, expression of an orthogonal pair of Saccharomyces cerevisiae suppressor $tRNA^{Tyr}$ and tyrosyl-tRNA synthetase also produced blue colonies as well as moderate increase of $\beta$-galactosidase activity. These data demonstrate that our reporter strain will provide an efficient method to assess amber suppression in both qualitative and quantitative manners.

Performance Evaluation of Advanced Municipal Wastewater Tretment by Phased Isolation Intrachannel Clarifier Ditch (침전지내장형 상분리 산화구공정에 의한 하수 고도처리특성 평가)

  • Phased isolation intrachannel clarifier ditch process developed in this study is an enhanced biological nutrient removal process employing two ditches with intrachannel clarifiers. Bench-scale phased isolation ditch process was used to evaluate the system performance on municipal wastewater and detailed assessment of internal behavior in a ditch and each reactions. When the system was operated at the HRTs of 6~12hours, SRTs of 9~31 days, and cycle times of 4hours, the system showed removals of BOD, TN, and TP as high as 88~97%, 73~78%, and 65~90%, respectively. The internal behavior were well matched on each reactions such as nitrification, denitrification, and phosphorus release and uptake. As the SRT became longer, TN removal increased gradually, whereas TP removal decreased contrarily. However, the system was capable of producing an effluent TP concentration 1mg/L or less even at longer SRTs except the case of solids discharge by malfunction of intra-clarifier occurred by its geometrical limit. The system performance slightly decreased by hydraulic shock loading(increasing of influent flowrate and decreasing of system HRT). However, the higher system performance could be achieved again after four cycles. Thus, the system reliability could be successfully achieved short-term hydraulic shock loading that occurred in medium- and small-sized wastewater treatment plants suffering fluctuation of influent quality and flowrate during wet season.

Expression of Human Thrombopoietin in Insect Cells by Polyhedrin-gp64 Dual Promoter-Based Baculovirus Vector System

  • A new baculovirus transfer vector (pPGP404) was constructed to increase the expression level of human thrombopoietin (hTPO) in insect cells. In pPGP404, hTPO was cloned next to the AcNPV polyhedrin-gp64 dual promoter and the leader sequence of hTPO was substituted with that of gp64. A recombinant baculovirus, AcPGP404, was constructed by using pPGP404 as a transfer vector. hTPO was expressed in AcPGP404-infected TN5 cells and it was observed that the expression levels of hTPO in TN5 cells increased three-fold ($6.0 {\mu}g/ml^{-1}$) compared to the level expressed under the control of the polyhedrin single promoter. These results indicate that the polyhedrin-gp64 dual promoter system would be useful for expression in large quantities of recombinant proteins in insect cells.

Long-term Paradigm Analyses of Chlorophyll a and Water Quality in Reservoir Systems

  • During the period of past fifteen years (1992~2006), variations of chlorophyll a in relation with water quality in freshwater reservoirs were investigated. This study compared total nitrogen (TN), total phosphorus (TP), chlorophyll a, Secchi depth (SD) and total suspended solids (TSS) between terrestrial freshwater reservoir and coastal freshwater reservoir systems based on their location. Regression analyses (linear and non-linear regressions) were applied for all study sites to examine relationship and interaction of these factors in the freshwater systems from in-land to coasts. The results demonstrated that chlorophyll a was significantly correlated to total phosphorus ($R^2=0.94$, P<0.0001) and was remarkably related to TSS increase ($R^2=0.63$, P<0.0001) in the selected reservoirs. The TN : TP ratio in the reservoir systems was higher than Redfield ratio (16 : 1) indicating that the reservoirs are potentially experiencing P limitation. Water quality of coastal freshwater reservoir system was more significantly decreased than the reservoirs located in in-land during the past fifteen years. The strict management of nutrient discharge into freshwater systems should implemented in the coastal reservoirs since the freshwater is introduced into coastal estuarine systems.
