TEV 검출에 의한 가스 절연 개폐기의 절연 진단 (Insulation Diagnosis of Gas-Insulated Switchgears by TEV Detection)

  • Transient earth voltage (TEV) signals propagate on metal surfaces when partial discharge (PD) occurs due to the deterioration of insulation performance in the operation of gas-insulated switchgears (GIS). A TEV sensor has advantages of high sensitivity and convenient installation for detecting PD defects. However, the TEV sensor depends on imports in domestic and detailed studies have not been conducted. In this study, a sensor was designed and fabricated by the TEV principle and its response characteristics were evaluated for detecting PD defects, which were simulated as protrusion on conductor (POC), protrusion on enclosure (POE), and free moving particle (FMP) defects. Finally, the PD-induced TEV signals were measured and phase-resolved partial discharge (PRPD) patterns were analyzed to identify the type of defect.

지하수자원의 경제적 가치 평가 적용과 관련한 최근동향 (Recent Trend for the Application of Total Economic Value (TEV) Estimation to Groundwater Resources)

  • Total Economic Value (TEV) provides a framework to estimate the economic value of water resources including groundwater with multiple applications to natural resource economics and environmental economics. Crucial to the application of economic analysis to natural resources are techniques to value the resources as an economic value that is expressed in monetary terms. On the other hand, the aim of TEV estimation is to determine the economic value of water resources including 'use' with production and recreation and 'non-use' such as existence values. TEV is used to assess the economic value of water resources for the multiple goods, and environmental 'services' that are provided by a water resource and also used to assess options for water use, for example balancing production values provided by water resource use against the cost of resource degradation by that use. The value of TEV can be assessed over time where pollution or unsustainable use may reduce the economic value of an environmental asset. Therefore, values are used to assess options of resource use, sometimes leading to policies on resource conservation or allocation. In conclusion, the application of TEV would be well adjusted over Jeju Island where groundwater resources account for more than 98% water resources and the budget of water demand/supply shows disparity over the Island.

Retrospective analyses of the bottleneck in purification of eukaryotic proteins from Escherichia coli as affected by molecular weight, cysteine content and isoelectric point

  • Experimental bioinformatics data obtained from an E. coli cell-based eukaryotic protein purification experiment were analyzed in order to identify any bottleneck as well as the factors affecting the target purification. All targets were expressed as His-tagged maltose-binding protein (MBP) fusion constructs and were initially purified by immobilized metal affinity chromatography (IMAC). The targets were subsequently separated from the His-tagged MBP through TEV protease cleavage followed by a second IMAC isolation. Of the 743 total purification trials, 342 yielded more than 3 mg of target proteins for structural studies. The major reason for failure of target purification was poor TEV proteolysis. The overall success rate for target purification decreased linearly as cysteine content or isoelectric point (pI) of the target increased. This pattern of pI versus overall success rate strongly suggests that pI should be incorporated into target scoring criteria with a threshold value.

  • The valuation of biodiversity is a fundamental step in conservation. The useful framework for analysing the economic value of biological resources is that of total economic value (TEV) and TEV comprises both use and non-use values, the former related to an actual use made of the resource, the latter to a willingness to pay for the resource independently of any use made of it. There are several valuation approacher in environmental economics literature. However, stated preference approach should be introduced for valuation of biodiversity because it can estimate non-use value as well as use value. Contingent Valuation and Conjoint Analysis are representative methods in stated preference and Conjoint Analysis can be more useful for valuation of biodiversity. Futhermore, the combination of ecology and economics to assess biodiversity leads to an integrated framework. Thus, interdisciplinary work is required, involving both economists and ecologists transferring elements or even theories and models from one discipline to another and transforming them for their specific, mutually consistent purpose.

