TCP10L negatively regulates alpha-fetoprotein expression in hepatocellular carcinoma

  • Alpha-fetoprotein (AFP) is one of the most commonly used and reliable biomarkers for Hepatocellular carcinoma(HCC). However, the underlying mechanism of AFP expression in HCC is poorly understood. In this study, we found that TCP10L, a gene specifically expressed in the liver, is down-regulated in HCC and that its expression inversely correlates with AFP expression. Moreover, overexpression of TCP10L suppresses AFP expression whereas knockdown of TCP10L increases AFP expression, suggesting that TCP10L might be a negative regulator of AFP. We found that TCP10L is associated with the AFP promoter and inhibits AFP promoter-driven transcriptional activity. Taken together, these results indicate that TCP10L negatively regulates AFP expression in HCC and that it could be a potential prognostic marker and therapeutic target for HCC.

  • Comparing 231 genes on chimpanzee chromosome 22 with their orthologous on human chromosome 21, we have found that 15 orthologs have indels within their coding sequences. It was rather surprising that significant number of genes have changed by indel, despite the shorter time since their divergence and led us hypothesize that indels and structural changes may represent one of the major mechanism of proteome evolution in the higher primates. Human T-complex protein 10 like (TCP 10L) is a representative having indel within its coding sequence. Gene structure of human TCP10L compared with chimpanzee TCP10L gene showed 16 base pair difference in genomic DNA. As a result of the indel, frame shift mutation occurs in coding sequence (CDS) and human TCP10L express longer polypeptide of 21 amino acid residues than that of chimpanzee. Our prediction found that the indel may affect to dramatic change of secondary protein structure between human and chimpanzee TCP10L. Especially, the structural changes in the C-terminal region of TCP10L protein may affect on the interacting potential to other proteins rather than DNA binding function of the protein. Through these changes, TCP10L might influence gene expression profiles in liver and testis and subsequently influence the physiological changes required in primate evolution.

Human Liver Specific Transcriptional Factor TCP10L Binds to MAD4

  • A human gene T-complex protein 10 like (TCP10L) was cloned in our lab. A previous study showed that it expressed specifically in the liver and testis. A transcription experiment revealed that TCP10L was a transcription factor with transcription inhibition activity. In this study, the human MAD4 was identified to interact with TCP10L by a yeast two-hybrid screen. This finding was confirmed by immunoprecipitation and subcellular localization experiments. As MAD4 is a member of the MAD family, which antagonizes the functions of MYC and promotes cell differentiation, the biological function of the interaction between TCP10L and MAD4 may be to maintain the differentiation state in liver cells. Also, we propose that the up-regulation of Myc is caused by the down-regulation of TCP10L in human hepatocarcinomas.

TCP10L synergizes with MAD1 in transcriptional suppression and cell cycle arrest through mutual interaction

  • T-complex protein 10A homolog 2 (TCP10L) was previously demonstrated to be a potential tumor suppressor in human hepatocellular carcinoma (HCC). However, little is known about the molecular mechanism. MAX dimerization protein 1 (MAD1) is a key transcription suppressor that is involved in regulating cell cycle progression and Myc-mediated cell transformation. In this study, we identified MAD1 as a novel TCP10L-interacting protein. The interaction depends on the leucine zipper domain of both TCP10L and MAD1. TCP10L, but not the interaction-deficient TCP10L mutant, synergizes with MAD1 in transcriptional repression, cell cycle G1 arrest and cell growth suppression. Mechanistic exploration further revealed that TCP10L is able to stabilize intracellular MAD1 protein level. Consistently, the MAD1-interaction-deficient TCP10L mutant exerts no effect on stabilizing the MAD1 protein. Taken together, our results strongly indicate that TCP10L stabilizes MAD1 protein level through direct interaction, and they cooperatively regulate cell cycle progression.