12주간 승마 운동이 초·중학생들의 Sit-to-Stand 동작 시 신체 이동 및 평형성 그리고 몸통의 굴신 운동에 미치는 영향 (Effects of the 12 Weeks Horse Riding Exercise on a Capacity for Locomotion of a Body and Bilateral Balance and Trunk Sway Velocity in Elementary and Middle School Student during the Sit-to-Stand)

  • The purpose of this study was to analyze the 12-week horse riding exercise effect on the locomotion of a body and bilateral balance and flexion/extension of trunk during the sit-to-stand(STS) in elementary and middle school students. One-hundred eleven participants were divided into three groups. Each group was subject to a different horse riding exercise: control group(n = 36), 1 time/week group(n=37), and 3 times/week group(n=38). Two-way repeated ANOVA was used to compare weight transfer time(WTT), trunk flexion velocity(TFV), trunk extension velocity(TEV), mean rising index(MRI), mean weight asymmetry( MWA) among the groups, and STS changes in before and after 12 weeks. There was a group interaction in WTT, TFV, TEV, MRI, and MWA with different responses to horse riding exercise participation. There was a significant increase in 1 time/week group and 3 times/week group of WTT, TFV, TEV, and MRI. Additionally, MWA(an index of a capacity for bilateral balance) in lower extremity was decreased in 3 times/week group. It seems that horse riding exercise positively affected musculoskeletal function of the trunk and lower extremity by accelerating locomotion and bilateral balance.

RT-PCR Detection of Five Quarantine Plant RNA Viruses Belonging to Potyand Tospoviruses

  • In order to detect quarantine plant viruses, we developed reverse transcription-polymerase chain reaction (RT-PCR) primer pairs for five single-stranded (ss) plant RNA viruses that are not currently reported in Korea but could be potential harmful plant viral pathogens. Three viruses such as Chilli veinal mottle virus (ChiVMV), Colombian datura virus (CDV), and Tobacco etch virus (TEV) belong to the genus Potyvirus while Chrysanthemum stem necrosis virus (CSNV) and Iris yellow spot virus (IYSV) are members of the genus Tospovirus. To design RT-PCR primers, we used reported gene sequences corresponding to the capsid protein and polyprotein for ChiVMV, CDV, and TEV while using nucleocapsid protein regions for CSNV and IYSV. At least two different primer pairs were designed for each virus. Fifteen out of 16 primer pairs were successfully applied in detection of individual quarantine virus with high specificity and efficiency. Taken together, this study provides a rapid and useful protocol for detection of five quarantine viruses.

Expression, Purification and NMR studies of SH3YL1 SH3 domain

  • SH3YL1, a novel protein containing one Src homology 3 domain at the carboxyl terminus was first detected in mouse anagen skin cDNA. This protein had a significant homology with YHRO 16c/Ysc 84, the yeast Src homology 3 domain-containing protein. The sequence identity was remarkable at the carboxyl and amino-terminal Src homology 3 domain, suggesting that the novel protein is a mouse homolog of the yeast protein and thus was termed as SH3YL1. SH3YL1 is composed of two domains, a DUF500 at N-termini and a SH3 domain at C-termini. In our study we cloned the SH3 domain in bacterial expression system in Escherichia coli using pET32a vector with TEV protease cleavage site and purified as a monomer using affinity chromatography. The N-terminal poly-Histidine tag was cleaved with TEV protease and target protein was used for backbone studies. Our study showed that SH3 domain primarily consists of $\beta$-sheet which is in consistence with previous result performed on the truncated SH3 domain of SH3YL1.

퍼지논리를 이용한 저온저장고의 온도제어시스템 개발 (Development of Temperature Control System for Cold Storage Room Using Fuzzy Logic)

  • Low temperature storage method is to increase the value of agricultural products by reducing quality loss and regulate consignment time by controlling respiration rates of agricultural products. Respiration rate of agricultural products depends on several factors such as temperature, moisture, gas composition and a microbe inside the storage room. Temperature is the most important factor among these, which affects respiration rate and causes low or high temperature damage. Fuzzy logic was used to control the temperature of a storage room ,which uses information of uncertain facts and mathematical model for room temperature control . Room temperature was controlled better by using fuzzy logic control method rather than on-off control method. Refrigerant flow rates and temperature deviations were measured for on-off system using TEV(temperature expansion valve) and for fuzzy system using EEV(Electrical Expansion Valve) . Temperature of the Storage room was lowered faster by using fuzzy system than on -off system. Temperature deviation was -0.6~+0.9$^{\circ}C$ for on-off system and $\pm$0.2$^{\circ}C$ for fuzzy system developed. Temperature deviation and variation of temperature deviation were used as inout parameters for fuzzy system. The most suitable input and output value were found by experiment. Cooling rate of the storage room decreased while temperature deviation increased for the sampling time of 20 sec.

Purification and NMR studies on Phosphatase domain of UBLCP1

  • UBLCP1 is composed of Ubiquitin Like domain and RNA Polymerase II Phosphatase I domain. Phosphatase domain (25.9KDa) has been cloned into the E.coli using pET32a vector with TEV protease cleavage site and successfully purified as a monomer using affinity chromatography and histidine tag was cleaved with TEV protease for structural studies. Our results indicated that the Phosphatase domain showed well-defined folded structure based on data from one-dimensional and two-dimensional NMR spectroscopy. Data form circular dichroism also suggested that Phosphatase domain consisted of both ${\alpha}$ -helix and ${\beta}$ -sheet. This information will be used for detailed structural study of UBLCP1.

새로운 신경전달물질 H2S 발생 효소, cystathionine γ-lyase의 대량발현 조건과 활성측정 (Overexpression and Activity Analysis of Cystathionine γ-Lyase Responsible for the Biogenesis of H2S Neurotransmitter)

  • 질병과 밀접한 관계가 있는 hCGL 단백질의 경우 대량 배양 시 유도체를 사용하지 않아도 발현이 되는 점과 유전자 측면에서 조작이 쉬운 E.coli를 이용하여도 발현이 된다는 점에 있어서 중요한 이점을 가지고 있다. 본 연구에서는 배양되는 온도와 발현에 관련 있는 유도체의 농도, 600 nm에서의 균 성장 정도에 따른 유도체의 첨가 그리고 배지의 양을 조절하면서 유입되는 aeration의 조건으로 hCGL 단백질 발현의 최적의 조건 확립을 목적으로 하였다. 또 각 발생되는 inclusion body의 양을 측정하면서 보다 많은 가용성 단백질을 발현시키는 조건을 확립하고자 하였다. hCGL 단백질은 저온에서 보다 많은 양의 단백질이 발현되며 inhibitor의 억제를 담당하는 유도체의 농도와는 상관없이 발현이 되었다. 또한 균의 성장 정도에 따라 유도체의 첨가시기를 달리 하였을 때, 발현 비율에 차이는 있었으나 전체적인 단백질 양과 비교해 보면, 이는 hCGL 발현에 큰 영향을 미치지 않는다. 배지의 양을 달리하여 살펴본 aeration에 따른 hCGL 발현 정도는 배지의 부피가 15%일 때 높은 aeration으로 균의 양은 많았으나 목적 단백질인 hCGL의 발현은 aeration이 되지 않는 조건에서 더 잘되는 것을 확인하였다. 그리고 His-TEV-hCGL의 활성은 야생형 hCGL의 활성을 기준으로 하였을 때, L-cystathionine을 기질로 하였을 경우 76%, L-cysteine을 기질로 하였을 경우 88% 수준으로 유사한 활성을 나타내었고, 이는 손쉽게 정제 가능한 His-TEV-hCGL을 야생형을 대신하여 사용할 수 있음을 시사한다. 또한 His-TEV-hCGL이 야생형 hCGL과 같이, 427 nm에서 흡광을 가지는 것으로 보아 보효소PLP를 포함하고 있음을 알 수 있었다. 이로써 homocysteine 대사연구에 필수적인 hCGL 효소를 다량 얻는 방법을 확립하고, 관련 연구에 기여하리라 사료된다.