Bending and Compressive Properties of Crystallized TCP/PLLA Composites

  • $\beta$-Tricalcium phosphate ($\beta$-TCP) particles reinforced bioresorbable plastics poly-L-lactide (PLLA) composites were prepared by injection molding. The nominal weight ratio of $\beta$-TCP was selected as 5, 10 and 15%. In order to clarify effects of the PLLA crystallinity on the mechanical properties, the specimens were heat treated isothermally. Results of differential scanning calorimetry indicated that the PLLA crystallinity increased with increasing heat treatment temperature. Bending and compressive tests were conducted on the specimen with different $\beta$-TCP contents and crystallinities. The results show that the bending and compressive moduli increased with increasing $\beta$-TCP contents and crystallinity. On the other hand, bending strength decreased with increasing $\beta$-TCP contents. Maximum bending strength was obtained at the heat treatment of $70^{\circ}C$ for 24 h, whereas compressive 0.2% proof strength increased with increasing heat treatment temperature. This difference is attributed to the difference in the microscopic damages.

가수분해 후 GC-ECD를 이용한 축산물 중 살균제 Prochloraz 및 그의 대사물의 동시분석 (Simultaneous determination for fungicide prochloraz and its metabolites in animal commodities with GC-ECD after hydrolysis)

  • 축산물 중 살균제 prochloraz 및 그의 대사체의 잔류량을 함께 정량분석할 수 있는 단성분 분석법을 개발하고자 하였다. 축산물 중 prochloraz의 잔류분의 정의가 'prochloraz와 2,4,6-trichlorophenol기를 포함하고 있는 대사산물의 합을 prochloraz로 함'이기 때문에 prochloraz 및 대사산물을 모두 2,4,6-trichlorophenol (2,4,6-TCP)로 분해시킨 후 GC/ECD로 정량하는 정밀 분석법을 확립하였다. 대표 축산물인 소고기, 돼지고기, 닭고기, 우유 및 계란 중 prochloraz 잔류분을 acetone으로 추출하고 2,4,6-TCP로 분해한 후 dichloromethane으로 분배하고 aminopropyl SPE로 정제한 다음 2,4,6-TCP를 정량하였다. 본 분석법을 통한 기기정량한계와 분석법정량한계는 prochloraz의 경우 각각 0.01 ㎍/mL과 0.02 mg/kg이었으며 2,4,6-TCP는 0.005 ㎍/mL과 0.01 mg/kg이었다. 2,4,6-TCP를 이용한 표준검량선은 0.005-0.2 mg/L 범위에서 결정계수(R2) ≥0.995으로 직선성을 확인하였다. 분석법의 회수율은 분석정량한계 및 정량한계의 10배 처리수준에서 실시하여 모두 90% 이상이었고 변이계수는 10% 이하이었다. 따라서, 본 연구에서 확립된 분석법은 축산물 중 prochloraz 및 대사체를 정량분석하는데 우수한 정확성 및 정밀도를 보였으며, 축산물 중 prochloraz 잔류분석을 위한 정밀시험법으로써 분석개발기준을 충족하였다.

민들레 분말을 이용한 쿠키의 품질과 항산화성 (Quality Characteristics and Antioxidant Activity of Cookies Prepared from Taraxacum coreamm Powder)

  • This study assessed the preparation and quality characteristics of cookies prepared from Taraxacum coreamm powder (TCP) (0, 2, 4, 6, 8%) substituting wheat flour. The pH of the cookie dough significantly decreased with increasing TCP. However the density of the cookie dough was not significantly different among the test groups. The baking loss rate of the groups with TCP was slightly lower, whereas the moisture content was higher than the control group. The spread factor of the cookies decreased significantly with increasing TCP. The hardness of the cookies increased as TCP increased. In Hunter's color value, the L and a value of the cookies decreased with increasing TCP, while the b value increased. The DPPH radical scavenging activity of the cookies significantly increased with increasing TCP. Based on the above results, this study suggested that TCP can be a good ingredient to increase the functionality of cookies.

Bufferbloat 환경에서 MPTCP 성능 개선을 위한 전송 상태 모니터링 방법 (Transmission status monitoring method for improving the performance of MPTCP in Bufferbloat environment)

  • Multipath TCP (MPTCP)는 다중 경로를 통해 데이터를 전송하므로 향상된 네트워크 성능을 기대할 수 있으나, 예고 없이 전송 경로에서 발생되는 Bufferbloat은 해당 경로의 성능뿐만 아니라 다른 경로의 성능 역시 저하시켜 단일 TCP 보다 오히려 좋지 않은 성능이 발생한다. 이와 같은 Bufferbloat 문제를 해결하기 위해 본 논문에서는 송신단에서 MPTCP 레벨의 전송 상태 모니터링 방법과 HoL 패킷 재전송 알고리즘을 제안하였다. 제안한 알고리즘은 송신단 독자적으로 Bufferbloat 감지를 가능하게 하며, 전송 상태 모니터링 버퍼에서 HoL 패킷을 식별하여 정상 경로로 재전송함으로 HoL Blocking문제를 해결할 수 있도록 하였다. NS-3기반 시뮬레이션 결과 제안한 알고리즘은 기존 MPTCP 대비 전송 성능은 최대 22.8%까지 향상되었고, 송신 버퍼 및 수신 버퍼에 대기하는 평균 패킷 수는 기존 대비 각각 44.3%와 9.2% 수준으로 감소되었음을 확인하였다.

  • 본 논문에서는 OPNET Modeler 9.0을 사용하여 IEEE 802.l1b 무선랜을 기반으로 한 핸드오프 시뮬레이터 개발 과정을 설명하고, 이를 이용하여 트랜스포트 계층(TCP, UDP)에 따른 핸드오프 성능을 분석해 본다. 기존의 OPNET Modeler 9.0에 제공되는 IEEE 802.l1b 무선랜 모델은 시뮬레이션의 초기에 설정된 하나의 BSS(Basic Service Set)를 기반으로 동작하기 때문에 MS(Mobile Station)가 다른 BSS로 이동할 경우 통신 단절이 발생한다. 따라서 논문에서는 기존의 무선랜 모델에 핸드오프 기능을 추가하고 이를 사용하여 핸드오프에 의한 여러 가지측면에서 성능 특성을 분석해 본다. 또한 결과를 통해 표준 핸드오프 알고리즘의 문제점을 분석하여 TCP Timeout 방지형 알고리즘을 제안하고 두 알고리즘의 성능을 비교한다.

  • Environmentally adapted synthetic base oils of polyalkylene glycols (PAGs) and polyol esters (POEs) show a high polarity because of their functional groups containing oxygen atom. The lubricating performance of these polar base oils was investigated by using a four-ball tribometer under boundary lubrication condition. Four polyalkylene glycols and five polyol ester base oils were used as sample base oils of high polarity. A mineral oil (MO) and alkylnaphthalene (AN) were used as low polarity base oils. Tricrecylphosphate (TCP) was added to all the base oils, in the range of 10 mmol/L-2000 mmol/L, as an antiwear additive. All the TCP-for-mutated base oils showed optimum concentration characteristics for minimizing wear. The order of optimum concentration of all the base oils was in a good accordance with the order of relative stability of TCP in base oils. The interaction model on solvation between additive and different polar base oils can expect the stability order of TCP. Thus, the model on solvation can explain well the order of optimum concentration of all the base oils, by using the effect of polarity (dielectric constant, $\varepsilon$) and molecular size (molecular weight, MW) of them on stability of TCP in polar base oils. Finally, a good correlation of the optimum concentration for all the base oils was obtained when it was arranged as a function of C∝(M $W_{Base Oil}$/M $W_{TCP}$)$^{-2}$.71/.($\varepsilon$$_{Base Oil}$)$^{3.38}$ by these two parameters.s..
